Rapid detection of beer-spoilage lactic acid bacteria: Modified hop-gradient agar with ethanol method

  • Hop-resistant lactic acid bacteria (LAB) are well-known, major beer-spoilage bacteria. The hop-gradient agar containing ethanol (c-HGA+E) method has been used to examine hop-resistance of beer-spoilage LAB. However, the selection of beer-spoilage bacteria by the c-HGA+E method is either too selective or too inclusive. Furthermore, it is accompanied by a complicated experimental procedure, high-cost and time. To overcome these disadvantages, the modified hop-gradient agar with ethanol (m-HGA+E) method was developed. The most remarkable modifications were the shape of the petri dish and the inoculation method for bacteria. The efficiency and validation of the m-HGA+E approach were proven by the formation of colonies at different hop concentrations in the bottom layer, co-culture with the bacteriocin producer and by PCR detection of hop-resistant genes. This study demonstrated that m-HGA+E is a rapid, economical, and easy method to monitor potential hop-resistant beer-spoilage LAB during the beer brewing process.

  • Most of precedent studies related to hip hop culture and fashion, which have been conducted till now were progressed under only theoretical background of hiphop, not the changed status of hiphop currently. Practical studies explaining the status of hip hop in popular culture, especially, the area of popular fashion are insufficient. Therefore, this study was conducted by focusing on cultural characteristics and musical characteristics by the stream of period, for which concept hip hop is accepted to people currently and which effect it makes on the area of fashion. Especially, the study was progressed by dividing periodical background into before and after the 2000s because there was the most noticeable change for the genre of hip hop in that period. Because of the appearance of 'Alternative hip hop' which is a sub-genre of hip hop music, hip hop music was changed to be more popular. Also, hip hop fashion also showed big changes. Hip hop fashion before 2000s had showed poor conditions, resistance, complex and alienation of them by their fashion. On the other hand, hip hop fashion after 2000s also showed popular characteristics because of high-level environment and social status. This study intended to understand and grasp hip hop as a kind of cultural situation coexisted by many-sided characteristics as well as redefine the changed concept and contents of new hip hop currently.

3D Quantitative and Qualitative Structure-Activity Relationships of the δ -Opioid Receptor Antagonists

  • Antagonists of the d -opioid receptor are effective in overcoming resistance against analgesic drugs such as morphine. To identify novel antagonists of the d -opioid receptor that display high potency and low resistance, we performed 3D-QSAR analysis using chemical feature-based pharmacophore models. Chemical features for d -opioid receptor antagonists were generated using quantitative (Catalyst/HypoGen) and qualitative (Catalyst/HipHop) approaches. For HypoGen analysis, we collected 16 peptide and 16 non-peptide antagonists as the training set. The best-fit pharmacophore hypotheses of the two antagonist models comprised identical features, including a hydrophobic aromatic (HAR), a hydrophobic (HY), and a positive ionizable (PI) function. The training set of the HipHop model was constructed with three launched opioid drugs. The best hypothesis from HipHop included four features: an HAR, an HY, a hydrogen bond donor (HBD), and a PI function. Based on these results, we confirm that HY, HAR and PI features are essential for effective antagonism of the d -opioid receptor, and determine the appropriate pharmacophore to design such antagonists.

Hull-form optimization of KSUEZMAX to enhance resistance performance

  • This paper deploys optimization techniques to obtain the optimum hull form of KSUEZMAX at the conditions of full-load draft and design speed. The processes have been carried out using a RaPID-HOP program. The bow and the stern hull-forms are optimized separately without altering neither, and the resulting versions of the two are then combined. Objective functions are the minimum values of wave-making and viscous pressure resistance coefficients for the bow and stern. Parametric modification functions for the bow hull-form variation are SAC shape, section shape (U-V type, DLWL type), bulb shape (bulb height and size); and those for the stern are SAC and section shape (U-V type, DLWL type). WAVIS version 1.3 code is used for the potential and the viscous-flow solver. Prior to the optimization, a parametric study has been conducted to observe the effects of design parameters on the objective functions. SQP has been applied for the optimization algorithm. The model tests have been conducted at a towing tank to evaluate the resistance performance of the optimized hull-form. It has been noted that the optimized hull-form brings 2.4% and 6.8% reduction in total and residual resistance coefficients compared to those of the original hull-form. The propulsive efficiency increases by 2.0% and the delivered power is reduced 3.7%, whereas the propeller rotating speed increases slightly by 0.41 rpm.

A study on the meanings of soul fashion in American pop culture (미국 대중문화에 있어서 소울 패션(Soul Fashion)의 의미)

  • The purpose of this study was to analyze the meaning of soul fashion in American pop culture. This study was conducted using a literature research method based on the prior theses, journals and relevant books. Soul as a concept, originated in African-American communities and evolved from the ideology of Black Power, which prompted Black Nationalism. Soul fashion, which took on two styles in African American culture began to embody black resistance and community pride in the late 20th century. One of these, hip-hop style represented the message of resistance and a sense of beauty outside the mainstream. The other, African-inspired fashion, which utilized a look inspired by African tradition, rejected white supremacy by expressing a proud dignity. As a result, the meaning of "soul" in soul fashion represented by American pop culture resulted in contrasting appearance due to different elements. First, one of its meanings is ironic and sarcastic, and it expressed historical trauma, cultural stereotypes, self-hatred, and self-degradation and, the self-mutilation of African-American by cynically distorting their silhouettes and, using modified materials and patterns, fantastic colors, and extraordinary accessories. Second, the other meanings is the pride and dignity of Black Power, which visualized the concentration of ideas implied by the tradition of African-American, through soul fashion by using fierce traditional of African costumes, unique patterns and accessories.

