Damage of Ibuprofen Suppository on Rectal Mucosa of Rats (이부프로펜 좌제의 흰쥐 직장점막 자극성 시험)

  • In the present study, quantitative and qualitative histology was used to assess the effects of ibuprofen suppositories with various treatments on the rectal mucosa of rats. Two suppositories were prepared with Witepsol W35 and compared with two commercial ibuprofen suppositories Reference I (Showa Pharm.ind., Tokyo, Japan), Reference II (P.Pharm., Seoul, Korea). Single and multiple dose(dosing interval 4 hr, n=4) studies were conducted. All suppositories significantly increased epithelial cell loss, but the extent of rectal irritation was variable. These studies showed that the incorporation of ibuprofen into the suppository bases increases the morphological change in rectal tissue both for the single and multiple administrations of suppositories, but which was significantly recovered within 24 hr although the interanimal variability in scores was very substantial. Multiple administration of ibuprofen suppositories caused significant damage to rectal mucosa, but it must be considered that these were under the severe condition, that is, interval of administration (4 hr) was three times shorter than normal interval of administration and dose was fifteen times larger than usual human dose. Aluminum oxide $(Al_2O_3)$, a dispersing agent, slightly increased the irritation of rectal mucosa in rats at 5 hr and 24 hr after multiple administration, but it was possible to ignore the difference of irritation in the data at 5hr and 24hr after single administration. Finally, it was concluded that Witepsol W35 and ibuprofen had a slight rectal mucosa-irritating effect on the usual human dose, and ibuprofen suppositories prepared with Witepsol W35 or Witepsol W35, $Al_2O_3$ showed almost similar extent of rectal irritation with commercial ibuprofen products.

Synergistic Interaction of Radiation with Pesticide on DNA Damage in Human Lymphocytes as Biological Information for Prevention of Environmental Disaster (환경재해 방지를 위한 생물정보로서의 사람 림프구 DNA 손상에 대한 방사선과 살충제의 상승작용)

  • Agricultural pesticides may cause certain biological risks since they are widely used to eradicate pests. Agricultural disasters may arise even from the possibility of their synergistic interaction with other harmful enviromnetal factors. The effect of pesticide on radiation-induced DNA damage in human blood lymphocytes was evaluated by the single cell gel electrophoresis (SCGE) assay. The lymphocytes, with or without pretreatment of the pesticide, were exposed to 0-2.0 Gy of $^60 CO$ gamma ray. Significantly increased tail moment, which was a marker of DNA strand breaks in SCGE assay, showed an excellent dose-response relationship. The present study confirms that the pesticide has the cytotoxic effect on lymphocytes and that it shows the synergistic interaction with radiation on DNA damage as well. The results may have a role of providing biological information necessary for the prevention of environmental disaster.

A DNA-Damage Response Gene Expression Analysis in MCF-7 followed by γ-Radiation (MCF-7 세포주의 γ선에 의한 DNA 손상 반응 유전자 발현 양상의 분석)

  • Cell response to genotoxic agents is complex and involves the participation of different classes of genes including cell cycle control, DNA repair and apoptosis. In this report, we presented a approach to characterize the cellular functions associated with the altered transcript profiles of MCF-7 exposed to low-dose in vitro gamma-irradiation. We used the method of human 2.4 k cDNA microarrays containing apoptosis, cell cycle, chromatin, repair, stress and chromosome genes to analyze the differential gene expression characterization that were displayed by radiation-exposed cell, human breast carcinoma MCF-7 cell line, such as 4 Gy 4 hr, 8 Gy 4 hr, and 8 Gy 12 hr. Among these genes, 66 were up-regulated and 49 were down-regulated. Specific genes were concomitantly induced in the results. Cyclin dependent kinase 4 (Cdk4) is induced for starting the cell cycle. This regulation is required for a DNA damage­induced G1 arrest. In addition to, an apoptotic pathways gene Bcl-w was concomitantly induced. Mismatch repair protein homologue-l (hMLH1), a necessary component of DNA mismatch protein repair (MMR), in G2-M cell cycle checkpoint arrest. The present study provides new information on the molecular mechanism underlying the cell response to genotoxic stress, with relevance to basic and clinical research.

  • The damage caused by natural and human disasters has been increasing year by year. While the damage from these disasters are unavoidable, reduction of damage level by proper establishment of control measures could be sufficiently achievable. Disaster prevention resources include resources introduced in the event of disaster, such as human, materials, equipments and facilities. Rapid yet proper inputs of these resources are the key factor to minimize any damages from the disaster. Currently, however, the standard and/or criteria for mobilization of the disaster prevention resources have not yet systematically established. Therefore, proper resource management as well as efficient resource input has not been in place. This research is an early stage construction of efficient mobilization resources, which had been irregularly loaded and applied before. Also this study has tried to provide a classification rule for efficient disaster prevention resource management and mobilization, and indeed, provide a foundation for efficient resource management system.

