Feline bocavirus (FBoV) is considered an emerging pathogen recently identified in domestic cats worldwide. To date, three species of FBoVs (FBoV-1, FBoV-2, and FBoV-3) have been reported, but there are no reports identifying FBoVs in Korea. In this study, we detected novel FBoVs for the first time in Korea in captive wild felids (four European lynx and a lion) kept at Seoul Zoo. In FBoV-positive fecal samples, not only singular infections but also dual or triple infections with three different species of FBoVs were confirmed, suggesting that three species of FBoVs are already introduced and co-circulated in susceptible host animals in Korea. These results will help expand our understanding of the geographical distribution and host susceptibility of novel FBoVs. Further studies are necessary to determine the infection status of FBoVs in domestic cats and the genetic characteristics of the viruses circulating in Korea.
The experiment was carried out to investigate Insecticide susceptibility of the spiraea aphid (Aphis citricola) in apple orchards In Korea, using dipping method. Although insecticIde susceptibility vaned with local and seasonal populations, the susceptibility to demeLon S-rnethyl and phosphamidons was different from that Lo the other insecticides. The LCso values were 10 ppm for deltamethrin and chloropyrifos, 103 ~ 629 ppm for demeton S-methy1, acephate, phosphamidon, monocrotophos and vamldothion, 12,200 ppm for EPN, and 1,745 ppm for pirimicarb. 'When Insecticide susceptibility was compared between the S-clone selected from a population on the host plant spiraea and the R-clone selected from a population on the apple tree, the resistance ratio as expressed by RIS ratio of $LC_{50}$ was 78 for phosphamidon and 546 for pirimicarb. Esterase zymogram determined by the agar gel electrophoresIs revealed a significant difference between the clones. The activity of the E2, E5, E6 and E7 of the R-clone was higher than that of the S-clone, It is suggesLed that the Increased esterase activity may be involved in the mechanism of insecticide resistance in the spiraea aphid, although the involvement of other factor(s) may not be ruled out.
To study the usefulness of Se301 cells, which is originated from Spodoptera exigua and has susceptibility to the Autographa californica NPV (AcNPV), as a host for the AcNPV-based expression vector system, we compared the characteristics of AcNPV in Se301 and Sf-21 cells. The symptom by viral infection was similar in both of cells, but the ratio of polyhedra released from the cell was higher in Se301 cells than in Sf-21 cells. The overall PIB productivity of AcNPV was similar in both cells but the size of polyhedra was larger in Se301 cells. While the polyhedrin expression efficiency was about 2.4 times higher in Se301 cells than in Sf-21 cells, the viral growth was higher in Sf-21 cells. These results suggested that Se301 cell is very useful in the AcNPV-based expression system as a host.
The studies were carried out to examine susceptibility or resistance of junipers as an inform ediate host of pear rust, to select effective fungicides to pear and juniper rust, and to determine their application time. The telia were formed abundanty on Juniperus chinensis var. kaizuka, moderately on J. chinensis, J. chinensis var. sargentii and J. virginiana, but little on J. chinensis var. globosa and J. utilis. The telia from the junipers were pathogenic to pear. Spraying juniper host with the fungicide Actidione in the 1st and End parts of April greaty inhibited swelling of telia as well as germination of teliospores. The two or three applications of Bayleton at the end of April to May 10 were remarkably effective for the control of pear rust. Fungicide sprays just before rain were more effective than those after rain.
Host cell death occurs during many, but not all, interactions between plants and the pathogens that infect them. This cell death can be associated with disease resistance or susceptibility, depending on the nature of the pathogen. The most well-known cell death response in plants is the hypersensitive response (HR) associated with a resistance response. HR is commonly regulated by direct or indirect interactions between avirulence proteins from pathogen and resistance proteins from plant and it can be the result of multiple signaling pathways. Ion fluxes and the generation of reactive oxygen species commonly precede cell death, but a direct involvement of the latter seems to vary with the plant-pathogen combination. Exciting advances have been made in the identification of cellular protective components and cell death suppressors that might operate in HR. In this review, recent progress in the mechanisms by which plant programmed cell death (PCD) occurs during disease resistance will be discussed.
