A Potent Anti-diabetic Agent from Kalopanax pictus

  • To search for the anti-diabetic principle from the stem bark of Kalopanax pictus, seven kinds of chemical constituents including hederagenin glycosides and phenolic glycosides wre isolated. The anti-diabetic evaluation of these isoltes in the streptozotocin-induced diabetic rats exhibited that kalopanaxsaponin A has a potent anti-diabetic activity in contrast to a mild activity of hederagenin. In addition, significant hypocholesterolemic and hypolipidemic activities of kalopanaxsaponin A and hederagenin were observed. The structure-activity relationship of kalopanaxsaponin A was also investigated in the present work.

Rhei Rhizoma and Chunghyuldan Inhibit Pancreatic Lipase

  • Pancreatic lipase-inhibitory activity of the rhizome of Rhei Rhizoma and its antihyperlipidemic activity were measured. Rhei Rhizoma inhibited pancreatic lipase with $IC_{50}$ value of 6.5 mg/ml (triolein as a substrate). Rhei Rhizoma significantly inhibited serum TG level in corn oil feeding-induced mice, and serum TG and cholesterol in Triton WR-1339-induced hyperlipidemic mice. However, Rhei Rhizoma did not show the hypolipidemic activity in high cholesterol diet-induced hyperlipidemic mice. When in vitro pancreatic lipase-inhibitory and in vivo antihyperlipidemic activities of Whangryunhaedoktang (WT) and Chunghyuldan (CD), which is consisted of ingredients of WT and Rhei Rhizoma, were measured, CD exhibited more potent inhibitory activities than WT. Therefore these results suggest that antihyperlipidemic activity of Rhei Rhizoma and CD may be more or less originated from the inhibition of pancreatic lipase.

벌나무(Acer tegmentosum Maxim) 추출물의 항산화 활성 및 고지방식이를 급이한 흰쥐의 지방간 개선 효과 (Antioxidant activity and improvement effect of Acer tegmentosum Maxim of dietary fatty liver in rat fed on a high-fat diet)

  • The effects on the radical scavenging activities and hepatic lipid levels in rats fed a high-fat diet (HFD) in the 70% ethanolic extract from Acer tegmentosum Maxim (ATM) were evaluated. Total phenol content of ATM was 168.60 mg catechin/g in the 70% ethanolic extract of Acer tegmentosum. The DPPH and ABTS radical scavenging activities were 18.32 mM TE/g and 32.25 mM TE/g, respectively. Food efficiency ratio was lower significantly in supplemented group with 150 mg/kg BW/day during 5 weeks (HFD+ATM) compared to HFD. Total cholesterol and triglyceride contents in liver tissue of HFD+ATM were lower significantly compared to those of the HFD. Supplementation of ATM significantly decreased lipid peroxide contents and increased radical scavenging activity in the liver tissue compared with that of HFD group. Moreover, the hepatocytes of HFD rats showed a typical fatty liver morphology showing the presence of cytoplasmic lipid droplets, whereas administration of ATM attenuated the number and the size of lipid droplets. In the liver tissue of ATM administrated HFD group, the mRNA levels of SREBP-1c, $PPAR{\gamma}$, and FAS were decreased. Therefore, these results suggest that Acer tegmentosum extracts could have antioxidant activities and the hypolipidemic effects in liver tissue by its phenolic compounds.

  • Plants of genus Dioscorea have long been served as important carbohydrate-stuffed foods in the tropical and subtropical regions, and utilized as traditional herb medicines to enhance digestive function, improve anorexia, and treat diarrhea in oriental countries. It is known that around 600 species of Dioscorea are distributed in the world including 107 species in Asia, but actually utilized Dioscorea species are restricted to small numbers. Phytochemical investigations for Dioscorea species have revealed a number of chemical components such as sapogenins, saponins, phenanthrenes, stilbenes, diterpenes and purine derivatives. According to recent pharmacological studies, Dioscorea species possess significant antioxidant, antibacterial and anti-inflammatory activities as well as anticancer, antidiabetic, cholesterol-lowering and hypolipidemic effects. Here, seven Dioscorea species (D. batatas, D. japonica, D. bulbifera, D. opposita, D. tokoro, D. nipponica and D. alata), mainly distributed and used in Korea, China, and Japan, are reviewed to provide their botanical, phytochemical and pharmacological properties were described.

