형질전환(形質轉換)된 포플러의 딱정벌레에 대한 저항성(抵抗性) 유전자(遺傳子)(Proteinase Inhibitor II) 발현(發現) (Gene Manipulation of Pin 2(Proteinase Inhibitor II) to the Cottonwood Leaf Beetle(Coleoptera : Chrysomelidae) in Transgenic Poplar(Populus deltodies × P. nigra))

  • 외래 저항성 유전자, Proteinase inhibitor II가 형질전환된 3계통의 벨기에 포플러를 대상으로 딱정벌레에 대한 유전자 발현정도가 기내에서 조사되었다. 포플러 계통은 선발 유전자로서 Nos-promoter와 Neomycin phosphotransferase gene에 의하여 조절되고 곤충에 대한 저항성 유전자로서 CaMV-35S와 Pin2(Proteinase inhibitor II)에 의한 형질전환체이다. 특히, 형질전환된 포플러의 내충성 저항력을 조기검정하기 위하여, 조직배양을 응용한 새로운 방법으로서 곤충의 알을 표면 살균하여 기내의 조직배양묘와 배양하는 동시배양 방법이 이용되었다. 형질전환된 포플러의 저항성은 기내에서 유충에 의해 섭취된 잎면적, 잎 섭취에 의한 유충의 무게 증감, 유충의 성장단계 등에 의하여 조사되었다. 특히, 잎면적은 각각의 LPI(Leaf plastochron index)별로 측정되었고, 잎면적, 유충의 무게, 곤충의 성장 속도는 형질전환체와 비형질전환체 간에 큰 차이를 보였다. 기내에서 무병상태로 배양된 알들이 부화된 후, 유충의 잎 섭취도는 LPI 4와 5사이에서 가장 높았다. 본 실험의 기내 배양법은 외래유전자를 삽입한 이후에 곧바로 발현을 빠른 시간내에 조기검정 할 수 있는 새로운 방법의 개발이라 할 수 있다.

반추위액 채취 시간이 사료작물의 In vitro 건물 소화율에 미치는 영향 (Effect of Collection Times of Rumen Fluid on In vitro Dry Matter Digestibility of Forage Crops)

  • 본 실험은 국립축산과학원에 초지사료연구센터에서 조사료의 IVDMD 분석을 위해 사용 중인 Holstein을 이용하여, 혼합건초를 자유 채식케 하는 Holstein에서 반추위액의 채취 시간이 사료작물의 IVDMD에 미치는 영향을 알아보기 위하여 실시하였다. 사료작물은 청보리 (Yong-yang, Yu-yeong), 크림손 클로버 (Tibbee), 이탈리안 라이그라스(Kospeed, Kogreen, Hwasan 101), 호밀(Wintergreen) 등 4작물 7품종을 공시재료로 이용하였으며, 반추위액의 채취 시간은 T1: 농후사료 급여 전, T2: 사료급여 직후(급여 1시간 후), T3: 사료급여 후 4시간, T4: 사료급여 후 8시간으로 하였다. 모든 사료작물에서 T1 처리구의 위액을 이용한 IVDMD가 T2, T3, T4 처리구에서 채취한 위액을 이용한 IVDMD 보다 높게 나타났다. 특히 IRG의 Kospeed의 경우 T1 처리구에서 T2 처리구보다, Kogreen의 경우 T1 처리구에서 T2 및 T3 처리구보다 IVDMD이 유의적으로 높게 나타났다(p<0.05). 하지만 나머지 모든 사료작물에서 처리구간에 IVDMD의 차이는 있었지만 유의성은 인정되지 않았다. 이러한 결과는 혼합건초의 자유채식이 사료급여 횟수를 증가시킴과 동시에 조사료를 충분히 급여하는 효과를 가져와 침 분비량이 증가하여, 배합사료 급여 후에도 제 1 위 내 pH가 정상범위를 유지하여 사료작물의 안정적인 소화에 영향을 미친 것으로 생각된다.

