Determination of Nutritive Value of Citrus Tree Leaves for Sheep Using In vitro Gas Production Technique

  • The nutritive values of leaves of Citrus grandis, Citrus aurantium, Citrus oranges, Citrus limon, and Citrus deliciosa were evaluated by chemical composition and in vitro gas production techniques. There were significant (p<0.001) differences among citrus species in terms of chemical composition. Crude protein (CP) contents ranged from 123.0 to 148.3 g/kg DM. Neutral detergent fibre (NDF) and acid detergent fibre (ADF) contents were varied with species in the range 219.4-355.4 and 215.0-278.8 g/kg DM respectively. Condensed tannin (CT) contents were ranged from 5.9 to 10.2 g/kg DM. The PEG addition significantly (p<0.001) increased the gas production and some estimated parameters of citrus tree leaves. However, species showed variable responses to polyethylene glycol (PEG) treatment. There were also significant (p<0.001) differences among species in terms of gas production and estimated parameters. The OMD and ME contents of citrus leaves without PEG supplementation were ranged from 66.5 to 73.3% and 9.8 to 10.9 MJ/kg DM respectively. The improvement in gas production, organic matter digestibility (OMD) and metabolizable energy (ME) with PEG emphasized the negative effect of tannins on digestibility. The increase (%) in the estimated OMD and ME contents ranged from 5.5 to 9.8% and 5.7 to 10.2% respectively. All citrus tree leaves studied in this experiment have potential nutritive values indicated by high crude protein content, OMD, ME and low fiber values.

The Effect of Variety and Growing Conditions on the Chemical Composition and Nutritive Value of Wheat for Broilers

  • The aim of this study was to examine the effect of variety and growing conditions of wheat on broiler performance and nutrient digestibility. One hundred and sixty-four wheat samples, collected from a wide range of different sources, locations, varieties and years, were analyzed for a range of chemical and physical parameters. Chemical and physical parameters measured included specific weight, thousand grain weight (TG), in vitro viscosity, gross energy, N, NDF, starch, total and soluble non-starch polysaccharides (NSP), lysine, threonine, amylose, hardness, rate of starch digestion and protein profiles. Ninety-four of the wheat samples were selected for inclusion in four bird trials. Birds were housed in individual wire metabolizm cages from 7 to 28 d and offered water and feed ad libitum. Dry matter intake (DMI), live weight gain (LWG) and gain:feed were determined weekly. A balance collection was carried out from 14 to 21 d for determination of apparent metabolizable energy (AME), ME:gain, DM retention, oil and NDF digestibility. At 28 d the birds were sacrificed, the contents of the jejunum removed for determination of in vivo viscosity and the contents of the ileum removed for determination of ileal DM, starch and protein digestibility. The wheat samples used in the study had wide-ranging chemical and physical parameters, leading to bird DMI, LWG, gain:feed, ME:GE, AME content and ileal starch and protein digestibility being significantly (p<0.05) affected by wheat sample. A high level of N fertilizer application to the English and NI wheat samples tended to benefit bird performance, with increases of up to 3.4, 7.2 and 3.8% in DMI, LWG and gain:feed, respectively. Fungicide application also appeared to have a positive effect on bird performance, with fungicide treated (+F) wheat increasing bird DMI, LWG and gain:feed by 6.6, 9.3 and 2.7%, over the non-fungicide treated (-F) wheats. An increase (p<0.1) of 9.3% in gain:feed was also observed at the low seed rate of 40 compared to 640 seeds/$m^2$. It was concluded that the type of wheat sample and environmental growing conditions significantly affects bird performance when fed wheat-based diets.

Preparation of Hypoallergenic Whey Protein Hydrolysate by a Mixture of Alcalase and Prozyme and Evaluation of Its Digestibility and Immunoregulatory Properties

  • Whey protein (WP) has nutritional value, but the presence of β-lactoglobulin (β-LG) and α-lactalbumin (α-LA) cause allergic reactions. In this study, hypoallergenic whey protein hydrolyate (HWPH) was prepared by decomposing β-LG and α-LA of WP using exo- and endo-type proteases. The enzyme mixing ratio and reaction conditions were optimized using response surface methodology (RSM). Degradation of α-LA and β-LG was confirmed through gel electrophoresis, and digestion, and absorption rate, and immunostimulatory response were measured using in vitro and in vivo systems. Through RSM analysis, the optimal hydrolysis conditions for degradation of α-LA and β-LG included a 1:1 mixture of Alcalase and Prozyme reacted for 10 h at a 1.0% enzyme concentration relative to substrate. The molecular weight of HWPH was <5 kDa, and leucine was the prominent free amino acid. Both in vitro and in vivo tests showed that digestibility and intestinal permeability were higher in HWPH than in WP. In BALB/c mice, as compared to WP, HWPH reduced allergic reactions by inducing elevated Type 1/Type 2 helper T cell ratio in the blood, splenocytes, and small intestine. Thus, HWPH may be utilized in a variety of low allergenicity products intended for infants, adults, and the elderly.

