In vitro Stimulation of NK Cells and Lymphocytes Using an Extract Prepared from Mycelial Culture of Ophiocordyceps sinensis

  • Ophiocordyceps sinensis is a natural fungus that has been valued as a health food and used in traditional Chinese medicine for centuries. The fungus is parasitic and colonizes insect larva. Naturally occurring O. sinensis thrives at high altitude in cold and grassy alpine meadows on the Himalayan mountain ranges. Wild Ophiocordyceps is becoming increasingly rare in its natural habitat, and its price limits its use in clinical practice. Therefore, the development of a standardized alternative is a great focus of research to allow the use of Ophiocordyceps as a medicine. To develop an alternative for wild Ophiocordyceps, a refined standardized extract, CBG-CS-2, was produced by artificial fermentation and extraction of the mycelial strain Paecilomyces hepiali CBG-CS-1, which originated from wild O. sinensis. In this study, we analyzed the in vitro immune-modulating effect of CBG-CS-2 on natural killer cells and B and T lymphocytes. CBG-CS-2 stimulated splenocyte proliferation and enhanced Th1-type cytokine expression in the mouse splenocytes. Importantly, in vitro CBG-CS-2 treatment enhanced the killing activity of the NK-92MI natural killer cell line. These results indicate that the mycelial culture extract prepared from Ophiocordyceps exhibits immune-modulating activity, as was observed in vivo and this suggests its possible use in the treatment of diseases caused by abnormal immune function.

  • Immunomodulation is a technique for the modulation of immune responses against graft material to improve surgical success rates. The main target cell for the immunomodulation is a macrophage because it is the reaction site of the graft and controls the healing process. Macrophages can be classified into M1 and M2 types. Most immunomodulation techniques focus on the rapid differentiation of M2-type macrophage. An M2 inducer, 4-hexylresorcinol, has been recently identified and is used for bone grafts and dental implant coatings.

Raw264.7 cells에서 바이칼린 함유 황금열수 추출물의 면역계 조절을 통한 항염증 효과 (Anti-inflammatory Effect of Scutellaria baicalensis Hot Water Extracts Containing Baicalin on Modulation of the Immune System in Raw264.7 Cells)

  • 체내 면역시스템은 내부의 돌연변이 세포에 의하여 발생된 암 뿐만 아니라 세균감염에 의하여 기인된 여러질환으로 부터 우리의 몸을 보호한다. 하지만 노화됨에 따라 비정상적인 면역시스템은 류머티스 관절염과 같은 만성염증을 유발시킨다. 그러므로, 본 연구의 목적은 효능성분으로 바이칼린을 함유한 황금의 항염증 효과를 조사한 것이다. 먼저 HPLC 분석에서 SBWE는 42.2 mg/g의 바이칼린을 함유하고 있는 것으로 관찰되었다. Raw264.7 세포에서 황금열수추출물(SBWE)의 세포독성을 평가하기 위하여 MTT assay를 수행하였다. SBWE는 160 ${\mu}g/ml$ 이하의 농도에서 세포에 대한 독성이 없는 것으로 나타났다. 또한 lipopolysaccharide로 자극된 대식세포에서 발생하는 nitric oxide 소거능 효과를 조사해 본 결과 40 ${\mu}g/ml$의 농도에서 SBWE는 40%의 억제효과를 나타내었다. 뿐만 아니라 SBWE는 zymosan으로 자극된 대식세포의 식세포작용도 억제 하였다. 더욱이 SBWE는 농도에 비례하여 대식세포에서 생성되는 $TNF{\alpha}$함량을 감소시켰다. SBWE는 또한 시간경과 실험에서 인터류킨 베타($IL-1{\beta}$)의 생성을 억제하였다. 더욱이 cyclooxynase-2의 발현수준은 20 ${\mu}g/ml$의 SBWE 처리에 의하여 현저하게 감소되었다. 그러므로 이상의 연구결과는 황금 열수 추출물이 면역조절을 통해 항염증을 효능을 발휘할 수 있다는 하나의 증거를 제공한다.