An Acoustic Analysis of Noise Environments during Mobile Device Usage

  • In contemporary modern society, people are constantly exposed to many kinds of noise, such as that from machinery, aircraft, construction sites, or road traffic. Noise is considered one of the most indispensable and influential parts of human life. This study investigates the acoustic characteristics of noise transfer from external sources to the human ear. For this study, we measured and analyzed various types of noise environments, installed monitoring speakers in a semi-anechoic room, and conducted intentional noise-filled experiments. In this environment, the size of the sounds generated by use of a portable device was also measured and the SNR (signal to noise ratio) calculated to study the influence of the noise. As sound is transmitted to the ear and the human body, it affects not only auditory damage but also other parts of the body. In this paper, we propose a proper SNR for noise emitted by portable IT equipment to prevent hearing loss when IT equipment is used.

Photoprotective Effects of Minerals from Korean Indigenous Ores on UVA-irradiated Human Dermal Fibroblast

  • The photoprotective effects of minerals from Korean indigenous ores, consisting mainly of sericite, on UVA-irradiated human dermal fibroblast (HDF) were examined. Zymographic analysis showed that the treatment of the minerals significantly reduced the UVA-enhanced MMP-1 activity and mRNA level. The minerals also showed strong inhibitory effect on MMP-2 activity and mRNA expression. Moreover, the minerals were better than polyphenol in reducing MMP-1 and MMP-2 expressions. Notably, the minerals significantly enhanced collagen biosynthesis in the HDF. Inhibition of the elastase activity and protection against the oxidatively damaged HDF cell were also found in the presence of the minerals. Taken together, the ore minerals may be used as the potent photo-protective and anti-skin-aging ingredients which can prevent skin cell damage by UVA.

Magnetic Impact Actuator for Robotic Endoscope (대장내시경을 위한 자기 충격 액츄에이터)

  • For robotic endoscope, some researchers suggest pneumatic actuators based on inchworm motion. But, the existing endoscopes are not seemed to be replaced completely because human intestine is very sensitive and susceptible to damage. We design and test a new locomotion of robotic endoscope able to maneuver safely in the human intestine. The actuating mechanism is composed of two solenoids at each side and a single permanent magnet. When the current direction is reversed, repulsive force and attractive at the opposition side propels permanent magnet. Impact force against robotic endoscope transfer momentum from moving magnet to endoscope capsule. The direction and moving speed of the actuator can be controlled by adjusting impact force. Modeling and simulation experiments are carried out to predict the performance of the actuator. Simulation experiments show that force profile of permanent magnet is the dominant factor for the characteristic of the actuator. The results of simulations are verified by comparing with the experimental results.

Lipopeptides Extract from Bacillus Amyloliquefaciens Induce Human Oral Squamous Cancer Cell Death

  • A lipopeptide extract of Bacillus amyloliquefaciens BACY1 (BLE) was found to induce cell death in human oral squamous cell carcinoma (OSCC) cell lines, SCC4 and SCC25, in this study. The results of MTT assay showed that BLE inhibited OSCC cell proliferation in a dose-dependent manner. BLE was also effective in increasing the sub-G1 phases. Furthermore, when membrane damage in SCC4 cells treated with BLE was monitored by LDH assay, release of LDH was significantly increased. The protein and mRNA levels of pro-apoptotic Bax, and caspase-3 were up-regulated by BLE. Taken together, these results suggest that BLE induces apoptosis and then inhibits the cell proliferation of human OSCC cells.

Effects of Eccentric Exercise on Torque-Angle Relationship of Human Tibialis anterior In-vivo (신장성 수축 운동에 의한 인체 하지 전경골근의 족배굴곡 토크-발목 각도 특성 변화)

  • The purpose of this study was to investigate how maximum-effort eccentric exercise over different contraction ranges affects the characteristics of torque-angle relationship of human ankle plantarflexor in-vivo. Subjects were randomly assigned in two groups. One group (n=6) performed 120 maximum-effort eccentric ankle dorsiflexion contractions at short muscle length (ankle range of motion from -5 to 15 deg) and the other group (n=6) at long (ankle range of motion from 10 to 30 deg) muscle length. Eccentric exercise decreased the maximum isometric ankle plantarflexion torque ${\sim}40%$. It was found that the optimum ankle joint angle changed from 7.5 deg to 11.1 deg and 10.1 deg, shifted toward the longer muscle length, regardless of the exercise range. The results of this study suggest that eccentric exercise alters the characteristics of torqueangle relationship of the muscle but there is no differential effect of the eccentric contraction range.

Hydrolysis of Tencel Fabrics by Cellulase Treatment (셀룰라아제 처리에 의한 텐셀직물의 가수분해)

  • Tencel fabrics were treated with cellulase after mechanical prefibrillation treatment. SEM analysis was carried out to study morphological change of the treated fabric. The cellulase-treated Tencel fabrics were evaluated for weight loss and tensile strength. X-ray diffraction method, moisture regain, and K/S value were used to elucidate crystalline structural changes occurred by cellulase treatment. Degree of polymerization and copper number of the cellulase-treated fabrics were also measured to estimate effect of hydrolysis. SEM analysis indicated that with treatment of prefibrillation and cellulase, fibrils were produced and damage occurred deep into the fiber. Increases in concentration and time of cellulase treatment increased weight loss and decreased tensile strength retention of the treated fabrics. As cellulase hydrolysis progressed, degree of crystallinity, moisture regain and K/S value were not much changed. (Korean J Human Ecology 2(1) : 142∼148, 1999)
