Change in composition of gut microbiota by exposure of natural medicines including Glycyrrhizae Radix in mice

  • Many of researches have revealed that human intestinal microbiota is related to health. Several diseases like obesity, diabetes, and hypertension are affected by the microbiota directly and indirectly. So, interventions with food and drug have been tried to change a composition of the microbiota to better condition. However, few natural medicines have elucidated to date. To understand an influence on microbiota by plant materials including Glycyrrhizae Radix, the extract of medicines were administered to mice and the feces were collected before and after the administration. The feces were analyzed by terminal restriction fragment length polymorphism (T-RFLP). The changes in composition of mice gut microbiota were detected and analyzed. The data could be utilized to further study about biological activities of the plant medicines.

Internal Amplification Control for a Cryptosporidium Diagnostic PCR: Construction and Clinical Evaluation

  • Various constituents in clinical specimens, particularly feces, can inhibit the PCR assay and lead to false-negative results. To ensure that negative results of a diagnostic PCR assay are true, it should be properly monitored by an inhibition control. In this study, a cloning vector harboring a modified target DNA sequence (${\approx}375bp$) was constructed to be used as a competitive internal amplification control (IAC) for a conventional PCR assay that detects ${\approx}550bp$ of the Cryptosporidium oocyst wall protein (COWP) gene sequence in human feces. Modification of the native PCR target was carried out using a new approach comprising inverse PCR and restriction digestion techniques. IAC was included in the assay, with the estimated optimum concentration of 1 fg per reaction, as duplex PCR. When applied on fecal samples spiked with variable oocysts counts, ${\approx}2$ oocysts were theoretically enough for detection. When applied on 25 Cryptosporidium-positive fecal samples of various infection intensities, both targets were clearly detected with minimal competition noticed in 2-3 samples. Importantly, both the analytical and the diagnostic sensitivities of the PCR assay were not altered with integration of IAC into the reactions. When tried on 180 randomly collected fecal samples, 159 were Cryptosporidium-negatives. Although the native target DNA was absent, the IAC amplicon was obviously detected on gel of all the Cryptosporidium-negative samples. These results imply that running of the diagnostic PCR, inspired with the previously developed DNA extraction protocol and the constructed IAC, represents a useful tool for Cryptosporidium detection in human feces.

A Membrane-Array Method to Detect Specific Human Intestinal Bacteria in Fecal Samples Using Reverse Transcriptase-PCR and Chemiluminescence

  • A membrane-based oligonucleotide array was used to detect predominant bacterial species in human fecal samples. Digoxygenin-labeled 16S rDNA probes were generated by PCR from DNA that had been extracted from fecal samples or slurries. These probes were hybridized to an array of 120 oligonucleotides with sequences specific for 40 different bacterial species commonly found in human feces, followed by color development using an alkaline phosphatase-conjugated antibody and NBT /BCIP. Twenty of the species were detected by this method, but E. coli, which was present at $\~$1 $\times 10$^5$ CFU per gram feces, was not detected. To improve the sensitivity of this assay, reverse transcriptase-PCR was used to generate probes from RNA extracted from fecal cultures. Coupled with a chemiluminescence detection method, this approach lowered the detection limit for E. coli from $\~1$ $\times 10$^6$ to ${\leq}$ 1 $\times 10$^5$ These results indicate that the membrane-array method with reverse transcriptase-PCR and chemiluminescence detection can simultaneously identify bacterial species present in fecal samples at cell concentrations as low as${\leq}$ 1 $\times 10$^5$ CFU per gram.

천연물중의 생리활성성분이 간해독기구에 미치는 영향 (Effects of Biologically Active Substances in Natural Products on the Hepatic Detoxication Mechanism)

  • Indolo[3,2-b]carbazole(ICZ) is a potent Ah receptor agonist with biological activities similar in several respects to those of the potent environmental toxin, TCDD. ICZ is produced during the oilgomerization of indole-3-carbinol(I3C), a breakdown product of the glucobrassicin present in food plants of the Brassica genus. In the present study we examined ICZ levels in tissues and excreta of rats treated with I3C or dietary cabbage of established glucobrasicin content, and in feces of conventional and germfree rats fed on a basal diet, and of humans. We also examined the levels of cytochrome P4501A1 induction, as determined by the ethoxyresorufin ο-deethylase assay, in tissues of animals that received cabbage-supplemented diets, or which were treated with purified I3C or ICZ. Our findings indicated that incorporation of either homogenized or whole freeze-dried cabbage in the feed led to large increases(16-60 fold) in the levels of ICZ in the feces and lower gastrointestinal tract of rats. We observed that whereas ICZ is readily detectable at about the same levels(2.00$\pm$0.50 ppb) in the feces of conventional rats fed on a purified diet and in human feces, levels of ICZ in the feces of germfree animals fed on the basal diet were at the limits of detection(0.40$\pm$0.20 ppb), indication that gut bacteria are important for the production of ICZ from essential dietary constituents in the basal diet. We showed that in contrast to the near 7000-fold difference in CYP1A1 inducing potencies of ICZ and TCDD in cells in culture, their inducing potencies differ by only about an order of magnitude in rats. Nonetheless, the levels of ICZ remaining in livers twenty hours after I3C treatment appear too low to account for the induced activity. This result indicates that ICZ may be rapidly cleared from the liver or that substances other than, or in addition to, ICZ be responsible for the enzyme-inducing activity of orally administered I3C or its precursors.