Genome-wide association studies using large case-control samples and several hundred thousand genetic markers efficiently and powerfully assay common genetic variations. The application of these studies to inflammatory bowel disease has led to the identification of susceptibility genes and affirmed the importance of innate and adaptive immunity in the pathogenesis of disease. Efforts directed towards the identification of environmental factors have implicated commensal bacteria as determinants of dysregulated immunity and inflammatory bowel disease. Host genetic polymorphisms most likely interact with functional bacterial changes to stimulate aggressive immune responses that lead to chronic tissue injury.
Azoles are currently the most widely used class of antifungal drugs clinically, and are effective for treating fungal infections. Target site of azoles is ergosterol biosynthesis in fungal cell membrane, which is absent in the mammalian host. However, the development of resistance to azole treatments in the fungal pathogen has become a significant challenge. Here, we report the identification and functional characterization of a UPC2 homolog in the human pathogen Cryptococcus neoformans. UPC2 plays roles in ergosterol biosynthesis, which is also affected by the availability of iron in Saccharomyces cerevisiae and Candida albicans. C. neoformans mutants lacking UPC2 were constructed, and a number of phenotypic characteristics, including antifungal susceptibility and iron utilization, were analyzed. No differences were found between the mutant phenotypes and wild type, suggesting that the role of C. neoformans UPC2 homolog may be different from those in S. cerevisiae and C. albicans, and that the gene may have a yet unknown function.
Among 120 U. maydis strains isolated in Korea 14 different strains containing specific viral dsRNA segments were analyzed for the distribution of dsRNA and the production of toxin protein. Several distinctive dsRNA patterns were identified, 9 cases of P type with typical H, M and L ds RNA and one case of non-P-type, the frequency of a specific isolate was decreased with increasing number of dsRNA segments. The presence of dsRNA had no effect on the cultural or morphological phenotype of the host. Two isolates containing P type dsRNA segments appeared to produce toxin protein (killer strains) which inhibited the growth of 4 isolates (sensitive strain) with different susceptibility. Two killer strains contain unique M dsRNA segment which may code for toxin protein. However, the presence of toxin-sensitive strains among dsRNA-free isolates was similar to that of ds RNA containing strains.
Immunosuppressive drugs are used to make the body less likely to reject transplanted organs or to treat autoimmune diseases. In this study, five immunosuppressive drugs including two glucocorticoids (dexamethasone and prednisolone), one calcineurin inhibitor (cyclosporin A), one non-steroid anti-inflammatory drug (aspirin), and one antimetabolite (methotrexate) were tested for their effects on viral proliferation using feline foamy virus (FFV). The five drugs had different cytotoxic effects on the Crandell-Ress feline kidney (CRFK) cells, the natural host cell of FFV. Dexamethasone-pretreated CRFK cells were susceptible to FFV infection, but pretreatment with prednisolone, cyclosporin A, aspirin, and methotrexate showed obvious inhibitory effects on FFV proliferation, by reducing viral production to 29.8-83.8% of that of an untreated control. These results were supported by western blot, which detected viral Gag structural protein in the infected cell lysate. As our results showed a correlation between immunosuppressive drugs and susceptibility to viral infections, it is proposed that immune-compromised individuals who are using immune-suppressive drugs may be especially vulnerable to viral infection originated from pets.
A small number of viable tuberculosis bacilli can reside in an individual with latent tuberculosis infection (LTBI) without obvious clinical symptoms or abnormal chest radiographs. Diagnosis and treatment for LTBI are important for tuberculosis (TB) control in public and private health, especially in high-risk populations. The updated 2014 Korean guidelines for TB recommend that tuberculin skin tests, interferon-gamma release assays, or a combination of the two can be used for LTBI diagnosis according to age and immune status of the host as well as TB contact history. The regimens for LTBI treatment include isoniazid, rifampicin, or isoniazid/rifampicin. However, results of drug susceptibility test from the index case must be considered in selecting the appropriate drug for recent contacts. Standardized LTBI diagnosis and treatment based on the new 2014 guidelines will contribute to the effective TB control in Korea as well as to the establishment of updated guidelines.
