The present study was aimed to study the effect of aqueous extract of Tinospora cordifolia and autologous plasma on uterine infections. Buffaloes in estrus, 40 days and above postpartum were checked for uterine infections. Animals having uterine infections were randomly divided into three groups of six animals each. Buffaloes in group I, II and III (control) were given intrauterine infusion of aqueous extract of Tinospora cordifolia (3,000 mg total dose), autologous plasma (150 ml) and phosphate buffer saline (150 ml) respectively, in three divided doses, once daily for three consecutive days, starting from the day of estrus. A fourth group (IV) comprising of six buffaloes having no uterine infection was also included in the present study. Buffaloes were inseminated artificially on next estrus following treatment and confirmed for pregnancy 60 days later. Bacterial population in CVM of buffaloes in group I, II and III was significantly (p<0.05) higher than group IV. After treatment there was a significant (p<0.01) reduction in bacterial population in group I (83.496$\pm$7.755%) and group II 80.233$\pm$5.799%) than group in III 7.557$\pm$33.551%) at next estrus. There was non-significant improvement in first service conception rate (CR, 33.33%) and overall conception rate (OCR, 27.27%) in group I, in comparison to group III (first service CR-16.67%; OCR-20.0%). No significant improvement was seen in OCR (22.22%) in-group II also in comparison to group III. The improvement in group I was however, nonsignificantly lower than normal animals of group IV (First service CR-16.67%; OCR-36.33%).
Two experiments were conducted to standardize in ovo injection of amino acids (AA) and to evaluate the effect of in ovo injection of limiting AA(s) on pre and post hatch growth performance, immune response and development of digestive organs. Combinations of essential and non-essential amino acids (Lys+Arg, Lys+Met+Cys, Thr+Gly+Ser, Ile+Leu+Val and Gly+Pro) were injected into 50 eggs in each treatment group at 14 d of embryonic age. Standardization of injection site, needle length and embryonic age revealed that when AA were injected in to the broad end of the egg with a 11 mm needle and at the narrow end with a 24 mm needle both at the 7$^{th}$ and 14$^{th}$ d of incubation there was poor hatchability. However, better hatchability was recorded when the AA were injected in the narrow end of the egg with a 11 mm needle and in the broad end with a 24 mm needle on the 14$^{th}$ d of incubation. The chick to egg weight ratio was higher (p<0.018) when AA were injected on the 14$^{th}$ d of incubation. When a combination of amino acids were injected a 63.6 or 63.2 g difference in body weight of bird at 21 d was recorded between uninjected control and Ile+Leu+Val or Gly+Pro group, respectively. Higher feed intake (p<0.047) was recorded in the AA injected groups and feed conversion ratio (FCR) was numerically better in Gly+Pro, Lys+Met+Cys AA injected groups than in the uninjected control. Significantly higher immune response to cell mediated (p<0.033) and humoral (p<0.002) immunity was observed in in ovo amino acid injected birds, especially in Lys+Met+Cys, Thr+Gly+Ser or Ile+leu+Val groups. The digestive organ weights at 21 d did not differ between specific AA injected groups and the uninjected control. In ovo injected amino acids may act as immunomodulators and their role in gastrointestinal development needs further research.
Anthrax lethal toxin, which consists of two separate protein, protective antigen (83 KDa) and lethal factor (85 KDa) is responsible for major symptoms and death from systemic infection of Bacillus anthracis. High concentrations of this toxin are cytolytic to macrophages, whereas sublytic concentrations of lethal toxin induce these cells to produce interleukin $1{\beta}$ ($IL-1{\beta}$). It is proposed that melatonin and dehydroepiandrosterone (DHEA) may play an important role in modifying immune dysfunction. In this study, we investigated whether or not melatonin and DHEA could prevent $IL-1{\beta}$ production that is induced by anthrax lethal toxin in mouse peritoneal macrophages. Treatment of melatonin or DHEA alone, as well as together, prevented the production of $IL-1{\beta}$ caused by anthrax lethal toxin. We found that melatonin at a concentration of $10^{-6}-10^{-7}$ M inhibits $IL-1{\beta}$ production induced by anthrax lethal toxin. As expect, treatment of DHEA at a concentration $10^{-6}-10^{-7}$ M also suppressed production of $IL-1{\beta}$ by lethal toxin stimulated macrophages. The results of these studies suggest that melatonin and DHEA, immunomodulators, may have an important role in reducing the increase of cytokine production in anthrax lethal toxin-treated macrophages.
