자연산 및 조직배양 사철쑥의 세포독성 및 면역활성 비교 (Comparison of Cytotoxin and Immune Activities between Natural and Tissue Cultured Plant in Artemisia capillaris Thunb.)

  • 자연산 인진호와 조직 배양체의 유용 생리활성을 살펴 보았다. 우선, 항암 효과를 살펴보기 위하여 인간 폐암세포(A549)와 위암세포(AGS), 인간 간암세포(Hep3B)와 유방암 세포(MCF7) 4가지 암세포주를 이용하여 세포 생육 저해능을 측정하였다. MTT assay의 경우 1.0g/l 의 농도에서 캘러스가 A549와 ACS에 대해 각각 약 45%와 42%로 모상근과 기내배양에 비해 높은 세포 생육 저해능을 나타내었고, 자연산 인진호는 약 45%와 38%로 캘러스와 비슷한 항암 활성을 나타내었다. SRB assay의 경우도 캘러스가 가장 높은 항암 효과를 나타내었는데 대체적으로 약 55% 이상의 암세포 저해율을 나타내었으며, 자연산 인진호의 항암 활성에 비해서도 높은 수치를 나타내었다. 면역세포를 이용한 생육도와 cytokine의 분비량을 측정한 결과 B 세포의 생육도는 6일째까지 계속적인 증가를 가져왔고, 이에 따라 cytokine의 분비량 또한 $TNF-{\alpha}$와 IL-6 모두 6일째까지 증가하는 경향을 나타내었다. 면역세포의 생육도의 경우는 자연산 인진호가 가장 높은 생육도를 나타내었고, 인진호의 기내배양, 모상근, 캘러스 중에서는 모상근에서 가장 높은 생육도를 나타내었다. Ctyokine의 경우도 $TNF-{\alpha}$에서 자연산 인진호의 분비량이 가장 많았고, 배양 방법에 따른 인진호의 경우 캘러스가 가장 많은 분비량을 나타내었다. IL-6의 경우는 모든 sample에서 control에 비해 높은 분비량을 나타내었고, 자연산 인진호와 기내배양, 모상근, 캘러스와 비교했을 때 큰 차이를 나타내지 않았다. HPLC 분석을 통한 추출물의 물질 비교 분석 결과 기내배양과 모상근에서는 자연산 인진호에 비해 주요 성분 peak인 artemisidin 면적이 모두 낮은 경향을 나타내었고, 캘러스의 경우는 일반 인진호에 비해 peak의 면적이 비슷한 경향을 나타법으로서, 캘러스가 다른 조직 배양체보다 인진호의 주요 성분인 artemisidin 성분의 함량이 더 높은 것으로 나타나 캘러스가 기내배양이나 모상근 보다 높은 항암 효과를 나타낸 것으로 사료되어진다. 이와 같이 자연산 인진호와 조직 배양체의 항암 효과나 면역활성에서 큰 차이를 나타내지 않았기 때문에, 이러한 조직배양체가 자연산 인진호를 대체 할 수 있는 하나의 방법으로서 환경적인 측면과 자원 보존적인 측면에서 볼 때 가치있는 결과라고 사료된다.

흑마늘(Black garlic) 추출물이 마우스 비장세포의 Cytokine 생성에 미치는 영향 (Effect of Black Garlic Extract on Cytokine Generation of Mouse Spleen Cells)