Effect of Different Ratios of Concentrate and Roughage on Lipid Synthesis by Rumen Microorganisms In Vitro

  • The effect of different feeding ratios of concentrate to roughage on ruminal lipid synthesis in vitro was examined. Three sheep fitted with a rumen fistula were fed three different ratios (8:2, 4:6 and 0:10) of concentrate and roughage, and their rumen liquor were used for incubation. $^{13}C$-labeled glucose or sodium acetate as substrate was added to cultures of rumen liquor, and they were incubated for 6 h. The total lipid in the culture of the rumen liquor was extracted, and the percentage of $^{13}C$ excess was analyzed. The percentage of $^{13}C$ excess recovered when incubated with glucose increased with increased ratio of concentrate in the diet. The values of cultures incubated with glucose were higher than those incubated with sodium acetate except the roughage-only feeding. In the roughage-only diet, the percentage of $^{13}C$ excess when incubated with sodium acetate was highest of all diets. The recovery percentage of $^{13}C$ from glucose increased with increased ratio of concentrate. The recovery percentage of $^{13}C$ from sodium acetate addition in only roughage feeding was highest among the three diets. The recovery percentage of $^{13}C$ from glucose was markedly higher than that of sodium acetate addition in all feedings. The results indicate that high concentrate feeding facilitates lipid synthesis by rumen microorganisms, and that glucose may be the precursor for lipid synthesis rather than acetic acid.

Phenolic Composition, Fermentation Profile, Protozoa Population and Methane Production from Sheanut (Butryospermum Parkii) Byproducts In vitro

  • Sheanut cake (SNC), expeller (SNE) and solvent extractions (SNSE) samples were evaluated to determine their suitability in animal feeding. The CP content was highest in SNSE (16.2%) followed by SNE (14.7%) and SNC (11.6%). However, metabolizable energy (ME, MJ/kg) was maximum in SNC (8.2) followed by SNE (7.9) and SNSE (7.0). The tannin phenol content was about 7.0 per cent and mostly in the form of hydrolyzable tannin (HT), whereas condensed tannin (CT) was less than one per cent. The in vitro gas production profiles indicated similar y max (maximum potential of gas production) among the 3 by-products. However, the rate of degradation (k) was maximum in SNC followed by SNE and SNSE. The $t^{1/2}$ (time taken for reaching half asymptote) was lowest in SNC (14.4 h) followed by SNE (18.7 h) and SNSE (21.9 h). The increment in the in vitro gas volume (ml/200 mg DM) with PEG (polyethylene glycol)-6000 (as a tannin binder) addition was 12.0 in SNC, 9.6 in SNE and 11.0 in SNSE, respectively. The highest ratio of $CH_4$ (ml) reduction per ml of the total gas, an indicator of the potential of tannin, was recorded in SNE (0.482) followed by SNC (0.301) and SNSE (0.261). There was significant (p<0.05) reduction in entodinia population and total protozoa population. Differential protozoa counts revealed that Entodinia populations increased to a greater extent than Holotricha when PEG was added. This is the first report on the antimethanogenic property of sheanut byproducts. It could be concluded that all the three forms of SN byproducts are medium source of protein and energy for ruminants. There is a great potential for SN by-products to be incorporated in ruminant feeding not only as a source of energy and protein, but also to protect the protein from rumen degradation and suppress enteric methanogenesis.

Three-step in vitro digestion model for evaluating and predicting fecal odor emission from growing pigs with different dietary protein intakes

  • Objective: The objective of this study was to select an effective in vitro digestion-fermentation model to estimate the effect of decreasing dietary crude protein (CP) on odor emission during pig production and to suggest potential prediction markers through in vitro and in vivo experiments. Methods: In the in vitro experiment, three diet formulations with different CP contents (170 g/kg, 150 g/kg, and 130 g/kg) but containing the same standardized ileal digestible essential amino acids (SID-EAA) were assessed. Each diet was evaluated by two different in vitro gastric-intestinal phase digestion methods (flask and dialysis), combined with fresh pig feces-ferment inoculation. Eighteen growing barrows (31.9±1.6 kg) were divided into three groups: control diet (180 g CP/kg, without SID-EAA adjustment), 170 g CP/kg diet, and 150 g CP/kg diet for 4 weeks. Results: The in vitro digestion results indicated that in vitro digestibility was affected by the gastric-intestinal phase digestion method and dietary CP level. According to the gas kinetic and digestibility results, the dialysis method showed greater distinguishability for dietary CP level adjustment. Nitrogen-related odor compounds (NH3-N, indole, p-cresol, and skatole) were highly correlated with urease and protease activity. The feeding study indicated that both EAA-adjusted diets resulted in a lower odor emission especially in p-cresol and skatole. Both protease and urease activity in feces were also closely related to odor emissions from nitrogen metabolism compounds. Conclusion: Dialysis digestion in the gastric-intestinal phase followed by fresh fecal inoculation fermentation is suitable for in vitro diet evaluation. The enzyme activity in the fermentation and the fecal samples might provide a simple and effective estimation tool for nitrogen-related odor emission prediction in both in vitro and in vivo experiments.

