Risk Assessment of Pesticide for Earthworms (농약의 지렁이에 대한 위해성 평가)

  • To assess the risk of pesticides on earthworm, the acute toxicities of 10 pesticides were investigated and their toxicity exposure ratios(TERs) were calculated. As the TERs of paraquat dichloride and pendimethalin were more than 100, their risks were rated negligible. Risk of benfuracarb, cadusafos, chlorpyrifos-methyl, endosulfan, isazofos and parathion which have TERs of $10\sim100$ were rated low. However, risk of imidacloprid and phorate which have TER of less than 10 were estimated highly to need a reproduction study. Earthworms were exposed to twenty two pesticides including dazomet 98% GR having PECs of more than $5mg{\cdot}kg^{-1}$ in artificial soil at standard and double dose for 14 days. All the earthworms exposed to dazomet 98% GR and metam-sodium 25% SL were died to show their high risk, while no serious adverse effects were observed in the soil treated with 15 pesticides, calcite 95% WP, calcium polysulfide 36% CF, chlorothalonil 75% WP, daminozide 85% WP, dichlonil 6.7% GR, etridiazole 25% EC, fosetyl-Al 80% WP, glyphosate 41 % SL, hymexazol 30% SL, iprodione 50% WP, machine oil 95% EC, mancozeb 75% WP, propineb 70% WP, terbuthylazine 80% WP and triazophos 40% EC. In case of thiophanate-methyl 70% WP, copper hydroxide 77% WP, dimethoate 46% EC, tolclofos-methyl 50% WP and propamocarb hydrochloride 67% SL, any effect did not show clearly, suggesting an additional subchronic toxicity study. The risk of thiophanate-methyl 70% WP to earthworm was estimated high, considering its subchronic effect, while effects of copper hydroxide 77% WP, dimethoate 46% EC, tolclofos-methyl 50% WP and propamocarb hydrochloride 67% SL to earthworms were negligible, considering no adverse effects in subchronic tests.

  • Incidence of rice stripe disease, caused by Rice stripe tenuivirus (RSV), was surveyed during 2002 to 2004. The incidence area and ratio of diseased fields were decreased gradually during those period. Rate of diseased plants were 45.8%, 45.0% and 43.7% in the susceptible cultivars Chucheong, Saechucheong and Ilpum, respectively. However, the rate was 4.4% in resistant cultivar Hwaseong. In addition, breakdown rate was also significantly high in the susceptible cultivars in Chucheong, Saechucheong and Ilpum showing 33.6, 33.2 and 31.9%, respectively. In Hwaseong, the breakdown rate was 0.8%. Collection efficiency was compared between two insect vector collection methods. Insect-sucking machine method was much more efficient than sweeping net method in collecting small brown plant hopper (SBPH). According to the survey of the population density of the insect vector during 2002 to 2004, the national average population density was gradually decreased year by year, 3.6, 2.3, and 1.3%, respectively. This result was significantly related with the decrease of the incidence of rice stripe disease. Control efficiency of rice stripe disease by treating several seedling box and water surface with insecticides against SBPH resulted that imidacloprid GR, Fipronil FG, Clothianidin+Probenazole GR and Thiamethoxam GR showed over 80% of control efficiency at 28 days after treatment at the early stage in nursery.

Biological Characteristic of Obolodiplosis robiniae and Control Effects of Some Insecticides (아까시잎혹파리(Obolodiplosis robiniae)의 생물학적 특성 및 약제 살충 효과)

  • Biological Characteristic of Obolodiplosis robiniae and insecticidal activity of some insecticides against larvae of O. robiniae were investigated. Egg was in oval shape, and its color was light orange and became red when close to hatch. Length of the major axis and the minor axis of egg was 0.4 mm and 0.1 mm, respectively. Larval color was milky and size was 2.6 mm. Pupa was deep brown and its size was about 3.2 mm. Wing and abdomen of adult was black and reddish, respectively. Size of female adult was about 3.3 mm, and larger than male adult. Number of eggs in the ovary was $192.3{\pm}50.7$. First emergence was from late April to late May, and second from late May to late June. Third emergence was from late June to late July. Newly emerged adult escaped from soil, and second and third emerged adult escaped directly from gall of Robinia pseudoacacia leaf. Egg parasitoid was identified as Platygaster robiniae and parasite rate was 51.6%. Among test insecticides, imidacloprid 10% WP and thiacoprid 10% FL showed very strong insecticidal activity against larvae of O. robiniae at 48h later after treatment.