  • 생리활성물질을 다량함유하고 있는 마늘의 발효산물인 흑마늘의 면역활성을 검증하기 위하여 C57BL6 마우스 비장세포를 이용하여 흑마늘이 비장세포의 활성화에 미치는 영향을 확인하였다. 흑마늘 추출물은 시판되는 남해 흑마늘 액기스를 농축하여 사용하였다. 그 결과 IL-2에서 흑마늘 추출물만 처리한 군에서 생성이 증가하였으며, LPS와 흑마늘 추출물을 함께 처리하였을 때 IL-2와 TNF-${\alpha}$, IFN-${\gamma}$의 생성이 LPS만 처리한 군보다 증가하여 대식세포나 T림프구의 발현에 의해 일어나는 세포성 매개 면역을 활성화를 유도하는 Th1 세포의 발현을 활성화 하였다. 그리고 IL-6는 흑마늘 추출물만 처리하였을 때 후기생성이 증가하였으며, LPS와 흑마늘 추출물을 함께 처리한 경우 LPS만 처리한 군보다 IL-4와 IL-6의 생성이 증가하였다. IL-10은 LPS와 흑마늘 추출물을 함께 처리하였을 때 후기 생성이 감소하였는데, 이는 B 림프구의 활성화에 따른 항체생성 면역을 활성화하며 Th1 세포로부터 유도되는 세포성 면역반응을 억제함으로서 항체유도 체액성 면역반응으로 전환을 효과적으로 조절하는 것을 확인하였다. 따라서 흑마늘 추출물은 마우스 비장세포에서 T 림프구의 활성화에 따른 Th1 세포와 Th2 세포가 활성화되어 면역계의 세포성 면역과 체액성 면역반응을 활성화하여 면역조절에 효과를 나타내는 것으로 사료된다.

Efficient Induction of Th1-type Immune Responses to Hepatitis B Virus Antigens by DNA Prime-Adenovirus Boost

  • Background: Chronic infection with hepatitis B virus (HBV) affects about 350 million people worldwide, which have a high risk of development of cirrhosis and hepatocellular carcinoma. Treatment of chronic HBV infection relies on IFN-${\alpha}$ or lamivudine. However, interferon-${\alpha}$ is effective in only about 30% of patients. Also, the occurrence of escape mutations limits the usage of lamivudine. Therefore, the development and evaluation of new compounds or approaches are urgent. Methods: We comparatively evaluated DNA and adenoviral vaccines expressing HBV antigens, either alone or in combined regimens, for their ability to elicit Th1-type immune responses in Balb / c mice which are believed to be suited to resolve HBV infection. The vaccines were tested with or without a genetically engineered IL-12 (mIL-12 N220L) which was shown to enhance sustained Th1-type immune responses in HCV E2 DNA vaccine. Results: Considering the Th1-type cytokine secretion and the IgG2a titers, the strongest Th1-type immune response was elicited by the DNA prime-adenovirus boost regimen in the presence of mIL-12 N220L. In addition, the codelivery of mIL-12 N220L modulated differentially the immune responses by different vaccination regimens. Conclusion: Our results suggest that the DNA prime-adenovirus boost regimen in the presence of mIL-12 N220L may be the best candidate for HBV vaccine therapy of the regimens tested in this study and will be worthwhile being evaluated in chronic HBV patients.

Association between serum fatty acid composition and innate immune markers in healthy adults

  • BACKGROUND/OBJECTIVES: Supplementation with n-3 polyunsaturated fatty acids (PUFAs) has been shown to generally decrease levels of innate immune markers and inflammatory cytokines, but the specific associations between blood levels of PUFAs and those of innate immune markers have not been investigated. Thus, the present study was conducted to test the hypothesis that innate immune markers as well as cytokines are negatively associated with n-3 PUFAs but positively associated with n-6 PUFAs in healthy adults. MATERIALS/METHODS: One hundred sixty-five healthy Korean adults aged 25-70 years old were included in this cross-sectional study. RESULTS: Serum levels of n-3 PUFAs, such as 18:3n3, 20:5n3, 22:5n3, and 22:6n3 were negatively correlated with eosinophil and basophil counts and $TNF-{\alpha}$, $IFN-{\gamma}$, IL-4, and IL-10 levels. Multivariate analysis also showed that serum levels of n-3 PUFAs were negatively associated with monocyte, eosinophil, and basophil counts and $TNF-{\alpha}$, $IFN-{\gamma}$, IL-4, and IL-12 levels. Additionally, the ratio of 20:4n6 to 20:5n3 was positively correlated with eosinophil counts and associated with $TNF-{\alpha}$, $IFN-{\gamma}$, and IL-4 levels. However, NK cell activity was not associated with serum fatty acid composition. CONCLUSIONS: Innate immune markers such as eosinophil, monocyte, and basophil counts were inversely associated with serum levels of n-3 PUFAs, but were positively associated with the 20:4n6/20:5n3 ratio in this population.