In vitro inhibition of 10-formyltetrahydrofolate dehydrogenase activity by acetaldehyde

  • Alcoholism has been associated with folate deficiency in humans and laboratory animals. Previous study showed that ethanol feeding reduces the dehydrogenase and hydrolase activity of 10-formyltetrahydrofolate dehydrogenase (FDH) in rat liver. Hepatic ethanol metabolism generates acetaldehyde and acetate. The mechanisms by which ethanol and its metabolites produce toxicity within the liver cells are unknown. We purified FDH from rat liver and investigated the effect of ethanol, acetaldehyde and acetate on the enzyme in vitro. Hepatic FDH activity was not reduced by ethanol or acetate directly. However, acetaldehyde was observed to reduce the dehydrogenase activity of FDH in a dose- and time-dependent manner with an apparent $IC_{50}$ of 4 mM, while the hydrolase activity of FDH was not affected by acetaldehyde in vitro. These results suggest that the inhibition of hepatic FDH dehydrogenase activity induced by acetadehyde may play a role in ethanol toxicity.

Evaluating feed value of native Jeju bamboo (Sasa quelpaertensis Nakai) for beef cattle

  • Objective: Recently, indigenous Korean grass Sasa quelpaertensis Nakai (SQ) has garnered much interest as a roughage source for livestock to mitigate its adverse effects on habitat diversity. Thus, the objective of the present study was to evaluate the ruminal fermentation, palatability, and nutrient digestibility of SQ for Korean native beef cattle (Hanwoo) using in vitro rumen fermentation, in situ rumen degradability, and in vivo feeding trials. Methods: Using in vitro tests with rumen fluid as the inoculum for 48 h, ruminal fermentation of SQ was evaluated and compared with that of other roughage sources commonly used in Korea (i.e., rice straw, Timothy hay, and Italian ryegrass [IRG]). Additionally, an in situ trial 96 h was performed using three cannulated Hanwoo steers. Further, an in vivo trial was performed using eight Hanwoo steers to compare the palatability of SQ with rice straw in total mixed ration (TMR) and forage-concentrate separate feeding conditions. Finally, an in vivo digestibility trial of SQ fed as TMR of two particle sizes was performed with four Hanwoo steers. Results: In vitro and in situ trials revealed that SQ was comparable or superior to rice straw in terms of the ruminal fermentation characteristics of pH, gas production, total volatile fatty acid content, and effective ruminal dry matter digestibility (DMD), although its fermentability was lower than that of Timothy hay and IRG. In the palatability test, steers showed a greater preference for SQ when given as TMR. The total tract DMD of SQ fed as TMR was 75.9%±1.37%, and it did not differ by particle size. Conclusion: The feed value of SQ as a roughage source for Hanwoo steers is comparable or superior to that of rice straw, particularly when provided as TMR.

지방의 공급형태와 Ca 및 Mg의 첨가가 In Vitro 발효 및 비육후기 한우의 성장성적에 미치는 영향 (Effects of Supplementation of Fat Sources, Ca and Mg on In Vitro Fermentation and the Performance of Finishing Hanwoo Bulls)

  • 본 실험은 지방의 공급형태에 따른 반추위발효성상, 영양소 이용효율 및 증체율을 관찰하기 위하여 in vitro 실험과 in vivo 사양 실험을 수행하였다. In vitro에서는 지방원료로서 tallow(T), 그리고 tallow에 2가 양이온은 MgO(T-MgCa), $CaCl_2$(T-Ca), MgO와 $CaCl_2$의 혼합(T-MgCa) 및 Ca-salt 처리 tallow(T-CaS)의 총 5개의 처리구를 두었다. In vitro 배양 시간은 0, 6, 12, 및 24 hr별로 두었다. In vivo 사양 실험은 in vitro 실험을 통하여 얻은 결과를 토대로 성적이 좋은 T-MgCa 구와 T-CaS구를 선택하여 지방 무첨가구를 대조구로 하여 비육후기 한우의 성장률 및 육질에 미치는 영향을 관찰하고자 처리구당 10두씩 3 처리구로 완전임의 배치하여 80일간 실시하였다. 실험 결과를 살펴보면 배양시간 6 hr 이후부터 처리구들간 변화를 보였는데 6 hr에서는 T-MgCa 구와 T-CaS 구에서의 pH 값이 다른 처리구들의 값보다 높게 조사되었고(P<0.05), 12 hr에서는 T와 T-Mg 구에서의 pH 값이 MgO와 $CaCl_2$ 첨가구 보다 높게 조사되었다(P<0.05). Ammonia_N의 농도는 6hr를 제외하고는 처리구들간 통계적 유의차이는 없었다. 6 hrdptjsm ammonia-N 농도는 tallow 단독 처리구에서의 농도가 다른 처리구들보다 낮았다(P<0.05). 총 VFA 생성량은 6 hr에서는 처리구들간 통계적 유의차이가 없었지만 12 hr와 24 hr에서는 T 처리구에 비하여 tallow에 2가 양이온 첨가 또는 Ca-salt를 혼합 첨가한 처리구들에서 높게 조사되었다(P<0.05). 영양소(DM, OM, CP 및 NDF)분해율은 12 hr 이후부터 tallow 단독 처리구보다 양이온이나 Ca-salt 처리 tallow 구에서 보다 높은 경향으 보였다. 용해성 지방산은 전 배양 시간대에 걸쳐 T-CaS 구에서 가장 낮았으나, 불용성 지방산 염 형성은 가장 높았다(P<0.05). 사료급여 30일까지(feeding period I)는 사료섭취량, 일당증체량 및 사료 이용효율이 처리구들간 통계적 유의차이가 없었다. 반면, feeding period II(31${\sim}$50일)와 III(51${\sim}$80일) 그리고, 전 급여기간 동안 일당 증체량과 사료이용효율은 T-MgCa>T-CaS>Control순으로 높았다(P<0.05). 결론적으로 사료 섭취량의 증가가 제한되는 비거세 한우의 비육 마무리기에 있어서 고 에너지 사료제조를 위한 우지의 첨가시 Ca 및 Mg 보충은 반추위 발효성상 및 성장성적의 개선효과가 있음이 확인되었다.

