Effects of Laminaran from Eisenia bicyclis on Serum Lipids in Rats Fed High Cholesterol Diet (대황유래 Laminaran이 고콜레스테롤 식이를 급여한 흰쥐의 혈청지질 성분에 미치는 영향)

  • This study was conducted to investigate the influence of laminaran from Eisenia bicyclis on serum lipid composition of rats fed high fat and cholesterol diets. Fourty male Sprague-Dawley rats weighing $70{\pm}2.5g$ of 4 weeks old were fed experimental diets for 6 weeks with high fat diet consisting of basal diet plus cholesterol (1%) and lard (10%) for the inducement of hyperlipidemia. The effect of laminaran supplements via drinking waters on serum lipid composition of rat were investigated for 5 weeks by administration of experimental diet group fed basal diet only as normal group, control group fed high fat diet, LL group fed high fat diet plus 0.25% laminaran containing water, and LH group fed high fat diet plus 0.5% laminaran containing water, respectively. As a results of experiments, it was found that LL and LH groups showed significant (p<0.05) decrease in body weight gain and liver weight as compared with control and it may caused by decreased FER. The weight of cecum and adipose tissue (EFP) of LL group showed a significantly (p<0.05) decreased patterns compared with control. It was also found that LL and LH diet groups affects the intestinal length and transit time of rat as significantly (p<0.05) increased in length of intestine and decreased in transit time. In addition, LL and LH diet groups showed a dramatic decrease in triglyceride, total and LDL-cholesterol, and significant increase in HDL-cholesterol compared with control diet group, by which results in decreased in AI. These results indicate that crude laminaran from Eisenia bicyclis has a strong hyperlipidemic and hypercholesterolemic activities in rat fed high fat and cholesterol diet.

The Effect of 3-(4-hydroxyl -33,53-dimethoxyphenyl)propionic Acid in Chinese Cabbage Kimchi on Lowering Hypercholesterolemia (배추김치의 활성성분인 3-(4-hydroxyl-33,53-dimethoxyphenyl)propionic acid의 고지혈증 치료 효과)

  • The active principle responsible for lipid lowering in Chinese cabbage kimchi, 3-(4-hydroxyl-3'5'-dimethoxyphenyl)propionic acid, of molecular weight 226, was chemically synthesized and then used for the treatment of hypercholesterolemia in male New Zealand white rabbit. Hypercholesterolemia in rabbits were induced by feeding 0.5% cholesterol added chow diet for 5 weeks. Each experimental group has four rabbits in it. for the 1st experiment,3-(4-hydroxyl-3'5'-dimethoxyphenyl)propionic acid or simvastatin was injected to the ear vein of rabbit every other day for 16 days (2 mg/3 kg/2 days) while normal chow diet was provided. Blood was drawn every 4th day. For the 2nd experiment, all the experimental condition was same as the 1st trial except 0.5% cholesterol diet was provided while 16 days. Plasma cholesterol level was decreased when cholesterol in the diet was removed. Decreased in cholesterol in kimchi and simvastatin groups were 18.65 and 29.67%, respectively compared to the control when the normal diet was given, and cholesterol increase was inhibited by 33.79 and 21.81% for kimchi and simvastatin groups, respectively, when 0.5% cholesterol diet was provided. The drop in LDL-C level by the active principle of kimchi and simvastatin was not significant when normal diet was given, however the changes was significant (p<0.05), approximately 130% decrease, when 0.5% cholesterol diet was given. The 105% and 62% decreased in triglyceride concentration were observed from 0.5% cholesterol diet fed kimchi and simvastatin groups respectively HDL cholesterol levels in experimental groups were not changed significantly from the both trials. The HMG-CoA reductase activity of kimchi and simvastatin groups were found to be higher than that of control to compensate the hypercholesterolemic condition induced by 0.5% cholesterol diet in these groups. In conclusion, diet is an important factor to control the hypercholesterolemia besides drug treatment. 3-(4-hydroxyl-3'5'-dimethoxyphenyl)propionic acid that is the active principle in Chinese cabbage kimchi seems a beneficial to the hypercholesterolemia and its effect is comparable to that of simvastatin.

