  • This study was done to evaluate the immunomodulatory effects of KSDBT. KSDBT significantly enhanced the lymphocyte proliferation through 3H thymidine uptake and also increased T and T helper cell. It upregulated IL-2, IL-12(p35, p40) and INF-${\gamma}$. NO and carbon clearance were significantly increased by KSDBT. These results indicate KSDBT exert antitumor activity by immunomodulation of cytokines.

Modulatory Effects of Chrysanyhemi Flos Pharmacopuncture on Nitric-oxide (NO) Production in Murin Macrophagy Cells

  • Objectives: Much evidence exists that herbs have effective immunomodulatory activities. Chrysanthemi Flos (CF) is effective in clearing heat, reducing inflammation, dropping blood pressure and treating headache and is used as a pharmaceutical raw material for an immune enhancer. The purpose of this study was to investigate the modulatory effect of Chrysanthemi Flos pharmacopuncture on nitric-oxide (NO) production in activating macrophages. Methods: After a murine macrophage cell line, RAW 264.7, was cultured in the presence of lipopolysaccharide (LPS), immune-modulating abilities of CF were evaluated by using NO, interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-${\alpha}$) production and phagocytic activity of macrophages. Results: CF enhanced the activities of macrophages by increasing the phagocytic activity and decreasing NO production. Especially, both LPS and CF, 200 ${\mu}g/ml$, treatment could significantly reduce the NO production, but did not change the production of IL-6 and TNF-${\alpha}$. Conclusion: The results of this study indicate that CF may be of immunomodulatory value, especially for adverse diseases due to increased NO production. It may have potential for use as immunoenhancing pharmacopuncture.

Oligopeptide derived from solid-state fermented cottonseed meal significantly affect the immunomodulatory in BALB/c mice treated with cyclophosphamide

  • In this study, the immunomodulatory activity of oligopeptide (CP) derived from solid-state fermented cottonseed meal were investigated in immunosuppressed BALB/c mice models by treatment with cyclophosphamide (CY). Results indicated that oligopeptide increased the thymus and spleen indices of CY-treated mice. The count of plague forming cells (PFC) and the content of half serum hemolysis ($HC_{50}$) in immunosuppressive mice were restored to the normal level in CP-10 and CP-20 groups while the cytokines interleukin (IL)-2, IL-6, and tumor necrosis factor alpha (TNF-${\alpha}$) were increased significantly in CP-20 group. Similar increasing the immunoglobulin of IgG and IgM content in the serum of CP-10 group were also observed. These findings indicated that oligopeptide derived from solid-state fermented cottonseed meal had a strong immune-enhancing activity as well as a protective effect against immunosuppression induced by cyclophosphamide in mice.

Effect of Differential Thermal Drying Conditions on the Immunomodulatory Function of Ginger

  • Thermal drying is a common process used in the food industry for the modification of agricultural products. However, while various studies have investigated the alteration in physiochemical properties and chemical composition after drying, research focusing on the relationship between different dehydration conditions and bioactivity is scarce. In the current study, we prepared dried ginger under nine different conditions by varying the processing time and temperature and compared their immunomodulatory effects. Interestingly, depending on the drying condition, there were significant differences in the immunestimulating activity of the dried ginger samples. Gingers processed at $50^{\circ}C$ 1h displayed the strongest activation of macrophages measured by $TNF-{\alpha}$ and IL-6 levels, whereas, freezedried or $70^{\circ}C$- and $90^{\circ}C$-dried ginger showed little effect. Similar results were recapitulated in primary bone marrow-derived macrophages, further confirming that different dehydration conditions can cause significant differences in the immune-stimulating activity of ginger. Induction of ERK, p38, and JNK signaling was found to be the major underlying molecular mechanism responsible for the immunomodulatory effect of ginger. These results highlight the potential to improve the bioactivity of functional foods by selectively controlling processing conditions.

