Effects of Rubus coreanus Extracts on Ultraviolet-A Irradiated Cultured Human Skin Fibroblasts (자외선이 조사된 인간피부섬유아세포에 복분자 추출물이 미치는 영향)

  • Sunlight, and in particular its UV component, is the major environmental trigger that underlies the major signs of human skin and skin cancer in general. Therefore, this study was carried out to investigate the UV protection effects of R. coreanus. R. coreanus was extracted by ultra high pressure extraction process at 500 MPa and $30^{\circ}C$ for 5 and 15 minutes. The cytotoxicity of the extracts extracted by ultra high pressure process on human dermal fibroblast cell CCD-986sk, human kidney normal cell HEK293, and human lung normal cell HEL299 was measured as 17.5%, 16.5% and 14.0%, respectively in adding $1.0\;mg/m{\ell}$ of the samples, which was much lower than that from conventional water extraction method at $100^{\circ}C$ as 23.2%, 22.5%, 21.2%. The secretion of $NO^-$ from macrophage showed $15.9\;{\mu}M$ on the R. coreanus extract from this process, which was higher than others. Prostaglandin $E_2$ ($PGE_2$) production from UV-induced human skin cells was also greatly decreased down to $510\;pg/m{\ell}$, compared to the control. From the results, we considered that the extracts from R. coreanus could be potent natural materials for skin anti-inflammation agent, and could be used as a potential anti-aging for the photo-damaged skin.

Enhancement of Cosmeceutical Activities of Berberis koreana Bark by High Pressure and Ultrasonification Extraction Processes (초고압 및 초음파 추출공정을 이용한 매자나무 수피의 향장활성 증진)

  • This study was performed to investigate the enhancement of cosmeceutical activities of Berberis koreana bark by different extraction processes. The extracts are WE (water extract at $100^{\circ}C$, control), USE (ultrasonification for 1 hours at $60^{\circ}C$ with water), HPE (high pressure for 5 minutes at $60^{\circ}C$ with water) and USE + HPE (ultrasonification process for 1 hours after high pressure for 5 minutes at $60^{\circ}C$ with water), respectively. The cytotoxicity of the extracts was in the range of 24.02~26.94% at 1.0 mg/ml concentration. The USE + HPE showed the lowest cytotoxicity. Compared to the WE, total phenolic and flavonoid contents in the USE + HPE increased to 121.5% and 154.2%. The USE + HPE showed the highest activity at 1.0 mg/ml concentration in 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging, inhibition activity of xanthine oxidase and superoxide dismutase (SOD)-like test, respectively. Tyrosinase inhibition of WE, USE, HPE and USE + HPE at 1.0 mg/ml concentration was measured as 17.72, 19.62, 22.83 and 24.16%, respectively. Hyaluronidase inhibition activities of the USE + HPE were higher than 20.8%~29.5% of the WE. Our results suggested that the extracts from ultrasonification process after high pressure extraction has relatively high cosmeceutical activities, and that the bark of Berberis koreana could be considered as a candidate of new functional cosmetic agents.

  • Hexafluoropropyleneoxide ($C_3F_6O$, HFPO) is highly expensive and it may be used as a raw material for the synthesis of various fluorine based compounds. Currently, extractive distillation method has gained considerable attention to collect the HFPO from a mixture of HFPO / hexafluoropropylene ($C_3F_6$, HFP). Optimized operating conditions are studied using a theoretical method for the extraction process. Among available solvents for the purification process, the use of 1,1-dichloro-1-fluoroethane exhibits a high purity of HFPO as a top product and minimize the required heat duty. Since the boiling point of the solvent increases as the pressure in the column increases, the enhanced extractive capability of the solvent led to the high purity of HFPO at the high pressure.