Effect of Kale Juice on Serum Lipid Levels & Phospholipid Fatty Acid Composition in Hypercholesterolemic Men (케일녹즙이 고콜레스테롤혈증 성인남자의 혈청 지질수준 및 인지질 지방산 조성에 미치는 영향)

  • The aim of this study was to evaluate the effect of kale (Brassica oleracea acephala) juice supplementation on serum lipid levels and phospholipid (PL) fatty acid compositions in hypercholesterolemic men. Thirty-two men with hypercholesterolemia (>200 mg/dl) were recruited among the faculty and staff at Y University after annual health examinations. The subjects consumed 150 ml of kale juice per day for a 12-week intervention period. Dietary and anthropometric assessments were performed before and after supplementation, respectively, to ensure that the subjects maintained their usual diet and lifestyle throughout the intervention. Serum concentrations of HDL-cholesterol and the ratio of HDL-cholesterol to LDL-cholesterol were significantly increased (p<0.001) after intervention. Serum LDL-cholesterol concentration and atherogenic index were significantly reduced (p<0.001). Levels of 12:0, 14:0, $18:1{\omega}9$, $18:3{\omega}6$, and sum of monounsaturated fatty acids (MUFA) in serum pholspholipid (PL) were significantly increased, while $22:4{\omega}6$ level was significantly reduced (p<0.05). It can be speculated that kale juice, containing large amounts of antioxidant nutrients, contributes to changes of serum-PL-fatty acid compositions and the improvements of serum lipid profiles. This study demonstrates the supplementation of regular meals with kale juice may favorably affect serum lipid profiles and serum-PL fatty acid compositions and, hence, could lower the risks of coronary artery disease in men with hypercholesterolemia.

Influence of Korean Red Ginseng Water Extract on Recovery of Hepatic Function in Hypercholesterolemic Mice Fed High Cholesterol Diet (홍삼물추출물이 고콜레스테롤 식이로 유도된 고콜레스테롤 혈증에서 간 기능 회복에 미치는 영향)

  • Hypercholesterolemia is associated with many pathological states such as fat deposit in the tendon and skin, hepatomegaly, pancreatitis and heart attack. The present study was focused on the effect of Korean red ginseng water extract (RGWE) on the recovery of hepatic function that was deteriorated in high-cholesterol diet fed mice. Mice were divided into 6 groups ; normal diet group (AIN-2WK), 4 week-high-cholesterol diet group (CHOL-4WK), 2 week-high-cholesterol + 2 week-normal diet group (CHOL-2WK), 2 week-high-cholesterol + 2 week-normal diet + RGWE treated group (R100, R300, R500). RGWE treated group received normal diet and RGWE at daily doses of 100, 300, 500 mg/kg b.w. for 2 weeks after the 2 weeks feeding of high-cholesterol diet. The results are as follows : 1. The hepatic weight and total hepatic lipid weight were markedly lower in RGWE treated group than those in the CHOL-4WK or CHOL-2WK group, indicating that ginseng inhibited hepatic hypertrophy. 2. Total hepatic cholesterol and HDL-cholesterol in normal diet conversion group (CHOL-2WK) were significantly reduced compared with the high cholesterol group (CHOL-4WK). The lowering effect was significantly reinforced by RGWE administration. 3. Hepatic triglyceride level of CHOL-4WK group was markedly lower than that of the normal group (AIN-2WK). However, the level was significantly increased in CHOL-2WK and RGWE treated group. The result indicated that ginseng administration and diet conversion exert normalizing effects of hepatic function. 4. The lipid peroxide levels in RGWE treated groups were markedly lower than CHOL-2WK group. These results suggest that Korean red ginseng water extract can be useful as a dietary supplement for the treatment of obesity or hypercholesterolemia by improving lipid metabolism and hepatic function.

Effects of Angelica keiskei Koidzumi and Turmeric Extract Supplementation on Serum Lipid Parameters in Hypercholesterolemic Diet or P-407-Induced Hyperlipidemic Rats (명일엽과 울금 추출물의 투여가 고콜레스테롤식이와 P-407로 유도한 고지혈증쥐의 혈중 지질 함량에 미치는 영향)

