Hydrolytic and Metabolic Capacities of Thermophilic Geobacillus Isolated from Litter Deposit of a Lakeshore (수변 낙엽퇴적층에서 분리한 호열성 Geobacillus의 물질 분해 특성)

  • To understand contribution of thermophilic microorganisms in decomposition of litter deposits on shore of lakes, we surveyed a lakeshore litter deposit for bacteria growing at $60^{\circ}C$. Ten thermophilic isolates were selected for in-depth characterization, based on their high capacity to degrade high molecular weight organic compounds. Based on phylogenetic analysis on their 16S rRNA gene sequences, all isolates were identified as Geobacillus. The optimal growth temperature and pH of the strains ranged $55{\sim}60^{\circ}C$ and 6.0${\sim}$8.0, respectively. Salinity was inhibitory to the growth of the isolates, showing marked decrease of growth rates at 3% salinity. Based on activities of hydrolytic enzymes and profiles of carbohydrate utilization (determined by API 50 CHB kit), three G. stearothermophilus strains showed patterns clearly distinctive from other isolates. Two G. kaustophilus strains also demonstrated distinctiveness in their metabolic pattern and ecological parameters. However, ecological and metabolic profiles of the other five isolates were more variable and showed some degree of digression from their phylogenetic classification. Therefore, it could be concluded that endospore-forming thermophilic bacteria in lakeshore litter deposits contribute to degradation of organic materials with diverse ecological niches while having successions similar to microbial flora in compost. We propose that the thermophilic isolates and/or their thermo-tolerant enzymes can be applied to industrial processes as appropriate mixtures.

Preparation of Pronase Hydrolysate from Alaska-pollack (명태단백 Pronase 가수분해물의 제조)

  • In order to enhance the utility of alaska-pollack, the optimum conditions for the preparation of pronase hydrolysate. The optimum temperature and pH for the hydrolysis of alaska-pollack by pronase were 4$0^{\circ}C$ and pH 7.0. The reaction time and enzyme concentration were 4 hr and 1,000 units per g of substrate. Under the above optimum conditions alaska-pollack was hydrolysed by pronase yielding a hydrolytic degree of about 89eye. The bitterness and hyrophobicity of pronase hydrolysate were decreased with increasing reaction time. Hydrophobic amino acids(Tyr, Met, Ala, flu, Leu, and Phe) were increased for 2 hr, but fur thor hydrolysis was showed decrease of hydrophobic amino acids content. Palatable amino acids (Asp, Glu, Pro, Ser, Thr and Gly) were increased with hydrolysis time.

  • To investigate changes of components in salt-fermented anchovy, Engraulis japonims sauce during fermentation, various chemical properties were examined at $1.5{\~}3$ months interval during 18 months fermentation. Moisture content and pH were decreased slightly, but the contents of VBN and crude protein, total and amino nitrogen, hydrolytic degree and absorbance at 453 nm were increased gradually during fermentation. On the other hand, ash content and salinity showed almost no change. Hk and uric acid were the most abundant in ATP related compounds, ranging from $80.1{\%}\;to\;92.7{\%}$ in salt-fermented anchovy sauce during the fermentation, After 18 months of fermentation the sauce was rich in free amino acids, such as glutamic acid, leucine, alanine, lysine, isoleucine, valine in that order.

  • To investigate changes of components in salt-fermented big eyed herring, Harengula zunasi sauce during fermentation, various chemical properties were examined at $2\~3$ months intervals during 18 months of fermentation. The hydrolytic degree increased sharply until 5 months of fermentation and showed the gentle increasement after that. On the other hand, the content of total and amino nitrogen increased gradually during 18 months of fermentation, The hypoxanthine and uric acid were the most abundant in ATP related compounds, ranging from $81.7\%\;to\;90.1\%$. After 18 months of fermentation, sauce was rich in free amino acids such as glutamic acid, aspartic acid, alanine, lysine, threonine in that order.

  • Characteristics on lintnerization of dry type (Suwon 147) and moist type (Hwangmi) sweet potato starches were investigated. Chain distribution of lintnerized starches was also studied by debranching with pullulanase. Hydrolytic patterns of two starches showed two distinct stages and hydrolysis extents of Suwon 147 starch were lower than those of Hwangmi starch. The relative crystallinities of Suwon 147 starches were higher than those of Hwangmi starch. The elution profiles of lintnerized starches were composed of two peaks about degree of polymerization (DP) 25 and DP 15. The elution profiles of debranched samples showed only one peak about DP 15 and peak DP of Suwon 147 lintnerized starch was higher than that of Hwangmi.

