Influence of Mucor mucedo immobilized to corncob in remediation of pyrene contaminated agricultural soil

  • In recent years, immobilization agents were introduced into organic contaminated soil remediation and more and more materials were screened and used as the immobilizing carrier. However, effect of the decomposition of the immobilizing carrier on the bioremediation was rarely concerned. Therefore, the decomposition experiment of immobilizing carrier -corncob was carried out in the lab with the efficient degradation fungi - Mucor mucedo (MU) existing, and polycyclic aromatic hydrocarbons (PAHs) residues E4/E6 of the dissolved organic matter and microbial diversity during the decomposition process were studied. The results showed that: a) during the decomposition, the degradation of pyrene (Pyr) was mainly in the first 28 d in which the content of extractable Pyr decreased rapidly and the highest decrease was in the treatment with only MU added. b) Anslysis of E4/E6 changes showed that rich microorganisms could promote aromatization and condensation of humus. c) From the diversity index analysis it can also be seen that there is no significant difference in effects of PAHs on the uniformity of microorganisms. These results will not only be useful to have a better understanding of the bioavailability of contaminants adsorbed to biodegradable carriers in PAHs contaminated soil remediation, but also be helpful to perfect the principle of immobilized microbial technique.

Cr (VI) separation by PolyHIPE membrane immobilized with Aliquat 336 by solvent-nonsolvent method

  • PolyHIPEs membrane prepared with styrene (St), divinylbenzene (DVB), and ethylhexyl acrylate (EHA) can yield a unique pore structure provided by large voids highly interconnected by many small window throats. With the advantageous pore structure, PolyHIPE presents a potential as a support for carrier facilitated transport membrane. Tricaprylmethylammonium chloride (Aliquat 336) can be efficiently incorporated into the PolyHIPE membrane by a two-step solvent-nonsolvent method to obtain an Aliquat 336-immobilized PolyHIPE membrane with good stability. The study of Cr (VI) transport through Aliquat 336-immobilized PolyHIPE membrane indicates that the membrane has high initial flux and maxima stripping flux ($J_f^o=15.01({\mu}mol/m^2s)$, $J_s^{max}=6.15({\mu}mol/m^2s)$). The reusability study shows that the Aliquat 336-immobilized PolyHIPE membrane can maintain high Cr(VI) recovery efficiency even after 15 cycles of operations. The developed membrane was also used in the separation of Cr (VI) from other anions (i.e., $SO_4{^{2-}}$ and $NO_3{^-}$) and other cations (i.e., Ni (II), Mg (II) and Cu (II)) with good selectivity.

Hydrogen Sulfide Removal by Immobilized Thiobacillus novellas on $SiO_2$ in a Fluidized Bed Reactor

  • The removal of hydrogen sulfide ($H_2S$) from aqueous media was investigated using Thiobacillus novellas cells immobilized on a $SiO_2$ carrier (biosand). The optimal growth conditions for the bacterial strain were $30^{\circ}C$ and initial pH of 7.0. The main product of hydrogen sulfide oxidation by T. novellus was identified as the sulfate ion. A removal efficiency of 98% was maintained in the three-phase fluidized-bed reactor, whereas the efficiency was reduced to 90% for the two-phase fluidized-bed reactor and 68% for the two-phase reactor without cells. The maximum gas removal capacity for the system was 254 g $H_2S/m^3/h$ when the inlet $H_2S$ loading was $300g/m^3/h(1,500ppm)$. Stable operation of the immobilized reactor was possible for 20 days with the inlet $H_2S$ concentration held to 1,100 ppm. The fluidized bed bioreactor appeared to be an effective means for controlling hydrogen sulfide emissions.

  • 가정용 혈중 cholesterol 측정시스템을 개발하고자, 그 성능을 조절할 수 었는 벤수들의 최적화가 수 행되었다. 시스템의 주요 구성성분은 분석에 요구되는 효소들 (cholesterol esterase, cholesterol oxidase, 그리고 horseradish peroxidase) 이 부동화된 nitrocellulose membrane strip과 비 이온세척제 (Tri­t ton X-1OO) 및 발색물질 (3,3'-diaminobenzidine) 이 포함된 시료운반용액이다. 시료와 운반용액이 훈 합된 수용액을 membrane의 하단으로부터 흡수시키 면 모세관현상에 의해 시료는 부통화된 효소층으로 전달되어 연속효소반응이 일어난다. 마지막 효소반 응에서 발색물질은 산화되고 그에 따라 cholesterol 농도에 비례한 특정색이 신호로써 발생된다. 그 신호세기에 대해 효소들의 부동화조건과 운반용액의 화학조성이 최적화되었다. 최적조건 하에서 구성된 측정시스템의 농도응탑곡선은 혈중 cholesterol 농도를 측정하기에 충분히 높은 민감도를 나타내었다.

  • 황산화박테리아인 Thiobacillus sp. IW를 사용한 황함유악취 제거장치를 제작하기 위한 기초자료로서 균의 최적성장조건, 활성탄과 bioceramics에서의 고 정화특성을 조사하였다. Thiobacillus sp. IW는 최척 성장조건이 pH 7, $30^{\circ}C$이고, 이때 세대시간이 38 분으로 다른 황산화균에 비해 아주 빠른 성장속도를 보여주었다. 균의 최적성장조건은 활성탄을 담체로 사용할 경우 pH 5, $35^{\circ}C$였으며, bioceramic의 경우 pH 7~8, $35^{\circ}C$로 액상배양시와는 상이한 성장조건 을 보여주었다. pH, 온도의 변화에 대한 균의 성장은 활성단보다 bioceramics에서 보다 안정적이었고, 총균수도 많았다. 전체적으로 균의 성장속도면에서 는 bioceramics가 활성탄보다 우수한 담체로 사료된다.