향신채소를 첨가한 어육 고음 추출물의 단백질 품질평가 (Protein Nutritional Qualities of Hydrocooked Fish Extracts Containing Spicy Vegetables)

  • 풍미향상과 고온 열처리로 인한 단백질 품질저하를 완화시킬 목적으로 세가지의 향신 채소 (마늘, 생강, 양파)를 첨가하여 $110^{\circ}C$에서 5시간동안 조리한 어육 고음추출물의 단백질의 품질을 평가하였다. 향신채소를 첨가한 붕어 어육가수분해물의 유효성 Iysine은 생육의 유효성 Iysine과 거의 같은 수준을 유지하였으며, 광어의 경우는 $66\~84\%$ 정도 유지되었다. 친유성 갈변물질 발생정도는 원료어육에 비해 붕어의 경우는 오히려 낮고, 광어는 높은 것으로 나타나 이런 종류의 갈변물질은 제거된 지방량에 따라 달라질 수 있을 것으로 판단되었다. TI생성은 원료어육과 별차이가 없고, 소화율은 원료어육보다 $4\~5\%$ 낮으나 대체로 고온 고음추출물에 비해 $3\~4\%$ 높게 나타나 단백질 품질 저하가 훨씬 완화된 결과를 보였다. 원료어육의 유리 아미노산의 총량이 $2,000mg\%$ (광어), $2,240mg\%$ (붕어) 정도였던 반면, 향신채소를 첨가한 고음추출물에는 $3,800mg\%$ (광어), $3,700mg\%$ (붕어) 정도의 유리아미노산이 함유되어 있었다. 붕어 고음추출물의 주요 유리아미노산은 histidine, taurine, glycine 및 alanine 이었으며, 광어의 경우는 taurine, alanine 및 asparagine이었다. 향신채소 첨가 어육 고음추출물 중 비교적 품질이 우수하다고 판단 된 생강첨가 추출물을 이용한 in vivo 단백질 품질 평가를 한 결과 in vivo 단백질 소화율은 표준단백질인 ANRC casein보다 $6\~7\%$낮았으나 고온 고압 처리 고음추출물보다 붕어의 경우 약 $10\%$이상 소화율이 높아졌다. Rat-PER은 약간 상승하였지만 실험 전기간 동안 실험동물의 체중증가는 거의 발생하지 않았고, 다만 고온 처리 시료에서 발생하였던 탈모현상은 볼 수 없었으며 분변의 상태도 약간 호전되었다.

Zinc Bioavailability in a Semolina/Soy Protein Mixture Was Not Affected by Extrusion Processing

  • Effects of extrusion processing on zinc bioavailability in a product made from 85% semolina and 15% soy protein concentrate were determined using in vitro and in vivo techniques. Soluble and dialyzable zinc contents of the extruded product were 15.3 and 13.0 $\mu$g/g, respectively, compared to 15.6 to 15.0$\mu$g/g in the raw (unprocessed) ingredients. Zinc bioavailability in diets in which all of the zinc (14ppm) was provided either by the extruded product or by its raw ingredients was determined in two groups of male Sprague Dawley rats. No differences were found in concentrations of zinc in plasma, liver and femur between rats fed the two experimental diets. Apparent zinc digestibility was similar for both diets. These results are consistent with the in vitro results showing no effect of extrusion processing on zinc bioavailability.

착유우 사료에 대한 Aspergillus oryzae 발효물질 첨가가 in vitro 건물, 섬유소 및 단백질 소화율과 발효액의 pH에 미치는 영향 (The Effects of a Fermentation Product by Aspergillus oryzae on the in vitro Digestibilities of Dry Matter, Fiber and Protein and pH in the Fermentation Fluid of Diets for Dairy Cows)