  • Crude cellulases freshly prepared from cultures of Aspergillus niger, Prnicillum motatum, Trichoderma vride 16273 and Trichoderma viride 16374 were assayed on 4 different substrates including Na-CMC, cellulose powder, starch and sucrose. Enzyme prepared from A. niger contained highly active hydrolytic enzymes of the 4 substrates assayed. P. notatum [yielded relatively lower amount of cellulase but the extracts were also highly reactive on starch and sucrose. Trichoderma viride 16274 yielded very little cellulase and invertase, but the extracts showed a high degree of amylase activity. Trichoderma viride 16374, however, yielded collulase comparable to that of Penicillium notatum, but lower activities of amylase and invertase were seen. Commercial cellulases prepared from Penicillium notatum (cellulase[K]) and Trichoderma viride(cellulase[J]) indicated enzyme activities closely parallel to the crude enzymes freshly prepared from fungus cultures. The optimum pH's of cellulolytic activities of cellulase[K] and cellulase[J] were 4.0 and 5.0 respectively. The optimum temperatures of the cellulolytic activities of cellulase[K] and cellualse[J] were 4.0 and 5.0 respectively. The optimum temperatures of the cellulolytic activities of cellulase [K] and cellulase [J] were $60{\circ}C$ and $50{\circ}C$ respectively. Assuming the average molecular weight of Na-CMC is about 115,000, the Km values of cellulase [K] and cellulase[J] were found to be $3.3{\times}10^{-5}/nM$ and $3.3{\times}10^{-4}/nM$ respectively.

  • To investigate a possible biological activity of iridoid glucosides, six compounds, aucubin, catalpol, gardenoside, geniposide, rehmannioside and swertiamarin, were studied in relation with their potential influences in RNA and protein biosyntheses in murine tumor cell, sarcoma 180, in vitro. Protein biosynthesis was slightly inhibited by aucubin, gardenoside and swertiamarin. Degree of inhibition of RNA biosynthesis by those iridoid appeared to be more sensitive than that of protein biosynthesis. When aucubin was pretreated with ${\beta}-glucosidase$ to produce its genin form and the sarcoma 180 cells were exposed to this aucubigenin, the protein and RNA biosyntheses in the cells were profoundly inhibited. The results indicate that a biologically active from of iridoid compounds is the hydrolytic products of glycoside, i.e. genin form. It is also suggested that sarcoma 180 cells used in the experiments appear to lack of ${\beta}-glucosidase$, since the inhibitory actions of iridoid glucosides were so slight that those glucosides were not hydrolysed by the enzyme to their genin forms.

  • Crosslinked poly(glycerol-co-malate)s were synthesized from L-malic acid in Krebs cycle and glycerol. The synthesized polymer was identified by FT-IR spectroscopy. Swelling degrees of the copolymer hydrogels were increased with an increase in pH of the aqueous solution. Hydrolytic behaviors of the crosslinked copolymers were investigated in various pH buffer solutions at 37${\circ}C$. The Hydrolysis of the copolymers proceeded faster with increasing pH of the aqueous solution. Releasing behaviors of the model drug such as diclofenac monosodium salt were also measured in various pH aqueous solutions at 37${\circ}C$. The release concentration of diclofenac monosodium salt from the hydrogel systems was increased with increasing pH. These facts indicate that the unreacted carboxyl and hydroxyl groups in the copolymers are greatly affected by pH in the conditions.

  • In order to enhance the processing quality and utility of alaska-pollack meat, the optimum conditions for the preparation of pronase hydrolysate and the synthesis of plastein were investigated. The optimum temperature and pH for the hydrolysis of alaska-pollack by pronase were $40^{\circ}C$ and pH 7.0. The reaction time and enzyme concentration were 4 hr and 1,000 units per g of substrate. Under the above optimum conditions alaska-pollack was hydrolyzed by pronase yielding a hydrolytic degree of about 89%. Pronase hydrolysate was employed as substrate for plastein synthesis. The 30% pronase hydrolysates were adjusted to pH 7 for fruit-bromelain and pH 5 for stem-bromelain, and then plastein were synthesized by 1% bromelain at $40^{\circ}C$ for 24 hr. The plasteins synthesized by fruit- and stem-bromelain were consisted of peptides having average peptide length of 22.6 and 20.8 under the optimum synthetic conditions. The plastein synthesis reaction reduced considerably the bitterness of pronase hydrolysate.

  • Interfacial properties and microfailure degradation mechanisms of the oxygen-plasma treated biodegradable poly(p-dioxanone) (PPDO) fiber/poly(L-lactide) (PLLA)composites were investigated for the orthopedic applications as implant materials using micromechanical technique and surface wettability measurement. PPDO fiber reinforced PLLA composite can provide good mechanical performance for long hydrolysis time. The degree of degradation for PPDO fiber and PLLA matrix was measured by thermal analysis and optical observation. IFSS and work of adhesion, $W_a$ between PPDO fiber and PLLA matrix showed the maximum at the plasma treatment time, at 60 seconds. Work of adhesion was lineally proportional to the IFSS. PPDO fiber showed ductile microfailure modes at We initial state, whereas brittle microfailure modes appeared with elapsing hydrolysis time. Interfacial properties and microfailure degradation mechanisms can be important factors to control bioabsorbable composites performance because IFSS changes with hydrolytic degradation.
