  • As a previous step for the development of immunostimulating substance from marine natural products the two new lectins, MLA-II and MLA-III, have been isolated and purified from the hemolymph of Meretrix lusoria in addition to the previously reported MLA-I. The new lectins molecules were characterized as pure by electrophoretic studies.

Immunostimulating Effect of a Well-known Thai Folkloric Remedy in Breast Cancer Patients

  • The study aimed to evaluate immune-stimulating effects of a well-known Thai folkloric remedy when used for adjuvant therapy with conventional chemotherapeutics for treatment of breast cancer. Immunostimulating influence of the remedy (215 mg/kg body weight per day) on NK cell activity and TNF-${\alpha}$ release from the monocytes/macrophages were investigated in a total of 15 healthy women and 13 female patients with breast cancer (Group 1). The effect of breast tumor surgery on NK cell activity was further investigated in 18 female patients with breast cancer (Group 2). NK cell cytotoxic activity was determined by chromium release cytotoxic assay using K562, an erythroleukemic cell line. TNF-${\alpha}$ release from monocytes/macrophages separated from blood samples was determined through a biological assay using actinomycin D-treated L929 mouse fibroblast cells in the presence and absence of LPS. Baseline NK cell activity of the monocytes/macrophages separated from Group 2 patients expressed as %cytotoxicity was significantly lower than in the healthy subjects at E:T ratios of 100:1 and 25:1. In healthy subjects, there was no change in NK cell cytotoxic activity (%cytotoxicity or LU) following 1 and 2 weeks of treatment with the remedy compared with the baseline at various E:T ratios but the binding activity (%binding) was significantly increased after 2 weeks of treatment. The addition of one or two conventional chemotherapeutic regimens did not significantly reduce the NK cytotoxic activity but did affect release of TNF-${\alpha}$ in both unstimulated and LPS-stimulated samples. Surgery produced a significant suppressive effect on NK cell activity. The use of the remedy as an adjunct therapy may improve therapeutic efficacy and safety profiles of conventional chemotherapeutic regimens through stimulation of the immune system in cancer patients.

참당귀 현탁세포배양에 의한 면역증강성 다당 생산 (Production of Immunostimulating Polysaccharide in Angelica gigas Nakai SusDension Cell Cultures)

  • 참당귀(Angelica gigas Nakai) 현탁세포의 면역 증강성 다당의 생산을 증진시키기 위하여 초기 당농도와 생장조절제인 2,4-D의 농도를 변화시켰다. 당의 농도를 높일수록 세포의 생장과 ECP의 생산은 각각 1.6배, 1.1배 증가하여 효과적이었다. 2,4-D는 첨가하지 않은 것이 세포의 생장이 대조군에 비해 1.2배 높았지만, ECP의 생산의 경우 2mg/L 2,4-D를 첨가한 경우 대조군에 비해 10% 이상 더 생산되었다. 또한 배양 중 여러 가지의 elicitor를 첨가함으로써 ECP의 생산을 증진시키기 위하여 fungal elicitor들을 처리한 결과 Verticillium dahliae 90 mg/L을 배양 4일째에 처리한 경우 세포의 생장을 다소 저해시킨 반면, ECP 생산량이 1.7배나 증가함을 확인하였다.

Extracelluar Signal-Regulated Kinase-Dependent Nitric Oxide Production from Macrophage-Like Cells by Lactic Acid Bacteria

  • Lactic acid bacteria (LAB) are considered as probiotics with immunostimulatory property. In this study, we investigated the molecular mechanism of its immunostimulating potency on macrophages using combined preparation of LAB (cpLAB). cpLAB is able to strongly stimulate nitric oxide (NO) production as well as inducible NO synthase (iNOS) expression from macrophage-like RAW264.7 cells. The cpLAB-induced NO release seemed to be mediated by extracellular signal-regulated kinase (ERK) but not p38 and C-Jun N-terminal kinase (JNK), since U0126, an ERK inhibitor, clearly suppressed NO production. cpLAB significantly diminished the binding of toll like receptor (TLR)-2 antibody up to 25%, implying that cpLAB-mediated activation of macrophages may be required for the functional activation of TLR-2, but not TLR-4. Therefore, our data suggest that cpLAB may directly allow macrophages to immunostimulating potency via activation of TLR-2 and ERK.

