Marked Expansion of CD11c+CD8+ T-Cells in Melanoma-bearing Mice Induced by Anti-4-1BB Monoclonal Antibody

  • 4-1BB (CD137), a member of the tumor necrosis factor receptor superfamily, is expressed on activated T-cells, and 4-1BB signaling due to interaction with 4-1BB ligand or ligation with anti-4-1BB monoclonal antibody (mAb) costimulates T cells. It has been shown that administration of anti-4-1BB mAb induces anti-tumor immunity in mice, but the nature of the cellular subsets responsible for this immunity is uncertain. In this study we found that anti-4-1BB mAb administration to B16F10 melanoma-bearing mice induced marked expansion of $CD11c^+CD8^+$ T-cells in parallel with suppression of pulmonary tumors. The mAb-treated mice produced higher levels of $IFN-{\gamma}$ in their tumor tissues, spleen and lymph nodes than mice exposed to control antibody. When the $CD11c^+CD8^+$ T-cells were purified and re-stimulated in vitro, they produced high levels of the Th1 cytokines, $IFN-{\gamma}$ and IL-2, but low levels of the Th2 cytokines, IL-4 and IL-10. Furthermore, they expressed high levels of 4-1BB and CD107a, a marker of activated cytotoxic T-lymphocytes. Our results suggest that $CD11c^+CD8^+$ T-cells play a role in the anti-tumor immunity induced by anti-4-1BB mAb.

  • This work was performed to investigate the effects of brazilin in vitro on mitrogen-induced proliferation, ConA-induced TCGF release and responsiveness to recombinant-induced proliferation, ConA-induced TCGF release and responsiveness to recombinant IL-2 using splenocytes from C57BL/6 female mice. Brazilin (29-80 ng/ml) caused a noticeable increase in TCGF production of splenocytes, but did not affect responsivness to recombinant IL-2, the expression of ConA-induced high affinity IL-2 receptor and mitogen-induced proliferation of splenocytes.

  • Angelica gigas Nakai (danggui) is a popular herb which has been used as a blood-building decoction for recovery from weakness in the Chinese medicine. Its demand increased in functional foods and pharmaceutical industries. For its hygiene, fumigation has been used, but the use of fumigants are going to be prohibited for food processing. In order to investigate gamma irradiation technique for hygiene of danggui, the immunomodulation activity of danggui after irradiation was examined. The water extract of irradiated danggui showed a strong mitogenic effect on splenocytes in vitro to the same level of lipopolysaccharide (LPS) and phytohemagglutinin (PHA). The effect was not different from that of non-danggui. It was tested whether there was any difference between irradiated and non-irradiated danggui in effects on the secretion of antibodies and graft versus host reaction in vivo. It turned out that intraperitoneal (i.p.) administration of the extract of irradiated danggui for 4 days remarkably increased the number of antibody-secreting cells in mice injected with sheep red blood cells (SRBC). Splenomegaly, due to graft versus host reacton, was also increased after 7 days i.p. administration of the extract of danggui in mice injected with allogeneic splenocytes. In these two in vivo test, the effect were not different from those of non-irradiated danggui. These results indicated that immunomodulation activity of danggui might be preserved after irradiation. In the other experiments (data not shown), the irradiated danggui was stable in active component analysis and safe in genetic toxicity test. In further research, the stability in other physiological activity of irradiated danggui will have to be proved before practical application of irradiation for hygiene.

닭의 맹장으로부터 분리한 Lactobacillus sakei L2와 L8의 특성 및 면역활성 (Characterization and Immunomodulation Activity of Lactobacillus sakei L2 and L8 Isolated from Chicken Cecum)

  • 건강한 닭의 맹장에서 분리한 Lactobacillus sakei LAB 2와 LAB 8의 생균제로 이용을 알아보기 위해 기본적인 특성과 면역활성을 시험하였다. LAB 2와 LAB 8은 높은 내산성과 내담즙성을 가지고 있으며, 1차 대사산물인 유기산에 기인한 항균활성을 보였다. Salmonella 종을 병원성균으로 이용하여 최대 항균력을 나타내는 시간을 시험한 결과 48시간으로 나타났으며, 생성된 유기산 중 젖산의 생산량은 다른 Lactobacillus 균주보다 높게 조사되었다. In vitro 모델로 알아본 면역증진효과는 면역관련 세포의 증식과 사이토카인의 생산량 증가로 확인하였다. 이에 Lactobacillus sakei LAB 2와 LAB 8은 유용한 생균제로 개발할 수 있는 균으로 생각된다.

Effects of Fucoidan on NO Production and Phagocytosis of Macrophages and the Proliferation of Neuron Cells

  • Fucoidans, high-molecular-weight sulfated polysaccharides extracted from brown seaweeds, have various biological activities. Here we examined whether fucoidan could ncrease the immunomodulation and proliferation capacity of cells in vitro. When peritoneal macrophages were treated with various concentrations of fucoidan $(1\~100{\mu}g/mL)$ for 24 hours, NO production was significantly increased. In addition, exposure of macrophages to $10{\mu}g/mL$ of fucoidan induced a phagocytic activity. Treatment of neuroblastoma cells (SK-N-SH) with fucoidan enhanced cell proliferation and NO production in a concentration-dependent manner. These results indicate that fucoidan has both immunomodulatory and cell proliferative properties, and may thus be a candidate for development as an immunomodulating agent.