귀비탕(歸脾湯)이 비소세포 폐암세포(NCI-H520) 이식 마우스에서 항암 및 면역 활성에 미치는 영향 (Anticancer and Related Immunomodulatory Effects of Kwibi-tang on Non-small Cell Lung Carcinoma, NCI-H520, Xenograft Mice)

  • Objectives : This study was to observe anticancer and related immunomodulatory effects of Kwibi-tang extracts (KBTe) on non-small cell lung carcinoma (squamous epithelial carcinoma), NCI-H520, xenograft Balb/c nu-nu nude mice. Methods : Three different dosages of KBTe, 50, 100 and 200 mg/kg were orally administered once a day for 42 days from 11 days after tumor cell inoculation. Six groups, each of 8 mice per group were used in the present study. Changes in body weight, tumor volume and weight, lymphatic organs (spleen and popliteal lymph node), serum interferon (IFN)-${\gamma}$ levels, splenocytes NK cell activity and peritoneal macrophage activities, splenic tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-$1{\beta}$ and IL-10 contents were observed with tumor mass and lymphatic organ histopathology to detect anticancer and immunomodulatory effects. The results were compared with a potent cytotoxic anticancer agent, 5-FU (5-Fluorouracil) 30 mg/kg, intraperitoneal treatment (3-day intervals for 42 days, the optimal effective treatment regimes already confirmed). Results & Conclusions : This study suggest that over 50 mg/kg of KBTe showed favorable anticancer effects on the NCI-H520 cell xenograft with immunomodulatory effects. Although relatively lower anticancer effects were observed in KBTe 200 mg/kg treated mice as compared with 5-FU 30 mg/kg treated mice, no meaningful favorable immunomodulatory effects were observed after 5-FU treatment in the present study.

Antimicrobial and Immunomodulatory Effects of Bifidobacterium Strains: A Review

  • Bifidobacterium strains can provide several health benefits, such as antimicrobial and immunomodulatory effects. Some strains inhibit growth or cell adhesion of pathogenic bacteria, including multidrug-resistant bacteria, and their antibacterial activity can be intensified when combined with certain antibiotics. In addition, some strains of bifidobacteria reduce viral infectivity, leading to less epithelial damage of intestinal tissue, lowering the virus shedding titer, and controlling the release of antiviral substances. Furthermore, bifidobacteria can modulate the immune system by increasing immunoglobulins, and inducing or reducing pro- or anti-inflammatory cytokines, respectively. In particular, these anti-inflammatory effects are helpful in the treatment of patients who are already suffering from infection or inflammatory diseases. This review summarizes the antimicrobial effects and mechanisms, and immunomodulatory effects of Bifidobacterium strains, suggesting the potential of bifidobacteria as an alternative or complementary treatment option.

꾸지뽕(Cudrania tricuspidata) 잎으로부터 분리된 다당류 추출물의 면역 활성 (Immunomodulatory Activity of Crude Polysaccharide Separated from Cudrania tricuspidata Leaf)

  • 본 연구는 꾸지뽕 잎으로부터 추출된 다당류인 CTP의 처리가 면역세포의 활성에 미치는 영향에 관하여 평가하였다. CTP는 에탄올 침전법에 의하여 추출하였고, 면역 활성능의 평가는 대식세포주인 RAW 264.7 세포와 미분화 골수세포로부터 유도 분화시킨 대식세포 및 마우스 비장으로부터 유리시킨 비장세포에 CTP를 농도별로 처리하여 관찰하였다. 선천면역계에서 중요한 역할을 수행하는 대식세포에 CTP를 처리하였을 때 세포 증식률, NO 및 cytokine 분비능이 CTP 처리 농도 의존적으로 증가하는 것으로 관찰되었을 뿐만 아니라 비장세포에서도 이와 유사하게 세포 증식률이 증가하고 Th 1 type의 cytokine 분비능 또한 유의적으로 증가하는 것으로 관찰되었다. 이상의 결과로 미루어보아 꾸지뽕다당류 추출물인 CTP는 다양한 면역세포의 활성을 증가시키는 것으로 생각하며 이를 활용하여 다양한 식품 및 건강보조식품을 개발한다면 그 경제적 가치가 매우 클 것으로 생각한다.

