Inhibition of Drug-metabolizing Enzyme and Drug Transporter by Major Components of Phellodendri cortex (황백의 주요 구성 화합물에 의한 약물대사효소 및 약물수송단백 저해능 평가)

  • We evaluated the potential of major components of Phellodendri cortex to inhibit the activities of CYP2D6 and p-glycoprotein. The abilities of berberine, palmatine, limonin, and rutaecarpine to inhibit CYP2D6-mediated dextromethorphan O-demethylation and calcein AM accumulation were tested using human liver microsomes and L-MDR1 cell, respectively. Berberine strongly inhibited CYP2D6 isoform activity, whereas limonin and reuaecarpine did not. The $IC_{50}$ value of berberine was reduced after preincubation with microsomes in the presence of NADPH generating system, suggesting that berberine is a mechanism based inhibitor. In addition, all chemicals tested, didn't show inhibitory effect on p-glycoprotein activity. These results suggest that berberine has potential to inhibit CYP2D6 activity in vitro. Therefore, in vivo studies investigating the interactions between berberine and CYP2D6 substrates are necessary to determine whether inhibition of CYP2D6 activity by berberine is clinically relevant.

Effect of Deep Sea Water on Phase I, Phase II and Ornithine Decarboxylase. (Phase I, phase II 효소 및 ornithine decarboxylase에 미치는 해양심층수의 영향)

  • Deep sea water was tested for cancer chemopreventive activity by measuring the activities of ${\beta}-$ naphthoflavone $({\beta}-NF)-induced$ cytochrome P 450 1A2 (CYP 1A2), quinone reductase (QR) and glutathione-S-transferase (GST), glutathione (GSH) levels, and ornithine decarboxylase (ODC) activity. The in vitro incubation of rat liver microsome with deep sea water (a hardness range of $100{\sim}1,000$) showed a hardness-dependent inhibition of CYP 1A2 activity. QR and GST activities were induced about $1.1{\sim}1.2$ fold with the treatment of deep sea water in murine hepatoma Hepa 1clc7 cells. In addition GSH levels were increased $1.3{\sim}1.4$ fold in a hardness range of $100{\sim}1,000$. The deep sea water showed 20.3 and 35.0% inhibition of 12-O- tetradecanoylphorbol-13-a-cetate (TPA)-induced ODC activity at hardness 800 and 1,000, respectively. Therefore, deep sea water is worth further investigation with respect to cancer chemoprevention or therapy.

The Inhibitory Effects of Acanthopeltis japonica on Melanogenesis (새발 추출물의 멜라닌 생성 저해 효과)

  • To develop the skin whitening agent, we investigated the effects of Acanthopeltis japonica, a rhodophyta on the coast of Jeju island, on melanogenesis. Dried A. japonica was refluxed with 70 % aqueous ethanol and the extract was evaporated to dryness. To validate the activity as a depigmenting agent, various in vitro tests, polyphenol contents, and free radical scavenging activity were performed. In addition, cellular tyrosinase activity and protein expression of p-ERT, tyrosinase, TRP-1, and TRP-2 were measured in B16/F10 murine melanoma cells. A. japonica had low polyphenol contents and low free radicals scavenging activities against 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical. A. japonica suppressed cellular tyrosinase activity up to 86.9 % at $100{\mu}g/mL$ with inhibition or tyrosinase and TRP-1 expression in ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH)-treated B16/F10 melanoma cells. Our results suggest that inhibitory effects of A. japonica on melanogenesis are due to inhibiting the pathways involving ${\alpha}$-MSH-induced ERK activation. Therefore, A. japonica nay be useful as a skin whitening agent associated with the suppressive effect of melanotrophin-induced signaling pathway to inhibit melanin synthesis.

Effect of exogeneous plant growth regulators on morphogenetic response in vitro by embryo and leaf cultures of Camellia sinensis(L.) O. Kuntze (차나무 잎과 배 배양에 있어서 식물 생장조절물질이 형태형성에 미치는 영향)

  • Morphogenetic responses were investigated by culturing embryo and leaf explants of Korean wild type tea plant, Camellia sinensis (L.) O. Kuntze. Induction of direct somatic embryogenesis as well as adventitious and/or axillary shoots was obtained from mature zygotic embryo cultures on Murashige and Skoog (MS) basal medium having 5 to $20\mu\textrm{M}$cytokinin a lone. Morphogenetic response was decreased dramatically by the addition of auxins tested. One hundred percent of induced and isolated shoots formed roots after four weeks of culture on half-strength MS or quarter-strength Schenk and Hildebrandt (SH) media supplemented with $10\mu\textrm{M}$indole-3-butyric acid (IBA). Immature zygotic embryos were shown to be a suitable explant for embryogenic callus formation in the presence of 2, 4-dichlorophenoxyacetic acid(2, 4-D) in basal medium. Mature zygotic embryo originated leaves were used to test their ability for mophogenesis by incorporating plant growth regulators such as IBA, naphthyl-1-acetic acid (NAA), and 6-benzylaminopurine (BAP). Apparently, the morphogenetic responses of the cultured explant sources on the types and/or levels of plant growth regulators tested were observed visually.

