Study on the Development of the Optimum Feeding Regimen for Pigs Weaned at 21 Days of Age

  • This experiment was conducted to investigate the effect of various feeding regimens on growth performance, nutrient digestibilities, nitrogen retention, blood urea nitrogen (BUN) concentration and feed cost in young pigs weaned at 21 days of age. One hundred twenty crossbred pigs (Landrace${\times}$Large White${\times}$Duroc, average 6.8 kg BW), weaned at 21 days of age, were allotted to 5 treatments in a 5 replicates by a randomized completely block (RCB) design. Treatments were as follow: 1) 1P (1-4 weeks: CP 23% and lysine 1.60%), 2) 2P-I (1 week: CP 23% and lysine 1.60%, 2-4 weeks: CP 21% and lysine 1.45%), 3) 2P-II (1-2 weeks: CP 23% and lysine 1.60%, 3-4 weeks: CP 21% and lysine 1.45%), 4) 2P-III (1-3 weeks: CP 23% and lysine 1.60%, 4 week: CP 21% and lysine 1.45%), 5) 3P (1 week: CP 23% and lysine 1.60%, 2-3 weeks: CP 21% and lysine 1.45%, 4 week: CP 19% and lysine 1.30%). Three different diets were formulated and supplied according to phase feeding programs. Diet 1 contains 23% crude protein and 1.60% lysine, diet 2 contains 21% crude protein and 1.45% lysine and diet 3 contains 19% crude protein and 1.30 lysine, respectively. Although there was no significant difference in growth performances, there was a beneficial effect of 3 phase feeding. The ADG was higher in 3P treatment than other treatments and it was observed clearly in late period (3-4 weeks) than in early period. Also, with increase in age, growth rate of pigs in 3P treatment was higher than that in 1P treatment approximately 37% (p=0.1379). There were no significant differences among all treatments in nutrient digestibility. The concentration of BUN was higher in pigs were fed diet containing 21% crude protein and 1.45% lysine (eg, 2P-1 and 3P) than those supplied diet containing high nutrient value at 2 week. The lowest feed cost/kg weight gain of pigs showed in 3P among treatments (p<0.05) whereas, high feed cost/kg weight gain of pigs was calculated in 1P and 2P-II treatments compared with 2P-I and 2 P-II (p<0.05), because of high milk products were used in those diet.

  • B. lactofermentum의 lysine 생합성에 있어서 DDH경로 및 ddh gene이 지닌 중요성을 조사하기 위하여, site-specific mutagenesis technique를 통하여 B. lactofermentum의 ddh gene을 disruption함으로서 DDH 경로를 차단시켰다. B. lactofermentum ddh mutant는 wild type 및 AEC내성 균주보다 성장이 매우 저조하였으며 lysine 생산량에서도 급격한 저하를 가져왔다. 이와 같이 B. lactofermentum이 DAP 경로만을 가졌을 때 세포의 성장 및 lysine 생산량에 있어서 극적인 저하를 가져왔기 때문에 B. lactofermentum에서의 DDH 경로는 meso-DAP 및 lysine 생합성에 있어 필수적인 경로로 작용한다는 것을 확인하였다. 그러므로 C. glutamicum과 B. lactofermentum과 같은 corynebacteria가 lysine을 많이 생산하는 것은 DDH 경로가 부가적으로 존재하기 때문이며, 이러한 DDH 경로는 metabolic flux가 증가되면 중간 대사물을 lysine으로 변화시키는 중요한 경로로 작용할 것이라 사료된다.

