A Study of the Phytoliths Analysis of Ga-dong Site in Gijang, Busan (가동 유적지의 식물규소체(Phytoliths) 분석 연구)

  • In order to understand the local vegetation history and the origin of rice cultivation, a phytoliths analysis was performed with sediments collected from archaeological site at Ga-dong in Gijang, Busan. From the analysis with the sediments of three sampling sites, fan-shape phytoliths of Oryza sativa were detected: sediment 8 ~ sediment 29 of sampling site 1, sediment 6 ~ sediment 20 of sampling site 2 and sediment 7 ~ sediment 33 of sampling site 3. On the bottom layers or near the bottom layers from the three sampling sites, high densities of Arundinoideae and Andropogoneae were detected. It indicates that these layers were wet soil environment when they were sedimented. From near bottoms up to surfaces, the densities of Arundinoideae and Andropogoneae were abruptly decreased, while the density of Oryza sativa was sharply increased. With these results, it should be considered that the rice cultivation was started from the Bronze Age on this region.

Aggregation of Human Eyelid Adipose-derived Stem Cells by Human Body Fluids

  • Fetal bovine serum (FBS) is the most frequently used serum for the cultivation of mammalian cells. However, since animal-derived materials might not be appropriate due to safety issues, allogeneic human serum (HS) has been used to replace FBS, particularly for the culture of human cells. While there has been a debate about the advantages of HS, its precise effect on human adult stem cells have not been clarified. The present study aimed to investigate the effect of HS on the human eyelid adipose stem cells (HEACs) in vitro. When HEACs were cultivated in a medium containing 10% HS, many cells moved into several spots and aggregated there. The phenomenon was observed as early as 9 days following 10% HS treatment, and 12 days following 5% HS plus 5% FBS treatment. However, the aggregation was never observed when the same cells were cultivated with 10% FBS or bovine serum albumin. To examine whether cell density might affect the aggregation, cells were seeded with different densities on 12-well dish. Until the beginning of aggregation, cells seeded at low densities exhibited the longest culture period of 16 days whereas cells seeded at high densities showed the shortest period of 9 days to form aggregation. The number of cells was $15.1{\pm}0.2{\times}10^4$ as the least for the low density group, and $29.3{\pm}2.8{\times}10^4$ as the greatest for the high density group. When human cord blood serum or normal bovine serum was examined for the same effect on HEACs, interestingly, cord blood serum induced the aggregation of cells whereas bovine serum treatment has never induced. When cells were cultivated with 10% HS for 9 days, they were obtained and analyzed by RT-PCR. Compared to FBS-cultivated HEACs, HS-cultivated HEACs did not express VIM, and less expressed GATA4, PALLD. On the other hand, HS-cultivated HEACs expressed MAP2 more than FBS-cultivated HEACs. In conclusion, human adult stem cells could move and form aggregates by the treatment with human body fluids.

The Yield and Quality of Rice for Early Transplanting Cultivation by Cultural Practices in Gyeongnam Plain Area (경남 평야지 벼 조기재배를 위한 재배방법에 따른 쌀 수량 및 품질특성)

  • This study was conducted to investigate the optimum transplanting method for increasing the rice yield and quality by early transplanting cultivation in Gyeongnam plain area. This examination used of the transplanting method of planting distance($30{\times}14$ and $30{\times}18cm$) with number of seedlings(3, 5 and 7 plants). In high planting density($30{\times}14cm$), the number of tillers was increased while plant height was decreased as the planting number increases. On the other hand, the plant height and number of tillers were increased as planting number increases in low planting density($30{\times}18cm$). However, the number of grain per panicle was increased while the number of panicle was decreased as the planting number decreases. The number of panicle and the number of grain per panicle was increased as the planting distance increase. The yield of rice, when transplanted in the distance of $30{\times}14cm$ with 3 plants, was higher than the other treatment. Head rice ratio and head rice yield was the highest when transplanted in the distance of $30{\times}18cm$ with 3 plants(403 kg/10 a). The protein content was decreased and Toyo taste value was improved when transplanted in the distance of $30{\times}18cm$. Taken together, the best transplanting method for early transplanting cultivation was low planting density($30{\times}18cm$) with 3 plants in Gyeongnam plain area.