Reduction Effects of Residual Pesticides using the Eco-friendly Soil Amendments in Agricultural Soil (환경친화적 토양개량제의 농경지 중 잔류농약 경감효과)

  • This study investigated the reduction of residual pesticide in soil by using eight kinds of soil amendments such as shell, quick lime, dolomite, silicic acid, leaf mold, oil cake, bed soil and husk. A total of ten pesticides most frequently detected in agricultural soil were selected (azoxystrobin etc.) and soil amendments were treated in soils at a 2% application rate and incubated for 7 days. Pesticides were analyzed by HPLC-MS/MS with QuEChERS-based sample preparation procedure and pH was measured on 1, 3 and 7 days. This study showed that, soil amemdments were effective in reducing pesticides after 7days of incubation. Over 90% of azoxystrobin, cadusafos, chlorpyrifos, fluquinconazole, imidacloprid, isoprothiolane and procymidone were decomposed from soils amended with 2% quick lime, whereas the concentration of boscalid, dimethomorph and triycyclazole were not decreased. The soil pH increased to 12.8-12.9 in soil amended with quick lime, but other soil conditioners did not change the soil pH. Quick lime was particularly effective in reducing residual pesticides.

Insecticidal Susceptibility and Emergence of First Instar Larvae of Pseudaulacaspis pentagona (뽕나무깍지벌레 1세대 부화 약충의 발생과 살충제 감수성)

  • The white peach scale (Pseudaulacaspis pentagona) is a significant pest of trees belonging to the Rosaceae family. We investigated the effect of ten insecticides on hatching rate of first instar white peach scale larvae (crawlers). The insecticides are Dinotefuran WG, Benfuracarb WG, Pyrifluquinazon WG, Clothianidin SG, Buprofezin WP, Buprofezin+Imidacloprid SC, Buprofezin+Etofenprox WG, Buprofezin+Thiacloprid SC, Benfuracarb+Buprofezin WG, Acetamiprid+Buprofezin EC. We observed that several crawlers emerged in early May, with the average number being 45.7. Hatching period was approximately 14 days, and the hatching success rate was approximately 77.7%. Overwintering females produced an average of 58.7 eggs. All insecticides achieved 100% mortality in crawlers. Our results suggest that these insecticides can effectively control white peach scale when used at the crawler stage.

  • Biological Characteristics of Lycorma delicatula and insecticidal activity of some insecticides against nymphs of L. delicatula was investigated. Nymph of L. delicatula had 4 instars, and color of body was black. There were white spots on the body of 1st-3rd nymph. Upper body became red at 4th nymph. Adult forewings were brownish, and had black spots. Color of hind wing were red. The egg mass was covered with a yellowish brown secretion. The adult of L. delicatula emerged once a year. Among test insecticides, deltamethrin 1% EC and fenitrothion 50% EC showed very quick and strong insecticidal activity against the 2nd-3rd nymphs of L. delicatula. Imidacloprid 4% SL and clothianidin 8% SC showed 100% insecticidal activity at 24h after treatment. Thiacoprid 10% SC revealed the weakest insecticidal activity among the insecticides tested.

Toxicity of Some Pesticides to Twospotted Spider Mite (Acari: Tetranychidae) and its Predator Amblyseius womersleyi (Acari; Phytoseiidae) (점박이응애(Tetranychus urticae Koch)와 긴털이리응애(Amblyseius womersleyi Schicha)에 대한 몇가지 농약의 선택독성)

  • Toxicity of 5 acaricides, 4 insecticides. and 6 fungicides to the twospotted spider mite, Tetranychus urticae Koch, and its predator Amblyseius womersleyi Schicha was assessed in a laboratory using a leaf disk bioassay. Dicofol was equally very toxic to aduly females of T. urticae and A. womersleyi, and caused 46% and 40% mortality of eggs of those two species, respectively. Adult females of A. womersleyi dipped in Abamectin solution showed low mortality(16.6%), while all T. urticae females died within 24 hours after dipping. Three ovicidal acaricides (Tetradifon, Clofentezine, and Hexythiazox) showed no mortality of predator eggs, but more than 90% mortality of T. urticae eggs. Four insecticides (Teflubenzuron, Triflumuron, Diflubenzuron, and Imidacloprid) and three fungicides (Propineb, Fenarimol, and Polyoxin-B captan) were non toxic to the A. womersleyi adult females. The other fungicides (Chlorothalonil, Nuarimol.Mancozeb, and Folpet) were a little toxic to adult females of the predator, showing$\leq$23.3% mortality, It may be suggested from these results that 4 insecticides, 4 acaricides, and 6 fungicides described could be incorporated into the integrated pest management system with A. womersleyi in apple orchard.