Broussonetia kazinoki Siebold stimulates immune response in ovalbumin-immunized mice

  • Objective: To evaluate the immune-stimulatory potential of extracts of Broussonetia kazinoki Siebold (BK) on specific cellular and humoral immune responses in ovalbumin (OVA)-immunized mice. Material and Methods: C57BL/6 mice were immunized intraperitoneally with OVA/alum ($100{\mu}g/200{\mu}g$) on days 1, 8, and 15. BK (100, 300 or 1000 mg/kg) was given to mice orally for 21 days (from day 1 to day 21). At day 22, OVA-, lipopolysaccharide (LPS)- and concanavalin A (Con A)-stimulated splenocyte proliferation and OVA-specific and total antibodies were measured in plasma. Further, the effects of BK on expression of cytokine mRNA in OVA-immunized mice splenocytes were evaluated by RT-PCR analysis. Results: BK significantly enhanced OVA-, LPS-, and Con A-induced splenocyte proliferation in OVA-immunized mice (p<0.01). BK also significantly enhanced total IgM and OVA-specific IgG1 levels in plasma compared with the OVA control group. Moreover, BK up-regulated significantly the expression of mRNA level of IL-2 and IFN-${\gamma}$ in splenocytes. Conclusions: BK has immune-stimulating activity in an OVA-immunized mouse model system, enhancing the Th1 immune response. BK showed no cytotoxicity in this system, suggesting that BK may be a safe and effective adjuvant in humans.

아토피피부염 동물 병태 모델에서 석자해기탕(石紫解肌湯)의 면역조절작용에 관한 연구 (Study on the Immune Modulatory Activity of Seokjahaeki-tang Using Atopic Dermatitis Animal Models)

  • In order to evaluate the efficacy of SHT against atopic dermatitis (AD), various immune related cytokines as well as histological comparison were performed in animal models, and the results are described. Clinical skin index of the SHT treated group decreased significantly in weeks 11 and 13, compared to the control group. Also, CD4+ immune cell ratio in the dorsal skin was significantly decreased to 69%, and both epidermal and dermal skin thickness was decreased. Serum IL-4, IL-5, IL-6, IL-13, and TNF-${\alpha}$, which are all important markers of inflammation, were decreased to 64%, 44%, 87%, 48%, and 45%, respectively. The expression of histamine, a chemical transmitter increasingly released during the progression of inflammation, was significantly decreased to 47%. The production of IgE immunoglobulin was significantly decreased to 16% compared to the control group. In conclusion, SHT pacifies the activation of T cells, leading to suppression of both Th2 cytokine overexpression and infiltration of immune cells into skin. As a result, relative thinning of both epidermis and dermis were observed. With the results obtained from in vitro studies, the immune modulatory effect of SHT in AD animal models was experimentally demonstrated. This study should provide solid information to construct EBM and for clinical practice.

Effect of 1-palmitoyl-2-linoleoyl-3-acetyl-rac-glycerol on Immune Functions in Healthy Adults in a Randomized Controlled Trial

  • We previously reported that 1-palmitoyl-2-linoleoyl-3-acetyl-rac-glycerol (PLAG) accelerates hematopoiesis and has an improving effect on animal disease models such as sepsis and asthma. The effects of PLAG supplementation on immune modulation were assessed in healthy men and women. The objective was to evaluate the effects of PLAG supplementation on immune regulatory functions such as activities of immune cells and cytokine production. A randomized double blind placebo-controlled trial was conducted. Seventy-five participants were assigned to one of two groups; all participants had an appropriate number of white blood cells on the testing day. The PLAG group (n=27) received oral PLAG supplements and the control group (n=22) received oral soybean oil supplements. IL-4 and IL-6 production by peripheral blood mononuclear cells (PBMC) were lower (p<0.001 and p<0.001, respectively) with PLAG than with soybean oil. However, the production of IL-2 and IFN-$\gamma$ by PBMC was unaltered with PLAG supplementation. The B cell proliferation decreased significantly in the PLAG group compared to the soybean oil control (p<0.05). The intake of PLAG in healthy adults for 4 weeks was deemed safe. These data suggest that PLAG has an immunomodulatory function that inhibits the excessive immune activity of immunological disorders such as atopic and autoimmune diseases. PLAG could improve the condition of these diseases safely as a health food supplement.

