The antioxidant and anti-diabetes effects were evaluated by the ethanol extracts from Astragalus membranaceus root classified by years using through DPPH radical scavenging activity, hydroxyl radical (${\cdot}OH$) scavenging activity, total phenolic and flavonoid contents, reducing power activity, $\alpha$-amylase and $\alpha$-glucosidase inhibitory activities. $IC_{50}$ values for DPPH radical scavenging activity of ethanol extract from 6 years old root ($749.25{\mu}g/mL$) was higher than 1 year ($1452.67{\mu}g/mL$) and 3 years old root ($1095.61{\mu}g/mL$). 6 years old root showed better effects in ${\cdot}OH$ scavenging activity ($IC_{50}$: $10.58{\mu}g/mL$), reducing power, total phenolic contents ($26.13{\pm}0.79\;Tan\;{\mu}g/mg$, $24.03{\pm}0.74\;Cat\;{\mu}g/mg$) $\alpha$-amylase ($33.33{\pm}0.55%$) and $\alpha$-glucosidase inhibitory activity ($49.71{\pm}1.01%$). On the other hand, total flavonoid compound contents were estimated much higher in 1 year old root ($44.93{\pm}1.35\;Que\;{\mu}g/mg$, $70.32{\pm}2.03\;Rut\;{\mu}g/mg$) than others. Based on these results, It was suggested that 6 years old root of A. membranaceus has a potential candidate for functional cosmetic and medicine.
발아에 의한 거대배아미와 유색미의 기능성 변화를 항산화 활성에 중심을 두고 조사하였다. 전반적으로 환원력은 일반현미보다 거대배아미와 유색미가 높았고, 발아처리는 환원력을 증가시키는 경향이 있었다. 지질과산화에 대한 억제효과를 조사한 결과, in vitro의 linoleic acid peroxidation system 및 ex vivo의 토끼 적혈구 막지질 과산화 system 모두에서 억제효과도 유색미가 가장 높았고, 그 다음이 거대배아미, 일반현미의 순서로 나타났다. Superoxide radical에 대한 소거활성은 유색미>거대배아미>일반현미의 순서로 낮아졌으며, 무발아 조건과 비교할 때 발아처리는 라디칼에 대한 소거활성을 증진시켰다. Hydroxyl radical 소거활성도 유색미가 가장 높았으며, 그 다음이 거대배아미, 일반현미의 순서였다. 발아에 의하여 일반현미의 hydroxyl radical 소거활성을 급격히 증가되었지만, 발아미간에 소거활성의 순위는 변하지 않았다. In vitro에서 조사된 ROS 소거활성과 같은 경향이 HL-60 세포에서 TPA로 자극으로 발생된 ROS에 대한 소거활성에서도 나타났다. 실험 결과, 유색미와 거대배아미에서는 발아와 연관되어 세포독성의 증가가 발견되지 않는 차별적인 ROS 소거활성의 증가 현상이 발견되었다.
This study was designed to investigate the protective effect of Sasa borealis leaf extract on 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH)-induced oxidative stress in LLC-PK1 cells (porcine kidney epithelial cells). The butanol fraction from Sasa borealis leaf extract (SBBF) was used in this study because it possessed strong antioxidant activity and high yield among fractions. Exposure of LLC-PK1 cells to 1 mM AAPH for 24 hr resulted in a significant decrease in cell viability, but SBBF treatment protected LLC-PK1 cells from AAPH-induced cell damage in a dose dependant manner. To determine the protective action of SBBF against AAPH-induced damage of LLC-PK1 cells, we measured the effects of SBBF on lipid peroxidation and antioxidant enzymes activities of AAPH treated cells as well as scavenging activities on superoxide anion radical and hydroxyl radical. SBBF had a protective effect against the AAPH-induced LLC-PK1 cellular damage and decreased lipid peroxidation and increased activities of antioxidant enzymes such as superoxide dismutase and glutathione peroxidase. Furthermore, SBBF showed strong scavenging activity against superoxide anion radical. The $IC_{50}$ value of SBBF was $28.45{\pm}1.28\;{\mu}g/mL$ for superoxide anion radical scavenging activity. The SBBF also had high hydroxyl radical scavenging activity ($IC_{50}=31.09{\pm}3.08\;{\mu}g/mL$). These results indicate that SBBF protects AAPH-induced LLC-PK1 cells damage by inhibiting lipid peroxidation, increasing antioxidant enzyme activities and scavenging free radicals.
