Applying the basic knowledge about regulation of pigmentation towards development of strategies for cutaneous hypopigmentation

  • 인체피부의 색소침착에 있어서 다양한 차이는 주로 표피의 멜라닌생성 세포에 의한 멜라닌 합성 비율, 합성된 eumelanin과 pheomelanin의 상대적인 양과 melanocyte에서 keratinocyte로 melanosomes의 이동 속도와 그 방법에서 기인된다. 색소침착은 유전적, 환경적으로 조절되어지는 복합적 특성이다. 많은 관심이 집중되었던 하나의 유전자인 melanocortin 1 receptor 유전자가 있다. 인간집단에서 이 유전자의 다앙한 polymorphism은 색소침착의 다양성에 있어서 중요하다. 자외선(UV)에 대한 노출은 다양한 성장 요인, cytokines과 호르몬의 합성이 증가되고, 표피에서 그들 수용체들의 환경적응등이 나타난다. 색소침착 조절에 관한 정보는 과다색소 침착된 피부손상의 임상치료를 위한 전략들에서 이끌어 냈다. 주된 3개의 전략은 다음과 같다. 1) melanin 합성경로를 방해하는 화합물의 사용 2) eumelanin 합성을 조절하는 호르몬의 구조에서 기인된 peptides 또는 peptide-mimetics개발 3) melanocytes에서 keratinocytes로의 melanosome 이동을 감소시키는 물질의 개발. 이 모든 3가지의 전략은 각각 전체 멜라닌 합성, eumelanin 생성 또는 피부의 멜라닌 단위를 억제시킴으로 미백작용을 유도시킬 것으로 기대되어 진다.

  • Epidermal growth factor receptor (EGFR) is a member of the ErbB family (ErbB1-4) of receptor tyrosine kinases (RTKs). EGFR controls numerous physiological functions, including cell proliferation, migration, differentiation and survival. Importantly, aberrant signaling by EGFR has been linked to human cancers in which EGFR and its various ligands are frequently overexpressed or mutated. EGFR coordinates activation of multiple downstream factors and is subject of various regulatory processes as it mediates biology of the cell it resides in. Therefore, many studies have been devoted to understanding EGFR biology and targeting the protein for the goal of controlling tumor in clinical settings. Endocytic regulation of EGFR offers a promising area for targeting EGFR activity. Upon ligand binding, the activated receptor undergoes endocytosis and becomes degraded in lysosome, thereby terminating the signal. En route to lysosome, the receptor becomes engaged in activating various signaling pathways including PI-3K, MAPK and Src, and endocytosis may offer both spatial and temporal regulation of downstream target activation. Therefore, endocytosis is an important regulator of EGFR signaling, and increasing emphasis is being placed on endocytosis in terms of cancer treatment and understanding of the disease. In this review, EGFR signaling pathway and its intricate regulation by endocytosis will be discussed.

대장암 세포에서 EGFR 저해제 Nimotuzumab의 방사선 병합 효과 (Combination Effect of Nimotuzumab with Radiation in Colorectal Cancer Cells)

  • 목 적: 대장암 세포에서 epidermal growth factor receptor (EGFR) 저해제인 nimotuzumab에 의한 방사선 민감도 증진 효과를 살펴보고자 한다. 대상 및 방법: 총 4종류의 인간 유래 대장암 세포주인 HCT-8, LoVo, WiDr, HCT-116를 nimotuzumab과 방사선을 병합 처리한 후 세포증식, 생존율, 세포주기 진행에 미치는 영향을 MTT, clonogenic survival assay, flow cytometry와 western blot을 통해 분석하였다. 결 과: 대장암 세포주에서 nimotuzumab에 의해 EGFR 인산화가 억제됨을 확인하였고 이러한 조건에서 nimotuzumab이 HCT-116을 제외한 나머지 3종류의 대장암 세포주의 방사선 민감도를 증진시킴을 확인하였다. 반면에, nimotuzumab은 방사선 조사와 무관하게 대장암 세포의 증식이나 세포 주기에는 아무런 영향을 미치지 않았다. 결 론: Nimotuzumab은 EGFR에 의한 세포 생존 신호 전달을 억제함으로써 대장암 세포의 방사선에 대한 민감도를 증가시켰다. 본 연구는 대장암의 방사선 치료에 EGFR 특이적 저해제인 nimotuzumab의 임상 적용 근거를 제공하였다.

