헛개나무 열수추출물로부터 항산화 및 항미생물 활성을 갖는 3-methoxy-4-hydroxybenzoic acid 와 3-methyoxy-4-hydroxycinnamic acid의 분리 및 동정 (Isolation and Identification of 3-Methoxy-4-hydroxybenzoic acid and 3-Methoxy-4-hydroxycinnamic acid from Hot Water Extracts of Hovenia dulcis Thunb and Confirmation of Their Antioxidative and Antimicrobial Activity)

  • 헛개나무 어린 줄기와 잎의 열수추출물에서 얻어진 MeOH 가용 획분에 항미생물 및 항산화 활성을 보여, 이들 활성 본체를 규명하고자 활성물질을 solvent fractionation, silica gel adsorption column chromatography, Sephadex LH-20 column chromatography, ODS column chromatography로 순차정제하고, HPLC등에 의해 두 종의 활성물질을 단리하여 LC-MS, GC-MS 분석에 의해 vanillic acid와 ferulic acid로 각각 동정하였다. 두 활성물질은 그람 양성 세균 및 그람 음성 세균 그리고 효모 등의 미생물에 항균 활성을 보였다. 또한, vanillic acid와 ferulic acid의 항산화 활성은 vanillic acid보다 ferulic acid가 강한 활성을 나타내, DPPH 자유라디칼 소거능을 나타내는 농도는 ferulic acid가 $14\;{\mu}g/mL(SC_{50})$을, 그리고 vanillic acid가 $8\;{\mu}g/mL(SC_{10})$이었다.

기와층버섯 자실체의 메탄올 및 열수추출물의 항산화, 항염증 및 아세틸콜린에스테라제 저해 활성 (Antioxidant, anti-inflammatory and anti-acetylcholinesterase activities of fruiting bodies of Phellinus xeranticus)

  • 본 연구에서는 기와층버섯의 자실체에서 메탄올과 열수를 이용해 추출한 물질의 항산화와 항염증, 항아세틸콜린에스테라제(anti-acetylcholinesterase)의 효과를 탐색하였다. DPPH 라디칼 소거능, 환원력 및 철 이온 제거능 등을 이용해 항산화 효과를 측정한 결과 양성대조군으로 사용한 BHT나 토코페롤에 비해 낮았지만 다른 종류의 버섯에 비해 효과가 우수한 것을 확인하였다. 철 이온을 제거하는 항산화 실험에서 기와층버섯의 메탄올 추출물의 효과는 양성대조군인 BHT나 토코페롤에 비해 월등하게 높아서 기와층버섯 자실체의 추출물은 높은 항산화 효과를 지닌 것으로 나타났다. 기와층버섯의 염증저해 효과 실험에서는 배양 중인 RAW 264.7 대식세포에 자실체의 메탄올과 열수추출물을 각각 전 처리 한 후 염증매개 물질인 LPS를 투여하여 추출물의 NO 생성 저해효과를 조사하였다. 실험 결과, 처리한 추출물의 농도가 증가함에 따라 생성된 NO의 양이 현저하게 감소하는 경향을 나타내었다. 또한 기와층버섯의 추출물을 이용해 carrageenan에 의해 흰쥐 뒷발에 유도된 부종을 저해하는 실험에서는 투여한 추출물의 농도가 증가함에 따라 흰쥐의 뒷발에 유도된 부종의 용적도 농도 의존적으로 감소되었다. 따라서 기와층버섯 자실체에 함유된 항산화, 항염증 및 항아세틸콜린에스테라제 성분은 천연 항산화제, 소염제 및 알츠하이머병 치료에 이용이 가능할 것으로 사료되었다.

Chemical Properties and Physiological Activities of Synnemata of Beauveria bassiana