외치방인 청기패독산(淸肌敗毒散)의 아토피피부염 동물 병태 모델에서의 면역조절작용에 관한 연구 (Study on the modulation of immune system of CPS in atopic dermatitis induced animal models)

  • In order to study the effect of CPS in the treatment of atopic dermatitis (AD), its role on various immune related cytokines were tested. Levels of liver and kidney function markers such as ALT, AST, BUN in NC/Nga mice were all normal indicating no toxicity in CPS treated group. Significant recovery from AD could be observed in CPS treated group through naked eye observation. Dermatitis index was significantly decreased after 11, 12, and 13 weeks of treatment. CD4+, CD8+, CD3+ /CD69+ immune cell ratio in DLN were decreased significantly by 37.2%, 49%, 20.8% in CPS treated group. CD4+ and CD11b+ /Gr-1+ immune cell ratio in dorsal skin were decreased significantly by 50.8% and 59.2% in CPS treated group. Expression of IL-4, IL-5, IL-6, IL-13 and TNF-${\alpha}$ in spleen were decreased by 78.8%, 97.8%, 64.7%, 73.6%, and 68.4%, respectively in CPS treated group. The results above strongly indicated the significant immune modulatory effect of CPS and thus clinical application of CPS on AD treatment.

면역세포 활성에 대한 Substance P의 영향 (Effects of Substance P on the Activities of Immune Cell)

  • The neuropeptide substance P(SP) has been recognized to modulate immune systems, with close proximity between peptidergic sensory nerve endings and immune cells. These include the macrophage and neutrophil activation, IL-2 production in T cell, augmentation of Ig synthesis, mast cell degranulation, $PGE_2$ and collagenase secretion in synoviocytes. In this study I examined SP-induced various biological activities such as antimicrobial action, cytokine production, and mast cell degranulation in the presence or absence of other inflammatory cell activators. Antimicrobial studies showed that undifferentiated HL-60 cells were not affected by SP. However, SP significantly enhanced antimicrobial action of TPA-treated or dbcAMP-treated HL-60 cells which had been differentiated into PMN or macrophage/monocyte. I could not find synergistic relationship between SP and LPS in parallel experiments of the above. SP did not induce IL-l production from murine macrophage cell line RAW264.7 whether costimulated with LPS or not. Mast cell degranulation was occured only when stimulated with high dose ($10^{-5}M$) of SP and the degree of this activation was slightly reduced by simultaneous application of $MIP-1{\alpha}$. In addition, CGRP which is known to be a common coexisting neuropeptide with SP within specific fibers did not augment the function of SP on mast cell degranulation. These results suggest that immunoregulatory activities of SP could be mediated through direct upregulation of various functions of immune cells and also upregulation of responsiveness of immune cells to other immune activators.

Restoration of Declined Immune Responses and Hyperlipidemia by Rubus occidenalis in Diet-Induced Obese Mice

  • Hyperlipidemia, which is closely associated with a fatty diet and aging, is commonly observed in the western and aged society. Therefore, a novel therapeutic approach for this disease is critical, and an immunological view has been suggested as a novel strategy, because hyperlipidemia is closely associated with inflammation and immune dysfunction. In this study, the effects of an aqueous extract of Rubus occidentalis (RO) in obese mice were investigated using immunological indexes. The mice were fed a high-fat diet (HFD) to induce hyperlipidemia, which was confirmed by biochemical analysis and examination of the mouse physiology. Two different doses of RO and rosuvastatin, a cholesterol synthesis inhibitor used as a control, were orally administered. Disturbances in immune cellularity as well as lymphocyte proliferation and cytokine production were significantly normalized by oral administration of RO, which also decreased the elevated serum tumor necrosis factor $(TNF)-{\alpha}$ level and total cholesterol. The specific immune-related actions of RO comprised considerable improvement in cytotoxic T cell killing functions and regulation of antibody production to within the normal range. The immunological evidence confirms the significant cholesterol-lowering effect of RO, suggesting its potential as a novel therapeutic agent for hyperlipidemia and associated immune decline.