누에 및 뽕나무 유래 물질의 인간(人間) 암세포주(癌細胞株)에 대한 세포독성(細胞毒性) (Cytotoxic Activity of Bombyx mori and Morus alba Derived Materials against Human Tumor Cell Lines)

  • 동결건조(凍結乾燥)한 누에 유래(4령유충(齡幼蟲), 암 수 번데기, 암 수 성충(成蟲)) 및 건조 뽕나무 유래 재료(잎, 오디, 상백피(桑白皮))의 메탄을 추출물, 백강잠(白彊蠶) 및 누에 4령유충(齡幼蟲) 잠분(蠶糞)의 메탄올 추출물의 5종 인간(人間) 암세포주(癌細胞株)(A549 lung, SK-OV-2 ovarian, SK-MEL-2 melanoma, XF-498 CNS, HCT-15 colon tumor cell lines)에 대한 세포독성(細胞毒性)을 sulforhodamine B법을 이용하여 in vitro 검정하였다. 공시시료중 잠분(蠶糞)의 70% 메탄올 열탕추출물(熱湯抽出物)은 이들 암세포주(癌細胞株)에 대하여 강한 세포독성(細胞毒性)을 나타내었으나, 잠분(蠶糞)의 메탄올 추출물 및 오디와 상백피(桑白皮)의 메탄을 추출물은 중간 정도의 활성을 보였다. 기타 물질들은 이들 암세포주(癌細胞株)에 거의 독성을 보이지 않았다. 70% 메탄올 열탕추출물(熱湯抽出物)이 강한 세포독성(細胞毒性)을 나타내어, 용매 분획한 결과 클로로포름과 에틸아세테이트 획분이 암세포주(癌細胞株)에 대하여 가장 강한 세포독성(細胞毒性)을 보였다. 결론적으로, 잠분(蠶糞), 상백피(桑白皮) 및 오디의 항암활성(抗癌活性)은 이들의 약리작용의 일부를 설명할 수 있을 것으로 생각된다.

Four Human Cases of Diphyllobothrium latum Infection

  • Diphyllobothrium latum infections in 4 young Korean men detected from 2008 to 2012 are presented. Three were diagnosed based on spontaneously discharged strobila of the adult worm in their feces, and 1 case was diagnosed by finding the worm at colonoscopy examination in a local clinic. The morphologic characteristics of the gravid proglottid and eggs were consistent with D. latum. All patients were treated with praziquantel 15 mg/kg, and follow-up stool examinations were done at 2 months after the medication. The main clinical complaints were intermittent gastrointestinal troubles such as indigestion, abdominal distension, and spontaneous discharge of tapeworm's segments in their feces. The most probable source of infection was the flesh of salmon or trout according to a patient's past history. These are the 45th to 48th recorded cases diagnosed by the adult worm in the Republic of Korea since 1971.

Plant Settlement Patterns and Their Effects on Breeding Sites of Little Terns (Sterna albifrons) on Sand Bars on Ganwol Lake

  • We analyzed soil characteristics, soil seed banks, and plant communities in a small islet in Ganwol Lake from May 2005 to August 2006 to examine the forces driving plant settlement on sand bars and the effects of plant settlement patterns on nesting sites of little terns (Sterna albifrons). The soil nutrients contents in a site where the feces of wintering birds accumulate (N: 15.4 mg/kg, P: 10.5 mg/kg, LOI: 0.51 %, pH: 6.8) and a site where organic sediments accumulate (N: 20.7 mg/kg, P: 16.4 mg/kg, LOI: 0.40%, pH: 6.6) were much higher those of a control site which was not affected by bird feces and organic sediments (N: 4.1 mg/kg, P: 5.4 mg/kg, LOI: 0.41%, pH: 6.7). However, a seed bank was formed only on the site with accumulated organic sediments. Plant settlement was accelerated by feces from wintering birds and organic sediment accumulation on sand bars in Ganwol Lake. The percentage of area disturbed by human activities increased from 0.2% in May 2005 to 13.9% in August 2006, and the percentage of annual communities increased from 27.5% to 43.3%, but the percentage of open area decreased from 55.2% to 28.0% from May 2005 to August 2006. These increases in disturbed area and annual communities decreased the open area for breeding of little terns. The enlargement of P. communis and T. angustata communities was suppressed by irregular flooding. These results provide useful information for the management of little tern breeding sites for conservation purposes.