  • Chemical properties and physiological activities of the freeze-dried synnemata of Beauveria bassiana were examined. A proximate analysis showed that the synnemata consisted mainly of carbohydrate (49.86%), protein 11.36%), and a moisture content of 30.64%. It contained a low amount of crude ash (4.76%) and fat (3.38%). The carbohydrate was composed mainly of mannose (52.3%), galactose (31.5%), glucose (13.2%), and rhamnose (3%). Trace amounts of arabinose, xylose, and fructose were present. Major amino acids In the synnemata were glutamic acid, glycine, aspartic acid, arginine, threonine, alanine, valine, leucine, lysine, and aspartic acid with the amounts of 30.42, 25.22, 17.17, 15.12, 12.65, 15.23, 12.47, 11.47, 14.24, and 17.17 mg/g, respectively. Among extracts from the synnemata, the hot-water extract showed 67% of anticomplementary activity compared to that of the positive control, followed by ethyl acetate extract (17%) and methanol extract (15%). The hot-water extract also had anticoagulant activity with 55 sec of coagulating time and this fraction exhibited the most potent Intestinal immune system modulating activity. The methanol extract showed the highest inhibitory activity (25%) on the 12-O-tetradecanoyl phorbol-13-acetate-induced superoxide ($O_2^-$) generation, followed by hot-water extract (18%) and ethyl acetate extract (10%). The data in the present study indicate that the extract of Beauveria bassiana synnemata contains some healthful chemical ingredients and it could provide beneficial physiological activities. These features of the synnemata should be of interest to the food industry as well as other industrial fields.

  • Wood chips of oak(Quercus mongolica) and larch(Larix leptolepis) were exploded with the optimum condition. Main components of exploded wood were separated with hot hot water and methanol. The hemicelluloses were purified from hot water extracts and alditol complexs were prepared from purified hemicellulose. And also, cellulose nitrate was prepared from extractive residue and characterized. The results can be summarized as follows. 1. Amounts of carbohydrate(72~79%) in the crude hemicellulose of larch wood was more than those of oak wood(55~66%). 2. The crude hemicelluloses were mainly composed of oligosaccharides in oak wood but those in larch wood contained about 50% monosaccharides. 3. Decolorization of hemicellulose was successful with activated charcoal and ion-exchange resin treatment. The alditol yields were 56.3~82.9%. 4. The degree of substitution(D.S.) of cellulose nitrate was 1.95~2.87 and it showed a good acetone solubility.

자목(柘木)의 콜라겐유도 생쥐 관절염에 대한 억제효과 (The Inhibitory Effect of Cudraniae Lignum on the Collagen-induced Arthritis in Mice)

  • Objectives : The present study purposed to examine the effect of Cudraniae Lignum, which is the wooden part of Cudrania tricuspidata Bureau (Moraceae), on cytokine secretion from the joint cells of mice with arthritis induced by collagen and verify its efficacy against rheumatoid arthritis. Methods : We prepared the hot-water extract, ethanol extract and methanol extract of Cudraniae Lignum, and tested their effects on cytokine secretion from the joint cells of mice with arthritis induced by collagen. Results : 1. Cudraniae Lignum reduced the secretion of inflammatory cytokines such as IL-1$\beta$, INF-$\alpha$ and IL-6 from joint cells. 2. The effect for reducing the secretion of IL-1$\beta$, TNF-$\alpha$ and IL-6 was strong in order of methanol extract, ethanol extract and hot water extract. 3. Considering the effect on cytokine secretion, the effective element is soluble in organic solvent, and is not volatile. 4. The ethanol extract and methanol extract of Cudraniae Lignum lowered the survival rate of cells significantly, but still the survival rate was over 92%, suggesting the low toxicity of the extracts. Conclusions : Cudraniae Lignum is considered effective for rheumatoid arthritis, and the effective element is considered soluble in organic solvent.

Changes in Antioxidant Activity with Temperature and Time in Chrysanthemum indicum L. (Gamguk) Teas During Elution Processes in Hot Water

  • Determining the elution of water-soluble substances from herbal teas is an important factor in their efficient use in terms of taste, perfume, and content of health-related components. The antioxidant activity and content of catechins in commercial Chrysanthemum indicum (gamguk) teas were determined for optimum elution conditions. The water extract of gamguk teas did not differ significantly in yield compared to methanol extracts and showed stronger antioxidant activity. Catechin contents in gamguk teas were 8-18% of the extracts when individual peaks in high-performance liquid chromatography analysis were compared to standard catechin peaks. Gamguk teas exhibited faster release of antioxidants, and the antioxidant activity was positively correlated with the thermal treatments. Gukhwacha (GC) was the best tea for rapid release (30 sec) of antioxidants with the $50^{\circ}C$ treatment, whereas antioxidants in other teas were relatively slower released.

