Genotoxicity of Capsaicin in Cultured Human Lymphocytes

  • The clastogenic activity of capsaicin, a major pungent and irritating constituent of hot chili pepper, was evaluated in cultured human lymphocytes. Capsaicin (125, 250, and 500 $\mu$M) caused cytogenetic damage as determined by increased frequency of chromosome/chromatid aberrations compared to the solvent control. The mitotic indices were also decreased in a concentration-related manner in capsaicin-treated cells. Moreover, capsaicin suppressed [$^3$]thymidine incorporation into lymphocytes. The clastogenicity and cytotoxicity of capsaicin towards human lymphocytes were evident without an external metabolic activation system. Taken together, these findings suggest that capsaicin is a genotoxic agent and may thus represent a potential health hazard in humans.

  • There have been 3E problems of energy, economy and environment since the earth has its history. Especially, as the industrial society is highly developing, human need in daily life has also changed drastically. With the introduction of 40 hour working week system, more households enjoy picnics on weekends. More gas accidents take place on Saturdays and on Sundays than any other days of week. Consequently, this study tries to find out the influence of flame caused by the explosion of butane canister on the adjacent combustibles and people by simulating relevant quantity of TNT. In addition, the damage estimation was conducted by using API regulations. If the scale of the radiation heat is known by calculating the distance of flame influence from the explosion site, the damage from the site can be easily estimated. And the accident damage was estimated by applying the influence on the adjacent structures and people into the PROBIT model. According to the pro bit analyze, the spot which is 50cm away from the flame has 97% of the damage probability by the first-degree burn, 8% of the damage probability by the second-degree burn and 4% of the death probability by the fire.

3D Terrain Model Application for Explosion Assessment

  • An increase in oil and gas plants caused by development of process industry have brought into the increase in use of flammable and toxic materials in the complex process under high temperature and pressure. There is always possibility of fire and explosion of dangerous chemicals, which exist as raw materials, intermediates, and finished goods whether used or stored in the industrial plants. Since there is the need of efforts on disaster damage reduction or mitigation process, we have been conducting a research to relate explosion model on the background of real 3D terrain model. By predicting the extent of damage caused by recent disasters, we will be able to improve efficiency of recovery and, sure, to take preventive measure and emergency counterplan in response to unprepared disaster. For disaster damage prediction, it is general to conduct quantitative risk assessment, using engineering model for environmentaldescription of the target area. There are different engineering models, according to type of disaster, to be used for industry disaster such as UVCE (Unconfined Vapor Cloud Explosion), BLEVE (Boiling Liquid Evaporation Vapor Explosion), Fireball and so on, among them.we estimate explosion damage through UVCE model which is used in the event of explosion of high frequency and severe damage. When flammable gas in a tank is released to the air, firing it brings about explosion, then we can assess the effect of explosion. As 3D terrain information data is utilized to predict and estimate the extent of damage for each human and material. 3D terrain data with synthetic environment (SEDRIS) gives us more accurate damage prediction for industrial disaster and this research will show appropriate prediction results.

Comparison of lymphocyte DNA damage levels and total antioxidant capacity in Korean and American diet

  • BACKGROUND/OBJECTIVE: This study aims to measure the in vitro antioxidant capacity of Korean diet (KD) with American diet (AD) as a control group and to examine the ex vivo DNA damage reduction effect on human lymphocytes. MATERIALS/METHODS: The KD applied in this study is the standard one-week meals for Koreans (2,000 kcal/day) suggested by 2010 Dietary Reference Intakes for Koreans. The AD, which is the control group, is a one-week menu (2,000 kcal/day) that consists of foods that Americans would commonly take in according to the National Health and Nutrition Examination Survey. The antioxidant capacity of each menu was measured by means of the total phenolic assay and 3 in vitro antioxidant activity assays (2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, trolox equivalent antioxidant capacity (TEAC), Oxygen radical absorbance capacity ($ORAC_{ROO{\cdot}}$)), while the extent of ex vivo lymphocyte DNA damage was measured by means of the comet assay. RESULTS: When measured by means of TEAC assay, the in vitro antioxidant capacity of the KD of the day was higher than that of the AD (P < 0.05) while there was no significant difference in total phenolic contents and DPPH and ORAC assays. The ex vivo lymphocyte DNA damage protective effect of the KD was significantly higher than that of the AD (P < 0.01). As for the one-week menu combining the menus for 7 days, the total phenolic assay (P < 0.05) and in vitro antioxidant capacity (P < 0.001, DPPH; P < 0.01, TEAC) of the KD menu were significantly higher than those of the AD menu. Likewise, the ex vivo DNA damage reduction rate of the Korean seven-day menu was significantly higher than that of the American menu (P < 0.01). CONCLUSION: This study demonstrates that the high antioxidant capacity and DNA damage protective effect of KD, which consists generally of various plant foods, are higher than those of typical AD.