돼지 오제스키바이러스 재조합단백질 gp50, gIII와 α-ADV을 이용한 Quil A-ISCOMs 접종시 백혈구아군 분포율에 관한 연구 (Effect on lymphocyte subpopulations of Quil A-ISCOMs with recombinant Aujeszkay's disease virus(ADV) gp50, gIII and α-ADV protein)

  • An effective candidate subunit vaccine was prepared by using the immunostimulating complexs(ISCOMs) with Quil A and recombinant protein(gp50, gIII and inactive $\alpha$-ADV) Aujeszky's disease virus(ADV). The weaned pigs were twice immunized with a ADV-ISCOMs, and followed by intramuscular challenge with $1{\times}10^4$ $TCID_{50}$ ADV(strain Yangsan). The unvaccinated pigs were also challenged with same dose of ADV. At 5 days after challenge, the control pigs have developed ADV clinical signs. Whereas, the vaccinated pigs protected them from ADV-induced acute symptoms and death. Also, to identify the lymphocyte subpopulation in peripheral blood with pigs from ADV-ISCOMs vaccinated and control group, lymphocyte reacted with a panel of monoclonal antibodies which are specific to swine leukocyte surface antigens and assayed by the flow cytometry. MHC class I, CD2, CD8, N cells, CD11a, and CD45 antigen positive cells were decreased after inoculating virulent ADV Yangsan strain in control group. The data indicated that ISCOMs technique was useful in ADV subunit vaccine preparation and demonstrated the importance of gp50, gIII as a component of ADV vaccine.

RAW264.7 Cell Activating Glucomannans Extracted from Rhizome of Polygonatum sibiricum

  • Water-soluble polysaccharides isolated from the rhizome of Polygonatum sibiricum and fractionated using ionexchange chromatography were investigated to determine their structure and immunostimulating activity. Crude and fractions ($F_1$ and $F_2$) consisted of carbohydrates (85.1~88.3%) with proteins (4.51~11.9%) and uronic acid (1.79~7.47%), and included different levels of mannose (62.3~76.3%), glucose (15.2~20.3%), galactose (4.35~15.3%), and arabinose (4.00~7.65%). The crude contained two peaks with molecular weights (Mw) of $151{\times}10^3$ and $31.8{\times}10^3$, but $F_1$ and $F_2$ exhibited one major peak with Mw of $103{\times}10^3$ and $628{\times}10^3$, respectively. Little immunostimulatory activity was observed by the crude; however, $F_1$ and $F_2$ significantly activated RAW264.7 cells to release nitric oxide and various cytokines, suggesting they were potent immunostimulators. The backbone of the most immunostimulating fraction ($F_1$) was ($1{\rightarrow}4$)-manno- and ($1{\rightarrow}4$)-gluco-pyranosyl residues with galactose and glucose attached to O-6 of manno-pyranoside.