  • In the previous report, we described the in vivo antitumor activity of PJ-4, a protein-polysaccharide fraction prepared from the culture filtrate of an insect-born fungus, Paecilomyces tenuipes DGUM 32001. In the present study, we elucidated the immunomodulating activity of PJ-4 on the BALB/c mouse splenic lymphocytes using flow cytometrical techniques. As a result, PJ-4 was found to stimulate the lymphocytes not only to form lymphoblasts but also to express CD25 (IL-2 receptor $\alpha$ chain) molecule, which is well known as a T cell activation marker. More interestingly, its T cell stimulatory activity was more strongly exerted on CD8$^{+}$ T cells than on CD4$^{+}$ T cells. All these data suggest that PJ-4 exerts its antitumor activity at least partly through stimulation of T cells which play major roles in the cell-mediated immune system.tem.

  • Mesenchymal stem cells are classified as multipotent stem cells, due to their capability to transdifferentiate into various lineages that develop from mesoderm. Their popular appeal as cell-based therapy was initially based on the idea of their ability to restore tissue because of their differentiation potential in vitro; however, the lack of evidence of their differentiation to target cells in vivo led researchers to focus on their secreted trophic factors and their role as potential powerhouses on regulation of factors under different immunological environments and recover homeostasis. To date there are more than 800 clinical trials on humans related to MSCs as therapy, not to mention that in animals is actively being applied as therapeutic resource, though it has not been officially approved as one. But just as how results from clinical trials are important, so is to reveal the biological mechanisms involved on how these cells exert their healing properties to further enhance the application of MSCs on potential patients. In this review, we describe characteristics of MSCs, evaluate their benefits as tissue regenerative therapy and combination therapy, as well as their immunological properties, activation of MSCs that dictate their secreted factors, interactions with other immune cells, such as T cells and possible mechanisms and pathways involved in these interactions.

시험관내에서 홍화의 물 추출물이 T 및 B 림프구의 활성에 미치는 영향 (Effect of Water Extract of Carthamus tinctorious L. on In Vitro Activity of T and B Lymphocytes)

  • Based on the traditional application of Carthamus tinctorious L. (CF) as a component of Korean medicinal decoctions, in the present study, we investigated in vitro an immunomodulatory activity of water extract of CF(WECF). Water extract of CF significantly increased the in vitro proliferative responses of spleen cells (SPC). However, addition of WECF during anti-CD3 activation resulted in a significant decrease in SPC proliferation. Flow cytometric analysis showed that WECF addition chanced T and B cell frequencies in anti-CD3-activated spleen cell populations. Using purified cells, it was revealed that WECF is mitogenic to B cells but rather inhibitory to T cell Proliferation. Upon anti-CD3 stimulation, high concentration (1 mg/ml) of WECF significantly inhibited T cell proliferation until day 2 of stimulation. At day 3, anti-CD3-activated cells exposed to WECF recovered their proliferation to the level comparable to control. Although B cell proliferation was also inhibited in proliferation at day 1, it recovered sooner and then was rather augmented by WECF at day 3. These data indicate that WECF down-regulates lymphocyte proliferation at early phase of activation but T cells are more vulnerable than B cells to WECF, However, CD4+ and CD8+ T cells did not differ in WECF-mediated immunotoxicity. Data of propidium iodide (PI) staining showed that WECF accelerates activated T cell, but not B cell, apoptosis and WECF concurrently inhibited cytokine production of activated T cells. Taken together, WECF exhibits B cell mitogenic activity and differential toxicity more pronounced to T cells, suggesting a possible in vivo application of WECF for specific control of T cells without alteration of B cell activity.

In vitro에서 Levamisole이 양식뱀장어의 면역조절작용에 미치는 영향 (Effect of Levamisole on Immunomodulation of Eels (Anguilla japonica) In Vitro)

  • Levamisloe(LMS)이 뱀장어의 면역반응에 미치는 영향을 측정하기 위해서 뱀장어의 혈액 및 신장으로부터 백혈구를 분리하여, 림프구의 증식능, MIF 생산능, 자연살해세포활성능, 탐식세포의 탐식능, superoxide anion, hydrogen peroxide 및 lysozyme 활성능 등을 측정하였다. LMS는 T-cell 마이토젠인 Con A 및 PHA 처리시에는 LMS 농도에 비례감소를 보인 반면에 B-cell 마이토젠인 LPS에는 LMS 처리와는 무관하게 큰 차이를 보이지 않았다. 한편 MIF 및 MAF의 생성에는 LMS 처리 농도에 비례 증가되었다. NK 세포의 활성은 LMS 첨가농도에 비례 증가하였는데, 이는 표적세포에 대한 결합율을 증가시킨 결과 NK 세포의 활성이 증가된 것으로 보인다. LMS는 백혈구의 탐식능, superoxide anion의 생성능, hydrogen peroxide 생성능 및 Iysozyme의 활성을 증가시켰다. 이러한 결과로부터 LMS가 어체의 면역작용에 관여하는 작용기전은 면역 작동세포수의 증가를 촉진시키기보다는 분화를 촉진시켜서 cytokine의 방출증가 및 세포의 기능활성을 증강시킴을 알수 있다.
