  • E. coli TGI/pDG7 $\alpha$ 와 빵효모의 혼합세포계에 의한 글루타치온 생산을 Aerated Slurry Bioreactor에서 수행하였다. 글루타치온 생산 효소는 34시간 동안 안정성을 보였으며, 글루타치온 수율은 4.6 mM을 유지함을 보였다. 고농도의 혼합세포를 이용한 Aerated Slurry Bioreactor에서 기질의 공급속도가 글루타치온 생산에 미치는 영향을 조사하였다. 세포가 회수되어 기질공급속도 5.2 mL/hr에서 글루타치온의 생산농도는 32시간에서 현탁반응보다 낮아짐을 보였고, 반면 세포가 회수되지 않은 기질공급속도 5.2 mL/hr에서는 22시간 동안 유지되었고 2.6 mL/hr에서는 42시간 동안 유지되었다. 글루타치온의 생산성은 기질공급속도가 10.4 mL/hr에서 25.7 mg/g wet $cell{\cdot}hr$이었으나, 2.6 mL/hr에서는 1.65 mg/g wet $cell{\cdot}hr$이었고 5.2mL/hr에서는 5.28 mg/g wet $cell{\cdot}hr$이었다. 계면활성제(Tween 80)을 사용한 경우, 글루타치온 생산시간을 25시간에서 45시간으로 연장되었다. 글루타치온 생산있어 산소가 세포계의 효소에 산소에 의해 영향을 미칠 것으로 판단되었고, 공기를 질소로 교체하여 글루타치온 생산시간을 40시간으로 연장시켰다.

In vitro rumen fermentation kinetics, metabolite production, methane and substrate degradability of polyphenol rich plant leaves and their component complete feed blocks

  • Background: This experiment aimed at assessing polyphenol-rich plant biomass to use in complete feed making for the feeding of ruminants. Methods: An in vitro ruminal evaluation of complete blocks (CFB) with (Acacia nilotica, Ziziphus nummularia leaves) and without (Vigna sinensis hay) polyphenol rich plant leaves was conducted by applying Menke's in vitro gas production (IVGP) technique. A total of six substrates, viz. three forages and three CFBs were subjected to in vitro ruminal fermentation in glass syringes to assess gas and methane production, substrate degradability, and rumen fermentation metabolites. Results: Total polyphenol content (g/Kg) was 163 in A. nilotica compared to 52.5 in Z. nummularia with a contrasting difference in tannin fractions, higher hydrolysable tannins (HT) in the former (140.1 vs 2.8) and higher condensed (CT) tannins in the later (28.3 vs 7.9). The potential gas production was lower with a higher lag phase (L) in CT containing Z. nummularia and the component feed block. A. nilotica alone and as a constituent of CFB produced higher total gas but with lower methane while the partitioning factor (PF) was higher in Z. nummularia and its CFB. Substrate digestibility (both DM and OM) was lower (P < 0.001) in Z. nummularia compared to other forages and CFBs. The fermentation metabolites showed a different pattern for forages and their CFBs. The forages showed higher TCA precipitable N and lower acetate: propionate ratio in Z. nummularia while the related trend was found in CFB with V. sinensis. Total volatile fatty acid concentration was higher (P < 0.001) in A. nilotica leaves than V. sinensis hay and Z. nummularia leaves. It has implication on widening the forage resources and providing opportunity to use forage biomass rich in polyphenolic constituents in judicious proportion for reducing methane and enhancing green livestock production. Conclusion: Above all, higher substrate degradability, propionate production, lower methanogenesis in CFB with A. nilotica leaves may be considered useful. Nevertheless, CFB with Z. nummularia also proved its usefulness with higher TCA precipitable N and PF. It has implication on widening the forage resources and providing opportunity to use polyphenol-rich forage biomass for reducing methane and enhancing green livestock production.

Response of Growth Performance, Cecal Fermentation Traits and In vitro Gas Production to Substitution of Soyhulls for Lignified Fiber in Rabbit Diets