BIOACTIVE PEPTIDES DERIVED FROM FOOD PROTEINS AND PREVENTION OF LIFE-STYLE RELATED DISEASES

  • Two opioid peptides, YPLDL and YPLDLF, were isolated from enzymatic digests of spinach ribulose-1, 5-bisphosphate carboxylase/oxygenase (RuBisCO) and named rubiscolin-5 and -6, respectively. These peptides were selective for delta-receptor and the latter was about 3 times more potent than the former. After oral administration in mice at the dose of 100 mg/kg, rubiscolin-6 showed analgesic activity in tail pinch test. It also stimutated learning performance at the same dose in passive avoidance experiment using step-through apparatus. An immunostimulating peptide, MITLAIPVNKPGR, was isolated from a trypsin digest of soybean protein and named soymetide. Immunostimulating activy of soymetide was mediated by fMLP receptor. Interestingly, after oral administration in rats at a dose of 300 mg/kg (po.), soymetide-4 (MITL) protected alopecia (hair-loss) induced by etoposide, a cancer chemotherapy agent. Stimulation of IL-1 release by the peptide was involved in the mechanism. Ovokinin(2-7), RADHPF, is a vasorelaxing peptide released from ovalbumin by the action of chymotrypsin. It lowered blood pressure of spontaneously hypersensive rats (SHR) after oral administration at a dose of 10 mg/kg. RPLKPW, which was designed by replacing 4 amino acid residues in ovokinin(2-7), exhibited hypotensive activity at a dose of 0.1 mg/kg (po.). This peptides was introduced into 3 homologous sites in soybean beta-conglycinin alpha' subunit by site-directed mutagenesis of the cDNA and expressed in E. coli. The minimum effective dose for hypotensive activity of the genetically modified beta-conglycinin alpha' subunit was 10 mg/kg (po.), which is about 1/200 that of ovalbumin.

마카 품종별 조다당 획분의 대식세포 활성 (Macrophage Stimulating Activity of Crude Polysaccharide on Maca (Lepidium meyenii) Varieties)

  • Maca roots (Lepidium meyenii) are an important medicinal herb that have long been used by the Andes-indigenous peoples and South Americans. In Korea, recently, it has attracted attention as a health food material because of nutritional values and physiological activities. The purpose of this study was to investigate the industrial applicability of maca (red and golden varieties; R&G) as immunostimulating materials. In the macrophage stimulating assay using RAW 264.7 cells at 125~500 ㎍/mL of non-cytotoxicity doses, G-HW showed the most potent production of TNF-α, IL-6 and nitric oxide compared to red maca or the other extracts. The general component analysis results showed that all extracts comprised more than 90% neutral sugars with small amounts of uronic acid and protein. Meanwhile, component sugar analysis showed the difference in the content of uronic acids of cold- and hot-water extract. Additionally, the further fractionation of G-HW into crude polysaccharide (G-CP) greatly enhanced the macrophage stimulating activity, and G-CP contained macromolecules over 144 kDa, comprised mainly of glucose and galacturonic acid (51.0 and 34.9%). In conclusion, crude polysaccharide from maca showed industrial applicability as immunostimulating material, and especially golden maca showed higher macrophage stimulating activity than red maca.

해조류 유래 후코이단(Fucoidan) 경구 투여가 조피볼락(Sebastes schlegelii)의 고수온 내성 및 Streptococcus iniae 감염 저항성에 미치는 영향 (Effect of Oral Administration of Seaweed-derived Fucoidan on the Resistance of the Rockfish Sebastes schlegelii to High Temperature and Streptococcus iniae Infection)

  • Immunostimulating effects of fucoidan administration to rockfish Sebastes schlegelii at a concentration of 20 g/kg of diet were evaluated under high water temperature condition. The oral administration of fucoidan mixed with feed at a concentration of 20 g/kg of diet for 2 weeks increased the interleukin 1β gene in the intestine and kidney of fish by 5.7 and 6.3 times, respectively. In addition, when the water temperature was gradually increased from 24 to 31.4℃ for 2 weeks, LT50 delayed by 24 h in the fucoidan treated group compared to that in the control group, and mortality also reduced. Streptococcus iniae infection at a concentration of 1.50×100 CFU/fish at 28℃ delayed LT50 by 12 h in the fucoidan-treated group. Furthermore, the overall survival rate was 0% in the control group and 20% in the fucoidan-treated group. This study confirmed the applicability of dietary additives such as fucoidan as an immune activator of rockfish under high temperature condition.