Kidney Toxicity Induced by 13 Weeks Exposure to the Fruiting Body of Paecilomyces sinclairii in Rats

  • Paecilomyces sinclairiis (PS) is known as a functional food or human health supplement. However concerns have been raised about its kidney toxicity. This study was performed to investigate the kidney toxicity of PS by 13 week-oral administration to rats. Blood urea nitrogen (BUN), serum creatinine, and kidney damage biomarkers including beta-2-microglobulin (${\beta}2m$), glutathione S-transferase alpha (GST-${\alpha}$), kidney injury molecule 1 (KIM-1), tissue inhibitor of matrix metalloproteinase 1 (TIMP-1), vascular endothelial growth factor (VEGF), calbindin, clusterin, cystatin C, neutrophil gelatinase-associated lipocalin (NGAL) and osteopontin were measured during or after the treatment of PS. BUN, creatinine and kidney damage biomarkers in serum were not changed by PS. However, kidney cell karyomegaly and tubular hypertrophy were observed dose-dependently with higher severity in males. KIM-1, TIMP-1 and osteopontin in kidney and urine were increased dose dependently in male or at the highest dose in female rats. Increased urinary osteopontin by PS was not recovered at 2 weeks of post-exposure in both genders. Cystatin C in kidney was decreased at all treatment groups but inversely increased in urine. The changes in kidney damage biomarkers were more remarkable in male than female rats. These data indicate that the PS may provoke renal cell damage and glomerular filtration dysfunction in rats with histopathological lesions and change of kidney damage biomarkers in kidney or urine. Kidney and urinary KIM-1 and cystatin C were the most marked indicators, while kidney weight, BUN and creatinine and kidney damage biomarkers in serum were not influenced.

A Seasonal Risk Analysis and Damage Effects Assessment by Gas Leakage of Chemical Plant using 3D Scan and FLACS (3D 스캔과 FLACS를 활용한 화학플랜트 가스 누출의 계절별 위험성 및 피해영향 평가)

  • The process and facilities of modern chemical plants are becoming increasingly complex, there is possibility of potential risk. Internal chemicals generate stress concentration when operated due to turbulence, laminar flow, pressure, temperature, friction, etc. It causes cumulative fatigue damage, which can damage or rupture chemical facilities and devices. The statistics of chemical accidents found that the highest rate of occurrence was in summer, and in the last five years statistics on chemical accidents, leakage incidents make up a decent percentage of accidents. Chemical leaks can cause serious human damage and economic damage, including explosions and environmental pollution. In this study, based on the leak accident of chemical plant, the risk analysis, and damage effects assessment were estimated using a 3D scanner and FLACS. As a result, if chemicals leak in summer, the risk is higher than in other seasons, the seasonal safety management measures, and countermeasure were estimated.

The Analysis of the Risk of Vehicle Fires in Korea and the Effectiveness of Initial Fire Extinguishing (국내 자동차 관련 화재발생 위험성 분석 및 초기소화의 효과성 연구)

  • Purpose: In this study, the status and the risk of fire occurrence in domestic automobile were evaluated and the damage reduction effect by using the fire extinguishing mechanism was evaluated. Method: The analysis of the risk of the occurrence of Vehicle Fires in Korea and the damage of human and property by the use of fire extinguisher applied to fire extinguisher were investigated and analyzed. Results: The annual damage rate of the damage caused by the automobile fire is 4.74%, which shows an annual increase. As a result of the analysis of the personal injury caused by the passenger car in the car fire situation, the damage amount per 100 cases of the fire occurrence increased from 424.65 million won to 473.06 million won in the past 5 years (2012 ~ 2016). Conclusion: When fire extinguishers are used in the case of passenger Vehicle Fires, the average amount of property damage per fire occurrence is estimated to be about 307.5 million, emphasizing the importance and necessity of the initial fire fighting.

Development of Estimation Functions for Strong Winds Damage Based on Regional Characteristics : Focused on Jeolla area (지역특성 기반의 강풍피해 예측함수 개발 : 전라지역을 중심으로)

  • Abnormal weather conditions have lately been occurring frequently due to the rapid economic development and global warming. Natural disasters classified as storm and flood damages such as heavy rain, typhoon, strong wind, high seas and heavy snow arouse large-scale human and material damages. To minimize damages, it is important to estimate the scale of damage before disasters occur. This study is intended to develop a strong wind damage estimation function to prepare for strong wind damage among various storm and flood disasters. The developed function reflects weather factors and regional characteristics based on the strong wind damage history found in the Natural Disaster Yearbook. When the function is applied to a system that collects real-time weather information, it can estimate the scale of damage in a short time. In addition, this function can be used as the grounds for disaster control policies of the national and local governments to minimize damages from strong wind.

Expressive Methods of Uncanny Image in Contemporary Fashion (현대패션에 나타난 언캐니 이미지의 표현방법)

  • The purpose of this study is to identify the expression methods of uncanny images shown in contemporary fashion through the visual expression methods of plastic arts. The study results are as following. First, there is a concept-forming element of 'The return of Repressed', which is a psychological mechanism for generating uncanny sets to the component of 'repression' and 'return'. 'Repression' herein consisted of 'unconsciousness', 'the other', and 'anxiety', while the return was composed of 'trauma' and 'repetition compulsion'. Second, there are visual expression methods of the uncanny in contemporary art. The subject of 'repressed things' was expressed as grotesque and having a terrible look. 'Fragmented body' was analyzed as 'dismantling' by mutilation, 'combination' placing cut body parts in other places, and 'damage' suffering from an illness or accident. 'Fear of death' was shown as 'anatomy' reminiscent of death and 'iconography of death' meaning return of the dead. 'Post-human' was a surrealistic being such as a monster and mechanized human, and was analyzed as 'gene variation' and 'cyborg'. Third, the methods of visual expression of the uncanny reflected on contemporary fashion was analyzed as 'fragmented body', 'fear of death', and 'post-human'. 'Fragmented body' appeared as the object reminiscent of the fragmented body, 'dismantling and combination of the body', and 'damage of the body' through the distortion of the clothing construction principle. 'Fear of death' visualized the characteristic elements of the subject reminiscent of death as 'iconography of death' and 'symbolic object of death' through the motif. 'Post-human' was the description of the subject beyond the body's function and form, and was shown as 'gene variation' and 'cyborg'.
