Common MCL1 polymorphisms associated with risk of tuberculosis

  • MCL1 expression has been found to be up-regulated during infection with virulent Mycobacterium tuberculosis. We investigated the genetic polymorphisms in MCL1 as potential candidate gene for a host genetic study of clinical TB infection. We have sequenced exons and their boundaries of MCL1, including the 1.5 kb promoter region, to identify polymorphisms, and eight polymorphisms were identified. The genetic associations of polymorphisms in MCL1 with clinical TB patients (n=486) and normal controls (n=370) were analyzed. Using statistical analyses, one common promoter polymorphism (MCL1-324C>A) which is absolutely linked with three other SNPs in the promoter and 3'UTR regions, were found to be significantly associated with increased risk of clinical TB disease. The frequency of the A-bearing genotype of -324C>A was higher in clinical TB patients than in normal controls (P=0.0008, OR=1.68). Our findings suggest that polymorphisms in MCL1 might be one of genetic factors for the risk of clinical tuberculosis development.

Efficacy of Korean Multipurpose Contact Lens Disinfecting Solutions against Acanthamoeba castellanii

  • Acanthamoeba keratitis has been increasing in recent years. Main risk factors are contact lens wear and their cleaning solutions. Most contact lens wearers use multipurpose disinfecting solutions (MPDS) for cleansing and disinfecting microorganisms because of its convenience. We determined amoebicidal effects of MPDS made in Korea and their cytotoxicity on human corneal epithelium cells. Fifteen commercial MPDS (A to O) were tested for their amoebicidal effects on Acanthamoeba castellanii trophozoites and cysts by using a most probable number (MPN) technique. Among them, 7 kinds of MPDS showed little or no amoebicidal effects for 24 hr exposure. Solutions A, B, G, H, L, and O showed positive amoebicidal effects, and solutions M and N killed almost all trophozoites and cysts after 24 hr exposure. However, 50%-N solution showed 56% cytotoxicity on human corneal epithelial cells within 4 hr exposure, and 50%-O solution also showed 62% cytotoxicity on human cells within 4 hr exposure. Solution A did not show any cytotoxicity on human cells. These results revealed that most MPDS made in Korea were ineffective to kill Acanthamoeba. The solutions having amoebicidal activity also showed high levels of cytotoxicity on human corneal epithelial cells. New formulations for improved MPDS that are amoebicidal but safe for host cells are needed to prevent Acanthamoeba keratitis.

  • Life table studies were conducted in the laboratory for the eulophid gregarious pupal parasitoid, Tetrastichus sp., on Hyphantria cunea Drury at a constant temperature of 28.2$\pm$2$^{\circ}C$, 50-60% RH to evaluate their impact on the host and their potential biological control. Development of immature stage took 20.1$\pm$2.7 d: adult females lived for 23.2$\pm$2.2 (range, 16-27)d and produced a mean of 53.6$\pm$26.6 adult progeny per female, with a sex ratio of 1: 9.5 (M:F). The intrinsic rate of natural increase (r$_{m}$) was 0.178/ female/day; the net reproductive rate (R$_{o}$), 46.74; the capacity for increase (r$_{c}$) 0.177; the finite rate of increase (λ) 1.19/female/day; thus each female contributed 46.74 individuals to the population in a mean generation time of 21.6 d. Biological factors in determining the life history trait variation of the parasitoid were discussed.d.d.d.

  • 곰팡이 종들은 유전체내에 대략 10% 정도의 다양한 전이인자와 함께 반복적인 염기서열을 갖는다. 이러한 전이인자들의 대부분은 유전체내에서 활발히 전이되며 곰팡이 병원균의 기주 범위와도 연관성을 갖으며 분포하는 것으로 알려져있다. 화본과 작물에 병을 일으키는 도열병에 분포하는 전이인자들은 활발히 전이하는 것으로 보이며, 특정 기주에 감염하는 개체군에 특이적으로 분포하는 경우가 많았다. 다수의 연구 보고에서도열병균의 전이인자가 비병원성 유전자의 기능을 상실하는데 작용하여, 이로인해 저항성 품종에 병을 일으켰다. 따라서, 도열병균의 전이인자들은 식물-곰팡이 사이의 상호 진화를 유도하는 원동력 중 하나일 수 있다. 본 총설에서는 도열병균에 존재하는 전이인자들의 종류와 생물학적인 기능에 관해 정리하였다.

소 모기매개 바이러스성 질병의 Vector 감염률 추정을 위한 표본추출 전략 (A Sampling Strategy for Estimating Infection Rate in Vector Mosquitoes of Mosquito-borne Bovine Viral Diseases)

  • Mosquitoes are the vectors of a number of viral diseases in cattle, such as Akabane disease, bovine ephemeral fever, Ainovirus infection, Chuzan virus infection, and Ibaraki disease. These diseases are transmitted from an infected animal to a non-infected host via the blood feeding of the vector. In Korea, the National Veterinary Research and Quarantine Services, Ministry for Food, Agriculture, Forestry and Fisheries is responsible for planning, implementation, laboratory investigations and reporting the results of the national surveillance program for mosquito-borne bovine diseases (MBD). The surveillance program, which was started in 1993, focused to determine the seroprevalence of each disease in cattle herds in space and time. From the epidemiological point of view, more important component of the surveillance program is to monitor infection rates in vectors for specific pathogens because this information is essential for a more precise understanding the dynamics of these diseases in a given environment and for determining risk of transmission. The aim of this study was to describe and compare methods for estimation of vector infection rates using maximum likelihood (MLE) and minimum infection rate in pooled samples. Factors affecting MLE such as number of pools, pooling size and diagnostic test performance are also discussed, assuming some hypothetical sampling scenarios for MBD.