Effects of Fermented Chub Mackerel Extract on Lipid Metabolism of Rats Fed a High-Cholesterol Diet

  • The present study was conducted to evaluate effect of fermented chub mackerel extract (FCME) on lipid metabolism of rats fed a diet supplemented with 1% cholesterol. Four week-old male rats were divided into three groups of 15 rats with 0, 0.1% or 0.2% FCME supplementation. In comparison with control, rats fed 0.2% FCME showed reduction of activities of acetyl-Coenzyme A carboxylase (p<0.05), 3-hydroxy-3-methyl glutaryl Coenzyme A reductase (p<0.01) and fatty acid synthetase (p<0.01). Rats fed 0.2% FCME also showed reductions in free cholesterol in the liver (p<0.05), and in concentrations of free cholesterol (p<0.05), LDL+VLDL-cholesterol (p<0.05), triglyceride (p<0.01) and phospholipid (p<0.01) in the plasma. Plasma HDL-cholesterol concentration was significantly (p<0.05) higher in treatment groups as compared with control group. Atherogenic index was also significantly lower in rats fed 0.1% or fed 0.2% supplement diet, whereas bile acid in feces was not significantly affected. The current study showed that 0.2% inclusion level of the fermented chub mackerel extracts might have hypolipidemic properties.

Effect of Resveratrol on Serum and Liver Lipid Profile and Antioxidant Activity in Hyperlipidemia Rats

  • The antioxidant activity of resveratrol in cholesterol-fed rats, along with its hypolipidemic effects was determined. Thirty two male Sprague-Dawley (SD) rats were randomly divided into three groups (Control, Res30 and Res70) and fed a hyperlipidemic diet for 4 weeks. Resveratrol was suspended in 0.3% carboxymethyl cellulose (CMC) solution and given to rats of the Res30 and Res70 groups once a day for 4 weeks by oral intubation at a dose of 30 and 70 mg/kg body weight, respectively. The control group received 0.3% CMC solution alone. Resveratrol significantly lowered serum lipid, hepatic cholesterol (TC) and triglyceride (TG) levels compared to the control. Excretion of bile acids was significantly enhanced by resveratrol. The overall potential of the antioxidant system was significantly enhanced by the resveratrol as plasma and hepatic thiobarbituric acid reactive substances (TBARS) levels were lowered while serum superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT) activities were increased in the cholesterol-fed rats. These findings suggest that resveratrol maintains an antioxidant efficacy as well as its anti-hyperlipidemic effect.

  • Since times immemorial, people have been dependant on plants for the various nutritional and pharamacological properties. Folk and traditional medicine recognizes thousands of plant species having miraculous benefits. Fig (Ficus carica Linn.) has been part of folk-lore since centuries. Ficus carica Linn. (Moraceae) is a huge deciduous tree, with more than 800 species. Different parts of Ficus carica like bark, root, leaves, fruit and latex have their own valuable importance and are frequently used for the treatment of various illnesses. Fruit of Ficus carica is commonly called as fig (anjeer) has various medicinal properties used in Unani, Ayurvedic and Chinese traditional system of medicines. Fig fruit is mostly used in gastro intestinal and respiratory disorders. In Unani medicine, fig is used as a diuretic, mild laxative and expectorant. Phytochemical studies on the leaves and fruits of the plant have shown that they are rich in Phenolics, Flavonoids, Vitamin C, Alkaloids, Saponins, Coumarins, tannins, organic acids, and volatile compounds due to which it is having great antioxidant property. Most interesting therapeutic effects include hypoglycemic, hepatoprotective, anticancer, antimicrobial and hypolipidemic activities.

3-(4′-hydroxyl-3′, 5′-dimethoxyphenyl) Propionic Acid Suppresses NO Production and Elevates GSH Levels in Murine Macrophages

  • Previous studies have shown that kimchi and kimchi-derived 3-(4'-hydroxyl-3', 5'-dimethoxyphenyl) propionic acid have anti-oxidative and hypolipidemic effects in rats and rabbits. This study was designed to investigate whether chemically synthesized 3-(4'-hydroxyl-3', 5' -dimethoxyphenyl) propionic acid (HDMPPA) may ameliorate oxidative stress through the regulation of nuclear factor KB (NFkB) activation in lipopolysaccharide (LPS)-stimulated RAW 264.7 murine macrophage cells. Treatment of RAW 264.7 cells with 400 uM of HDMPPA significantly reduced LPS-stimulated nitric oxide (NO) production. Treatments with HDMPPA at 100 uM to 400 uM concentrations significantly elevated glutathione (GSH) level. However, cell viability and thiobarbituric acid-reactive substances (TBARS) concentrations were not affected by the concentrations of HDMPPA used. The specific DNA binding activities of NFKB, a transcription factor which is sensitive to oxidative stress, were not down-regulated by HDMPPA treatments. These results suggest that HDMPPA may have weak anti-oxidative activity against LPS challenge by scavenging NO and stimulating GSH production.

