Detection of porcine encephalomyocarditis virus by in situ hybridization (In situ hybridization에 의한 돼지 뇌심근염 바이러스의 검출)

  • The purpose of this study was to establish a rapid, reliable diagnostic method detecting Encephalomyocarditis virus(EMCV) RNA in formalin-fixed, paraffin-embedded tissues of EMCV naturally infected pigs by cDNA probe of EMC $K_3$, the EMCV strain isolated from Korea. Using a biotin-labelled nick translated probe for the cDNA marker. We made up for some defects of radiolabeled method. In sits hybridization(ISH) technique, differently from the other nucleic acid hybridization methods, is able to detect the virus genome specifically in the state of the intact shapes of cells and/or tissues. We succeeded in performing the experiment to detect the EMCV within 1~2 hours using the $MicroProbe^{TM}$ capaillary action system. In this study, we observed highly specific positive signals of red color by staining the paraffin-embedded tissue sections of naturally EMCV-infected pig organs or tissues, including brain, heart, kidney and lacrimal gland with the Fast Red TR salt/Naphtol phosphate chromogen. The results suggested that this ISH method is considered as a highly sensitive and reliable tool for molecular biologic diagnosis of the EMC viral disease.

  • Compaction is a process of increasing soil density using physical energy. It is intended to improve the strength and stiffness of soil. In embankment, degree of compaction affects the construction time, money, also method of soil improvement. In large scale embankment project, difficulties of embankment should change due to uncertainty of settlement. So it is very important to predict the final settlement and factor of safety induced by embankment. In many construction site, there are primarily design of high embankment using in-situ soil. Therefore numerical analyses are necessary for valid evaluation of the settlement prediction. But due to the construction cost and schedule, there were lacking in properties of soil and also limited number of in-situ test were performed. So we proposed the method that can easily estimate the proper soil parameters and suggest the proper method of numerical analysis. From this, two-dimensional finite-difference numerical analysis was conducted to investigate the settlement and factor of safety induced by embankment with various case of compaction rate and embankment height.

  • This study was carried out to increase the efficiency of site investigation through the evaluation of the applicable methods of investigation for cutting slope design. In an effort to find out the applicability of the method employed during the slope investigation, some tests were carried out on 6 subjects i.e location of weak zones, characteristics of discontinuities, distribution of strata, in situ tests for geo-technical properties, laboratory test and estimation of weathering. The method was highly applicable and produced expected results during the test of weak zones, discontinuities and distribution of strata. In order to apply the method to in- situ test for geo-technical properties and laboratory test on soil and rock slope, a statistic analysis of the existing data were required in advance. However its design applicability on rock slope was good although actual cases of application were not many due to limitation of the cases for investigation. The method was frequently referenced but not actually applied for anti seismic design test and estimation of weathering.

The Effects of Sample Disturbance on Undrained Properties of Yangsan Clay (양산점토의 비배수 특성에 대한 시료교란의 효과)

  • It is important to estimate the mechanical properties of clay since it is directly related to the design and the construction of geotechnical structures. Site exploration, which is composed of boring, sampling, in-situ, or laboratory tests, is preformed to estimate the mechanical properties. However, mechanical properties of clay measured from laboratory test may be different from in-situ properties due to disturbances occurred during sampling, transportation, storage, and trimming. In this study, the degree of disturbance according to sampling method was estimated with the test results of CK/sub o/U triaxial compression test on Yangsan clay. The soil samples were obtained by three types of sampling method, j.e., 76mm-tube sampler, 76mm-piston sampler, and block sampler. In order to evaluate the quality of samples, volumetric strain, undrained shear strength, secant Young's modulus, and pore pressure coefficient at peak measured from each sample were compared with one another. From the test results, it was observed that mechanical properties of the block and piston samples were more reliable than those of tube samples. But it was observed that the water content of piston was similar to that of tube samples at given depths while the water content of block samples was 14.3∼15.8% smaller than that of piston and tube samples. In addition to the evaluation of the quality of samples, relationship between c/sub u// σ/sub vc/'and OCR was established from the results of the CK/sub o/U triaxial compression tests, which were carried out using SHANSEP method. And also undrained shear strength was analyzed using the in-situ test data such as Cone Penetration Test(CPT), Dilatometer Test(DMT), and Field Vane Test(FVT) and was compared with that evaluated from CK/sub o/U triaxial compression test.

Effect of Surface Morphology and Adhesion Force on the Field Emisson Properties of Carbon Nanotube Based Cathode (탄소나노튜브 캐소드의 전계방출 특성에 대한 표면 형상과 부착력의 영향)

  • The effects of the field emission property in relation to the surface morphology and adhesion force were investigated. The single-wall-nanotube-based cathode was obtained by use of an in-situ arc discharge synthesis method, a screen-printing method and a spray method. The morphologies of the formed emitter layers were very different. The emission stability and uniformity were dramatically improved by employing an in-situ arc discharge synthesis method. In this study, it was confirmed that the current stability and uniformity of the field emission of the cathode depend on the surface morphology and adhesion force of the emitters. The current stability of the field emission device was also studied through an electrical aging process by varying the current and electric field.

