Plant growth promoting fungi(PGPF) obtained from zoysiagrass rhizosphere offer dual advantages - induse systemic disease resistance response in cucumber to C. orbiculare infection and cause enhancement of plant growth and increase yield. PGPF protected plants either by colonizing roots or by their metabolites. PGPF offer an advantage by protecting plants for more than 9 weeks and 6 week in the greenhouse and field. PGPF-induced plants limited pathogen spore germination and decreased the number of infection hyphae on the leaf, and increased lignification at places of attempted pathogen infection, thus reducing the pathogen spread. PGPF elicited increased activities of chitinascs, glucanases, peroxidase, polyphenol oxidase, and phenylalanine ammonia lyase to C. orbiculare infection in cucumber plants. The role of PGPF in elevating cucumber defense response to pathogen infection suggests potential application of PGPF as biological control agents.
Selection of fungal strains with high virulence against the developmental stages of Bemisia tabaci was performed using internal transcribed spacer regions. The growth rate of hyphae was measured and bioassay of each developmental stage of B. tabaci was conducted for seven days. All of the fungal strains tested were identified as Lecanicillium spp., with strain 4078 showing the fastest mycelium growth rate (colony diameter, 16.3 $\pm$ 0.9 mm) among the strains. Compared to strain 4075, which showed the slowest growth rate, the growth rate of strain 4078 was increased almost 2-fold after seven days. Strains 4078 and Btab01 were most virulent against the egg and larva stages, respectively. The virulence of fungal strains against the adult stage was high, except for strains 41185 and 3387. Based on the growth rate of mycelium and level of virulence, strains 4078 and Btab01 were selected as the best fungal strains for application to B. tabaci, regardless of developmental stage.
The strain BY-445 which produces new inhihitors of ${\alpha}$-amylase was isolated from a soil sample and identified. The aerial hyphae of this strain develope in the from of open spirals. The spore chain of BY-445 strain appears in spiral shape with spiny surface. Melanoid and soulble pigments were not observed. Gelatin was liquefied, and skin milk and starch was also hydrolyzed. The isolate contained LL-diaminopimelic acid in its cell wall hydrolysate. The content of fatty acid 16:iso, 15:0 anteiso and 16:0 was 25:30, 16.19 and 13.16%, respectively. BY-445 strain was closely related to Streptomyces violaceusinger but it was different from this strain in some cultural and physiological characteristics. This strain was, therefore, designated as Streptomyces sp. BY 445.
In order to select multifunctional powerful antagonistic biocontrol agent against red-pepper rotting fungi Phytophthora capsici, we isolated an indigenous antagonistic bacterium which produces antifungal substances and siderophores from a local soil of Kyongju, Korea. The isolated strain was identified as Pseudomonas fluorescens biotype F. The antibiotic produced from P. fluorescens 2112 inhibited hyphae growth and the zoospore germination of Phytophthora capsici. The favorable carbon, nitrogen source and salts for the production of antibiotic from P. fluorescens 2112 were glycerol, beef extract and LiCi at 1.0%, 0.5% and 5 mM, respectively. And antagonistic activity of P. fluorescens 2112 was confirmed against P. capsici in vivo.
Aspergillosis outbroken in I to 2-week-old chicks in a poultry-farm at Tae-gu district were observed in april, 1959. Clinical and pathological manifestations of the cases were as follows: 1. Clinically, inappetence, drowsiness, and increased thirst were observed in the early stage. In the later stage, extreme dyspnea, emaciation and retarded growth were main features. There was diarrhea in a few cases. 2. Macroscopically, tubercle-like nodules in the lung and cloudy swelling of the parenchymatous organs were present. Histologically, the lung was shown suppurative bronchitis and bronchopneumonia. Hyphae, myceilia, sterigmata and free spores of aspergillus were observed in the center of the nodules and the bronchial lumen.