  • Relationship between monoclonal antibody (MAb) productivity and growth rate, and effects of high cell density on MAb production rate increased with increasing specifis growth rate in both suspended and immobilized continuous cultures indicate a positively growth-associated relationship between MAb productivity and growth rate. moreover, the specific production rate was higher in the immobilized cell culture than that in suspended one at all dilution rates. In order to clarify these phenomana, MAb mPNA experession and cell cycle distribution were investigated in bacth cultures with immobilized cells and suspended cells. RT-PCR was used for observation of MAb mRNA expression and a two-color bromodeoxyuridine (BrdU)/propidium iodide (PI) flow cytometry method for determination of cell cycle distribution. The results revealed that MAb nRNA expression until dead phase, which was longer than in suspended cell. The cell cycle distribution patterns were observed almost the same for both immobilized and suspended cells. Such results may imply that a high cell density state has positive influence on the mRNA expression and on growth-associated Mab productivity of T0405 cells.

고정화 글루코아밀라제의 성질과 응용 (Characteristics and Applications of Immobilized Glucoamylase)

  • Glucoamylase를 $ZrO_2$로 피복된 96% porous glass에 azo-linkage를 형성시켜 결합하게 한후, 2.5% glutaraldehyde로 처리하여 효소를 고정화시켰다. 효소기질로는 용해도가 높고 점도가 낮은 30% enzyme thinned cornstarch (dextrose equivalent 값 : 24)를 사용하여 plug flow-column reactor에서 연속반응시켰다. 반응 최적 pH는 수용성효소의 5.0보다 alkaline 쪽으로 기울어져 7.0으로 나타났고, 고정화반응에 따라 열안정성이 높아지고 $40{\sim}60^{\circ}C$에서 최적 온도범위를 가리키며, Km값은 수용성 효소의 1.25mM보다 낮은 1.04mM값을 보여 주었다. 따라서, pH 7.0, $45^{\circ}C$에서 160시간 동안 corn starch를 기질로 효소반응을 시켜 glucose 90.3%, maltose 8.0%인 DE값 94.0인 전분당분해산물을 획득할 수 있었다.

  • 본 연구에서는 푸마르산 또는 숙신산을 생산/소비하는 생물공정에서 각 물질의 농도를 온라인 모니터링을 하기 위한 흐름주입분석기술 (FIA)을 개발하였다. 각 효소반응에 필요한 효소 (Fum/MDH, ICL/ICDH)를 VA-Epoxy Biosynth E3-carrier에 공유 결합에 의해 고정화하였고 소형 고정화 효소반응기를 흐름주입분석 장치에 도입하였다. 푸마르산-FIA 및 숙신산-FIA 시스템에서 고정화 효소 반응기의 특성 (예를 들면, 고정화 Fum/MDH 효소 반응기와 ICL/ICDH 효소 반응기는 최적 pH가 9.0이었으며 35$^{\circ}C$의 반응온도에서 최대 활성을 보임) 을 연구하였고 실제 생물공정에의 적용 가능성을 조사하기 위해 각 FIA 시스템의 성능을 조사하였다. 또한 모사공정과 실제 발효공정에서 푸마르산과 숙신산의 농도를 온라인 모니터링하여 오프라인 분석값과 비교하였고 잘 일치함을 볼 수 있었다.

  • In this study, we have developed a microbe-carrier that combined Desulfovibrio desulfuricans and zeolite for removal of Zn and As in contaminated seawater. Desulfovibrio desulfuricans, one of the sulfate-reducing bacteria (SRB) microorganism was exhibited stable growth characteristics in highly salted water and strong resistance to Zn and As contaminated seawater. Moreover, zeolites are one of the most useful carrier to remove heavy metals from wastewaters. The results showed that SRB immobilized zeolite carrier can enhance removal ratio of Zn and As. In addition, heavy metals tended to be better removed in medium at conditions of $37^{\circ}C$. In case of heavy metal concentration, they were effectively removed ranging from 50 to 100 ppm. These results show that SRB-zeolite carriers hold great potential to remove cationic heavy metal species from industrial wastewater in marine environment.

  • The use of carrier systems for the delivery of drugs to areas in the body in need of pharmacological intervention is now the subject of intense research in many laboratories. Because of its obvious advantages (e.g. protection of drugs from hostile environments, facilitated target penetration and avoidance of side effects), drug delivery is expected to ease the pressure and expense of new drug development by making better use of drugs in existence. Generally, carrier-mediated delivery has been envisaged either as direct transport of drugs to a biological target by a carrier that will associate with it selectively, or as release of drugs from a carrier circulating in the blood or immobilized in tissues, at rates compatible with optimal action. One system that has attracted considerable attention is the use of liposomes as carriers of pharmacologically active agents. 154 references were reviewed with special emphasis on the targeting of drugs by use of liposomes in this respect. Recent advances in the other carrier systems and in methods for the preparation of liposomes were also reviewed briefly.