Repeated injections of low-doses of mercuric chloride in rats or mice induce polyclonal activation which includes the induction of anti-glomerular basement membrane (GBM) antibodies and circulating immune complex and it results in nephritis. Because this disease is autoimmune mediated disease resulted from immune dysfunction, immunomodulators are used to control the symptoms or to cure the disease. Irpex lacteus Fr. is a kind of new medicinal fungus. The polysaccharide fraction extracted from submerged fermentation of Irpex lacteus Fr. decreased the serum agglutinin, serolysin and IgM plaque forming cells in normal mice. The hitherto obtained clinical results suggested that it significantly improved the oligourea, edema, and hypertension in patients who have nephritis. To elucidate the action-mechanisms of Irpex lacteus Fr., we established the experimental model of HgCl$_2$induced polyclonal activation by intraperitoneal administrations of HgCl$_2$to mice. To assess the immunomodulating effect of Irpex lacteus fraction, we Investigated its effects on the mitogen induced proliferation and IgM PFC counts of splenic lymphocytes in mice during the treatment of HgCl$_2$. The Irpex lacteus polysaccharide reduced the abnormally increased mitogen induced Iymphocyte proliferation and IgM PFCs to almost normal levels. And the Irpex lacteus polysaccharides prevented the increasement of serum immunoglobulin level induced by HgCl$_2$. These data suggested that the Irpex lacteus polysaccharides might have the immunomodulating activity to prevent and /or improve the HgCl$_2$ induced autoimmune disease.
Alopecia areata (AA) is a common and tissue-specific autoimmune disease of hair follicle resulting in the loss of hair on the scalp and elsewhere on the body. Hair follicles is a unique organ because it has its own immune system and hormonal milieu and has a different immune state at each hair cycle stage. The collapses of anagen-dependent hair follicle immune privilege arise autoimmune attack, inducing ectopic MHC class I expression in the hair follicle epithelium and autoantigen presentation to autoreactive CD8+T cells, which results in AA. Clinical and experimental studies have pointed that psychological stress may also influence the hair follicle immune/hormone systems and contribute to the induction of AA. The key pathogenesis of AA is associated with immune privilege guardians (including ACTH, ${\alpha}-MSH$, and $TGF-{\beta}$), natural killer group 2D-positive (NKG2D+) cells (including NK and CD8+T cells), and stress hormones (including CRH and substance P). Effective treatments for AA are still demanded. One of the future targets of treatment will be the modification of hair follicle immune privilege including stress. Recent studies have reported that JAK inhibitors and immunomodulators used in other autoimmune disease, such as psoriasis, atopic dermatitis, and rheumatoid arthritis, Tregs, platelet-rich plasma therapy, statins, and prostaglandin anaolgues are effective for AA. Here the article reviews the recent understanding in the pathogenesis associated with perifollicular endocrine/immunology and new treatments of AA.
Two kinds of complement activating (anti-complementary) polysaccharides, which were expected to be immunomodulators were purified from Arecae Pericarpium (the pericarps of Areca catechu), and their action modes have been studied. The active polysaccharides, AC-2-IIIa and AC-2-IIIc from Arecae Pericarpium showed dose-dependent anti-complementary activities on $TCH_{50}$. The anti-complementary activities of AC-2-IIIa and AC-2-IIIc in metal ion-free condition were completely decreased in comparison with control whereas in case of $Ca^{2+}$-free condition, these activities were maintained, considerably. Also AC-2-IIIa and AC-2-IIIc showed relatively potent alternative complement pathway activities. Furthermore, after incubation of the normal human serum with polysaccharide of Arecae Pericarpium in the absence of $Ca^{2+}$ ion, a cleavage of C3 in the serum was found to have occurred through immunoelectrophoresis (IEP) with anti-human C3. Also, from the results of IEP using anti-human whole serum, the ratios of the height of 3rd peak to ${\alpha}2-M$ peak by AC-2-IIIa and AC-2-IIIc proved to be $1.50{\pm}0.04$ and $1.22{\pm}0.08$, respectively. These results indicate that the modes of complement activation by AC-2-IIIa and AC-2-IIIc from Arecae Pericarpium are via both the classical pathway and the alternative pathway.