보기익혈 복합방, 바이오 큐의 면역조절 효과 (Immunomodulatory Effect of Bio-Q, Bokiikhyul Prescription)

  • Bio-Q is a modified prescription with the activities of supplementing Qi and blood in human body. In the present study, immunomodulatory effect of Bio-Q was examined. After oral administration of Bio-Q for 7 days to Balb/c mice, splenocytes were isolated and immunological experiments were performed. Bio-Q significantly increased the proliferation of splenocytes exposed to concanavalin A (Con A), while it did not in case of lipopolysaccharide (LPS) stimulation. Bio-Q also significantly increased CD3/CD19, CD4/CDB and NK cells by flow cytometric analysis. In addition, Bio-Q significantly enhanced the level of $INF-\gamma$ in splenocytes, but not $TNF-\alpha$ by ELISA. These results strongly suggest that Bio-Q has immunomodulatory activity through the regulation of T cell mediated immune pathway.

Lactobacillus gasseri 505를 사용한 꾸지뽕잎 추출물 첨가 신바이오틱스 발효유의 면역활성 조절 연구 (Immunomodulatory Effects of Fermented Milk based on Synbiotic Interaction between Cudrania tricuspidata Leaf Extract and Lactobacillus gasseri 505)

  • This study examined the immunomodulatory effects of milk fermented with based on the synbiotic interaction between Cudrania tricuspidata leaf extract (CT) and Lactobacillus gasseri 505. Raw 264.7 macrophages were treated with milk (Control), milk supplemented with 0.2% CT (CT-milk), or CT-milk fermented with L. gasseri 505 (FCT). FCT-treated cells showed higher NO production than the other groups. ELISA revealed that FCT produced the highest enhancement of the production of pro-inflammatory cytokines such as $TNF-{\alpha}$, $IFN-{\gamma}$, $IL-1{\beta}$, IL-6, IL-2, IL-12, and IL-10. Furthermore, FCT also increased the gene expression of pro-inflammatory mediators such as iNOS and COX-2 in a concentration-dependent manner. Collectively, these results indicate that the synbiotic interaction between CT and L. gasseri 505 might enhance their immunomodulatory effects of fermented milk.

Bifidobacterium adolescentis P2P3, a Human Gut Bacterium Having Strong Non-Gelatinized Resistant Starch-Degrading Activity

  • Resistant starch (RS) is metabolized by gut microbiota and involved in the production of short-chain fatty acids, which are related to a variety of physiological and health effects. Therefore, the availability of RS as a prebiotic is a topic of interest, and research on gut bacteria that can decompose RS is also important. The objectives in this study were 1) to isolate a human gut bacterium having strong degradation activity on non-gelatinized RS, 2) to characterize its RS-degrading characteristics, and 3) to investigate its probiotic effects, including a growth stimulation effect on other gut bacteria and an immunomodulatory effect. Bifidobacterium adolescentis P2P3 showing very strong RS granule utilization activity was isolated. It can attach to RS granules and form them into clusters. It also utilizes high-amylose corn starch granules up to 63.3%, and efficiently decomposes other various types of commercial RS without gelatinization. In a coculture experiment, Bacteroides thetaiotaomicron ATCC 29148, isolated from human feces, was able to grow using carbon sources generated from RS granules by B. adolescentis P2P3. In addition, B. adolescentis P2P3 demonstrated the ability to stimulate secretion of Th1 type cytokines from mouse macrophages in vitro that was not shown in other B. adolescentis. These results suggested that B. adolescentis P2P3 is a useful probiotic candidate, having immunomodulatory activity as well as the ability to feed other gut bacteria using RS as a prebiotic.