  • Stainless steel fibers with a diameter of $17\;{\mu}m$ and 630 nm were produced from stainless steel wires by the drawing/annealing/exfolitation process. The suitable sheath material to draw the core stainless steel wires to fibers was the Cu coating. The low melting metal of Zn was not a suitable sheath coating. Also, an attempt was made to produce $20\;{\mu}m{\Phi}Ti$ fibers from the core titanium wires. The main obstacles in producing Ti fibers were their resistance to deformation owing to the Ti's hop structure, and high reactivity of Ti with the exfolitation solution.

Immunization of Mice with Recombinant Brucella abortus Organic Hydroperoxide Resistance (Ohr) Protein Protects Against a Virulent Brucella abortus 544 Infection

  • In this study, the Brucella abortus ohr gene coding for an organic hydroperoxide resistance protein (Ohr) was cloned into a maltose fusion protein expression system (pMAL), inserted into Escherichia coli, and purified, and its immunogenicity was evaluated by western blot analysis using Brucella-positive mouse sera. The purified recombinant Ohr (rOhr) was treated with adjuvant and injected intraperitoneally into BALB/c mice. A protective immune response analysis revealed that rOhr induced a significant increase in both the IgG1 and IgG2a titers, and IgG2a reached a higher level than IgG1 after the second and third immunizations. Additionally, immunization with rOhr induced high production of IFN-γ as well as proinflammatory cytokines such as TNF, MCP-1, IL-12p70, and IL-6, but a lesser amount of IL-10, suggesting that rOhr predominantly elicited a cell-mediated immune response. In addition, immunization with rOhr caused a significantly higher degree of protection against a virulent B. abortus infection compared with a positive control group consisting of mice immunized with maltose-binding protein. These findings showed that B. abortus rOhr was able to induce both humoral and cell-mediated immunity in mice, which suggested that this recombinant protein could be a potential vaccine candidate for animal brucellosis.

  • tRNA and 7SL RNA based antisense vehicles were prepared by inserting conserved anti-viral and anti-viroid domains. Anti-PVS coat protein leader sequence (ACPL) and antistructural antihairpin domain of PSTVd (AHII) were inserted in tRNA cassette; anti- zing finger domain of PVS, AHII and anti hop latent viroid ribozyme were inserted in 7SL RNA gene isolated from A. thaliana. These constructs were shown to be transcribed both, in in vitro and in in vivo conditions. However, it followed from our work that closely linked position of PoIII reference genes and PoIIII antisense genes within T-DNA lead to the impairment of RNA expression in transgenic plants. To assay in vivo transcription of antisense genes, hairy root potato cultures were established using h. tumefaciens A4-24 bearing both, Ri plasmid and PoIII-promoterless plant expression vectors with antisense RNA genes. Expression of antisense RNA in transgenic potato tissues was proven by specific RT-PCR reactions.

The Bactericidal Effect of High Temperature Is an Essential Resistance Mechanism of Chicken Macrophage against Brucella abortus Infection

  • Knowledge of avian host responses to brucellosis is critical to understanding how birds resist this infection; however, this mechanism is not well established. On the other hand, temperature has a major involvement in the physiology of living organisms, and cell death induced by heat is attributed to protein denaturation. This study demonstrates the direct bactericidal effect of a high temperature ($41^{\circ}C$) on Brucella abortus that resulted in the gradual reduction of intracellular bacteria and inhibited bacterial growth within avian macrophage HD11 in an increasing period of time. On the other hand, this study also revealed that high temperature does not affect the rate of bacterial uptake, as confirmed by the bacterial adherence assay. No significant difference was observed in the expression of target genes between infected and uninfected cells for both temperatures. This study suggests the susceptibility of B. abortus to bacterial death under a high temperature with an increased period of incubation, leading to suppression of bacterial growth.

Development of Multipoint Simultaneous Full-duplex Team Communication Module for SCBA (SCBA 면체용 다자간 동시 양방향 팀 통신모듈 개발)

  • This study presents the design and manufacture of a self-contained breathing apparatus SCBA wireless communication module with a multipoint simultaneous full-duplex communication system to enable communication between team members wearing the SCBA system. It is necessary for fire-fighters to wear the SCBA system during extinguishing and rescue work at the fire site. Evaluation of the team communication module confirmed the feasibility of communication over more than 500 m in the test condition based on the line of sight. By implementing the Ad-hoc function, it was confirmed that the communication distance could be extended to 128 m by automatic routing up to 3 hoc. The vertical distance inside the building for successful communication was up to the 5th floor in the open staircase and up to the 3rd floor in the partitioned staircase. Furthermore, the performance testing of the communication module assuming a fire situation, confirmed that five team members correctly recognized the standard abbreviation of fire and wireless communication without a separate PTT key operation. In addition, the flame resistance was verified by exposing the module to a flame at 950 ± 50 ℃ for 5 s and then immediately extinguishing the flame.