BACKGROUND/OBJECTIVES: A variety of immunomodulators can improve the efficacy of low-dose chemotherapeutics. Active hexose correlated compound (AHCC), a mushroom mycelia extract, has been shown to be a strong immunomodulator. Whether AHCC could enhance the antitumor effect of low-dose 5-fluorouracil (5-FU) via regulation of host immunity is unknown. MATERIALS/METHODS: In the current study Hepatoma 22 (H22) tumor-bearing mice were treated with PBS, 5-FU ($10mg{\cdot}kg^{-1}{\cdot}d^{-1}$, i.p), or AHCC ($360mg{\cdot}kg^{-1}{\cdot}d^{-1}$, i.g) plus 5-FU, respectively, for 5 d. $CD^{3+}$, $CD^{4+}$, $CD^{8+}$, and NK in peripheral blood were detected by flow cytometry. ALT, AST, BUN, and Cr levels were measured by biochemical assay. IL-2 and $TNF{\alpha}$ in serum were measured using the RIA kit and apoptosis of tumor was detected by TUNEL staining. Bax, Bcl-2, and TS protein levels were measured by immunohistochemical staining and mRNA level was evaluated by RT-PCR. RESULTS: Diet consumption and body weight showed that AHCC had no apparent toxicity. AHCC could reverse liver injury and myelosuppression induced by 5-FU (P < 0.05). Compared to mice treated with 5-FU, mice treated with AHCC plus 5-FU had higher thymus index, percentages of $CD^{3+}$, $CD^{4+}$, and NK cells (P < 0.01), and ratio of $CD^{4+}$/$CD^{8+}$ (P < 0.01) in peripheral blood. Radioimmunoassay showed that mice treated with AHCC plus 5-FU had the highest serum levels of IL-2 and $TNF{\alpha}$ compared with the vehicle group and 5-FU group. More importantly, the combination of AHCC and 5-FU produced a more potent antitumor effect (P < 0.05) and caused more severe apoptosis in tumor tissue (P < 0.05) compared with the 5-FU group. In addition, the combination of AHCC and 5-FU further up-regulated the expression of Bcl-2 associated X protein (Bax) (P < 0.01), while it down-regulated the expression of B cell lymphoma 2 (Bcl-2) (P < 0.01). CONCLUSIONS: These results support the claim that AHCC might be beneficial for cancer patients receiving chemotherapy.

We studied the antioxidative and immunoregulatory properties of 70% ethanol extracts from the leaves, branches, and fruit of 13 species of the Rhamnaceae collected during the bearing season (Berchemia berchemiifolia, Berchemia floribunda, Hovenia dulcis, Paliurus ramosissimus, Rhamnella franguloides, Rhamnus crenata, Rhamnus davurica, Rhamnus koraiensis, Rhamnus parvifolia, Rhamnus ussuriensis, Rhamnus yoshinoi, Sageretia thea, and Ziziphus ujube). Total phenolic and flavonoid content values were determined using calibration curves of gallic acid and rutin, respectively. ABTS and DPPH radical scavenging assays were performed to determine antioxidative potential. Anti-inflammatory effects were studied using the nitric oxide (NO) inhibition assay in LPS-induced RAW264.7 cells. Extracts of Berchemia berchemiifolia, Sageretia thea, and Ziziphus jujuba had the highest total phenol and flavonoid content values. Extracts of Berchemia berchemiifolia, Paliurus ramosissimus, Rhamnella franguloides, and Sageretia thea had the highest ABTS radical scavenging activities. Berchemia berchemiifolia and Sageretia thea had the highest DPPH radical scavenging activities. Furthermore, extracts of Berchemia berchemiifolia, Berchemia floribunda, Paliurus ramosissimus, Sageretia thea, and Ziziphus jujuba showed strong anti-inflammatory activity through inhibition of NO production. The results of this study suggest that Rhamnaceae extracts can be used as natural antioxidants and immunomodulators.
