Vitamin A: a key coordinator of host-microbe interactions in the intestine

  • The human intestine is home to a dense community of microbiota that plays a key role in human health and disease. Nutrients are essential regulators of both host and microbial physiology and function as key coordinators of host-microbe interactions. Therefore, understanding the specific roles and underlying mechanisms of each nutrient in regulating the host-microbe interactions will be essential in developing new strategies for improving human health through microbiota and nutrient intervention. This review will give a basic overview of the role of vitamin A, an essential micronutrient, on human health, and highlight recent findings on the mechanisms by which it regulates the host-microbe interactions.

MicroRNA-451 Inhibits Growth of Human Colorectal Carcinoma Cells via Downregulation of Pi3k/Akt Pathway

  • MicroRNAs (MiRNAs) play important roles in coordinating a variety of cellular processes and abnormal expression has been linked to the occurrence of several cancers. The miRNA miR-451 is downregulated in colorectal carcinoma (CRC) cells, suggested by several research groups including our own. In this study, synthetic miR-451 mimics were transfected into the SW620 human CRC cell line using Lipofectamine 2000 and expression of miR-451 was analyzed by real time PCR, while expression of CAB39, LKB1, AMPK, AKT, PI3K and Bcl2 was analyzed by Western blot, and cell growth was detected by MTT assay. In comparison to the controls, a significant increase in the expression of miR-451 was associated with significantly decreased expression of CAB39, LKB1, AMPK, AKT, PI3K and Bcl2. The capacity of cell proliferation was significantly decreased by miR-451 expression, which also inhibited cell growth. Our study confirmed that miR-451 has a repressive role in CRC cells by inhibiting cell growth through down-regulating the P13K/AKT pathway.

Propagation Characteristics from Ingested Sources in Human Body (인체 내부 소스에 의한 전파 특성)

  • This paper presents the propagation characteristics from ingested sources in human body using the human model simulator. The simulator applies the FDTD method to the human data consisting of the human CAD and tissue data. After the accuracy of the simulator is verified, the received fields at one horizontal layer including the small intestine among the digestive organs are calculated in case that the electric field source is implanted in the center of the small intestine. The human propagation characteristics are illustrated by calculating the path loss per unit length according to various received positions from the simulated results.

  • We have cloned and functionally expressed a sodium dependent human nucleoside transporter, hCNT2, from a CNS cancer cell line U251. Our cDNA clone of hCNT2 had the same predicted amino acid sequence as the previously cloned hCNT2 transporter. Of the several cell lines studied, the best hCNT2 transport function was obtained when transiently expressed in U251 cells.(omitted)

Korean pine nut oil replacement decreases intestinal lipid uptake while improves hepatic lipid metabolism in mice

  • BACKGROUND/OBJECTIVES: Consumption of pine nut oil (PNO) was shown to reduce weight gain and attenuate hepatic steatosis in mice fed a high-fat diet (HFD). The aim of this study was to examine the effects of PNO on both intestinal and hepatic lipid metabolism in mice fed control or HFD. MATERIALS/METHODS: Five-week-old C57BL/6 mice were fed control diets containing 10% energy fat from either Soybean Oil (SBO) or PNO, or HFD containing 15% energy fat from lard and 30% energy fat from SBO or PNO for 12 weeks. Expression of genes related to intestinal fatty acid (FA) uptake and channeling (Cd36, Fatp4, Acsl5, Acbp), intestinal chylomicron synthesis (Mtp, ApoB48, ApoA4), hepatic lipid uptake and channeling (Lrp1, Fatp5, Acsl1, Acbp), hepatic triacylglycerol (TAG) lipolysis and FA oxidation (Atgl, Cpt1a, Acadl, Ehhadh, Acaa1), as well as very low-density lipoprotein (VLDL) assembly (ApoB100) were determined by real-time PCR. RESULTS: In intestine, significantly lower Cd36 mRNA expression (P<0.05) and a tendency of lower ApoA4 mRNA levels (P = 0.07) was observed in PNO-fed mice, indicating that PNO consumption may decrease intestinal FA uptake and chylomicron assembly. PNO consumption tended to result in higher hepatic mRNA levels of Atgl (P = 0.08) and Cpt1a (P = 0.05). Significantly higher hepatic mRNA levels of Acadl and ApoB100 were detected in mice fed PNO diet (P<0.05). These results suggest that PNO could increase hepatic TAG metabolism; mitochondrial fatty acid oxidation and VLDL assembly. CONCLUSIONS: PNO replacement in the diet might function in prevention of excessive lipid uptake by intestine and improve hepatic lipid metabolism in both control diet and HFD fed mice.

The Effects of Sa-Am Acupuncture on Radial Pulse in Healthy Subjects: A Comparative Study of Large Intestine Tonifying and Sedating (대장정격 및 승격 자침이 정상 성인의 맥파에 미치는 영향 비교 연구)

  • Objectives : The purpose of this study is to find parameters to compare the effects of Sa-Am acupuncture with large intestine tonifying and sedating on radial pulse in healthy subjects. Methods : Sixty healthy subjects participated in this study, divided into large intestine tonifying acupuncture group, large intestine sedating acupuncture group and control group. Radial pulse was measured by 3 dimensional pulse imaging system(DMP-3000) before, right after, 30 minutes after and 60 minutes after acupuncture at Cun, Guan, and Chi in the acupuncture group and in the control group at the same time points. Results : 1. Angle of main peak and magnitude of fourier component significantly changed by integrated analysis of Cun, Guan and Chi. 2. Amplitude of H1, Pulse Power volume / min, elasticity, AIx / HR, Frequency and magnitude of fourier component significantly changed by analysis of Cun, Guan, and Chi. Conclusions : The effect of Sa-Am acupuncture with large intestine tonifying and sedating in healthy human may be observed on time, amplitude, pulse area, augmentation index and fourier components parameters. The parameters analysed in this study may be used to differentiate the effects between Sa-Am acupuncture with large intestine tonifying and sedating on radial pulse. Further studies on the effects of Sa-Am acupuncture using radial pulse are needed.