  • A growth trial (Expt. 1) and an in vitro fermentation experiment (Expt. 2) were conducted to determine the response of growth performance, cecal fermentation characteristics and in vitro gas production to incremental levels of substitution of digestible fiber for lignified fiber in the diet of weaned rabbits. Three diets, formulated by substituting soyhulls (SH; used as digestible fiber source) for soybean straw (used as lignified fiber source) at substitution levels of 0, 25 and 50%, were used in a factorial design. In the growth trial (Expt. 1), increasing levels of SH substitution resulted in a quadratic increase in daily body weight gain rate (p<0.04) and feed conversion efficiency (p<0.02), but in a numerical decrease in dietary DM intake (p=0.15). When SH were included in the diet at 25% substitution level, rabbits had the highest rate of liveweight gain and feed conversion efficiency. As SH substitution level increased, pH values and ammonia-N of cecal contents linearly (p<0.001) decreased, but total VFA concentration linearly (p<0.03) increased. With incremental levels of SH substitution, the percentage of acetate and butyrate linearly (p<0.05) reduced, but the percentage of propionate and minor acids linearly (p<0.03) increased. Increasing the SH substitution levels tended to increase incidence of diarrhea. In the in vitro fermentation experiment (Expt. 2), regardless of origin of substrates fermented, increasing SH substitution level resulted in increased maximal gas production (p<0.001) and shortened gas production lag time, but had no effect on gas production rate (p>0.2). These observations suggest that incrementally feeding SH to rabbits could stimulate their cecal microbial activity, allowing cecal fermentation to shift towards favoring fiber digestion. In conclusion, digestible fiber from soyhulls may partially substitute for more lignified fiber, soybean straw, without having an adverse effect on cecal fermentative and microbial activity and growth performance. For growing rabbits, about 73% of total dietary NDF should be supplied by effective NDF, the remainder could come from digestible NDF, such as soyhulls.

Effects of harvest time and added molasses on nutritional content, ensiling characteristics and in vitro degradation of whole crop wheat

  • Objective: Wheat is an alternative to corn silage for ruminant feeding in northern China. This study examined the effects of harvest time and added molasses on nutritional content, ensiling characteristics and in vitro degradation of whole crop wheat (WCW). Methods: Fresh WCW at the milk-ripe stage was harvested at 0700 h (i.e., in the morning [Mo]) and 1700 h (i.e., in the afternoon [Af]), and then immediately used to prepare silage and make hay. Commercial molasses was added to Af WCW at 0%, 2%, 4%, and 6% (fresh weight) proportions. The WCW treated with molasses was mixed thoroughly prior to ensiling. Results: Dry matter (DM), neutral detergent fiber, water soluble carbohydrate (WSC) content (p<0.01), accumulative gas production in 72 h ($GP_{72h}$, 77.46 mL/g vs 95.15 mL/g) and dry matter disappearance in vitro (69.15% vs 76.77%) were lower (p<0.05), while crude protein (CP) content was higher for WCW silage (WCWS) compared to WCW (p<0.01). The propionic acid and butyric acid concentrations in WCWS from Mo WCW were 1.47% and 0.26%, respectively. However, the propionic and butyric acid concentrations were negligible, while the ammonia nitrogen/total nitrogen ($NH_3-N/TN$, p<0.01) concentration was lower and the rate of gas production at 50% of the maximum (17.05 mL/h vs 13.94 mL/h, p<0.05) was higher for Af WCWS compared to Mo WCWS. The incubation fluid's $NH_3-N$ concentration was lower in WCWS and Af WCW compared to Mo WCW (p<0.05). The CP and WSC content increased with increasing molasses levels (p<0.05). Furthermore, the pH (p<0.01) and time when gas production was 50% of the maximum (2.78 h vs 3.05 h, p<0.05) were lower in silage treated with 4% molasses than silage without molasses. Conclusion: Harvesting wheat crops in the afternoon and adding molasses at 4% level to WCW optimally improved ensiling characteristics, leading to well-preserved silage.

The Effects of Thyme and Cinnamon Essential Oils on Performance, Rumen Fermentation and Blood Metabolites in Holstein Calves Consuming High Concentrate Diet

  • Essential oils have been shown to favorably effect in vitro ruminal fermentation, but there are few in vivo studies that have examined animal responses. The objective of this study was to evaluate the effects of thyme (THY) and cinnamon (CIN) essential oils on feed intake, growth performance, ruminal fermentation and blood metabolites in feedlot calves fed high-concentrate diets. Twelve growing Holstein calves ($213{\pm}17kg$ initial BW) were used in a completely randomized design and received their respective dietary treatments for 45 d. Treatments were: 1-control (no additive), 2-THY (5 g/d/calf) and 3-CIN (5 g/d/calf). Calves were fed ad libitum diets consisting of 15% forage and 85% concentrate, and adapted to the finishing diet by gradually increasing the concentrate ratio with feeding a series of transition diets 5 wk before the experiment started. Supplementation of THY or CIN did not affect DMI and ADG, and feed efficiency was similar between treatment groups. There were no effects of additives on ruminal pH and rumen concentrations of ammonia nitrogen and total VFA; whereas molar proportion of acetate and ratio of acetate to propionate decreased, and the molar proportion of propionate increased with THY and CIN supplementation. Rumen molar concentration of butyrate was significantly increased by adding CIN compared to control; but no change was observed with THY compared with control group. No effects of THY, or CIN were observed on valerate, isobutyrate or isovalerate proportions. Plasma concentrations of glucose, cholesterol, triglyceride, urea-N, ${\beta}$-hydroxybutyrate, alanine aminotransferase and aspartate aminotransferase were not changed by feeding THY or CIN. Results from this study suggest that supplementing a feedlot finishing diet with THY or CIN essential oil might be useful as ruminal fermentation modifiers in beef production systems, but has minor impacts on blood metabolites.