Synthesis and biological activities of Chloronicotinyl derivatives (Chloronicotinyl 유도체의 합성 및 생물활성 검정)

  • Chloronicotinyl derivatives were synthesized by substitution of amino in 3-pyridylmethylamine with phosphite groups and their insecticidal and fungicidal activities were determined. At 500 ppm, compound 4 with methyl and butyl group in phosphonate and compound 5, 6, 7, and 8 with two butyl, 2,2,2-trifluorotehtyl, 2-ethylhexyl, phenyl, respectively, in phosphonate showed 90% insecticidal activities against brown plant-hopper (Nilaparvate lugens). These compounds showed, however, poor insecticidal activities against diamond-back moth (Plutella xylostella) and two-spotted spider mite (Tetranychus urticae) (<65%), suggesting that insecticidal activity of chloronicotinyl derivatives containing phosphorus moieties are species-dependent. Newly synthesized chloronicotinyl derivatives with halogen and/or heterocycle (compound $10{\sim}21$) did not show insecticidal activities. We also determined fungicidal activity of the synthesized chloronicotinyl derivatives against rice sheath blight (Pyricularia grisea), cucumber gray mold (Bortytis cinerea), tomato late blight (Phytophthora infestans), wheat leaf rust (Puccinia recondita), and barley powdery mildew (Erysiphe graminis). Compound 10 with butyl and 4-nitrophenyl in phosphonate at 10 ppm showed 85% fungicidal activity against rice blast, suggesting that chloronicotinyl derivatives containing phosphorus moieties could be developed as a fungicidal agent of a novel chemical structure.

Monitoring and Risk Assessment of Pesticide Residues in Commercial Environment-Friendly Agricultural Products Distributed Using LC-MS/MS in Seoul Metropolitan Area (LC-MS/MS를 이용한 수도권에 유통되는 친환경 농산물의 잔류농약 모니터링 및 안전성 평가)

  • This survey was carried out in 2014 to estimate the pesticide residue levels in commercial environment-friendly agricultural products in Seoul metropolitan area. Pesticide residues in 27 commodities were examined and analyzed using LC-MS/MS, which can simultaneously detect up to 85 pesticides. A total of 441 samples were collected and analyzed. Among the detected samples, the residue levels in 1.2% of organic agricultural products and 2.2% of pesticide-free agricultural products exceeded the maximum residue limits (MRLs); hence, 1.8% of the total samples exceeded the MRLs. The safety of the detected pesticides was assessed by monitoring the acceptable daily intake level (ADI) and acceptable dietary exposure (ADE) to the pesticides via consumption of the commodities. ADI and ADE are found to be 0.07728-9.46530% and 0.00141-0.17210%, respectively, which means that the residual pesticides in the environmental-friendly agricultural products in Korea are below the safe limit and therefore, pose no potential public health risks.

Gut Bacterial Diversity of Insecticide-Susceptible and -Resistant Nymphs of the Brown Planthopper Nilaparvata lugens Stål (Hemiptera: Delphacidae) and Elucidation of Their Putative Functional Roles

  • Knowledge about the gut bacterial communities associated with insects is essential to understand their roles in the physiology of the host. In the present study, the gut bacterial communities of a laboratory-reared insecticide-susceptible (IS), and a field-collected insecticide-resistant (IR) population of a major rice pest, the brown planthopper Nilaparvata lugens, were evaluated. The deep-sequencing analysis of the V3 hypervariable region of the 16S rRNA gene was performed using Illumina and the sequence data were processed using QIIME. The toxicological bioassays showed that compared with the IS population, IR population exhibited 7.9-, 6.7-, 14.8-, and 18.7-fold resistance to acephate, imidacloprid, thiamethoxam, and buprofezin, respectively. The analysis of the alpha diversity indicated a higher bacterial diversity and richness associated with the IR population. The dominant phylum in the IS population was Proteobacteria (99.86%), whereas the IR population consisted of Firmicutes (46.06%), followed by Bacteroidetes (30.8%) and Proteobacteria (15.49%). Morganella, Weissella, and Enterococcus were among the genera shared between the two populations and might form the core bacteria associated with N. lugens. The taxonomic-to-phenotypic mapping revealed the presence of ammonia oxidizers, nitrogen fixers, sulfur oxidizers and reducers, xylan degraders, and aromatic hydrocarbon degraders in the metagenome of N. lugens. Interestingly, the IR population was found to be enriched with bacteria involved in detoxification functions. The results obtained in this study provide a basis for future studies elucidating the roles of the gut bacteria in the insecticide resistance-associated symbiotic relationship and on the design of novel strategies for the management of N. lugens.