Optimal Lysine:DE Ratio for Growing Pigs of Different Sexes

  • This study was conducted to evaluate changes in the lysine to digestible energy (DE) ratio on performance, apparent ileal and fecal nutrient digestibilities as well as blood urea nitrogen (BUN), and to estimate optimal lysine:DE ratios for growing pigs of different sexes. A total of 150 pigs ($(Landrace{\times}Yorkshire){\times}Duroc$, 16.78 kg average body weight, 75 barrows and 75 gilts) was randomly allotted into a $2{\times}3$ (sex by diet) factorial design. Three diets were formulated to contain a crude protein level of 19%, a DE level of 3.5 Mcal/kg with three lysine:DE ratios of 3.2 (low), 3.5 (middle) and 3.8 (high) g lysine/Mcal DE per kg diet for both barrows and gilts throughout the study. With increasing dietary lysine:DE ratio, the average daily gain (ADG) of barrows decreased but there was no significant difference among treatments (p>0.05). However, ADG was significantly higher in gilts fed the diet containing the high lysine:DE ratio (p<0.05), followed by the middle and low lysine:DE ratio dietary groups. No significant effects of lysine:DE ratios on feed intake (ADFI) and feed conversion (F/G) were observed for barrows and gilts during overall period (p>0.05), while the optimal F/G was found in barrows fed diets of low and in gilts fed high lysine:DE ratio. Blood urea nitrogen had a positive relationship with growth rate. The results showed that the optimal lysine:DE ratios were 3.2 and 3.8 g lysine/Mcal DE per kg diet for barrows and gilts of 16 to 57 kg body weight, respectively.

  • Cadaverine (1,5-diaminopentane) is an important industrial chemical with a wide range of applications. Although there have been many efforts to produce cadaverine through fermentation, there are not many reports of the direct cadaverine production from lysine using biotransformation. Whole-cell reactions were examined using a recombinant Escherichia coli strain overexpressing the E. coli MG1655 cadA gene, and various parameters were investigated for the whole-cell bioconversion of lysine to cadaverine. A high concentration of lysine resulted in the synthesis of pyridoxal-5'-phosphate (PLP) and it was found to be a critical control factor for the biotransformation of lysine to cadaverine. When 0.025 mM PLP and 1.75 M lysine in 500 mM sodium acetate buffer (pH6) were used, consumption of 91% lysine and conversion of about 80% lysine to cadaverine were successfully achieved.

Performance and Carcass Composition of Broilers under Heat Stress : II. The Effects of Dietary Lysine

  • An experiment was conducted to determine the effect of lysine on performance and carcass composition of broilers under heat stress during the grower period (3-6 weeks). A factorial arrangement of three levels of dietary protein (18, 20, and 22%), three levels of dietary lysine (1.26, 1.39, and 1.52%), and two rearing temperature regimens were used in this study. Birds were kept under either moderate temperature ($24{\pm}1^{\circ}C/24h$) or hot cycling temperature ($26-34^{\circ}C/6h$, $34{\pm}1^{\circ}C/12h$, and $34-26^{\circ}C/6h$). Body weight (BW), weight gain (WG), feed intake (FI), feed conversion (FE), carcass weight (CW), carcass yield (CY), and percentages of breast meat (BM), abdominal fat (AF), drumsticks (DS), and thighs (TH) were determined at the end of experiment. Exposure to high ambient temperature significantly (p<0.05) decreased BW, WG, FI, FE, CW, BM, AF, and increased CY, DS, and TH. High dietary protein significantly (p<0.05) decreased AF and TH, and improved CW only under moderate temperature, resulting in significant (p<0.05) protein by temperature interaction. High dietary lysine significantly (p<0.05) decreased BW, WG, FI, CW, CY and AF, while BM was reduced only when high dietary protein was fed, resulting in significant (p<0.05) protein by lysine interaction. It is concluded that increasing dietary lysine adversely affected broilers' performance and carcass composition irrespective of rearing temperature.

Effects of Dietary Metabolizable Energy and Lysine on CarcassCharacteristics and Meat Quality in Arbor Acres Broilers

  • An experiment was performed to evaluate the effects of dietary metabolizable energy (ME) and lysine on carcass characteristics and meat quality in Arbor Acres (AA) broilers from 1 to 56 days of age. A total of 2,970 1-d-old male broiler chicks were randomly allocated to nine dietary treatments (three ME levels in combination with three lysine levels), and dietary ME and lysine concentrations were formulated by varying corn, soybean meal, tallow, and L-lysine sulfate concentrations. Live body weight (BW), carcass weight (CW), dressing percent, breast muscle weight (BMW), yield of breast muscle, muscle color (CIE L*, a*, and b*), pH values 45 min and 24 h postmortem ($pH_{45}$, and $pH_{24}$), meat shear force value (SFV), and water loss rate (WLR) were evaluated. Results showed that live body weight and dressing percent increased (p<0.05) as dietary energy increased. Higher dietary lysine content improved breast muscle weight. Neither carcass weight nor yield of breast muscle was affected by dietary energy or lysine content. Higher ME increased the b* value (p = 0.067) and $pH_{24}$ value (p<0.05), whereas it decreased SFV (p<0.05) and WLR (p = 0.06). Only water loss rate was influenced (p<0.01) by dietary lysine, which was higher in broilers from the high lysine diet as compared to those from medium or low lysine diets. The $pH_{45}$ value and L* value of breast muscle were not affected by ME or lysine. Significant interaction of dietary ME and lysine was found on a* value of breast muscle. These results indicated that dietary ME and lysine had important effects on breast muscle growth and meat quality, however their effects were different. Different concentrations of dietary ME and lysine might be considered to improve meat quality.