Suppressive Effect of Organic Farming Materials on the Development of Tomato Gray Mold (토마토 잿빛곰팡이병에 대한 유기농업자재의 억제효과)

  • Botrytis cinerea infects stems, leaves and fruits of greenhouse tomato and can cause serious economic losses. This study was conducted to develop organic farming control method against tomato gray mold. Twenty two organic farming materials including mineral and plant extracts were screened for the suppressive activity against Botrytis cinerea, in vitro and in vivo. Among the organic farming materials, sulfur, copper, Chinese twinleaf extract and rhubarb extract decreased by 51.7-90% of the spore germination of Botrytis cinerea. Also, gray mold incidence was reduced more than 90% on tomato stems by treating sulfur, seaweed extracts, rhubarb root extracts and Chinese twinleaf extract. After the selected four organic farming materials were applied on tomato cultivated in greenhouse, their control effects against the tomato gray mold were tested. When the water soluble sulfur was foliar-sprayed on the tomato leaves infected by artificial inoculation with spore suspension of Botrytis cinerea, it showed 87.9% of control value. Also, control activity of the water soluble sulfur was paralleled with chemical fungicide, diethofencarb+carbendazim. The above mentioned results indicate the sulfur formulation can be used as chemical fungicide alternatives for controlling tomato gray mold in the greenhouse.

Studies on Biological Activity of Leaves from Korean Rosa davurica Pall. (한국산 생열귀나무(Rosa davurica Pall.) 잎의 생리활성 기능 검색에 관한 연구)

  • The objective of this study was carried out to investigate biological activities effects of Korean leaf from Rosa davurica Pall. in vitro. They were extracted with methanol, ethanol, chloroform and water. Methods of the antimutagenic used in this experiment were well-known bacterial short term tests which include Ames test and the antigenotoxic used in this experiment was DPPH radical scavenge. All extracts (ethanol, methanol, water) except chloroform extract exhibited 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity with IC$\_$50/ of 11.5 $\mu\textrm{g}$/mL, 6.4 $\mu\textrm{g}$/mL, 4.8 $\mu\textrm{g}$/mL. In Ames test, most of extracts had strong antimutagenic effects against the mutagenesis induced by 4-nitroquinoline-1-oxide (4NQO), N-methyl-N’-nitro-N-nitrosoguanidine (MNNG), 3-amino-l,4-dimethyl-5H-pyrido[4,3-b]indol(Trp-P-I) and benzo(${\alpha}$)pyrene(B(${\alpha}$)P). The extracts of leaves (200 $\mu\textrm{g}$/plate) showed approximately 60∼80% inhibitory effect on the mutagenesis induced by 4NQO, Trp-P-1 and B(${\alpha}$)P against TA98 strain, whereas 60∼80% inhibition were observed on the mutagenesis induced by MNNG, 4NQO, Trp-P-1 and B(${\alpha}$)P against TA100 strain. respectively.

  • This study aimed to evaluate the effects of oral rinsing solution on the color stability, surface microhardness and surface roughness change of composite resin. In this in-vitro study, 80 disc-shaped specimens were fabricated of Filtek P60 and Filtek Z250(A2 shade). The samples of each group were randomly divided into eight subgroups (n=10). The baseline color values ($L^*$, $a^*$, $b^*$) of each specimen were measured according to CIE LAB system using a colorimeter. After baseline color measurements, the control samples were immersed in distilled water and the test groups were immersed colorless, green and purple mouthrinses three times a day for thirty minutes. This process was repeated for two weeks. Green and purple oral rinsing solutions displayed color, microhardness and roughness change of all composite resin after immersion in the mouthrinses. Therefore, prescription of oral rinsing solution for a minimum of two weeks is a common practice, which may cause discoloration of aesthetic composite restorations of patients.

