PAA와 HPMC을 이용한 Metformin HCl 함유 GR 타입 정제에 의한 용출율 개선 (Enhanced Dissolution Rate of Metformin HCl via GR-type Tablet with PAA and HPMC)

  • To develop a novel metformin HCl-loaded GR type tablet, various tablets were compressed with poly acrylic acid (PAA) and hydroxypropylmethyl cellulose (HPMC) using a wet granulation, and their physical properties such as swelling rate, hardness and dissolution were then investigated. Among the formulae tested in this study, the tablet prepared with PAA 971 and 974 as disintegrants showed fastest dissolution rate and swelling rate. Furthermore, the tablets prepared with PAA and HPMC improved the swelling rate, hardness and dissolution compared to those prepared with only HPMC. Our results suggested that the tablets prepared with PAA 971, 974 and HPMC might be a potential candidate for gastric retention type tablets.

  • The purpose of this study is to minimize the PCH(Propylene CHlorohydrins) as a by-product in HPMC(HydroxyPropyl MethylCellulose) manufacturing process. HPMC is made of cellulose which is natural high polymer. And HPMC is applicable to several industrial areas. Especially it can be used in food industry as an effective additive. PCH is the by-product which is generated in chemical reaction in HPMC manufacturing process. So it is essential to eliminate PCH for the improvement of product quality. Therefore we have studied to minimize the amount of PCH. It is expected that the application of HPMC could be enlarged as the result of this study.

고강도 시멘트 경화체의 특성에 미치는 미세골재 및 혼화재료의 영향 (Effect of Micro-aggregate and Admixtures on the Properties of High Strength Hardened Cement Paste)

  • 보통 포틀랜드 시멘트와 수용성 고분자 물질인 hydroxy propyl methyl cellulose(HPMC), 미세골재 및 혼화재료를 사용하여 W/C=0.1이 되도록 물을 첨가하고 twin roll mill로 혼합 성형한 후 60일간 양생하여 고강도 시멘트 경화체를 제조하였다. 이 경화체에 대한 휨강도 및 파괴인성을 검토하였다. SiC 분말 및 백색시멘트를 첨가한 경우 휨강도는 약 100MPa정도, 탄성계수는 80-95GPa의 값을 나타내었다. 실리카흄을 첨가한 경우 휨강도는 80MPa, 탄성계수는 60GPa의 값을 나타내었다.

식용코팅이 미숙 자두의 연화에 미치는 효과 (Effect of Edible Coatings on Softening of the Stored Immature Plums)

  • 칼슘을 첨가한 메틸셀룰로오스(CaMC)와 하이드록시프로필 메틸셀룰로오스(CaHPMC-15)의 식용코팅을 미숙자두에 처리하여 자두의 연화 현상을 알아보았다. 비코팅 자두는 저장 기간 동안 중량 및 호흡률 감소 현상을 나타내었고, 적정산도는 감소하였으며, pH는 증가하였다. 그러나 CaMC와 CaHPMC-15를 코팅하였을 때 초기의 중량감소와 호흡률감소 효과를 나타내었다. 일반적으로 연화의 지표로 사용되는 수용성 펙틴 함량과 polygalacturonase(PG)의 활성은 저장 중 증가하였으며, pectin esterase(PE)의 존재 또한 확인되나 CaMC와 CaHPMC-15식용코팅에 의하여 연화현상을 억제하였다. 저장 기간 경과에 따라 기계적 경도가 감소되었으나 식용코팅에 의하여 자두의 경도 유지 효과가 있었으며, 관능검사 결과 저장 기간 중 자두 조직의 연화가 감지되었고 코팅자두의 경우 비코팅자두보다 더 단단한 것으로 평가되었다. 결과적으로 미숙자두에 CaMC와 CaHPMC-15를 코팅함으로서 수확 후 연화현상을 억제할 수 있었다.