Hamcho (Salicornia herbacea) extracts were evaluated for total polyphenol content, antioxidant and pro-oxidant activities. The total polyphenol content was 1.81 g and 0.72 g per 100 g of dried sample in water and ethanol extracts respectively. Both water and ethanol extracts of Hamcho significantly exhibited antioxidant activity. The scavenging activity of hydroxyl radical was 13.8~26.2% and 14.2~16.0% in water and ethanol extracts respectively. The inhibitory effect of conjugated diene formation was 24.6~39.1% and 28.4~39.6% in water and ethanol extracts respectively. However, pro-oxidative effect was also observed in the Hamcho extracts. The Hamcho water extract showed the pro-oxidant effect by enhancing the formation of hydroxyl radical and conjugated diene. The ethanol extract of Hamcho induced conjugated diene formation at 0.5 mg/mL but not at 1 mg/mL. The hydroxyl radical formation was not induced by the Hamcho ethanol extract. Taken together, these results show that Hamcho extracts can act as pro-oxidants by generating hydroxyl radical or conjugated diene in addition to their antioxidant properties. Therefore, this study suggests that the physiological properties of Hamcho and its use as food materials should be considered with caution because antioxidant dietary plants such as Hamcho possess possible adverse effects induced by pro-oxidant activity.
The antioxidative effect of Ecklonia cava, a brown marine alga, was investigated on radical scavenging, including 1,1-diphenyl-2-picrylhydrazyl (DPPH), and hydroxyl and alkyl radicals, using an electron spin resonance (ESR) technique, and on the inhibition of $H_2O_2$-induced DNA damage using comet assay. E. cava was enzymatically hydrolyzed with five food industrial proteases (Alcalase, Flavourzyme, Kojizyme, Neutrase and Protamex) to prepare water-soluble extracts. All the proteolytic hydrolysates exhibited strong dose-dependent radical scavenging activities (above 80%) at a concentration of $2.5\;{\mu}g/mL$. Kojizyme extract (obtained by proteolytic hydrolysation of E. cava with Kojizyme) showed the highest hydroxyl radical scavenging activity of around 98%. In addition, the $H_2O_2$-induced DNA damage was determined using a comet assay, which was quantified by measuring the tail length. Reduction of DNA damage increased with increasing concentrations of Kojizyme extract from E. cava. These results indicated that E. cava has a potential as a valuable natural antioxidative source.
Objectives : This study was performed to investigate the antioxidant activity of water and ethanol extracts from Sasa borealis leaves, stems and roots. Methods : Sasa borealis leaves, stems and roots extract were prepared using water and ethanol. The total polyphenol and total flavonoid contents were analyzed. 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical scavenging activity, SOD like activity, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)(ABTS), hydroxyl radical scavenging activity, and Nitrite scavenging activity assays were carried out to determine the antioxidant activities. Results : The antioxidant activities of the Sasa borealis appeared higher in ethanol extract than water extracts. Total polyphenol and total flavonoids contents in ethanol extracts of leaves were $24.6mg/m{\ell}$ and $14.3mg/m{\ell}$, respectively, which were much higher than those of any other parts. SOD like activity was 70% ethanol extract of the leaves was highest with 15.68%. Electron donating ability was 70% ethanol extract of the leaves had the highest 59.07%. It exhibited high electron donating ability than BHT(45.68%). Nitrite scavenging activity of 70% ethanol extract was higher than the water extract at pH 2.5 and pH 4.2. Nitrite scavenging activity was 70% ethanol extract of the leaves was the highest 75.2%. Hydroxyl radical scavenging activity was 70% ethanol extract of the leaves was highest with 16.16%, showed very low activity than BHT(61.56%). Conclusions : These results suggest that 70% ethanol extracts from Sasa borealis leaves, exhibited higher antioxidant activities than those of root and stem, and can be potentially used as proper natural antioxidants.