Histone Deacetylase Inhibitor Trichostatin A Enhances Antitumor Effects of Docetaxel or Erlotinib in A549 Cell Line

  • Background and Objective: Histone deacetylase (HDAC) inhibitors represent a promising class of potential anticancer agents for treatment of human malignancies. In this study, we investigated the effect of trichostatin A (TSA), one such HDAC inhibitor, in combination with docetaxel (TXT), a cytotoxic chemotherapy agent or erlotinib, a novel molecular target therapy drug, on lung cancer A549 cells. Methods: A549 cells were treated with TXT, erlotinib alone or in combination with TSA, respectively. Cell viability, apoptosis, and cell cycle distribution were evaluated using MTT (3- (4, 5-dimethylthiazol-2-yl) -2, 5-diphenyltetrazolium bromide) assay, Hochst33258 staining and flow cytometry. Moreover, immunofluorescent staining and Western blot analysis were employed to examine alterations of ${\alpha}$-tubulin, heat shock protein 90 (hsp90), epidermal growth factor receptor (EGFR), and caspase-3 in response to the different exogenous stimuli. Results: Compared with single-agent treatment, co-treatment of A549 cells with TSA/TXT or TSA/erlotinib synergistically inhibited cell proliferation, induced apoptosis, and caused cell cycle delay at the $G_2/M$ transition. Treatment with TSA/TXT or TSA/erlotinib led to a significant increase of cleaved caspase-3 expression, also resulting in elevated acetylation of ${\alpha}$-tubulin or hsp90 and decreased expression of EGFR, which was negatively associated with the level of acetylated hsp90. Conclusions: Synergistic anti-tumor effects are observed between TXT or erlotinib and TSA on lung cancer cells. Such combinations may provide a more effective strategy for treating human lung cancer.

Feasibility Study of Docetaxel and Cyclophosphamide Six- Cycle Therapy as Adjuvant Chemotherapy for Japanese Human Epidermal Growth Factor Receptor 2-Negative Breast Cancer Patients

  • Background: We compared treatment completion rates and safety of docetaxel and cyclophosphamide sixcycle therapy (TC6) with docetaxel followed by 5FU, epirubicin and cyclophosphamide (T-FEC) therapy in Japanese patients with human epidermal growth factor receptor 2 (HER2)-negative breast cancer. Materials and Methods: We administered TC6 q3w or T-FEC q3w to HER2-negative breast cancer patients. The primary endpoint of this trial was toxicity. As second endpoints, the treatment completion rate and relative dose intensity were evaluated. Results: The TC6 and T-FEC group consisted of 22 and 21 patients, respectively. Concerning hematological toxicity, grade 3 or higher adverse reactions included neutropenia and febrile neutropenia. As non-hematological adverse events, exanthema and peripheral neuropathy were frequently reported in the TC6 group, whereas more patients of the T-FEC group reported nausea and vomiting. In TC6, the treatment completion rate was 86.4% and the relative dose intensity of docetaxel was 93.2%. In T-FEC, the values were 95.2% and 98.9%, respectively. Conclusions: These results suggest that TC6 is tolerable in Japanese, and that this regimen can also be performed in outpatient clinics. However, with the TC6 regimen, the compliance was slightly lower than with the T-FEC regimen, and supportive therapy needs to be managed appropriately.

ERK1/2 activation by the C. elegans muscarinic acetylcholine receptor GAR-3 in cultured mammalian cells involves multiple signaling pathways

  • Extracellular signal-regulated kinases 1/2 (ERK1/2) play important roles in a variety of biological processes including cell growth and differentiation. We have previously reported that GAR-3 activates ERK1/2 via phospholipase C and protein kinase C, presumably through pertussis toxin (PTX)-insensitive Gq proteins, in Chinese hamster ovary (CHO) cells. Here we provide evidence that GAR-3 also activates ERK1/2 through PTX-sensitive G proteins, phosphatidylinositol 3-kinase (PI 3-kinase), and Src family kinases in CHO cells. We further show that in human embryonic kidney (HEK293) cells, epidermal growth factor receptor and Ras are required for efficient ERK1/2 activation by GAR-3. Taken together, our data indicate that GAR-3 evokes ERK1/2 activation through multiple signaling pathways in cultured mammalian cells.

게잡이 원숭이에서 Recombinant Human Erythropoietin의 4주간 투여 후 비장 유전자 발현 연구 (Effects of Recombinant Human Erythropoietin Treatment in Male Cynomolgus (Macaca fascicularis) Monkeys (II): Gene Expression Profiling in Spleen)

  • We investigated effects of recombinant human erythropoietin (rHuEPO) on profiles of mRNA transcripts in 6 male cynomolgus (M. fascicularis) monkey's spleen for 4 weeks. Six monkeys, composed of control and treatment group (Control : M1, M2, M3: Treatment : M4, M5, M6) were intravenously administered 3 times per week without or with a dose of rHuEPO 2730 IU/0.1 ml/kg. After 4 weeks rHuEPO treatment, spleen was removed for RNA isolation. Splenic gene expression was assessed using Affymetrix U133A 2.0 arrays containing 18,400 transcripts and variants, including 14,500 well-characterized human genes. Gene expression pattern was very different between individuals even in same treatment. In rHuEPO treated groups showed number of genes were up- or down-regulated (M4: 79: M5: 48; M6: 73 genes). Six genes (epidermal growth factor receptor, calgranulin A, estrogen receptor binding site associated antigen, matrix metalloproteinase 19, zinc finger and BTB domain containing 16, progestin and adipoQ receptor) were commonly expressed in rHuEPO treated group. The different individual response could be major considering factor in monkey experiment. Further study is needed to clarify the different individual response to rHuEPO in molecular level. This study will be valuable in the fundamental understanding and validation of molecular toxicology for bio-generic drugs including rHuEPO in cynomolgus monkey.