Anti-adherence of Antibacterial Peptides and Oligosaccharides and Promotion of Growth and Disease Resistance in Tilapia

  • Four hundred and fifty tilapias ($6.77{\pm}0.23$ g) were assigned randomly to six groups to evaluate the feasibility of the tested antibacterial peptides (ABPs) and oligosaccharides as substitutes for antibiotics. The control group was fed with a commercial tilapia diet; other five groups were fed with the same commercial diet supplemented with konjac glucomannan (KGLM), cluster bean galactomannan (CBGAM), and three animal intestinal ABPs derived from chicken, pig and rabbit at 100 mg/kg respectively. After 21 days of feeding, growth, disease resistance, and in vivo anti-adherence were determined. Furthermore, the inhibitory effect of tested agents on adhesion of Aeromonas veronii biovar sobria (A.vbs) strain BJCP-5 to tilapia enteric epithelia in vitro was assessed by cell-ELISA system. As a result, the tested agents supplemented at 100 mg/kg show significant benefit to tilapia growth and disease resistance (p<0.05), and the benefit may be correlated with their interfering in the contact of bacteria with host mucosal surface. Although none of the tested agents did inhibit the growth of BJCP-5 in tryptic soy broth at $100{\mu}g/ml$, all of them did inhibit the adhesion of A.vbs to tilapia enteric epithelia in vivo and in vitro. In vitro mimic assays show that three ABPs at low concentrations of $25{\mu}g/ml$ and $2.5{\mu}g/ml$ have the reciprocal dose-dependent anti-adherence effect. The inhibition of ABPs may be correlated with a cation bridging and/or receptor-ligand binding, but not with hydrophobicity. The KGLM and CBGAM inhibited the adherence of BJCP-5 to tilapia enteric epithelia with dose-dependent manner in vitro, and this may be through altering bacterial hydrophobicity and interfering with receptor-ligand binding. Our results indicate that the anti-adherence of the tested ABPs and oligosaccharides may be one of the mechanisms in promoting tilapia growth and resistance to A.vbs.

Study on the rumen fermentation, growth performance and carcass characteristics according to the supplementation of lupin flake in Hanwoo steers

  • This study was conducted to determine the rumen fermentation dynamics of lupin flakes and elucidate the effects of lupin flake supplementation on the growth performance, blood metabolites, and carcass characteristics of Hanwoo steers. In vitro and in situ trials of lupin grains and lupin flakes were conducted using three Hanwoo cows with rumen fistulas. The feeding trial included 40 early-fattening Hanwoo steers randomly divided into four groups: control, T1, T2, and T3. Their formula feed contained 0%, 3%, 6%, and 9% lupin flakes, respectively. In vitro rumen pH and ammonia concentrations were lower in the lupin flake group than in the lupin grain group after 6 and 24 h of incubation, respectively (p < 0.05). Concentrations of propionate, butyrate, and total volatile fatty acids were higher in the lupin flake group than in the lupin grain group after 12 h of incubation (p < 0.05), as was the crude protein disappearance rate at 9 and 12 h of rumen fermentation (p < 0.05). Supplementation with lupin flakes did not affect the average daily gain. Compared to that in the control group, dry matter intake was lower in the lupin flake-supplemented groups (p < 0.05); the feed conversion ratio was lower in T2 and T3 (p < 0.05); and plasma total protein concentration in 29-month-old steers was lower in T1 and T3 (p < 0.05). Plasma triglyceride concentration was lower in the lupin flake-supplemented groups than in the control group (p < 0.05). The incidence rate of yield grade A was higher in T1 and T2 than in the control group; the incidence rate of meat quality 1+ grade or higher was highest in T2. The carcass auction price was higher in T2 than in the other groups. Overall, compared to whole lupin grains, lupin flakes seem to more substantially affect rumen ammonia concentrations and crude protein disappearance rate. Additionally, we suggest that supplementation with 6% lupin flake formula feed exerts positive effects on the feed conversion ratio, yield grade, and quality grade of Hanwoo steers.