Purpose: The aim of this study is to report the efficacy of infliximab, a monoclonal antibody directed against tumor necrosis factor alpha which is used for both treatment of refractory pediatric Crohn disease (CD) and induction of remission. Methods: Among pediatric patients who were diagnosed with CD at Samsung Medical Center between March 2001 and August 2007, a total of 16 patients were given infliximab to treat conventional therapyresistant refractory CD and severe active CD for induction of remission. Patients needing maintenance therapy were treated with an infliximab infusion every 8 weeks, and fistulizing CD patients occasionally received the infusion upon the condition that a fistula developed. The efficacy of treatment was assessed by comparing the Pediatric Crohn Disease Activity Index (PCDAI), Hct, ESR, CRP, and serum albumin levels using paired t-test. Results: The male/female ratio was 13:3, and the median age was 13 years (range, 21 months~15 years). The patients included 7 cases of therapy-resistant refractory CD, 7 cases of severe active CD, and 2 cases of fistulizing CD. Mean PCDAI before infliximab therapy was 34.19${\pm}$14.96, and mean follow-up PCDAI within 2 to 4 weeks after the last infusion was significantly lower, at 6.88${\pm}$10.31 (p=0.000). Hematological markers such as ESR (p=0.000), serum albumin (p=0.016), and CRP (p=0.009) also improved significantly after infusion. Remission was achieved in 2 of 4 patients refractory to conventional therapy. Among 3 steroid-dependent patients, 2 were able to discontinue steroid therapy, and dose reduction was possible in 1 patient. Remission after top-down therapy without prior use of other immunomodulators was achieved in 6 weeks in all 7 of the patients who had severe CD. Nine of ten refractory fistulizing CD patients also showed improvement after infliximab therapy. Conclusion: Infliximab was effective in pediatric refractory CD for induction of remission and maintenance therapy, as well as in severe CD for top-down induction therapy. Furthermore, infliximab has contributed to steroid cessation and dose reduction. Long-term follow-up evaluation is needed to determine safety and efficacy of infliximab in the future.
This study was conducted to test the efficacy of Ulva pertusa kjellman as immunomodulators under lipopolysaccharide (LPS) challenge in broilers by investigating their effects on serum superoxide dismutase (SOD) like ability, immunoglobulin concentration, and splenic cytokine mRNA expression. A total of ninety six1-d-old male broiler chicks (Ross 308) were divided into 4 treatment groups with 8 replicates of 3 birds in each group. NC (negative control, no immune substances), PC (positive control, ${\beta}$-glucan 25 ppm), Ulva P (Ulva pertusa kjellman Powder 3%), and Ulva E (Extract form Ulva pertusa kjellman 0.3%) were added in feed with respective substance. Heparinized venous blood and spleens were collected from five birds per dietary treatment at 5 wk of age. The SOD like activities in Ulva P and Ulva E were significantly increased in comparison with other groups (P<0.05). The immunoglobulin M content was lower in the Ulva E than other groups (P<0.05). Expression patterns of splenic cytokine mRNA in the IL-$1{\beeta}$, IL-2 and IL-6 were similar. Expression rate of IL-$1{\beta}$, IL-2 and IL-6 in Ulva pertusa kjellman (Ulva P, Ulva E) were decreased (P<0.05) in comparison to other groups. Expression rate of IL-$1{\beta}$, IL-2 and IL-6 were significantly lower in groups treated with Ulva E than Ulva P. In conclusion, dietary supplementation of Ulva pertusa kjellman improved SOD like activity and affect immune system by inhibiting inflammatory response in broiler chicks. In addition, it is more effective to use Ulva pertusa kjellman extract than powder for immunomodulatory function. These results suggest the possibility that Ulva pertusa kjellman could be used as the immunomodulator for inflammatory response in broiler chicks.

제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