In this study, we evaluated the protective effects of supercritical extracts and two step ethanol extracts after supercritical extraction from Schizandra chinensis on antioxidant activities and oxidative DNA and cell damages. Supercritical extracts removed DPPH (1,1-diphenyl-2-picryldrazyl) radical by 85.5% at 200 ${\mu}g$/ml, but showed low activities of scavenging and chelating the hydroxyl radical and ferrous iron. However, two step ethanol extracts showed low activities of scavenging the DPPH radical, but removed the hydroxyl radical by 86% at 200 ${\mu}g$/ml. In addition, we tested the activities of extracts for reducing hydroxyl radical-induced DNA and cell damage. Two step ethanol extracts showed protective effect against the oxidative DNA damage by reducing DNA segmentation, inhibiting DNA migration and decreasing the expression of phospho-H2AX. Also, two step ethanol extracts showed protective effect against the oxidative cell damage by inhibiting lipid peroxidation and increasing the expression of p21 protein. Taken together, we suggest that two step ethanol extracts from S. chinensis have a role as useful inhibitors against oxidative damages.
In this study, polyphenol and flavonoid contents were measured, and DPPH, OH, H2O2 radical scavenging activity, and the α-amylase inhibitory activity were measured to study the antioxidant activity of 70% ethanol extract from Morinda citrifolia L. The polyphenol and flavonoid contents of noni 70% ethanol extract were 29.52 GAE/g and 12.48 CE/g, respectively. Also, the IC50 values of DPPH, hydroxyl radical, and hydrogen peroxide scavenging activity of 70% ethanol extract from noni were 18.70 mg/mL, 26.45 mg/mL, and 35.67 mg/mL, respectively. Measurement of the α-amylase inhibitory activity of 70% ethanol extract from noni showed 45% inhibitory activity at 10 mg/mL.
갈변된 표고버섯을 건강기능성 식품산업의 소재로 활용할 수 있는 가능성을 탐색하는 일환으로서 갈변된 표고버섯의 생리활성을 항산화 활성을 지표로 평가하였다. 본 연구에서는 DPPH 라디칼에 대한 전자공여능과 superoxide radical 및 hydroxyl radical에 대한 소거활성, 그리고 linoleic arid 자동산화반응에 대한 억제활성을 조사하여 각 추출물의 항산화 활성을 평가하였다. 그 결과, 갈변 표고버섯의 전자공여능은 생 표고버섯보다 추출방법에 따라 2-4배 정도 높았지만, 건조 표고버섯보다는 높거나 비슷한 수준을 보였다. Superoxide radical에 대한 소거활성도 DPPH에 대한 전자공여능과 유사한 경향을 보였는데, 갈변 표고버섯의 소거활성은 생 표고버섯보다 2배 정도 높았다. 또한 이와 같은 갈변화에 따른 superoxide radical 소거활성의 증가는 직접적인 radical 소거작용의 결과일 가능성이 제시되었다. Hydroxyl radical 소거활성의 경우, 추출방법에 관계없이 소거활성은 갈변 표고버섯 > 건조 표고버섯 > 생 표고버섯의 순서로 낮아졌으며, superoxide radical 소거작용과 마찬가지로 시료가 직접적으로 hydroxyl radical을 소거했을 가능성이 암시되었다. Linoleic acid 자동산화반응을 이용한 지질과산화에 대한 억제활성을 측정한 결과, 전반적으로 70% 에탄올추출물의 활성이 열수추출물보다 높았고, 억제활성도 갈변 표고버섯 > 건조 표고버섯 > 생 표고버섯의 순서로 낮아지는 경향을 보였다. 이상의 결과는 상품성을 상실한 갈변 표고버섯이 건강기능성을 지향한 다양한 가공식품의 개발을 위한 적절한 소재임을 보여주었다.