Cimentidine에 의(依)한 면역반응조절(免疫反應調節) (Modulation of Immune Response by Cimetidine)

  • 최근(最近) 히스타민이 면역반응(免疫反應)을 조절(調節)함이 구명(究明)되고 있으나 생체내(生體內) 실험(實驗) 특(特)히 마우스에서의 연구보고(硏究報告)를 희소(稀少)하다. 본(本) 실험(實驗)에서는 histamine-2-receptor antagonist($H_2$ 차단제(遮斷劑))인 cimetidine과 히스타민이 마우스의 면양적혈구(緬羊赤血球)(SRBC)에 대(對)한 면역반응(免疫反應)에 미치는 영향(影響)을 실험(實驗)하였다. 마우스를 매일(每日) 14일간(日間) 여러가지 농도(濃度)의 cimetidine 으로 전처리(前處理)하고 여러가지 농도(濃度)의 SRBC($10^6,\;10^7$$10^8$ 세포(細胞))로 면역(免疫)하고 4일(日) 후(後)에 마우스 족척(足蹠)에 SRBC로 야기주사(惹起注射)하여 Arthus반응(反應)과 지연성과민반응(遲延性過敏反應)(DTH)를 족척종창반응(足蹠腫脹反應)으로 측정(測定)하였으며, 체액성면역반응(體液性免疫反應)은 적혈구응집소가(赤血球凝集素價)를 측정(測定)하였다. 수(數) 종(種) 농도(濃度)($10^{-1}M,\;10^{-3}M$, 및 $10^{-5}M$)의 히스타민을 야기주사(惹起注射)와 동시(同時)에 주사(注射)하여 24시간(時間)-DTH를 측정(測定)하여 히스타민 효과(效果)를 평가(評價)하였다. Cimetidine은 DTH를 항진(亢進)시켰으며 그 항진(亢進)은 250 ${\mu}g$의 cimetidine을 투여(投與)하였을 때 현저(顯著)하였다. 그러나 Arthus 반응(反應)과 혈청항체가(血淸抗體價)는 cimetidine 전처리(前處理) 군(群)과 대조군간(對照群間)에 유의(有意)한 차이(差異)가 없었다. 히스타민은 SRBC에 대(對)한 DTH를 투여량-의존성(投與量-依存性) 유형(類型)으로 억제(抑側)하였으며 그 억제(抑制)는 저농도(低濃度)의 항원량(抗原量)($10^6$$10^7$ SRBC)일때 더 현저(顯著)하였다. 그러나 외인성(外因性) 히스타민은 $10^8$ SRBC로 면역(免疫)하였을 때늘 DTH를 감소(滅少)시키지 않았다. 이상(以上)의 본(本) 실험결과(實驗結果)는 cimetidine이 세포성(細胞性) 면역반응(免疫反應)을 항진(亢進)시키나 체액성(體液性) 면역반응(免疫反應)은 증가(增加)시키지 않으며 내인성(內因性) 및 외인성(外因性) 히스타민 즉시형과민반응(卽時型過敏反應)뿐만 아니라 세포성(細胞性) 면역반응(免疫反應) 조절(調節)에 관여(關與)함을 강력(强力)히 시사(示唆)하는 증거(證據)라고 사료(思料)되었다.

Modulation of senoinflammation by calorie restriction based on biochemical and Omics big data analysis

  • Aging is a complex and progressive process characterized by physiological and functional decline with time that increases susceptibility to diseases. Aged-related functional change is accompanied by a low-grade, unresolved chronic inflammation as a major underlying mechanism. In order to explain aging in the context of chronic inflammation, a new integrative concept on age-related chronic inflammation is necessary that encompasses much broader and wider characteristics of cells, tissues, organs, systems, and interactions between immune and non-immune cells, metabolic and non-metabolic organs. We have previously proposed a novel concept of senescent (seno)-inflammation and provided its frameworks. This review summarizes senoinflammation concept and additionally elaborates modulation of senoinflammation by calorie restriction (CR). Based on aging and CR studies and systems-biological analysis of Omics big data, we observed that senescence associated secretory phenotype (SASP) primarily composed of cytokines and chemokines was notably upregulated during aging whereas CR suppressed them. This result further strengthens the novel concept of senoinflammation in aging process. Collectively, such evidence of senoinflammation and modulatory role of CR provide insights into aging mechanism and potential interventions, thereby promoting healthy longevity.

Postbiotics Enhance NK Cell Activation in Stress-Induced Mice through Gut Microbiome Regulation

  • Recent studies have revealed that probiotics and their metabolites are present under various conditions; however, the role of probiotic metabolites (i.e., postbiotics in pathological states) is controversial. Natural killer (NK) cells play a key role in innate and adaptive immunity. In this study, we examined NK cell activation influenced by a postbiotics mixture in response to gut microbiome modulation in stress-induced mice. In vivo activation of NK cells increased in the postbiotics mixture treatment group in accordance with Th1/Th2 expression level. Meanwhile, the Red Ginseng treatment group, a reference group, showed very little expression of NK cell activation. Moreover, the postbiotics mixture treatment group in particular changed the gut microbiome composition. Although the exact role of the postbiotics mixture in regulating the immune system of stress-induced mice remains unclear, the postbiotics mixture-induced NK cell activation might have affected gut microbiome modulation.