  • The effects of administration alone or mixed of Angelica keiskei and turmeric extract on blood lipids were evaluated in hypercholesterolemic diet or P-407-induced hyperlipidemic rats. In the study 1, female Sprague-Dawley rats weighing 160-180 g were divided into each groups and fed high cholesterol diets for 8 weeks. Experimental groups were administered with following diets: basal diet (Normal), high cholesterol diets (1% cholesterol). We did the oral administration for evaluation in experimental groups: C (vehicle), A (angelica extract), T (turmeric extract), AT (angelica extract, turmeric extract/ 1:1 complex). The concentrations of serum total cholesterol, and LDL-cholesterol were decreased by 6.8%, 9.8% in A group, by 22.1%, 28.8% in C group and by 28.2%, 35.6% in AT group, compared to the C group, respectively. HDL-cholesterol levels were not different among the experimental groups. In the study 2, we induced the hypertriglyceridemia in rats by intraperitoneal injection of P-407 (0.5 g/kg) once per three days. From the next day after P-407 injection beginning, we did the oral administration as the study 1. Angelica keiskei extract, turmeric extract and complex extract decreased serum triglyceride by 17.2%, 19.7% and 48.3%, respectively. These results suggested that Angelica keiskei and turmeric extract complex might have synergistic effect in lowering total cholesterol, LDL-cholesterol and triglyceride in hyperlipidemic rats.

Effects of Duck Oil on Serum and Organ Lipid Composition in Mature Rats (오리기름이 성숙쥐의 혈청 및 장기의 지질 조성에 미치는 영향)

  • This study was designed to evaluate lipid component of serum and organ in matured rats fed oil extract from ducks. Male Sprague-Dawley rats weighing 595g(52-week-old) were divided into 3 groups and fed experimental diets for 8 weeks. Rats were fed soybean oil(control group) diet, duck oil(DO group) diet and duck oil supplemented with 1% cholesterol(DOC group) diet. Serum triglyceride concentration was significantly lower in DO and DOC groups than that in the control group. Serum total cholesterol concentration was significantly higher in DOC group than those in the other groups, while serum HDL-cholesterol concentration was significantly lower in DOC group than those in the other groups. Hepatic total lipid, triglyceride and total cholesterol concentrations in DOC group were significantly higher than those in the other groups. These studies indicate that the duck oil decreases the concentrations of triglyceride and shows almost same concentrations of total cholesterol compared to the soybean oil supplemented group in serum and liver. But hypercholesterolemic effect of duck oil was not improved.

Effects of Ethylacetate Fraction of Onion on Lipid Metabolism in High Cholesterol-Fed Rats (양파 에틸 아세테이트 분획이 고콜레스테롤식이를 섭취한 흰쥐의 지질대사에 미치는 영향)

  • To investigate effects of ethylacetate fraction of onion(EFO) on serum lipid metabolism in rats fed high cholesterol diet, four groups of Sprague-Dawley male rats weighing about 100 g were given a high cholesterol diet of 1% cholesterol and 0.25% sodium cholate and EFO containing three concentrations (1%, 3% and 10%), respectively for 6 weeks. growth rate of the hypercholesterolemic group (control group) was higher than the normal group, whereas the groups given EFO showed a decreasing trends, compared with the control group, especially the most excellent effect in 10% of EFO, but any differences were not found between groups in feed efficiency ratio. Serum concentrations of total cholesterol and LDL-cholesterol were lower, whereas HDL-cholesterol concentration was significantly higher in EFO suplemented-grups than control group in dose dependent manner. EFO increased HDL-cholesterol/total cholesterol ratio and lowered atherogenic index. Free cholesterol and triglyceride concentrations were not decreased significantly with in 6 weeks, but cholesteryl ester concentration was significantly decreased in EFO suplemented groups than control group, and in case of serum phospholipid concentration, EFO was not showed significantly effect, but it gradually increased the level, compared with the control group. Therefore, it might be expected that ethylacetate fraction of onion is believed to be a possible protective or curative effects for the fatty liver and hyperlipidemia-induced by a high cholesterol diet.