EGFR and HER2 Expression in Papillary Thyroid Carcinoma

  • Purpose: The epidermal growth factor receptor (EGFR) family plays a crucial role in the growth of malignant tumors. EGFR and human EGFR 2 (HER2) protein overexpression are associated with an unfavorable prognosis and are important therapeutic targets in breast cancer. The aim of this study was to evaluate the relationship between EGFR and HER2 expression and clinicopathological factors in papillary thyroid carcinoma (PTC) at a single institution. Methods: A total of 129 consecutive patients with PTC were enrolled in this study and underwent thyroid surgery between October 2013 and February 2015. EGFR and HER2 protein expression was evaluated in the 129 primary tumors by immunohistochemistry, and the results were compared with the clinicopathological features. Results: Of the 129 PTC tumors, 20 (15.5%) were HER2 positive, and 109 (84.5%) were HER2 negative. Moreover, EGFR positivity were observed in 111 (86%) tumors. The mean age of the patients was $46.3{\pm}11.9years$ (range, 20-74 years), and the mean tumor size was $1.08{\pm}0.75cm$ (range, 0.2-3.5 cm). Tumor size, extrathyroidal extension, histological subtype, and TNM stage were not significantly associated with EGFR or HER2 expression. Meanwhile, high Ki-67 labeling index was significantly associated with EGFR expression (P=0.002), HER2 expression was significantly associated with younger age (${\leq}45years$) and cervical lymph node metastasis. Conclusion: Based on our data, it is not clear whether EGFR and HER2 expression is associated with tumor aggressiveness in PTC.

HER-2/neu 단백질이 개 유방암에서의 발현분석 (HER-2/neu Protein Expression in Canine Mammary Adenocarcinoma)

  • 개에서의 유선 종양진단은 총 49 case 중에서 Human epidermal growth factor receptor (HER-2/neu, c-erbB-2), Epidermal growth factor receptor (EGFR), Activated leukocyte cell adhesion molecule (ALCAM) 등 면역조직화학적염색법을 실시하였다. 우선 49 case를 두 그룹으로 즉: 양성종양그룹 (22 case)과 악성종양그룹 (27 case)으로 구분하였다. 면역조직화학적염색법의 분석결과 HER-2/neu의 발현은 양성종양에서는 31.8% (7/22), 악성종양에서는 29.6% (8/27)의 발현율을 보였고, EGFR의 발현은 양성종양에서는 27.3% (6/22), 악성종양에서는 22.2% (6/27)의 발현율을 보였으며, ALCAM의 발현은 양성종양에서는 40.9% (9/22), 악성종양에서는 7.4% (2/27)의 발현율을 보였다. 결론적으로 개에서의 유선종양진단의 발현율은 사람에서 보고된 것($25%{\sim}30%$)과 비슷하게 나타났으며 임상진단분야에서 HER-2/neu항체로 개에서의 유선종양진단에서 유용한 평가수단으로 적용될 수 있으리라 사료된다.

Mechanistic insights into differential requirement of receptor dimerization for oncogenic activation of mutant EGFR and its clinical perspective

  • The epidermal growth factor receptor (EGFR), a member of the ErbB family (EGFR, ErbB2, ErbB3 and ErbB4), plays a crucial role in regulating various cellular responses such as proliferation, differentiation, and survival. As a result, aberrant activation of EGFR, mostly mediated through different classes of genomic alterations occurring within EGFR, is closely associated with the pathogenesis of numerous human cancers including lung adenocarcinoma, glioblastoma, and colorectal cancer. Thus, specific suppression of oncogenic activity of mutant EGFR with its targeted drugs has been routinely used in the clinic as a very effective anti-cancer strategy in treating a subset of tumors driven by such oncogenic EGFR mutants. However, the clinical efficacy of EGFR-targeted therapy does not last long due to several resistance mechanisms that emerge in the patients following the drug treatment. Thus, there is an urgent need for the development of novel therapeutic tactics specifically targeting mutant EGFR with the focus on the unique biological features of various mutant EGFR. Regarding this point, our review specifically emphasizes the recent findings about distinct requirements of receptor dimerization and autophosphorylation, which are critical steps for enzymatic activation of EGFR and signaling cascades, respectively, among wildtype and mutant EGFR and further discuss their clinical significance. In addition, the molecular mechanisms regulating EGFR dimerization and enzymatic activity by a key negative feedback inhibitor Mig6 as well as the clinical use for developing potential novel drugs targeting it are described in this review.