Corynebacterium glutamicum 균주 개량 및 발효 공정 최적화에 의한 L-lysine 생산성 증진 (Enhancement of L-lysine Productivity by Strain Improvement and Optimization of Fermentation Conditions in Corynebacterium glutamicum)

  • 본 연구에서는 Corynebacterium glutamicum I 균주에 salt tolerance를 도입하여 L-lysine 생산량을 증가시키고자 하였다. I 균주를 이용하여 mutagenesis를 수행한 후 모균주가 생장하는 못하는 9%의 NaCl이 포함된 배지에서 빠르게 생장하는 C14-49-3-15-7-3-20 균주를 선별하였다. flask 배양으로 L-lysine 생산을 조사한 결과 모균주 I의 경우 L-lysine 농도가 53.3 g/L, 수율이 51.6%인 반면 변이주 C14-49-3-15-7-3-20의 경우에는 L-lysine 농도 61.2 g/L, 수율 61.0%로 나타났다. 그리고 5 L 발효조에서 유가식 배양법으로 배양하여 L-lysine 생산량을 조사하였다. 그 결과 모균주는 113.0 g/L의 L-lysine을 생산하였고 수율은 41.8%이었다. 하지만 변이주의 경우에는 $33^{\circ}C$로 유지하여 배양한 후 PCV가 7.5%가 되는 시점에 배양 온도를 $35^{\circ}C$로 올려주고 배양하였을 때 L-lysine 생산량이 130.6 g/L, 수율이 48.6%로 모균주보다 많은 양의 L-lysine을 생산하였다. L-lysine 생산과 균주의 생장에 대한 osmotic pressure의 영향을 조사하기 위해 변이주 C14-49-3-15-7-3-20을 고농도의 NaCl과 당이 포함되어 있는 배지에 각각 배양하여 균체 생장 및 L-lysine 생산량을 조사하였다. 그 결과 모균주는 균체 생장이 느리고 생산량도 낮은 반면 변이주 C14-49-3-15-7-3-20의 경우에는 균체 생장 정도가 높고 생산량도 모균주보다 높았다. 그리고 2%의 NaCl이 포함되어 있는 배지에 osmoprotectant 를 첨가하였을 경우 모균주는 균체 생장 및 L-lysine 생산량이 높아졌다. 하지만 C14-49-3-15-7-3-20 균주의 경우에는 proline의 영향을 받지 않았다. 이러한 결과로 Corynebacterium glutamicum 균주에 salt tolerance를 도입하면 L-lysine 생산성을 크게 향상시킬 수 있음을 확인하였다.

Brevibacterium lactofermentum에서 ddh 유전자의 증폭에 의한 L-Lysine의 생산 (L-Lysine Production by Amplification of the ddh Gene in a Lysine-producing Brevibacterium lactofementum.)

  • 본 연구는 B. lactofermentum에서 ddh유전자의 증폭이 lysine 생성량에 미치는 영향을 조사하기 위하여, 먼저 E. coli-B. lactofermentum shuttle vector pEB1 및 pEB2를 제조하였으며, 제조된 shuttle vector에 ddh 유전자를 ligation하여 재조합 plasmid pRK1 및 pRK2를 구축하였다. B. lactofermentum에서 ddh 유전자를 증폭시키기 위하여 재조합 plasmld를 B. lactofermentum으로 도입하여 DDH 활성을 측정한 결과 대조균보다 7배 정도 증가하였다. 또한 lysine 생성량의 비교 분석에서는 재조합 plasmid를 함유한 균주의 경우 48시간 이후부터 control 균주보다 lysine 생성량이 증가하기 시작하여 72시간 때에는 최대치를 나타내었으며 그 이후는 오히려 감소하였다. 최대치를 나타낸 72시간 때의 lysine 생성량은 대조균주가 4.30∼4.38 g/l를 나타내었으며 pRK1 및 pRK2를 함유한 균주는 각각 5.34 g/l 및 5.07 g/l이었다. 이상의 결과로 미루어 볼 때 ddh유전자의 증폭에 의한 B. lactofermentum의 lysine 생성량은 대조균주에 비하여 18∼20% 정도 증가하였다.