The Effects of Gangzitongmaekeum on Antioxidation Activity and Dietary Hyperlipidemia-induced Mice (강지통맥음(降脂通脈飮)이 항산화 활성과 고지혈(高脂血) 유발(誘發) 백서(白鼠)에 미치는 영향(影響))

  • This experiments was performed to determine the effects of Gangzitongmaekeum(降脂通脈飮 : GTE) on antioxidation activity and hyperlipidemia induced by hypercholesterolemic diet in mice. The results obtained were as follows : 1. GTE showed a safety in cytotoxicity and toxicity of human fibroblast cells and liver. 2. GTE showed DPPH scavenging activity, superoxide dismutase (SOD)-like activity and inhibitory effect on reactive oxygen species (ROS). 3. GTE treated mice showed body and liver weight decrease, compared to the hyperlipidemia-induced control group. 4. GTE decreased total cholesterol and LDL cholesterol levels significantly, but HDL cholesterol levels not significantly. 5. GTE decreased triglyceride levels significantly. 6. Glucose levels in GTE treated mice significantly decreased compared to the hyperlipidemia-induced control group. 7. Albumin levels in GTE treated mice were similar with the hyperlipidemia-induced control group. 8. The lipophagy in liver compared with the control group tended to be decreased in GTE treated mice. In the change of aorta, the cell was regular and boundary of vessel wall was clear, compared to the hyperlipidemia-induced control group. 9. TBARS levels in GTE treated mice significantly decreased compared to the hyperlipidemia-induced control group. 10. The change of SOD and catalase activity significantly increased compared to the hyperlipidemia-induced control group. 11. ACAT mRNA level and HMG-CoA reductase mRNA levels in GTE treated mice significantly decreased compared to the hyperlipidemia-induced control group. Conclusion : These results suggest that Gangzitongmaekeum is effective in antioxidation activity and dietary hyperlipidemia-induced mice.

Treatment Effect of Mulberry Root-Bark (Mori radicis Cortex) Extract on Hypercholesterolemia in Rabbits

  • This study reports that the effect of Mori radicis cortex, Mulberry root-bark, on the lipid peroxidation products formation in the liver, blood serum and atherosclerosis of hypercholesterolemia rabbits fed Mulberry root-bark for eight weeks. The rabbits were permitted a fixed quantity to extract of Mulberry root-bark mixed hypercholesterol fodder. The lipid peroxidation products, malondialdehyde (MDA) was significantly diminished. Total cholesterol in serum worthily decreased. Also, triglyceride and high density lipoproteins (HDLs) increased after feeding Mori radicis cortex. Besides, thicky atheromatous plaque on the wall of aorta of rabbits fed on hypercholesterol diet were markedly attenuated by Mulberry root-bark extract. These results support and substantiate the extract of Mulberry root-bark as protection against lipid peroxidation in the liver, blood serum and atherosclerosis.

Effect of Curcuminoids and Natural Plants Extract Mixture on the Cardiovascular System in Rats (흰쥐에서 Curcuminoid 및 이를 함유한 천연식물 혼합물이 심혈관계에 미치는 영향)

  • Antiplatelet aggregation, anticoagulant and lipid-lowering drugs are clinically widely used for secondary preventive purpose in the cardiovascular patients, but there is no primary preventive agents to prevent these diseases. With the aim of developing effective primary agents for cardiovascular diseases, we tried to formulate an optimized mixture of natural plants extract containing Theae sinensis, Camelliae sinensis, Vitis vinifera, Gingko folium and curcuminoids from Curcuma longa and to evaluate its anti-thrombotic and anti-hypercholesterolemic effects in vivo. The inhibitory effect of curcuminoids on vascular smooth muscle cell proliferation and migration were also investigated in vitro. in the animal experiments treated with hyperlipidemic diet, oral treatment of curcuminoids and natural plants extracts mixture (100 mg/kg) into male Sprague Dawley rats for 7 week simultaneously inhibited platelet aggregation as well as improved lipid profile in the blood. Compared to control group, both of curcuminoids-treated and mixture-treated groups revealed significantly decrease of total cholesterol (24.4%, 28.6%), free cholesterol (25.1%, 24.0%), cholesterol ester (14.6%, 29.0%), LDL-cholesterol (27.0%, 32.0%) and triglyceride (15.0%, 31.0%), respectively. However, both groups showed increase of HDL-cholesterol (46.6% and 51.5%) . In particular, atherogenic index of curcuminoids and mixture treatment group was significantly decreased to 47.0% and 56.0%, respectively. Furthermore, oral treatment of curcuminoids and mixture significantly inhibited collagen-induced platelet aggregation (21.1% and 29.1%, respectively), compared to control group. The anti-thrombotic values of mixture was almost similar to that of aspirin treatment (100 mg/kg) group. These results suggest that the oral treatment of curcuminoids-based natural plant extract mixture improved cardiovascular conditions in hyperlipidemic rats.