In Vitro and In Vivo Antioxidant Activity of Aged Ginseng (Panax ginseng)

  • Fresh ginseng roots were aged in an oven at $80^{\circ}C$ for 14 d. The in vitro and in vivo antioxidant activities of this aged ginseng, in comparison with those of the white and red ginsengs, were evaluated. In in vitro antioxidant assays, the ethanolic extracts from aged ginseng showed significantly higher free radical scavenging activity and reducing power than those of the white and red ginsengs. In in vivo antioxidant assays, mice were fed a high fat diet supplemented with white, red, or aged ginseng powders. High fat feeding resulted in a significant increase in lipid peroxidation and a substantial decrease in antioxidant enzymes activities in the animals. However, diet supplementation of ginseng powders, particularly aged ginseng, markedly reduced lipid peroxidation and enhanced the antioxidant enzymes activities. The results illustrate that the aged ginseng has greater in vitro and in vivo antioxidant capacity than the white and red ginsengs. The aged ginseng also showed considerably higher total saponin, phenolic, and flavonoid contents, indicating that its antioxidant capacity may have been partly due to its high levels of antioxidant compounds. This new ginseng product may be useful as a functional food with strong antioxidant potential.

Chemical Composition and Feeding Value of Chopped Date Fronds (CDF) as Affected by Urea and Date Syrup Extract Treatment

  • In the first of two experiments, chemical composition (Ash, CP and CF contents), nutritive value (in vitro organic matter disappearance "lVOMD" and energy content "ME") and nitrogen retention of chopped date fronds (CDF) as affected by chemical treatment (CT) and storage duration (SD) were investigated. The experimental design was a completely randomized block, arranged in a $3{\times}4$ factorial, with 3 (CT) and 4 (SD) as the main factors using 2 replications / treatment. Three chemical treatments, were used : Control (With zero or no chemical treatment); treatment with 4% urea solution (w /w) and treatment with 4% urea + Date syrup extract (DS). The 4 (SD) were: Control (immediately after opening - without any storage or zero time storage); one month storage period; two months storage period and three months storage period. Both (CT) and (SD) had significant effects (p < 0.05) on CP and ash contents of the CDF, however there were no significant effects (p > 0.05) due to (CT) and (SD) on CF and nutritive value. High retention value (> 80%) for nitrogen was reported for the treated CDF, immediately after opening the incubated material (zero SD). However, nitrogen retention decreased with increasing (SD) to 67% after one month (SD) but tended to stay at a fairly constant level of 67% until 3 months of storage. In experiment 2, the feeding value of treated CDF was evaluated in comparison to Rhodes grass hay, using growing goats and sheep. A $4{\times}2$ factorial design was used (Four roughage sources were used : Rhodes grass hay, untreated CDF, urea-treated CDF and CDF treated with urea + DS with sheep and goats as two animal species, using three animals / treatment). Untreated CDF had a similar feeding value to Rhodes grass hay. However treatment of CDF with urea alone or with urea + DS depressed animal performance of both goats and sheep, apparently due to depression of feed intake (appetite). Feeding untreated CDF decreased feeding cost by 29% and cost/kg gain by 23%. CDF showed a good potential as a cheap local roughage and emergency feed for ruminants in the Sultanate.