전생활혈탕가미방(全生活血湯加味方)의 면역조절 작용을 통한 항관절염 효능에 관한 연구 (Anti-arthritis Effects of Jeonsaenghwalhyeoltanggamibang through Immune Modulation Modulation)

  • Purpose: The purpose of this study is to investigate the anti-arthritis effects of Jeonsaenghwalhyeoltanggamibang(JHTG) on collagen-induced arthritis(CIA) in mice. Methods: To assess the effects of JHTG on CIA in mice, we conducted several experiments such as analysis of arthritis index, cell count of draining lymph node(DLN) and paw joint, measurement of serum antibody levels and observation of the histological changes of joint. Results: 1. JHTG extract had a suppressive effect on the arthritis index of paw joints in CIA mice. 2. JHTG extract increased the total cell number of DLN, and decreased the total cell number of paw joints in CIA mice. 3. JHTG extract increased the absolute number of various cell surface receptors in DLN, and decreased the absolute number of B220+/CD23+ cells in DLN in CIA mice. 4. JHTG extract decreased the absolute number of CD3+, CD4+, CD11b+/Gr-1 cells in paw joint in CIA mice. 5. JHTG extract didn't decrease the absolute number of CD4+/CD25+ cells in paw joints in CIA mice. 6. JHTG extract decreased levels of total IgM in the serum of CIA mice, but had no effect on levels of collagen II specific antibody. 7. JHTG extract decreased the destruction of articular cartilages and collagen fibers and the proliferation of synovial cells in paw joints from CIA mice. Conclusion: These results indicate that JHTG has clinical potential for the treatment of rheumatoid arthritis by modulating the immune response.

Use of Prebiotics, Probiotics and Synbiotics in Clinical Immunonutrition

  • It is a recent observation that about 80 per cent of the body's immune system is localized in the gastrointestinal tract. This explains to a large extent why eating right is important for the modulation the immune response and prevention of disease. In addition it is increasingly recognized that the body has an important digestive system also in the lower gastrointestinal tract where numerous important substances are released by microbial enzymes and absorbed. Among these substances are short chain fatty acids, amino acids, various carbohydrates, poly-amines, growth factors, coagulation factors, and many thousands of antioxidants, not only traditional vitamins but numerous flavonoids, carotenoids and similar plant- and vegetable produced antioxidants. Also consumption of health-promoting bacteria (probiotics) and vegetable fibres (prebiotics) from numerous sources are known to have strong health-promoting influence. It has been calculated that the intestine harbours about 300,000 genes, which is much more than the calculated about 60,000 for the rest of the human body, indicating a till today totally unexpected metabolic activity in this part of the GI tract. There are seemingly several times more active enzymes in the intestine than in the rest of the body, ready to release hundred thousand or more of substances important for our health and well-being. In addition do the microbial cells produce signal molecules similar to cytokines but called bacteriokines and nitric oxide, with provide modulatory effects both on the mucosal cells, the mucosa- associated lymphoid system (MALT) and the rest of the immune system. Identification of various fermentation products, and often referred to as synbiotics, studies of their role in maintaining health and well-being should be a priority issue during the years to come.

족삼리(足三里)의 전침자극(電鍼刺戟)이 흰쥐의 중추신경계(中樞神經系)에서 Interleukin-6 의 활성(活性)에 미치는 영향(影響) -구심성(求心性) 체감각(體感覺) 정보전달(情報傳達)을 중심(中心)으로- (Differential Modulation of ST36 Stimulation on Interleukin-6-Induced Changes of Afferent Somatosensory Transmissionto the SI Cortex of Rats)

  • Objectives : Acupuncture is expected to have somewhat like the efficacy parallel increasing activity of immune system in Western modem medicine. There, already, are many animal researches on activating effect of acupuncture for the immune system in peripheral organs. So, we carried out this experiment to see whether acupuncture has controlling effect on interleukin-6(IL-6) activity in rat's brain. Methods and Results : We had topical application of IL-6(1U=lpg, $10{\mu}l$) on brain of rat. It reduced afferent sensory transmission to the primary somatosensory(SI) cortex from periphery. Whereas, electrical stimulation(ES, 2Hz, 1.5V, 15min) of ST36(足三里) with application of IL-6 prominently activated afferent sensory transmission. ES of non-acupoint(proximal tail) with IL-6 showed suppression of afferent transmission. ES of ST36 without IL-6 application also exerted facilitation of afferent transmission to the SI cortex. Conclusions : Electoacupuncture(EA) on ST36 has noticeable influences on modulating activation of IL-6 in central nervous system, which do major role in immune system.