Variation of Medicinal Plants Species Richness along Vertical Gradient in Makawanpur District, Nepal

  • The research attempted to analyze the medicinal plant species richness in the vertical gradient from lower to the highest elevation of Makawanpur, focusing on the relationship between species richness and elevation which is very important for conservation and management of species diversity. Inventory was carried out in the study area by taking sampling intensity of 0.5% in the effective area. Altogether, 42 sample plots were laid in the field with the help of GIS software maintaining 50 m altitude difference. High species diversity was found in the herbs species whereas shrubs have comparatively low species diversity. The maximum species richness is found in herbs and poles whereas shrubs and trees have relatively low species richness. Research showed that species richness of medicinal plants increased with altitudinal gradient. While analyzing the species richness from 350 to 2,550 m (msl), the highest species richness was received with the elevation ranges from 1,800 m to 2,300 m. There was a positive relationship between species richness and altitudinal gradient in the study area. In addition, we have recorded the high value medicinal plants after 1,800 m altitude and rarely within 1,000 m. Medicinal plants correlated both positive and negative relationships with the increased altitude. The altitudinal response has positively seen except density (n/ha) of Shrubs. Domestication and cultivation of high value medicinal plants should be promoted in community forest including private lands. Training, workshops and awareness programs should be conducted to make people aware about medicinal plants resource utilization, conservation and commercialization of available medicinal plants.

Breeding of 'Jinmani' Cultivar of Gomchwi with Disease Resistance, High Quality and Yield

  • Gomchwi using stuffed leaves is many cultivated for 'Gondalbi(Ligularia stenocephala)' among Gomchwi species. 'Gondalbi' species like to be cultivation on farm because of low the incense and the bitter taste, and high yield. But 'Gondalbi' caused to curtail yield that susceptibility of powdery mildew disease and shriveling and death of plant on summer season. To solve this problems, we crossed a Gomchwi and 'Handeari-gomchwi' to have resistance of powdery mildew disease and high yield. A new Gomchwi cultivar 'Jinmani' was bred by crossing between Gomchwi (Ligularia fischeri (Ledeb.) Turcz.) and Handaeri-gomchwi (Ligularia fischeri var. spiciformis Nakai). The selection and investigation of growth and yield characteristics were conducted from 2007 to 2020 in field and greenhouse of Highland Agriculture Research Institute, NICS, Rural Development Administration. The color of petiole ear was purple. trichome of petiole and leaf back non-existed, and luster of leaf back existed. Density of leaf vein was 4 degree among 1-5 degree in a newly developed cultivar 'Jinmani'. Plant height, leaf length, leaf width and petiole length were 55.7, 21.8, 22.2, and 33.9 cm, respectively in the 2nd year of growth characteristics. Plant size was similar with that of 'Gommany'. Bolting and flowering time were Aug. 5 and Sept. 5, respectively, and Bolting and flowering time of 'Gommany' showed similar to Aug. 8 and Sept. 1, respectively. 'Jinmani' showed higher number of leaves (202 ea.) per plant compared to 'Gommany' (159 ea.). Furthermore, yield was 67.9% higher in 'Jinmani' (2,569 g/plant) than in 'Gommany' (1,530 g/plant). 'Jinmani' showed lower leaf thickness (0.66mm) than 'Gommany' (0.69 mm), and consequently showed more hardness in leaf characteristics (25.1 kg/2) compared to 'Gommany' (24.3kg/cm2). 'Jinmani' showed similar strong resistance compared to 'Gommany' in the susceptibility of powdery mildew disease.

Characteristics and pedigree selection of a shortened cultivation period strain in Lepista nuda (재배기간이 짧은 민자주방망이버섯 우량계통 선발 및 특성)