A Study on the High-Flowing Concrete with Low Unit Weight of Cement

  • Most compressive strengths commonly used in the construction field are in a range of 240 to 300 kgf/$\textrm{cm}^2$ at 28 days. To get this rage of strengths, however, high-flowing concrete requires cementitious binders more than 400 to 450 kg/$\textrm{cm}^2$ for preventing segregation and sedimentation of aggregates. This amount of cementitious binder generates a large emission of excessive hydration heat, which may consequently induce harmful cracks in concrete structure. In order to reduce excessive hydration heat, thus, this paper aims at fabricating a high-flowing concrete under the condition that cement content is kept as low as 350kg/$\textrm{cm}^3$ by using viscose agents. In a parametric study, effects of cement types such as a ternary blended cement and Type V on he physical characteristics of high-flowing concrete were evaluated. In addition, the influence of viscosity was also investigated by applying two different viscose agents, one in a range of 6,000 to 10,000 cps and the others of 10,000 to 14,000 cps. In terms of chemical admixtures used in concrete mixture, the superplasticizer was Sulfonated Melamine-Formaldehyde Condensate with about 30,000 of molecular weight, and main component of viscose agent was HPMC (Hydroxy Propyl Methyl Cellulose). Slump flow was fixed at 50cm with different dosages of superplasticizer in weight.

메밀가루를 이용한 제빵적성 연구 (Study on Bread-Making Quality with Mixture of Buckwheat-Wheat Flour)

  • The possibility of buckwheat-wheat flour mixture as bread was studied by adding 10, 20, and 30% of buckwheat to wheat flour. To improve bread-making quality of the mixture, gluten, ascorbic acid, and hydroxy propyl methyl cellulose (HPMC) were added to 30% buckwheat-wheat flour and sensory evaluation was also exercised on it. The weight of bread increased but the volume of that decreased as the percentage of buckwheat to wheat flour increased, and improved bread-making quality by adding gluten, ascorbic acid, HPMC to 30% buckwheat-wheat composite flour. When it comes to color, color of the bread got darker as the percentage of buckwheat increased, got brighter when gluten and HPMC were added and showed little difference when ascorbic acid was added. Texture of bread increased in maximum weight, strength, hardness but decreased in springiness and cohesiveness. The use of additives showed influence to the some results. In terms of the aging process, enthalpy increased with storage time, and it could be slowed down by the use of the additives. The sensory evaluation shwoed that 10% buckwheat-wheat bread was most excellent, 20% bread was similar to other bread made from wheat flour, and the bread made by mixing additives were better than just 30% buckwheat-wheat flour in terms of quality.

고체 당밀정화제와 종속영양 탈질미생물을 이용한 질산염 제거 (Denitrification by a Heterotrophic Denitrifier with an Aid of Slowly Released Molasses)

  • This study was conducted to determine the potential applicability of slowly released molasses (SRM) to treat nitratecontaminated groundwater. SRM was made by dispersing molasses in hydroxy propyl methyl cellulose-silicamicrocrystalline cellulose matrix. Column test indicated that SRM could continuously release molasses with slowly decreasing release rates of $64.6mg-COD/L{\cdot}h$ up to 65 hrs, $12.1mg-COD/L{\cdot}h$ up to 215 hrs, and $4.4mg-COD/L{\cdot}h$up to 361 hrs. A batch test using an isolated indigenous heterotrophic denitrifier Pseudomonas sp. KY1 having nitrite reductase (nirK) and liquid molasses demonstrated that the bacterium decreased 100 mg-N/L of nitrate to less than 10 mg-N/L at the C/N ratio of 10/1 in 48 hours. In a Pseudomonas sp. KY1-attached Ottawa sand column which continuously received molasses from a SRM-containing reservoir, the bacterium successfully removed nitrate from 20 mg-N/L to 3 mg-N/L during the 361 hours of column operation. The results showed the possibility that SRM can be used as a reliable, longterm extra carbon source for indigenous heterotrophic denitrifiers.