  • To study on the assignment of the eight trigrams (八卦) in human body being based on based the number of HADO(河圖)-NAGSEO(洛書). Being based on the number of HADO(河圖)-NAGSEO(洛書) and the BEOK-HEE's diagram of eight trigrams order(伏羲次序圖), human being organ (five-Jang organs (liver, heart, spleen, lung and kidney), five organs (ear, eye, mouth and nose), provisional namely six entera for the process on intake, digestion, excretion of food (mouth, stomach, small intestine, large intestine, rectum, anus) are assigned eight trigrams (八卦). Eight trigrams(八卦) are attached to the based on the number HADO(河圖)-NAGSEO(洛書): 1-GEON(乾), 2-TAE(兌), 3-LEE(離), 4-JIN(震), 9-SON(巽), 6-GAM(坎), 7-GAN(艮), 8-GON(坤). And (八卦) are attached to the human being organs (five-Jang organs (live, heart, spleen, lung and kidney), five organs (ear, eye, mouth, nose, anus) : 1-GEON(乾)-left lung-left nose, 6-GAM(坎)-right kidney-right ear, 7-GAN(艮)-left heart-anus, 8-GON(坤)-right liver-right ear. Eight trigrams(八卦) are attached to the provisional namely six entera for the process on intake, digestion, excretion of food : the air(GEON 乾), the month(TAE 兌), the stomach(LEE 離), the small intestine(JIN 震), the large intestine(Son 巽), the rectum(GAM 坎), the anus(GAN 艮), the excrements(GON 坤). The BEOK-HEE's diagram of eight trigrams order(伏羲次序圖) means immutable order naturally. The process of the human being organs( five-Jang organs, five organs and six entera's digestion) is also unchangeable in region and program. Therefore we can set up the five-Jang organs, five organs and the process of six entera's digestion as the category of observation on the basis of BEOK-HEE's diagram of eight trigrams order(伏羲次序圖), and then we can arrange eight trigrams (八卦) on the five-Jang organs, five organs and the process of six entera's digestion.

Toxicity of the Puffer fish, Takifugu pardalis (Jolbok) and Takifugu niphobles (Bokseom) from Coastal Area of Korea (한국 연안산 졸복(Takifugu pardalis)과 복섬(Takifugu niphobles)의 독성)

  • The toxicity of two species of puffer fish, Takifugu pardalis and T. niphobles, collected from the coastal regions of Korea was determined using a mouse bioassay. In T. pardalis collected at Tongyeong, the proportion of toxic specimens containing ${\geq}10MU/g$ exceeded 90% for the skin, fins, liver, intestine, ovary, and gallbladder, 11.1% for the testis, and 6.9% for the muscle. In each of the organs, the highest toxin levels were several tens (14-39) of mouse units (MU) per gram in the muscle, testis, and eyeball, but thousands (1,444-5,755) of MU per gram in the skin, liver, intestine, ovary, and gallbladder. The organs of T. pardalis exhibited remarkable variation in toxicity. In T. niphobles, the proportion of toxic specimens exceeded 90% for the ovary and skin, 60-80% for the fins, liver, intestine, and gallbladder, and 4.5% for the muscle; no toxicity was detected in the testis or eyeball using the mouse bioassay. The highest toxin levels were thousands (2,291-7,777) of MU per gram in the liver, intestine, ovary, and gallbladder, hundreds(146-328) of MU per gram in the skin and fins, and 18 MU/g in the muscle. Takifugu niphobles toxicity also exhibited remarkable regional variation. The toxicity in the edible muscle of T. pardalis and T. niphobles was at acceptable levels for human consumption, while the toxicity of the skin of both species of puffer fish was very high, so that care must be taken when used for human consumption.

  • Simple spring-damper-mass models have been widely used to investigate whole-body vortical biodynamic response characteristics of the seated vehicle driver. Most previous models have not considered the effect of wobbling masses; i.e. heart, lungs, liver, intestine, etc. In this study, 4 -DOF seated driver model including one non-rigid mass representing wobbling visceral mass, 5-DOF model including intestine, and 10-DOF model including five lumbar vertebral masses were proposed. The model parameters were identified by a combinatorial optimization technique. simulated annealing method. The objective function was chosen as the sum of error between model response of seat-to-head transmissibility and driving point mechanical impedance and those of experimental data for subjects seated erect without backrest support. The model response showed a good agreement with the experimental response characteristics. Using a 10-DOF model, calculated resonance frequency of lumbar spine at 4Hz was matched well with experimental results of Panjabi et al.

  • The ability of microbial strains to confer health benefits to human and animal hosts is not a feature of many organisms. Lactic acid bacteria are the most commonly used bacteria applied as probiotics and there is now strong evidence that certain strains confer tangible benefits to the host. In terms of preventing infection, the ability of probiotic lactobacilli to colonize the tissue site, even temporarily, and inhibit growth and adhesion of pathogens, has been documented. Using molecular tools, such as RAPD and DGGE, probiotic organisms can be tracked through the intestine and in the vagina, and their impact on the flora assessed. Arguably, strains L. rhamnosus GG and GR-l are the most studied probiotic strains in terms of human application to the gut (GG and GR-l) and vagina (GR-1). Combined with L. fermentum RC-14, GR-l provides a two-pronged therapeutic for the intestine and urogenital tract. Care in manufacturing and distribution is essential to ensure that optimal doses of probiotics are accessible to consumers and patients.