  • This study was conducted to cultivate new Lepista nuda varieties with shorter cultivation period and better fruiting body compared to that of wild strains, for mass production and commercial application. Eighteen genetic resources of L. nuda were collected and grown in boxes using rice straw-fermented growth medium. Four lines with fruiting bodies were formed and selected as cross-breeding lines. Although 657 combinations were crossed through monospore crossing, only 17 combinations were bred between the 'CBMLN-19' line and the 'CBMLN-30' line. Among them, 8 lines with fast mycelial growth and high density were selected. After inoculating the rice straw-fermented growth medium with 14 genetic resources and 8 cross-breeding lines, their incubation period was investigated. Six of the cross-breeding lines completed their incubation in 20 days, while 7 of the 14 genetic resources took more than 40 days to complete their incubation, reducing the incubation period by more than 20 days in most cross-breeding lines. After the incubations were completed, the clay loam soil was covered with for post-cultivation, and when the mycelial cultivation was complete, the formation of fruiting bodies was induced after scraping the mycelial bodies under these environmental conditions: 14℃, 95% relative humidity or higher, and 1,500 to 2,000 ppm CO2 concentration. The temperature was reduced to 6℃ at night, resulting in a low temperature shock. Thus, 4 lines of fruiting bodies occurred from two genetic resources 'CBMLN-31' and 'CBMLN-44' and two cross-bred lines 'CBMLN-96' and 'CBMLN-103'. After inoculation, the longest period for fruiting bodies to occur was 100 days for the control:, the genetic resource 'CBMLN-31', and the shortest period (45 days) was observed for the cross-breeding line 'CBMLN-103'. The result of the investigation of the fruiting body characteristics shows that the cross-bred line 'CBMLN-103' showed a small form with 1.9 g of individual weight and 123validstipes per box, which was the highest incidence among the four lines. Another cross-bred line, 'CBMLN-96', had an individual weight of 5.5 g, which is larger than that of 'CBMLN-103'; however, the number of valid stipes per box was 30 less than that of 'CBMLN-103'. Quantity analysis showed that the control, 'CBMLN-31', had the highest quantity of 783 g per box, followed by the cross-bred line, 'CBMLN-96' with 165 g per box, and then the 'CBMLN-103' with 232 g. The quantity of the two crossbred lines was lower than that of the control 'CBMLN-31'; however, the amount of fruiting bodies was higher, and the cultivation period was shortened by 32 to 33 days. Therefore, these two lines would be selected as superior lines.

Effects of microorganism density and mushroom yields according to the sterilization of casing soils at the cultivation of button mushrooms (복토살균 조건에 따른 양송이 재배과정별 복토내 미생물 밀도 및 수량 특성)

  • This study was conducted to set the proper sterilization standards of casing soil for the stable production of button mushroom(Agaricus bisporus) from mushroom disease that occurs in infection of casing soil material. Changes of aerobic bacteria are increased as the longer grow-out period and sharply increased after second flushes. Fluorescence Psuedomonas showed high density at high sterilization temperature and $100^{\circ}C$ treatment has extremely high density at 30 min and 60 min in casing 22 days. Density of thermophilic actinomyces is sharply increase from casing with soil and the highest density at 22 days of casing and rapidly decrease after first flushes. Sterilizing temperature of casing soil affects quality and quantity of button mushroom. Treatment of 60 min, 90 min at $80^{\circ}C$ and 30 min at $100^{\circ}C$ produced the highest mushroom yields, especially mushrooms yields of A grads were the highest at treatment of 90 min at $80^{\circ}C$. Treatment of 60min at $100^{\circ}C$ products many yields, however, this treatment has low economic feasibility for its yields. Sterilizing temperature of casing soil has an effect on generating diseases and insect pests. Treatment of 60 min, 90 min at $80^{\circ}C$ and 30 min $100^{\circ}C$ showed lower incidence than the other treatment. Although treatment of 30 min at $100^{\circ}C$ causes low diseases and mushroom fly damage, it has low mushroom yields. Furthermore, although treatment of 60 min at $100^{\circ}C$ has high mushroom yields, it causes high diseases and mushroom fly damage. Therefore the best conditions for the sterilization of casing soils was 60 min and 90 min at $80^{\circ}C$.

Production of Mother Bulbs with High Quality Using Bulbils of Garlic (Allium sativum L.) (마늘의 주아(珠芽)를 이용(利用)한 우양종구생산(優良種球生産))

  • This study was carried out to determine the promising method for the production of mother bulbs using garlic bulbs at the experimental field of Ange high school, Eusung in Korea from 1996 to 1998. The bulbs and bulbils were taken from a local cultivar 'Eusung' garlic. The growth of bulbil and yield potential were compared with different harvesting times and cultivation methods of bulbil. The harvesting time of bulbils which were capable of producing the mother bulbs was ranged from 15 days to 30 days after the emergence of flower stalks. The best results based on the yield and number of bulbs were obtained from the bulbils sampled at the just before garlic harvesting, 30 days after flower-stalk emergence. The mulching with transparent polyethylene(PE) film in bulbil cultivation was more effective for the plant growth and bulb formation than that with black PE film. The PE mulching from 30 days after seeding of bulbils to 30 days (late-April) before bulb harvesting was the moot effective for the increase of bulb yield. In the cultivation of garlic bulbils, application of lime (150 kg/10a) and compost (3,000 kg/10a) without any chemical fertilizers produced more bulbs in comparison with the standard level of fertilizer (N - $P_2O_5$ - $K_2O$ - compost - lime = 24.8 - 8.0 - 19.2 - 3,000 - 150 kg/10a). It was determined that the involucre seeding and assemble seeding of bulbils was effective and labour-saving methods for the production of mother bulbs from garlic bulbils. Input of labour force for the involucre seeding was decreased up to 61% as compared with the conventional drill seeding of bulbils. Mean yield from the cultivation of bulbil-derived single cloves was remarkably increased up to 95% in comparison with that of the common cloves. The larger size of single clove produced more yields, and the optimum planting density of single clove was $20{\times}10cm$. The cloves differentiated directly from bulbil showed to have higher yield potential than that from common cloves.