유동층 코팅기를 이용한 염산알푸조신의 서방형 과립 설계 및 제조 (Formulation and Preparation of Sustained Release Pellet for Alfuzosin HCI Using Fluid-bed coater)

  • Alfuzosin, an Alphal-adrenoceptor antagonist is used for the treatment of patients with voiding and in a lesser extent storage lower urinary tract symptoms (LUTS) associated to benign prostatic hyperplasia (BPH). The objective of this study was to formulate sustained release alfuzosin HCl granules and assess their formulation variables. The $Eudragit^{(R)}$ as a polymer, sustained release membrane, and dibutyl sebacate (DBS) as a plasticizer were used. Multi-coated alfuzosin HCl delivery systems composed of sugar sphere, various excipients, $Eudragit^{(R)}$ and HPMC (hydroxy propyl methyl cellulose), Cellulose Acetate were prepared by fluid-bed coater. Membrane layer were used $Eudragit^{(R)}$ RS PO and NE 30D. And the alfuzosin HCl coated beads were coated immediate release drug layer for initial burst. Its dissolution test was carried out compared to conventional products ($XATRAL^{(R)}$ XL). The release rate of drug from coated beads was higher than that from $XATRAL^{(R)}$ XL in pH 6.8.

멜라토닌이 함유된 다층 코팅 펠렛의 방출 및 특성분석 (Release and Characterization of Multiple Coated Pellets Containing Melatonin)

  • Melatonin (MT) is an indole amide pineal hormone. It has not only very short half-life but also pH-sensitive property. The sustained release dosage form which delivers MT in a circadian fashion over 8 h is clinical value. The purpose of this study is to prepare sugar beads using multiple coating methods and enteric-coated in a sustained release to evaluate in vitro release characteristics in simulated gastric and intestinal fluids. The $Eudragit^{\circledR}$ as a polymer, sustained release membrane, and triethylcitrate (TEC) as a plasticzer were used. Multi-coated melatonin delivery system was composed of sugar, various excipients, $Eudragit^{\circledR}$ and enteric materials (e.g. hydroxy propyl methyl cellulose phthalate, HPMCP), and prepared by fluid bed coater. The dissolution test was carried out using the basket method at a stirring speed of 100 rpm at $37^{\circ}C$ in simulated gastric (pH 1.2) and intestinal fluid (pH 7.4). The released amount of MT was determined by High performance liquid chromatography method. The morhologies of surface and cross section of multi-coated beads were observed by scanning electron microscope. Size of multi-coated sugar beads was ranged over $1000{\sim}1300\;{\mu}m$. The release rate of MT from coated beads was limited in simulated gastric fluid (pH 1.2), but it was sustained in intestinal fluid (pH 7.4) during $3{\sim}8$ hours. The MT beads may provide small-intestine-targeted device for oral delivery. Studies on animal and relative experiment are in process.

PEO와 HPMC를 이용한 티니다졸 정제의 용출율 개선 (Dissolution Rate Improvement of Tinidazole Tablets using PEO and HPMC)

  • A novel polymeric tablet of Tinidazole was formulated to treat Helicobacter pylori and Giardia lambria more efficiently, It was possible to reduce hepatotoxicity by controlling the release of Tinidazole after peroral administration. A gastric retentive formulation made of naturally occurring carbohydrate polymers and containing Tinidazole was tested in vitro for swelling and dissolution characteristics. Tinidazole tablets containing various concentration of either PEO or HPMC were prepared by the wet granulation method. In vitro release of Tinidazole at pH 1.2 and pH 6.8 buffer solutions was observed at $37^{\circ}C$ by using a KP dissolution method and an UV (313 nm) spectrophotometer. Compared to a commercial Tinidazole tablet, in vitro release of Tinidazole at both pH 1.2 and pH 6.8 buffer solutions significantly decreased as the concentration of PEO or HPMC in the tablet increased up. And the gastric retentive formulation hydrated and swelled back to about 50% of its original size in 30 min. Thus, it was possible to control the release of Tinidazole by changing the content of PEO or HPMC in the tablet, thereby manipulating the release rate and the retention of Tinidazole.