위해성평가와 관리를 고려한 유전자변형작물 개발의 필요성 (Development of genetically modified crops based on considerations of risk assessment and management)

  • Over the last five years, we have conducted research on risk assessment of domestically developed genetically modified (GM) crops and found a number of factors which could delay risk assessment process. In this review, we described such cases and discussed the problem of transgene cassette integration, the lack of information on vectors, the poor quality control in seed production and absence of bioinformatic analysis on amino acid sequence homology before GM crop development. To solve these problems, we have suggested the introduction of the screening system of elite event before risk assessment process and quality control strategies for GM seed production. In addition, we suggested that the developers of GM crops should understand the importance of risk assessment and management for the commercialization of those crops and consider the biological and ecological characteristics of host plants. Consistent communications may need to be established between GM crop developers, risk assessors and risk managers at the initial stages of GM crop development to reduce trial-and-errors.

Isolation and Physiological Characterization of a New Algicidal Virus Infecting the Harmful Dinoflagellate Heterocapsa pygmaea

  • Dinoflagellates are considered one of the most abundant and diverse groups of marine microplankton and viruses are recognized as one of the significant factors affecting the plankton dynamics. Here, we report basic characteristics of a new dinoflagellate-infecting virus, Heterocapsa pygmaea DNA virus (HpygDNAV) which infects a toxic dinoflagellate, H. pygmaea. HpygDNAV is a polyhedral large virus (ca. 160-170 nm in diameter) propagating in its host's cytoplasm. Because of the virion size, appearance in thin sections, and propagation characteristics, HpygDNAV is assumed to harbor a large double-stranded DNA genome; i.e., HpygDNAV is most likely a nucleocytoplasmic large DNA virus (NCLDV) belonging to the family Phycodnaviridae. Its infectivity is strain-specific, rather than species-specific, as is the case for other algal viruses. The burst size and latent period are estimated to be roughly 100-250 infectious units $cell^{-1}$ and < 96 h, respectively.

IP계층에서의 VPN 전송성능에 관한 연구 (Secure VPN Performance in IP Layers)

  • 본 논문에서는 IPsec을 리눅스에서 구현하여 IP 계층에서 AH, ESP 프로토콜 사용시 노드간 성능을 측정하여 네트웍에서의 처리 성능에 영향을 미치는 인자에 대하여 분석을 하였다 IPsec에서 사용하는 AH와 ESP는 인증 데이터의 계산, 비교, 암호화에 의해서 IP프로토롤의 전송성능에 영향을 준다. 이에 AH, ESP 프로토콜에 대하여 응용 프로토롤(FTP, Telnet, SMTP)을 이용해 전송 데이터량을 증가시키며 Non IPsec과 IPsec의 처리성능을 평가하여 보았다. 성능평가 결과 전송패킷의 크기와 터널링에 사용되는 암호, 인증 함수. 호스트의 CPU속도, IPsec 구현방식이 전송성능에 영향을 주는 인자로 나타났으며, 대용량의 트래픽에서는 Non IPsec에 비하여 10 여배 이상의 전송지연이 발생하여 IPsec 전송에는 적합하지 않았다.

Epigenetic regulation of fungal development and pathogenesis in the rice blast fungus

  • Fungal pathogens have huge impact on health and economic wellbeing of human by causing life-threatening mycoses in immune-compromised patients or by destroying crop plants. A key determinant of fungal pathogenesis is their ability to undergo developmental change in response to host or environmental factors. Genetic pathways that regulate such morphological transitions and adaptation are therefore extensively studied during the last few decades. Given that epigenetic as well as genetic components play pivotal roles in development of plants and mammals, contribution of microbial epigenetic counterparts to this morphogenetic process is intriguing yet nearly unappreciated question to date. To bridge this gap in our knowledge, we set out to investigate histone modifications among epigenetic mechanisms that possibly regulate fungal adaptation and processes involved in pathogenesis of a model plant pathogenic fungus, Magnaporthe oryzae. For functional and comparative analysis of histone modifications, a web-based database (dbHiMo) was constructed first to archive and analyze histone modifying enzymes from eukaryotic species whose genome sequences are available. Based on the database entries, we carried out functional analysis of genes encoding histone modifying enzymes. Here I provide examples of such analyses that show how histone acetylation and methylation is implicated in regulating important aspects of fungal pathogenesis. Current analysis of histone modifying enzymes is followed by ChIP-seq and RNA-seq experiments to pinpoint the genes that are controlled by particular histone modifications. We anticipate that our work will provide not only the significant advances in our understanding of epigenetic mechanisms operating in microbial eukaryotes but also basis to expand our perspective on regulation of development in fungal pathogens.

Association of the Genetic Polymorphisms for CD247 Gene and Tuberculosis Case

  • Tuberculosis is airborne disease caused by Mycobacterium tuberculosis (MTB). Host genetic factors of these tuberculosis play an important role in determining individual difference in susceptibility or resistance to infectious diseases including tuberculosis. CD247 is named CD3zeta chain or CD3ζ. CD247 gene is a protein-coding gene involved in phagocytosis and signal transduction of the T cell receptor (TCR). Also, downregulation of the CD3ζ chain has been associated to chronic inflammation. The aim of this study was to research association of the genetic polymorphisms for CD247 gene and tuberculosis. We analyzed association of CD247 and Mycobacterium tuberculosis using 149 imputed single nucleotide polymorphisms (SNPs) with Korean population. And the results of this study show that seven SNPs of CD247 were identified to associate with tuberculosis. The most significant SNP was rs858545 (OR=1.22, CI: 1.05~1.42, P=0.009481). This study suggests that polymorphisms of CD247 may affect the T cell receptor signaling pathway, which may associate the infection of tuberculosis.
