A study of the metabolites for 7-keto-DHEA-acetate in human urine (뇨시료에 함유된 7-keto-DHEA-acetate의 대사체에 관한 연구 (I))

  • 7-keto-dehydroepiandrosterone-acetate (7-keto-DHEA-acetate) is an anabolic steroids, and we studied basically to the metabolites of it after human dosing. We tested the matrix effect from human urine to detect the 7-keto-DHEA-acetate. And LC/ESI/MS and GC/MSD was used to detect the metabolites in dosed urine. We found the some unknown compound from dosed urine (M1, M2, M3, M4 and M5), and from these results, we supposed that these compounds have the more than 3 hydroxyl and/or ketone group. Metabolite M1 was supposed that molecular weight is 302 and 3-,17-diketone and 7-hydroxyl compound (7-OH-androstendione). Metabolite M2 was supposed that the molecular weight was same to M1 and 7-,17-diketone and 3-hydroxyl compound (7-keto-DHEA).

  • Human and animals are living by acclimation to environmental changes like high and cold temperature, nose, confinement, etc. If the above changes reach a defined levels, some physiological abnormal state could appear, which we call them as stress state. Catecholamines are excreted by the sympathetic-adrenomedullary system in free from in urine. Catecholamines are derived from the adrenal medulla and urinary epinephrine can be taken as a rough estemation of the activity of this gland. Many scientist reported the endocrinological change, excretion of catecholamine and its metabolites, stomach ulcer formation, etc. under the condition of the confinement and high temperature. In this study author gave restraint, electric shock and immersion stress to rats by administrating by HPLC and got the following results. 1. In the restriant experiment, epinephrine contents in control rat was 194.7 ng, but in Bohyulanshintang administered rat urine 198.9 ng of epinephrine was found. 2. In the electrical shock experiment, 199.5 ng of epinephrine was found in the control rat urine, but in Bohyulanshintang administered rat urine epinephrine content was 142.4 ng. 3. Dopamine contents in control rat urine the immersion environment was 118.9 ng, but in Bohyulanshintang administered rat urine only 55.2 ng of dopamine was found. 4. Incontrol rat stomach there appeared focal erosion and inflamatory exudate, but in experimental group these symptom were turned to mild condition.

  • Tissue 02 tension is an important guide in detection of the general condition in critical patients. The tissue 0, is more difficult to measure with 0, sensor in skeletal muscle and subcutaneous tissues to present. But it is much easier to measure 0, tension in bladder urine with Censini catheter in Foley catheter than in tissue. We have measured 0, tension in bladder urine, main pulmonary artery and radial artery in 16 patients in chest surgical department of Yonsei University. College of Medicine from September 26 to December 22, 1981. Six patients were male and ten patients were female. Their ages ranged from 8 to 43 years. The correlation equation between the simultaneously measured PuO2 and PvO2 was found to be Ypvo2=4.04 + 0.88 Xpuo2 [r=0.88, p<0.0001] in regression curve with computer [HP/3,000, Program: SPSS] in the Yonsei University. Measurement of 0, tension in bladder urine and MPA will be rather simple, rapid and reproducible method than that of the 0, tension in tissues. But the speed of 0, consumption in urine is fast and so the 0, tensions in bladder urine were measured as soon as possible after they were collected. They were no complications or morbidity during measurement of 0, tension in these procedures except spontaneous removal of radial arterial cannulas in 2 patients.

Effect of Ultrasonic Pretreatment on Analysis of Potassium Ion in Human Urine Using 15-Crown-5-Anthracene-based Membrane (15-Crown-5-Anthracene 막을 이용한 요 중의 칼륨이온 분석에 미치는 초음파 전처리의 효과)