  • Steam explosion process is an efficient pretreatment method for sparating and utilizing wood main components has attracted attention in utilization of ligno-cellulosic biomass. In order to obtain the effective pretreatment condition. this study was made explosion apparatus. examined the composition. extraction of exploded wood. Wood chips of pine(Pinus densiflora oak (Quercus serrata) and birch wood (Belula platyphylla var. japonica) were treated with a high pressure steam(20-30 kg/$cm^2$, 2-6 minutes). The results can be summarized as follow; In analysis of exploded wood(EXW). It was found arabinose residues rapidly decreased with increasing of steaming time and pressure. Extractives of EXW with sodium hydroxide increased with increasing of steaming-time and- pressure especially extractives 1% sodium hydroxide has higher than other extracted method extractives of hard wood(oak, birch) has higher than pine wood. In EXW extracted with sodium hydroxide and methanol lignin was partially delignified alkali extraction was more delignified than methanol extraction hardwood than pine wood.

  • Poly(acrylic acid) (PAA) microspheres is one of the widely-used polymeric materials for the bio-field application and the electric materials. For the synthesis of PAA microspheres, the polymerization technique using surfactants is applied. After the synthesis, the purification and separation processes are required for the removal of surfactant. When general organic solvents were used, many problems, such as huge amount of waste solvent, additional separation processes, and the possibility of residual media, were occurred. Thus, High-pressure Soxhlet extraction using liquid $CO_2$ was developed to solve these problems. In this study, High-pressure Soxhlet extraction of the synthesized PAA microspheres using liquid $CO_2$ was conducted for the removal of Monasil PCA which is used for the dispersion polymerization of acrylic acid in compressed liquid Dimethyl ether (DME). The morphology of the extracted PAA particles was checked by field emission scanning electron microscopy (FE-SEM) and the residual concentration of Monasil PCA was analyzed by inductively coupled plasma - Optical Emission Spectrometer (ICP-OES). For studying the effect of the solvent effect, Soxhlet extraction was conducted using n-hexane, liquid DME, and liquid $CO_2$. In case of n-hexane, some extracted PAA microspheres were produced. However, deformation was also occurred due to the high thermal energy of n-hexane vapor. Liquid DME could not remove Monasil PCA. When using liquid $CO_2$, the extracted PAA microspheres which were free for the residual solvent were produced without deformation. For finding the optimum operating condition, high-pressure Soxhlet extraction was conducted for 8 hours with changing the temperature of reboiler and condenser. When the extractor temperature is $19.6{\pm}0.2^{\circ}C$ and the pressure is $51.5{\pm}0.5$ bar, the best removal efficiency was obtained.

Enhancement of Anticancer Activity of Acer mono by High Pressure Extraction Process (고로쇠 수피 초고압 추출물의 항암활성 증진)

  • We investigated a method to improve anticancer activities of Acer mono by ultra high pressure extraction process. The extract yields by ultra high pressure were 9.49% and 9.87% for 5 min and 15 min processing time, respectively, which were relatively higher than 3$\sim$4% of conventional extraction processes due to their resid bark structure. The extract for 15 minutes extraction (HPE15) showed higher potent scavenging effect as 94.56% than the control, BHA as 93.24%. On SOD-like test, HPE15 also showed the highest activity as 38.6% at 1.0 mg/mL concentration. The cytotoxicity of HPE15 on normal human lung and kidney cell were below 23.54% in adding 1.0 mg/mL. Generally, human cancer cell growth stomach adenocarcinoma (AGS), lung adenocarcinoma (A549), breast adenocarcinoma (MCF-7), colon adenocarcinoma (Caco-2) and liver adenocarcinoma (Hep3B) were inhibited up to 75% with higher selectivity of above 4.0. High antioxidant activity of HPE15 resulted in high anticancer activity, and its activity was also due to higher yields of Acer mono by ultra high pressure extraction process. It was also proved by HPLC comparison analysis.