홍국(Monascus purpureus)쌀을 첨가한 고콜레스테롤 식이가 흰쥐의 항산화 활성에 미치는 영향 (Effect of Red Yeast (Monascus purpureus) Rice Supplemented Diet on Lipid Profiles and Antioxidant Activity in Hypercholesterolemic Rats)

  • 고콜레스테롤 식이에 홍국쌀 분말을 0.2%, 1% 및 5% 첨가한 식이를 4주간 섭취한 동물에서 홍국쌀의 지질 개선 효과와 함께 항산화 효과를 혈액과 간의 항산화 효소 활성, 항산화 효소의 유전자 발현 및 DNA 손상에 미치는 영향으로 분석하였다. 홍국쌀 분말 첨가 식이를 섭취한 후의 체중 변화, 식이섭취량, 식이 효율 및 간 무게는 대조군과 유의한 차이가 없었다. 혈장에서 총 콜레스테롤은 대조군에 비해 0.2% 첨가군에서 24% 감소하였으며, HDL 콜레스테롤은 5% 첨가군에서 20% 증가하였고 LDL 콜레스테롤은 0.2% 첨가군에서 42% 감소한 것으로 나타났다. 항산화 효소 활성에서는 SOD 활성이 감소하거나 유의성이 없었으나 적혈구에서 GPx와 CAT의 활성이 대조군에 비해 유의성 있게 증가하는 것으로 나타났으며, 간의 TBARS는 5% 첨가군에서 대조군에 비해 19% 유의적으로 감소한 것으로 나타났다. 항산화 효소의 유전자 발현에서는 5% 첨가군에서 CAT의 발현이 대조군에 비해 7.9배 유의성 있게 증가하였다. 홍국쌀 분말 섭취로 인한 DNA 손상은 관찰되지 않았으며, $H_2O_2$로 산화 스트레스를 가했을 때 DNA 손상이 농도 의존적으로 억제되는 것으로 나타났다. 이상의 결과로부터 홍국쌀의 섭취가 혈액과 간의 지질 대사 개선 효능을 가지며, 항산화효소의 활성화를 통해 ROS에 의한 세포 손상을 억제할 뿐 아니라 LDL 콜레스테롤의 산화도 억제할 것으로 예상되므로 심혈관계 질환에 대한 예방 효과가 있을 것으로 사료된다.

과루인 에탄올 추출물의 혈관신생 억제효과 (Inhibitory Effect of the Ethanolic Seed Extract of Trichosanthes kirilowii on Angiogenesis in Human Umbilical Vein Endothelial Cells)

  • The seeds of Trichosanthes kirilowii (STK) used in traditional Oriental medicine for the treatment of dry cough and constipation have diverse pharmacological activities, including hypolipidemic, antioxidant, immunosuppressive, and anticancer effects. However, the effect of STK on angiogenesis has not been studied yet. In this study, we investigated whether the ethanolic extract of STK (ESTK) can regulate the migration and tube formation of human umbilical vein endothelial cells (HUVECs) and explored the underlying mechanism. Results of transwell assay showed that ESTK treatment dose-dependently suppressed the migration of HUVECs. The conditioned medium collected from H1299 human lung cancer cells was used as a chemoattractant. Our observation suggests that ESTK would inhibit the recruitment of endothelial cells into tumors. In addition, ESTK treatment significantly reduced the tube formation of HUVECs. As a molecular mechanism, we found that vascular endothelial growth factor (VEGF)-induced phosphorylation of VEGF receptor 2 (VEGFR2) was completely blocked by ESTK treatment. The expression of angiogenic factors, including VEGFA, fibroblast growth factor 2 (FGF2), angiopoietin, placental growth factor (PGF), platelet derived growth factor (PDGF), angiogenin, and tumor necrosis factor (TNF)-α, was commonly decreased by ESTK treatment in H1299 cells, indicating that ESTK would reduce the production of angiogenic factors from cancer cells. Taken together, our results clearly demonstrated that ESTK exhibited anti-angiogenic effects in HUVECs, which provides another possible mechanism underlying the anticancer activities of STK.