  • $_L-Lysine$ 발효산업에 이용되고 있는 B. lactofermentum의 L-lysine 생합성은 succinylase 경로와 dehydrogenase 경로를 통하여 일어난다. 특히 lysine 생산 균주에 부가적으로 존재하는 dehydrogenase 경로는 lysine 생합성에 있어서 필수적인 경로로 작용하며 이때 meso-DAP-dehydrogenase (DDH)를 암호화하는 ddh gene이 관여한다. 그러므로 B. lactofermentum의 lysine 발효에 있어서 ddh gene의 over expression에 의한 lysine 생성량을 비교 조사하기 위하여, shuttle vector pEB1 및 pJC1으로 ddh gene을 삽입하여 재조합 plasmid pRK1 및 pRK31을 구축하였고 이를 B. lactofermentum으로 도입시켜 DDH 활성을 측정한 결과 pRK1을 함유한 균주는 대조균주보다 7배 정도, pRK31을 함유한 균주는 14배 정도 증가하였다. 또한 Shuttle vector를 함유한 대조균주와 재조합 plasmid를 함유한 균주간의 성장 비교에서는 서로 비슷한 수준을 나타내었으며 플라스크 배양에서 lysine 생성량의 비교 분석에서는 재조합 plasmid를 함유한 균주의 경우 48시간 이후부터 대조균주보다 lysine 생성량이 증가하기 시작하여 72시간때에는 최대치를 나타내었으며 그 이후는 오히려 감소하였다. 최대치를 나타낸 72시간 때의 lysine 생성량은 대조균주가 4.38g/L를 나타내었으며 pRK1 및 pRK31을 함유한 균주는 각각 5.34g/L 및 5.21 g/L이었다. 이상의 결과로 미루어 볼 때 B. lactofermentum내에서 ddh gene의 증폭에 의한 lysine 생성량은 pRK1 및 pRK31에서 대조균주보다 각각 20% 및 19% 증가하였다. 또한 발효조 배양에서의 lysine 생성량도 재조합 균주가 대조균주보다 23% 정도 증가를 나타내어 B. lactofermentum에서 ddh gene의 증폭으로 lysine 생성량이 증가하였음을 확인하였다.

Elk종의 녹용과 부산물의 이화학적 특성에 관한 연구 (A Study on Physico-chemical Property and Composition of Velvet Antler and Byproducts in Elk.)

  • 녹용 및 녹혈, 녹미, 녹신, 녹건 등의 유리 아미노산 함량을 보면 녹용에서는 glutamic acid, lysine, taurine, 녹혈에는 histidine, glycine, lysine, 녹미에는 glutamic acid, lysine, alanine, 녹신에는 histidine, glutamic acid, taurine, 녹건에는 taurine, alanine, histidine등이 많이 함유되었다 무기물의 함량을 보면 녹용은 $K^{+}$, $Ca^{2+}$, $P^{+}$가 각각 888.4, l1533.1, 147.2 (mg/100g)였고 F $e^{3+}$ , $K^{+}$, $P^{+}$가 각각 344.1, 1023.6, 157.2 (mg/100g). 녹미에는 $K^{+}$, $Mg^{2+}$, $P^{+}$가 각각 1420.6, 118.4, 1105.2 (mg/100g), 녹신에는 $K^{+}$, $Ca^{2+}$, $P^{+}$ 이 각각 294.4, 330.5, 514.3 (mg/100g)였으며 녹건에는 $K^{+}$, $Ca^{2+}$, $P^{+}$가 각각 444.84, 166.61, 242.87 (mg/100g)이였다. 유리 양이온 함량을 보면 $Na^{+}$$K^{+}$는 사슴의 혈액에서 가장 많았고 $Mg^{2+}$는 꼬리에서 그리고 $Ca^{+}$은 녹용에서 가장 많았으며 비교적 다른 부산물 보다 혈액에서 양이온 함량이 많았다.