  • 본 연구는 Aspergillus oryzae 배양물이 in vitro 건물, 섬유소 및 단백질 소화율과 pH에 미치는 영향을 구명하고자 실시하였다. 균종 Aspergillus oryzae는 한국 생명공학연구원에서 분양받았으며, 본 연구에 사용된 사료는 시중 유통중인 착유우용 배합사료와 TMR 사료로서, 일반성분 분석은 (주)우성사료 중앙실험실에서 실시하였고, 건물, 섬유소 및 단백질 소화율 실험은 충남대학교 실험실에서 실시하였다. 배합사료와 TMR에 Aspergillus oryzae 배양물 0, 1.0, 2.0 및 3.0%를 첨가한 후, Holstein종 착유우로부터 채취한 반추위액에 시료 2.0g 내외를 넣어 진탕배양기에서 24, 48 및 72시간 소화시켰다. 침전물에 0.2% pepsin이 들어있는 0.1N HCl 30ml를 넣고 $39^{\circ}C$ incubator에서 48시간 소화시키고 나서, 마지막으로 침전물을 $60^{\circ}C$의 건조기에서 48시간 건조시켰다. 실험은 3회 반복하여 실시하였다. 건물 소화율은 Aspergillus oryzae 발효물을 첨가하지 않은 대조구에 비해 24시간 배양시킨 배합사료의 경우 2.1%(63.1%), 9.7%(68.5%) 및 9.0%(68.0%) 개선되었고, TMR은 4.8%(60.0%), 6.4%(61.1%) 및 2.9%(58.8%)의 개선효과가 있었다. 이와 대조적으로, 48시간 및 72시간 배양시킨 시험구의 건물 소화율에 Aspergillus oryzae 배양물이 미치는 영향은 24시간 배양시보다 상대적으로 낮은 경향을 보였다. 섬유소 소화율에서 Aspergillus oryzae가 조섬유, ADF 및 NDF 소화율을 유의적으로 개선시킨다는 것을 알 수 있었다. 배합사료에서 3% 첨가구의 경우 24시간 배양시 13.3%(53.3%), 72시간 배양시 2.4%(54.6%)까지 증가되었다. 하지만, 첨가수준에 따라 소화율이 높아졌음에도 불구하고 첨가수준별 개선효과에는 유의적인 차이가 없었다. 단백질 소화율은 Aspergillus oryzae 배양물 첨가에 의해 배합사료에서 0.4%(79.7%)에서 9.4%(71.8%)의 유의적인 개선효과를 나타내었지만, 2.0%와 3.0% 첨가구간의 유의적인 차이는 인정되지 않았다. TMR의 경우에는 4.0%(70.4%)에서 6.3%(65.1%)의 유의적인 소화율 향상을 나타내었지만 2.0%와 3.0% 첨가구간의 유의적인 차이는 인정되지 않았다. 본 연구에서, pH 처리구간 유의적인 차이는 없었으나, Aspergillus oryzae 배양물 첨가에 따라 pH가 약간 감소하는 경향을 보였다. 그러므로 본 발효물은 pH에는 영향을 미치지 않으나 건물, 섬유소 및 단백질 소화율 향상에는 다소 효과가 있 는 것으로 결론을 내릴 수가 있다.

Effects of Polyurethane Coated Urea Supplement on In vitro Ruminal Fermentation, Ammonia Release Dynamics and Lactating Performance of Holstein Dairy Cows Fed a Steam-flaked Corn-based Diet

  • Three experiments were conducted to investigate the effects of polyurethane coated urea on in vitro ruminal fermentation, ammonia release dynamics and lactating performance of Holstein dairy cows fed a steam-flaked corn-based diet. In Exp. 1, a dual-flow continuous culture was run to investigate the effect of polyurethane coated urea on nutrient digestibility, rumen fermentation parameters and microbial efficiency. Three treatment diets with isonitrogenous contents (13.0% CP) were prepared: i) feedgrade urea (FGU) diet; ii) polyurethane coated urea (PCU) diet; and iii) isolated soy protein (ISP) diet. Each of the diets consisted of 40% steam-flaked corn meal, 58.5% forages and 1.5% different sources of nitrogen. PCU and FGU diets had significantly lower digestibility of NDF and ADF (p<0.01) than the ISP diet. Nitrogen source had no significant effect (p = 0.62) on CP digestibility. The microbial efficiency (expressed as grams of microbial N/kg organic matter truly digested (OMTD)) in vitro of the PCU diet (13.0 g N/kg OMTD) was significantly higher than the FGU diet (11.3 g N/kg OMTD), but comparable with the ISP diet (14.7 g N/kg OMTD). Exp. 2, an in vitro ruminal fermentation experiment, was conducted to determine the ammonia release dynamics during an 8 h ruminal fermentation. Three treatment diets were based on steam-flaked corn diets commonly fed to lactating cows in China, in which FGU, PCU or soybean meal (SBM) was added to provide 10% of total dietary N. In vitro $NH_3-N$ concentrations were lower (p<0.05) for the PCU diet than the FGU diet, but similar to that for the SBM diet at all time points. In Exp. 3, a lactation trial was performed using 24 lactating Holstein cows to compare the lactating performance and blood urea nitrogen (BUN) concentrations when cows were fed PCU, FGU and SBM diets. Cows consuming the PCU diet had approximately 12.8% more (p = 0.02) dietary dry matter intake than those consuming the FGU diet. Cows fed the PCU diet had higher milk protein content (3.16% vs. 2.94%) and lower milk urea nitrogen (MUN) concentration (13.0 mg/dl vs. 14.4 mg/dl) than those fed the FGU diet. Blood urea nitrogen (BUN) concentration was significantly lower for cows fed the PCU (16.7 mg/dl) and SBM (16.4 mg/dl) diets than the FGU (18.7 mg/dl) diet. Cows fed the PCU diet had less surplus ruminal N than those fed the FGU diet and produced a comparable lactation performance to the SBM diet, suggesting that polyurethane coated urea can partially substitute soybean meal in the dairy cow diet without impairing lactation performance.