생감자의 열탕 추출물을 첨가한 감자 김치의 이화학적 특성 및 In Vivo에서의 항암 효과 (Physico-chemical Properties and In Vivo Anti-cancer Effects of Potato Kimchi Prepared by adding Hot Water Extracts of Potato)

  • In this study, potato kimchi was prepared by applying heat to raw potatoes, and then the physico-chemical properties and anti-cancer effects of the kimchi were analyzed. The texture results indicated the potato kimchi had very good hardness and springiness attributes. During th late storage period, total vitamin C content of the kimchi slowly increased. In addition, the potato kimchi had non-volatile organic acid changes that promoted early aging; however, after the complete aging period, it was comparatively similar to other types of kimchi. Using the methanol extracts of various kimchi samples, the potato kimchi(solid 100%) showed the highest anti-carcinogenic effects in terms of anti-tumor activity in tumor bearing Balb/c mice with sarcoma-180 cells. In addition, the effects of the methanol extracts on hepatic glutathione S-transferase content were $289.76\;{\mu}mol/mg$ protein/min, $250.97\;{\mu}mol/mg$ protein/min, $251.20\;{\mu}mol/mg$ protein/min, $219.53\;{\mu}mol/mg$ protein/min, $183.79\;{\mu}mol/mg$ protein/min, for control kimchi, mul kimchi, and two potato kimchis [(solid 100%) and(solid 60%+kimchi juice 40%)], respectively. The in vivo anti-cancer effects of the potato kimchi were investigated using AGS human gastric adenocarcionoma cells and HT-29 human colon adenocarcionoma cells. Overall, an MTT assay revealed that the methanol extract of the potato kimchi showed the highest anti-carcinogenic effects.

조릿대 잎차의 영양성분 분석 및 기능성 평가 (Analysis of Nutritional Components and Evaluation of Functional Activities of Sasa borealis Leaf Tea)

  • 조릿대 잎을 이용한 가공품 개발에 대한 기초자료로 활용하기 위하여 잎차의 영양성분 및 기능성을 평가하였다. 일반성분은 수분 5.68%, 조단백 16.38%, 조지방 4.68%, 가용성 무질소물 32.37%, 조섬유 32.36% 및 회분 8.53%로 각각 나타났다. 주요 무기성분으로는 K 2,133.83, Ca 1,144.09 및 P 543.00 mg/100 g이었으며, 아미노산중 proline이 1,275.26 mg/100 g으로 가장 높은 함량을 보였고, cystine이 71.49 mg/100 g으로 가장 낮은 함량을 보였다. 주요 지방산으로는 linoleic acid(50.52%), palmitic acid(18.52%) 및 oleic acid(14.16%)였으며, 관능검사 결과 $80^{\circ}C$에서 추출한 조릿대 잎차가 가장 높은 기호도를 보였다. 80% 메탄올 및 열수추출물의 총 페놀과 플라보노이드 함량을 측정한 결과 15.09, 7.69 mg/g과 12.03$\pm$0.67, 6.12$\pm$0.29 mg/g으로 각각 나타났다. DPPH와 $ABTS^+$ 라디칼 소거활성을 측정한 결과 80% 메탄올추출물 농도 1.25 mg/mL에서 각각 86.87%와 83.85%의 소거활성을 보였으며, 환원력 및 acetylcholinesterase(퇴행성뇌신경질환의 일종인 치매 유발 효소) 저해활성은 농도 의존적인 경향을 나타내었다.

Antioxidant and inflammation inhibitory effects from fruiting body extracts of Ganoderma applanatum

  • In this study, the antioxidant and anti-inflammatory effects of methanol extract (ME) and hot water extracts (HE) from the fruiting bodies of Ganoderma applanatum were investigated. The 1,1-diphenyl-2-picryl-hydrazy (DPPH) radical scavenging activity of 2.0 mg/mL ME (94.83%) was comparable to that of butylated hydroxytoluene (96.97%), the reference standard. The hydroxyl radical scavenging activities of ME and HE were similar to that of BHT at 2.0 mg/mL, whereas lipid peroxidation activity of the ME and HE were significantly lower than that of BHT. High-performance liquid chromatography analysis showed that the G. applanatum fruiting bodies contained nine phenolic compounds, which might contribute to antioxidant and anti-inflammatory activities. The survival rate of RAW 264.7 macrophages treated with 2.0 mg/mL ME and HE were 65.23 to 68.12% at 2.0 mg/mL, thereby indicating that the extracts were slightly cytotoxic at the concentration tested. The extracts also inhibited the nitric oxide (NO)-mediated expression of inducible nitric oxide synthase (iNOS) protein in lipopolysaccharide-induced RAW 264.7 macrophages and carrageenan-induced paw edema in rats. The study results demonstrated that the fruiting bodies of G. applanatum possessed good antioxidant and anti-inflammatory activities, which might be used to develop novel anti-inflammatory agents.