Antimicrobial activity of sophorolipid produced by Candida bombicola ATCC 22214 was investigated, Minimum inhibitory concentration of sophorolipid to Propionibacterium acne and Bacillus subtilis were 0.5 ppm and 4 ppm, respectively. However no antimicrobial activity to E. coli was detected. Indicating the antimicrobial activity of sophorolipid only toward gram positive bacteria. Antifungal activity of sophorolipid was also detected in the hyphae growth assay for Botrytis cinerea. Malate dehydrogenase, a cytoplasmic enzyme, of B. subtilis was remarkably increased with the sophorolipid treatment, indicating the leakage of membrane by sophorolipid. This result shows the potentials of sophorolipid as mild, non-toxic antimicrobial agent.
Seasonal distribution of ectomycorrhizal associations in various types of forest in a boreal forest in Manitoba. Canada was investigated. Alsohe relationship between ectomycorrhizal growth and pine pollen nutrients was examined. In four different forest stands, ectomycorrhizas tended to be lower in the spring than in the summer and fall samples. In addition. a mature jack pine (Pinus banksiana) stand showed higher mycorrhizal activities than a young jack pine stand. Growth of Suillus brevipes hyphae wa ts stimulated by additions of pollen representing mean pollen deposition in Mistik Creek study area after 30 and 70 days of growth with dextrose availability. This result suggests that the peak ectomycorrhizal activity is followed by pollen deposition in the study region and therefore, addition of pine and spruce pollen in early or middle of June in the boreal forest can be an important seasonal nutrient source for ectomycorrhizal growth.
[ $(+)-Lyoniresinol-3{\alpha}-O-\beta-D-glucopyranoside$ ] (1) was isolated from an ethyl acetate extract of the root bark from Lycium chinense Miller, and its structure was determined using 1D and 2D NMR spectroscopy including DEPT, HMQC, and HMBC. $(+)-Lyoniresinol-3{\alpha}-O-\beta-D-glucopyranoside$ exhibited potent antimicrobial activity against antibiotic-resistant bacterial strains, methicillin-resistant Staphylococcus aureus (MRSA) isolated from patients, and human pathogenic fungi without having any hemolytic effect on human erythrocytes. In particular, compound 1 induced the accumulation of intracellular trehalose on C. albicans as stress response to the drug, and disrupted the dimorphic transition that forms pseudo-hyphae caused by the pathogenesis. This indicates that $(+)-Lyoniresinol-3{\alpha}-O-\beta-D-glucopyranoside$ has excellent potential as a lead compound for the development of antibiotic agents.
In 2002, sooty mold was observed on persimmon (Diopyros kald) from the Jinju Agricultural Products Wholesale Market in Gyeongnam, Korea. The lesion of the sooty mold usually appeared as scars or wounds formed on the surface of the fruits. The symptom started with water soaking lesion, then the fruit softened rapidly. Colony of the causal pathogenic fungus was dark green in color on potato dextrose agar. The diameter of growing hyphae was 1-2 $\mu\textrm{m}$. The mostly one-celled conidia were ovoid, lemon or cylinder in shape, blastophores, and sized 46-132 $\mu\textrm{m}$. Erected conidiophores were long-branched chains and their lengths varied from 24 to 346 $\mu\textrm{m}$ with 2-5 $\mu\textrm{m}$ width. The optimum temperature for mycelial growth was about 2$0^{\circ}C$. Based on their mycological characteristics, the fungus was identified as Cladosporium cladosporioides. This is the first report in Korea of the sooty mold of persimmon caused by the said fungus.
In 2005 and 2007, a basal stem rot of Disporum smilacinum caused by Sclerotium rolfsii occurred sporadically in a herb farm at Hamyang, Korea. The symptom initiated with water-soaking lesion and progressed into stem rot and wilt of a whole plant. Severely infected plants were blighted and died eventually. White mycelial mats appeared on the lesion at early stage and a number of sclerotia were formed on the stem near the soil line. The sclerotia were globoid in shape, 1-3 mm in size and white to brown in color. The optimum temperature for the growth and sclerotia formation was 30 on PDA and the hyphal width was measured $3-8{\mu}m$. The typical clamp connections were observed in the hyphae of the fungus grown on PDA. On the basis of symptom, mycological characteristics and pathogenicity to the host plant, this fungus was identified as Sclerotium rolfsii Saccardo. This is the first report on the stem rot of D. smilacinum caused by S. rolfsii in Korea.