Rapid and Easy Detection of Porcine Epidemic Diarrhea Virus (PEDV) by in situ Hybridization (In Situ Hybridization에 의한 돼지 유행성설사증 (Porcine Epidemic Diarrhea)의 진단)

  • Molecular diagnostic techniques have been used to identify porcine epidemic diarrhea virus (PEDV), a causative agent of acute enteritis in swine, but they were difficult to be petformed and time-consuming. To detect PEDV in a rapid and easy way, we developed biotinylated cDNA probe for N gene encoding the nucleoproteins of PEDV. Formalin-fixed and paraffin-embedded tissues from 24 naturally infected pigs were used for the experiment. The ISH produced a positive reaction in all cases. When intestinal tissues were hybridized with PEDV probe, strong signals were seen in the villus enterocytes of the jejunum and ileum. Hybridization signals were also found in the duodenum from one pig and in colon from dnother. In conclusion, ISH with a biotinylated cDNA probe was provided to be a useful diagnostic method for detecting PEDV effectively in routinely processed tissue sections.

The Genetic Diversity of Bacterial Communities in the Groundwater (지하수 세균 군집의 유전적 다양성)

  • In order to characterize the genetic diversity of bacterial community in groundwater, samples were collected from used for drinking water and polluted with heavy metal wastewater in Seoul city and natural cave of Kangwondo. The DNA was amplified with 165 rDNA-based primers by use of the PCR, and then analysed ARDRA (amplified ribosomal DNA restriction analysis). Restriction endonuclease analysis patterns of amplified 165 rDNA in drinking water and wastewater relatively showed high genetic diversity in situ and drinking groundwater. The number of DNA fragments varied with in situ and drinking water. This method of ARDRA of bacterial communities in groundwater could be used for a quick assessment of genotypic changes between different locations reflecting different environmental conditions and the diversity reflected pollution of groundwater (natural cave water>drinking water>waste water, as in order of grade). An experimental procedure for evaluating the consolidation state of marine clay deposits using shear wave velocity

  • In marine clay deposits, naturally formed or artificially reclaimed, the evaluation and monitoring of the consolidation process has been a critical issue in civil engineering practices due to the time frame required for completing the consolidation process, which range from several days to several years. While complementing the conventional iconographic method suggested by Casagrande and recently developed in-situ techniques that measure the shear wave, this study suggests an alternative experimental procedure that can be used to evaluate the consolidation state of marine clay deposits using the shear wave velocity. A laboratory consolidation testing apparatus was implemented with bimorph-type piezoelectric bender elements to determine the effective stress-shear wave velocity (${\sigma}^{\prime}-V_s$) relationship with the marine clays of interest. The in-situ consolidation state was then evaluated by comparing the in-situ shear wave velocity data with the effective stress-shear wave velocity relationships obtained from laboratory experiments. The suggested methodology was applied and verified at three different sites in South Korea, i.e., a foreshore site in Incheon, a submarine deposit in Busan, and an estuary delta deposit in Busan. It is found that the shear wave-based experimental procedure presented in this paper can be effectively and reliably used to evaluate the consolidation state of marine clay deposits.

토착 미생물의 활성에 의한 유류오염 토양 정화 실험

  • Many methods have been developed for the remediation of contaminated soil and groundwater. Among those technologies, in-situ bioremediation is most likely to be cost-effective method for petroleum hydrocarbon contamination. But the in-situ bioremediation can require more time to remediate hydrocarbon-contaminated soil and groundwater than other methods. Therefore we intended to save time of in-situ bioremediation using a biological additive to activate indigenous microbes in soil. The additive, 'Inipol EAP 22' stimulates the growth of specific flora, significantly accelerating the speed at which hydrocarbons are biodegraded. And it hans been tested in accordance with protocol approved by the USEPA and is registered on the National Contingency Plan Product Schedule List. In the experiment, three soil samples contaminated with fuel oil were prepared in the same concentration. Inipol EAP 22 was not added to one sample and was added to the other two samples with 5% and 10% of hydrocarbon by weight respectively. And $CO_2$gas derived from bacterial respiration was analyzed in each samples for 15 days. As a result, 145% and 153% of $CO_2$ evolution (microbial respiration) against the sample without 'Inipol EAP 22' occurred in samples with 'Inipol EAP 22' addition of 5% and 10%, respectively

Sex Determination of In Vitro Fertilized Bovine Embryos by Fluorescence In Situ Hybridization Technique

  • Sexing from bovine embryos which were fertilized in vitro implicate a possibility of the sex-controlled cattle production. This study was carried out to investigate the possibility of determining of embryo sex by fluorescence in situ hybridization (FISH) technique. FISH was achieved in in vitro fertilized bovine embryos using a bovine Y-specific DNA probe which constructed from the btDYZ-1 sequences. To evaluate Y-chromosome specificity of the FISH probe, metaphase spreads of whole embryos and lymphocytes were prepared and tested. A male-specific signal was detected on 100% of Y chromosome bearing metaphase specimens. Using the FISH technique with a bovine Y-specific probe, 232 whole embryos of 8 cell- to blastocyst-stage were analyzed. Observing the presence of the Y-probe signal on blastomeres, 102 embryos were predicted as male, and 130 embryos as female. The determining rate of embryo sex by FISH technique was about 93% regardless of embryonic stages. In conclusion, the FISH using a bovine Y-specific DNA probe is an accurate, reliable and quick method for determining the sex of bovine embryos.