  • The effect of ultrasonic decomposition was introduced to develop a pretreatment method for the analysis of potassium ion in human urine by potentiometry. N-(4’-benzo-15-crown-5)-anthracene-9-imine, which has a good selectivity coefficient for potassium against ammonium, was used as an ion-selective material for the determination of potassium in urine with relatively high concentration of $NH_4{^+}$. Protenis in urine be removed by 85.1% when the sample acidified with 1.0 M $HNO_3$ was preteated for 100 s by sonication. Potential response of the membrane electrode in the pretreated urine had a slope of 54.6(${\pm}0.2,\;n=5$) mV/decade over the linear range of log $[K^+]$=-5~-1(r=0.9997). When an oxidant, $H_2O_2$, was addwd to the urine sonicated with $HNO_3$, the deproteinization increased 10% more than that in case if only $HNO_3$ and then the maximum ratio of ca. 95% was obtained. Moreover, the Nernstian slope for $K^+$ added to the urinary sample increased to 56.7(${\pm}0.1,\;n=3$) mV/decade. When the calibration curves were measured, the slopes did not vary even after the electrode was successively used 20 times with ultrasonic cleaning. The results showed that an ultrasonic pretreatment method provides simplicity in use, reduced treatment time and improved potentiometric characteristics of the membrane as the method effectively removes ca. 95% of proteins in urine.

  • This study was carried out to investigate the correlation between toluene in air and hippuric acid in human urine, which is based in the results of the health check-in and measure of working environment, was investigated for five years. Toluene in air and hippuric acid in urine were checked 116 workers who exposed to toluene in five type of industry. The Mean concentration of toluene in air by type of industry and every year and sex distinction, had positive correlation.(p<0.01) The Mean concentration of hippuric acid in urine by type of industry, every year, sex distinction, disparity of age and term of duty had positive correlation.(p<0.01).

  • An attempt is made to diagnose cancer of eight persons' urine by measuring the areas of the characteristic proton NMR signals observed in most cancer urine other than by using the previously developed reagent. The results of the attempt show that the former and ther latter give respectively one false diagnosis and four false diagnoses out of the persons' urine tested.

  • Hypertonicity (hypernatremia) of extracellular fluid causes water movement out of cells, while hypotonicity(hyponatremia) causes water movement into cells, resulting in cellular shrinkage or cellular swelling, respectively. In most part of the body, the osmolality of extracellular fluid is maintained within narrow range($285-295 mOsm/kgH_2O$) and some deviations from this range are not problematic in most tissue of the body except brain. On the other hand, the osmolality in the human renal medulla fluctuates between 50 and $1,200 mOsm/kgH_2O$ in the process of urine dilution and concentration. The adaptation of renal medullary cells to the wide fluctuations in extracellular tonicity is crucial for the cell survival. This review will summarize the mechanisms of urine concentration and the adaptation of renal medullary cells to the hyper tonicity, which is mediated by TonEBP transcription factor and its target gene products(UT-A1 urea transporter etc.).

  • A sensitive method for the quantitation of oxandrolone in urine was developed using GC/MS. After oral administration of 10mg oxandrolone, oxandrolone excreted in urine as unchanged form was extracted in ether and derivatized to its O-TMS. Oxandrolone excreted in urine as glucuronide conjugated form was extracted after enzymatic hydrolysis and derivatized to its O-TMS. The amounts of oxandrolone-O-TMS was measured in GC/MS with selected ion monitoring. Calusterone, a structurally similar anabolic steroid, was employed as internal standard.

  • A simple and sensitive high performance liquid chromatography for analysis of Putrescine, Spermidine and Spermine in urine is described. The benzoyl chloride derivatives of Putrescine, Spermidine and Spermine are separated on a ${\mu}$ Bondapak $C_{18}$ reverse-phase column with methanol/$H_2O$(50/50v/v) as the mobile phase. The results show that the mean level of polyamines in uterine cancer patients urine is much higher than in normal human urine.

Analysis of Matrine Alkaloids in Human Urine by Hollow Fiber Liquid-phase Microextraction with High-performance Liquid Chromatography (고성능 액체 크로마토그래피의 중공섬유 액상 미세추출에 의한 인간뇨의 Matrine Alkaloids의 분석)

  • A sensitive quantitative method for the determination of matrine alkaloids in human urine was developed based on hollow fiber liquid-phase microextraction (HF-LPME) combined with HPLC. The influence of the different factors on the HF-LPME efficiency including the pH and ion strength of the donar solution, the pH of the acceptor solution, stirring rate and extraction time were examined. The best HF-LPME conditions were as follows: 1-octanol impregnated in the pores of the hollow fiber, 100 mmol/L of $H_3PO_4$ at pH 1.50 as the acceptor solution injected into the lumen of the hollow fiber, 1 mol/L NaOH used to adjust the pH of the donor solution, stirring rate of 600 rpm and extraction time of 60 min. The LPME method was applied successfully to the analysis of matrine and sophocarpine in real urine samples.