방사선 조사된 재구성 돈육포의 단백질 품질 (Nutritional Quality of Restructured Pork Jerky with Electron Beam and Gamma Ray Irradiation)

  • To evaluate the effect of irradiation on restructured pork jerky containing paprika and Japanese apricot extracts, the quality of protein was determined in vitro based on the formation of trypsin indigestible substrate inhibitor (TIS) and the computed protein efficiency ratio (C-PER) as determined based on the protein digestibility and amino acid analysis. In addition, we compared the effects of electron beam irradiation to those of gamma irradiation. Approximately 3% of the moisture content of pork jerky was reduced in response to irradiation with 3kGy administered using an electron beam however, no additional reduction was observed in samples that were subjected to higher doses of irradiation. In addition, there were no notable differences in the crude protein and fat content of pork jerky samples that were subjected to irradiation, regardless of dose. Furthermore, the total amino acids profiles did not change in response to electron beam irradiation. However, the in vitro protein digestibility increased by 7% in response to 3kGy of electron beam irradiation and 5kGy of gamma irradiation, but no significant changes in digestibility were not observed in response to treatment with higher doses. TIS quantified as trypsin inhibitors were formed in response to irradiation using the electron beam (3kGy) and gamma rays (5kGy), although there was a slight reduction in the production of TIS inhibitors in samples irradiated with higher doses. Moreover, only samples irradiated with 10kGy (electron beam and gamma ray) showed higher TBA values than those of the control samples. Finally, the C-PERs $(2.50{\sim}2.60)$ were greater in all of the irradiated pork jerky samples than in the control samples (2.22). Taken together, these results suggest that electron beam irradiation and the incorporation of extracts (paprika and Japanese apricot) may be useful methods of improving the nutritional quality of pork jerky.

가열 처리한 미꾸라지 단백질의 품질 (Protein Qualities of Loach as Affected by Cooking Methods)

  • To explore the possibility of using freeze dried loach for instant choo o tang(Korean traditional loach soup), protein qualities and fatty acid composition were evaluated on boiled and steamed loach. Total lipid and ash content were lowered in both heated(boiled and steamed) loaches due to deboning and eviscerating during cooked meat preparation. Profiles of total amino acids were not changed seriously by the type of cooking, but the amount of essential amino acids were comparable in all samples. Two times more free amino acids were quantified in cooked samples compared to raw meat. Available lysine was marginally decreased by cooking, and that caused some measurable change in typsin indigestible substrate(TI) in streamed whole loach. In vitro protein digestibility of the heated loaches was not altered drastically and the protein quality determined as computed protein efficiency ratio(C PER) was similar for the raw, boiled and steamed loach. The ratios of unsaturated to saturated fatty acids changed measurably in heated whole loach. The results shows that heating caused apparent oxidative deterioration of the polyunsaturated fatty acids.

Study on Nutritive Value of Tropical Forages in North Sumatra, Indonesia

  • This study was conducted to evaluate nutritive value of forages commonly used as ruminant feeds in North Sumatra, Indonesia. Seven species of grasses and five species of legumes were collected during the rainy season. The results showed that chemical composition, in vitro digestibility of dry matter (DMD), organic matter (OMD) and crude protein (CPD), in vitro gas production and metabolizable energy (ME) content greatly varied among the species of grass and legume forages. The CP content ranged from 6.6 to 16.2% in grass and from 17.5 to 29.1% in legumes; while NDF content of grass and legume ranged from 57.2 to 66.2% and from 24.4 to 55.6%, respectively. The DMD, OMD and CPD of grass ranged from 49.1 to 62.2%, 51.9 to 64.4% and 50.5 to 60.3%; while in legumes the values ranged from 59.1 to 71.8%, 65.2 to 72.0% and 68.2 to 71.6%, respectively. The ME content of grass varied from 6.4 to 9.3 MJ/kg and from 6.5 to 8.3 MJ/kg for legumes. In general, within species of grass Cynodon plectostachyus contained higher CP but was lower in NDF that resulted in much higher digestibility; a similar result was also found in Leucaena leucocephala for the legumes. The two forages also contained much higher ME than the others. In conclusion, the nutritive value of forages in North Sumatra, Indonesia during the rainy season was relatively high as ruminant feed, with the best quality noted for Cynodon plectostachyus and Leucaena leucocephala.