카드뮴 투여 흰쥐에서 키토산 섭취가 무기질 대사에 미치는 효과 (Effects of Chitosan on Mineral Metabolism in Rats Exposed to Cadmium)

  • This study investigated the effects of chitosan on mineral metabolism in rats exposed to cadmium by oral administration. Six week-old Sprague-Dawley rats were divided into eight groups. Four groups were fed AIN-93G based 3% ${\alpha}$-cellulose diets and the other four groups were fed 3% chitosan diets for four weeks with the oral administration of 0, 0.5, 1.0, 2.0 mg Cd/2ml distilled water three times per week, respectively. The essential mineral contents of serum, liver, kidney and bone (femur and lumbar), and the excretion of calcium in feces and urine were determined. There was no significant difference in weight gain and food intake among groups. The cadmium administration significantly decreased calcium in serum, iron in blood, calcium and iron in liver and iron contents in kidney. In contrast, calcium and zinc contents in kidney increased by the administration of cadmium. The weight, length and breaking forces of the femur and lumbar were not significantly different due to cadmium administration and chitosan among the groups. The fecal excretion of calcium was increased by the administration of cadmium. On the other hand, calcium absorption and the absorption rate were decreased by Cd administration. In the groups without Cd administered (N, N-Chi groups), chitosan significantly decreased the absorption rate of calcium by increasing the excretion in feces (p<0.05). These results suggest that cadmium administration may facilitate the decline of essential minerals in rats and also, chitosan may have a conflicting effect between cadmium and the essential minerals of tissues.

Arthrostoma miyazakiense (Nematoda: Ancylostomatidae) infection in raccoon dogs of Korea and experimental transmission to dogs

  • Arthrostoma miyazakiense (Nematoda: Ancylostomatidae) is a hookworm species reported from the small intestines of raccoon dogs (Nyctereutes procyonoides) in Japan. Five Korean raccoon dogs (N. procyonoides koreensis) caught from 2002 to 2005 in Jeollanam-do (Province), a southeastern area of South Korea, contained helminth eggs belonging to 4 genera (roundworm, hookworm, whipworm, and Capillaria spp.) and cysts of Giardia sp. in their feces. Necropsy findings of 1 raccoon dog revealed a large number of adult hookworms in the duodenum. These hookworms were identified as Arthrostoma miyazakiense based on the 10 articulated plates observed in the buccal capsule and the presence of right-sided prevulval papillae. Eggs of A. miyazakiense were $60-65{\times}35-40{\mu}m$ (av, $62.5{\times}35{\mu}m$), and were morphologically indistinguishable from those of Ancyiostoma caninum. The eggs were cultured to infective 2nd stage larvae via charcoal culture, and 100 infective larvae were used to experimentally infect each of 3 mixed-bred puppies. All puppies harbored hookworm eggs in their feces on the 12th day after infection. This is the first report thus far concerning A. miyazakiense infections in raccoon dogs in Korea, and the first such report outside of Japan.

한국에서의 농산물 및 환경시료에서 노로바이러스와 위생지표세균의 모니터링 (Monitoring of norovirus and indicator microorganisms from agricultural products and environmental samples in Korea)

  • 노로바이러스는 국내에서도 지속적으로 유행성 바이러스성 위장염을 일으키고 있다. 노로바이러스 저감화사업단(NOROTECL)의 1중 2세부과제에서는 농산물의 노로바이러스 오염 원인을 추적조사하여 노로바이러스 전파를 예방하고 저감화하기 위한 과제를 수행하였다. 2015년 1월부터 11월 사이에 노로바이러스와 관련이 있을 것으로 추정되는 80개 농산물, 80개 토양, 78개 인체분변, 3개 가축분변, 80개 농업용수, 80개 하천수 시료를 대상으로 노로바이러스, Male specific coliphage (MSC), 위생지표세균(Coliform, E. coli) 세 가지 검사를 통해 오염현황을 조사하였다. Semi-nested PCR과 DNA sequencing을 통해 18개의 Genogroup I과 3개의 Genogroup II 노로바이러스가 총 18개의 시료에서 발견되었다. Genogroup이 확정된 노로바이러스에 대하여 RT-PCR을 진행하였다. 대장균군과 대장균은 농산물에서 68%와 1%, 토양에서 88%와 7.5%, 인체분변에서 44%와 12.8%, 가축 분변에서 67%와 67%, 농업용수에서 74%와 30%, 하천수에서 96%와 51%의 검출율을 각각 나타냈다. MSC 결과에서는 14개의 시료가 양성으로 나타났다.