Korean Red Ginseng (Panax ginseng) Potentiates the Inhibitory Actions of Testosterone on Obesity and Adipogenesis in High Fat Diet-Fed Castrated Mice

  • It has been suggested that ginseng is beneficial for ameliorating the aging males' symptoms, such as weight gain, fatigue, erectile dysfunction, and depression, in elderly men with testosterone deficiency. We thus investigated the effects of Korean red ginseng (Panax ginseng C.A. Meyer; Araliaceae) on obesity in a mouse model of testosterone deficiency (castrated C57BL/6J mice). The effects of ginseng extract (GE) and/or testosterone on obesity and adipogenesis in high-fat diet (HFD)-fed castrated C57BL/6J mice and 3T3-L1 adipocytes were examined using in vivo and in vitro approaches. After feeding mice a HFD for 8 weeks, we found that mice also receiving GE and/or testosterone showed decreased body weight, adipose tissue mass, adipocyte size, and hepatic lipid accumulation compared with untreated HFD-fed mice. Expression of adipogenic genes ($PPAR{\gamma}$, $C/EBP{\alpha}$, and aP2) was decreased by GE and/or testosterone in adipose tissues. Consistent with the in vivo data, lipid accumulation and the mRNA expression of adipogenesis genes in 3T3-L1 adipocytes were decreased by GE, ginsenosides, and testosterone. The inhibitory effects of GE (or ginsenosides) were comparable to those of testosterone, and the effects of co-treatment with GE (or ginsenosides) and testosterone were greater than those of testosterone alone in vivo and in vitro. Our results indicate that ginseng may be able to potentiate the inhibitory effects of testosterone on obesity and adipogenesis in HFD-fed castrated mice, providing possible therapeutic implications in men with testosterone deficiency.

Nutritive Value of Wheat Straw Treated with Pleurotus Fungi

  • Soaked and pasteurised wheat straw was inoculated with five species of Pleurotus fungi (coded P-21, P-30, P-41, P-60 and P-90), packed in polyethylene bags and incubated in a fermentation chamber for 21 days. The chemical composition, in vitro digestibility and in sacco degradability of the treated and untreated straw were estimated using a complete randomised design consisting of six treatments and four replicates. In a feeding trial, in vivo digestibility and voluntary intake were determined in bulls, using a $3{\times}3$change over design. Dietary treatments were: 1) untreated wheat straw (UWS) as control; 2) fungal treated (P-41) wheat straw before mushroom formation (FTWS); 3) spent wheat straw (SPWS) after mushrooms were harvested. Apart from P-90, fungal treatment significantly (p<0.05) increased the crude protein (CP) and reduced the cell wall components of the straw. The in vitro dry mater and organic mater digestibility significantly (p<0.05) increased in the treated straw particularly with the treatments of P-41 and P-60. The in situ degradability and in vivo digestibility of DM and OM were significantly (p<0.05) increased in treated straws with the highest values observed for treatment P-41. The intake of DM, OM and digestible organic mater (DOM) were significantly (p<0.05) increased in cows fed FTWS.

  • 진딧물 방제제 개발을 위해 In vitro 경엽살포 검정방법이 널리 사용되고 있다. 이러한 신소재 진딧물 방제 제형은 종합방제와 화학농약의 대안으로 많은 연구가 진행되고 있다. 하지만, 경엽살포 검정방법은 환경이 조절되는 실내에서도 진딧물의 증식과 살충에 영향을 받는다. 본 연구에서는 담배를 기주로 하여 솜과 한천방법을 이용하여 진딧물 방제제 검정을 위한 최적 경엽살포 확립하고자 하였다. 진딧물 검정 챔버에 솜과 한천을 넣은 후 담배 잎과 진딧물 3-4령 약충을 접종하였다. Water-sensitive paper를 이용하여 경엽살포 시에 가장 표면 피복이 높은 최적 경엽살포 거리와 살포량을 확립하였다. 대조구로 물을 처리한 구에서 한천 방법이 솜 방법에 비해 살충율이 낮고, 증식율이 높았다. 솜 검정 방법에는 곤충 검정 챔버의 상대습도를 60% 이상 유지시켰을 때 가장 최적 조건이었지만, 한천 검정 방법에서는 한천의 농도에 상대습도 차이가 없었다. 최적화된 조건하에서 대조화학 농약, Sulfoxaflor, 경엽살포 시 솜 방법에서 살충율이 한천방법보다 빨랐지만, 최종 살충율은 통계적으로 유의하지 않았다. 본 연구는 살진딧물 물질을 검정 시 재현성과 활용성이 가능한 최적화된 증식율과 살충율 검정 조건을 제시하였다.