Assessment of In vitro Antioxidant, Antidiabetic and Cytotoxic Activities of Sphaeranthus africanus Extracts

  • Sphaeranthus africanus is commonly used as a traditional remedy for sore throats and pain treatment in Vietnam. The aerial parts have been studied for its anti-inflammatory and anti-proliferative properties. However, the antioxidant and antidiabetic potential of the plant has not been explored. In this work, hydrophilic extracts of the plant's aerial parts were prepared in order to investigate its antioxidant and anti-diabetic properties. Also, the cytotoxicity of the root was evaluated and compared to that of the aerial parts. All of the extracts inhibited lipid peroxidation with IC50 values ranging from 2.05 to 3.56 ㎍/mL, indicating substantial antioxidant activity. At an IC50 value of 4.80 ㎍/mL, the 50% ethanol extract exhibited the most potent inhibition of α-glucosidase. The cytotoxic activity of root extracts is 2 to 5-fold less than that of the aerial parts. Nevertheless, dichloromethane and ethyl acetate extracts of the root demonstrated a selective effect on leukemia cells, with no harm towards the normal HEK-293 cell line. This work provides a scientific support for the antioxidant and antidiabetic activity of the plant. Hence, it may find a promising material for the development of novel antioxidant and antidiabetic agents. More research can be conducted on the phytochemistry and anticancer activities of the plant's root.

Control Efficacy of Mixing Application of Microbial and Chemical fungicide against Phytophthora blight of red-pepper (미생물농약과 유기합성 살균제 혼용에 따른 고추 역병 방제 효과)

  • This study was conducted to reduce the using amount of chemical fungicides for the control of red-pepper Phytophthora blight. Effect of combination application of two microbial fungicides and two chemical fungicides for the control of red-pepper Phytophthora blight was examined in vitro, in greenhouse and under field conditions. Each microbial fungicides and chemical fungicides was two-fold diluted and mixed-soil drenched. In the greenhouse pot assay, the mixed application of B. pumilus QST2808 and a mixture of dimethomorph + ethaboxam (De) among four mixed applications of two microbial fungicides (B. pumilus QST2808, P. polymyxa AC-1) and two chemical fungicides showed the highest control effect against Phytophthora blight. Also, control effect of mixed application of B. pumilus QST2808 and De was similar to that of single application of De (dimethomorph + ethaboxam) or Mo (mancozeb + oxadixyl). In the field test, when the microbial fungicides (B. pumilus QST2808, P. polymyxa AC-1) and the chemical fungicide(De) for the control of Phytophthora blight of red pepper were mixed-soil drenched four times at 7~10 day-intervals, the control values were in the range of 78.8% to 82.0%. On the other hand when each of the two chemical fungicides (De, Mo) were soil drenched four times at 7~10 day-intervals, the control value were 65.7% to 85.8%. Consequently, the mixed application of the microbial fungicides and chemical fungicides could be recommended as a control method for reducing the using amount of chemical fungicides.

Effect of Maturity at Harvest and Wrap Colors on the Quality of Round Baled Rye Silage (수확시 숙기 및 비닐색이 호밀 라운드베일 사일리지 품질에 미치는 영향)

  • This experiment was conducted to evaluated the effect of maturity at harvest and wrap colors on the quality of round baled rye(Secal cereale L.) silage at the forage experimental field, Grassland and Forage Crops Division, National Livestock Research Institute, Suwon in 1998. The experiment was consist of split-plot design with three repications. The main plots were three different harvesting stages : boot, heading, and flowering stages, the subplots were wrap colors : white, black, and light green color. Acid detergent fiber(ADF) and neutral detergent fiber(NDF) of rye silage were increased with delayed harvesting date, but there were no significant differdence among wrap colors. However, average in vitro dry matter digestibility (IVDMD) of rye silage with white color wrap tended to have higher than those of other colors. The silage pH of heading stage was highest (5.12), and that of flowering stage was lowest (4.57) in different harvest stages. Among tested wrap colors, white color wrap resulted on lowest pH than others (p<0.05), but there were no significant difference between black and light green color. Dry matter (DM) content of rye silage were increased as harvest stage progressed (p<0.05). However, DM content of white color was highest among warp colors, but there was signifiant difference (p<0.05). Ammonia-N content of silage harvested at boot stage was the highest in harvest stage, but there was no difference between heading and flowering stages. Among wrap colors tested, ammonia-N content of black color was highest, but there was no significant difference. The number of lactic acid bacteria of white colors was highest in different colors, and that of flowering stage was highest among harvest stage. Acetic and butyric acid contents were decreased as harvest stage progressed, and lactic acid was increased from 6.33 to 7.98%. However, wrap colors did not influence lactic acid concentration. Among different wrap colors, outside temperature of rye silage was affected by air temperature, but effect of inside temperature was minimal. Black color wrap increased inside temperature by $3{\sim}5^{\circ}C$. The results of this study indicate that rye should be harvested for round bale silage after heading stage. The quality of rye silage wrapped white and light green color will increase slightly.