Effects of Different Nitrogen Levels and Planting Densities on the Quality and Quantity of 'Nunkeunheugchal' Rice (시비량과 재식밀도 변화에 따른 '눈큰흑찰'의 품질 및 수량변화)

  • 'Nunkeunheugchal' is a waxy black rice variety that has a large embryo. The quality of black rice depends on the anthocyanin content of the rice seed coat, which is mainly determined by cultivation environment. Factors that affect the anthocyanin content include nitrogen level, planting density, transplanting date and harvesting date. This study was carried out to investigate the optimum black rice cultivation conditions by examining the effects of different nitrogen levels and planting densities. An initial study was conducted to determine the optimum nitrogen level in which four levels of nitrogen were applied to the field (0, 4, 8 and 12 kg/10a). As the nitrogen contents were increased up to 8 kg/10a, there was a concomitant increase in rice yields. However, nitrogen levels greater than 8 kg/10a, the yield was maintained at the same level. Correlation analysis indicated that the optimum nitrogen level for maximum yield was 9.6 kg/10a. In addition, anthocyanin levels showed a trend similar to that of yield, with correlation analysis indicating that the optimum nitrogen level for maximum anthocyanin content is 10.6 kg/10a.On the basis of these results, a second study was conducted to determine the optimum combination of planting density and nitrogen level. The planting densities investigated were $30{\times}12$, $30{\times}14$, $30{\times}16$ and nitrogen levels were 7, 9 and 12 kg/10a. A high planting density ($30{\times}12cm$) was shown to produce higher numbers of tillers and yield. As calculated in the first study, a nitrogen level of 9 kg/10a shown to produce the highest anthocyanin content and yield. Collectively, the results of this study indicate that a planting density of $30{\times}12cm$ and a nitrogen level of 9 kg/10a is the optimal combination in terms of maximizing both rice yield and anthocyanin content.

Selective Toxicity of Pesticides to the Predatory Mite, Phytoseiulus persimilis and Control Effects of the Two-spotted Spider Mite, Tetranychus urticae by Predatory Mite and Pesticide Mixture on Rose (칠레이리응애에 대한 농약의 선택독성과 장미에서 천적과 농약의 혼용에 의한 점박이응애의 방제효과)

  • Toxicities of 42 pesticides (13 acaricides, 13 insecticides, 13 fungicides and 3 adjuvants) commonly used to control rose insect, mite, and disease pests were evaluated to the two-spotted spider mite, Tetranychus urticae egg and adult, and its predator Phytoseiulus persimilis egg, nymph and adult at the recommended concentration. The effect of density suppression of T urticae by predatory mite and pesticide mixture on the rose in the greenhouses was also investigated. Among 13 acaricides tested, acequinocyl, bifenazate, fenbutatin oxide and spirodiclofen showed much less toxicity to P. persimilis than to T urticae. Among insecticides, acetamiprid, imidacloprid, spinosad, thiamethoxam and acetamiprid+etofenprox showed low toxicity to P. persimilis. and T ruticae. Among 13 fungicides, azoxystrobin, kresoxim-methyl, myclobutanil, nuarimol, triadimefon, triflumizole and oxadixyl+mancozeb had a negligible effect on P. persimilis and T. urticae. Among three adjuvants, cover and siloxane expressed high toxicity, while spreader showed very low toxicity to P. Persimilis. In the greenhouses experiments, the density of T urticae before treatment was 65.3 mites per leaf. However, their density after release about 30 predatory mites per rose abruptly decreased from 3.8 mites at 11th day to zero mite at 20th day. During survey periods, four treatments of fungicides (kresoxim-methyl, myclobutanil, nuarimol, triflumizole) for the control of Sphaerotheca pannosa and one treatment of insecticide (spinosad) the control of Frankliniella occidentalis were applied, and these treatments had no the pesticides had no effect on the predatory mite density. It may be suggested from these results that four acaricides, five insecticides, seven fungicides, and one adjuvant could be incorporated into the integrated T. urticae management system with P. persimilis on rose cultivation.