Enhancement of Cosmeceutical Activity from Codonopsis lanceolata Extracts by Stepwise Steaming Process (증숙 및 초고압 증숙 공정을 통한 더덕의 향장활성 증진)

  • In general, stepwise hot steaming process is known to be effective in improving its biological activities; however, not much employed in processing Codonopsis lanceolata due to its hardness. The complex processed C. lanceolata showed highest free radical scavenging acitivity as 45.21%. Total phenol and flavonoid content were of complex processed C. lanceolata higher than conventional extract and alone steaming process. It was showed the lower melanogenesis rate on melanin production test by B16F10 cells as 27.46%. High inhibitory of tyrosinase was also measured as 28.61% by adding steamed Codonopsis lanceolata extracts by high pressure extraction of 1.0 $mg/m{\ell}$. And anti-wrinkle activity were 39.08%. In comparing phenolic acids profiles in the extract, in general higher amounts of polyphenol were obtained possibly by easy release of active components during thermal processing, which results in better antioxidant activities than that of general extract. This findings can also be supported by result that the extract by steaming process showed better activities than the general extraction extract.

  • Using the high-pressure heating extraction process suitable for mass-production, crab stock(CS), red crab stock(RCS) and fish stock(FS) were manufactured and their quality and organic features were compared. The results are as follows. In mineral content, the content of essential amino acids, the content of delicious amino acids and preference, CS and RCS were rated higher than FS, and especially RCS had the most abundant essential amino acid content and had the highest rating in organic evaluation. Also, in terms of cost, RCS was more economical than regular CS, which is relatively expensive. Therefore, it is believed that replacing fish bones with RCS in making stock will be more cost-saving and have higher preference.

Quality characteristics of Hijikia fusiforme extracts with different extraction method (추출방법에 따른 톳 추출물의 품질특성)

  • The physiological properties of water extracts from Hizikia fusiformis extracted using different extraction methods (water extraction, WE; autoclave extraction, AE; high pressure extraction, HPE) were investigated. The freeze-dried powder yields from HPE, AE and WE were 29.33, 27.84 and 23.63%, respectively. The $L^*$ and $b^*$ color values were higher in WE, while the $a^*$ color values were higher in WE and AE. The total sugar content of AE (60.14%) was higher than those of WE (47.10%), HPE (40.97%). The reducing sugar content (7.88%) and protein content (42.83%) of AE was higher than those of WE, and HPE. The uronic acid (5.04%), total free amino acid (785.19 mg/g), taurine (19.16 mg/g), aspartic acid (66.63 mg/g), asparagine (204.84 mg/g), alanine (188.87 mg/g) and ammonium chloride (243.91 mg/g) contents, however, were the highest in HPE. Additionally, the crude polysaccharide yield was higher in HPE (4.75%) than in AE and WE, and the crude saccharide (fucose, galactose, glucose, xylose and fucose) yields were higher in AE. It can be concluded that optimum conditions for the efficient extraction of Hizikia fusiformis depending on components are high pressure and a lower temperature than in the typical process.

A Study on the Variable Condition Debinding Process in Supercritical CO2 for Removing Binder from Thick Ceramic Injection Molded Parts (두꺼운 세라믹 사출성형체로부터 효율적인 결합제 제거를 위한 초임계 CO2 가변조건 탈지공정 연구)

  • The purpose of this study is to remove paraffin wax binder effectively from powder injection molded part using supercritical fluids in powder injection molding process. For a thin powder injection molded part about 1-2 mm thickness, paraffin wax binder can be removed rapidly without any defect by traditional supercritical extraction process which has fixed high temperature and pressure condition. But, for a thick powder injection molded part, there are limitations in removing paraffin wax binder by the fixed high process condition because crack occurs at the beginning step. Therefore, here we studied variable condition debinding process that starts with mild process condition at the beginning step and then increase the process conditions simultaneously at each step. To find out the initial process condition that has the highest extraction yield without any defect for each sample thickness, we investigated various supercritical debinding conditions using 1-4 mm thickness ceramic injection molded sample. By using the variable condition debinding process that starts with the initial process condition at the first step and then increasing process conditions simultaneously at each step (temperature from 333.15 to 343.15 K, pressure from 12 to 27 MPa, and $CO_2$ flow rate from 1.5 to 10 L/min), over 95% of paraffin wax binder was removed from the 4 mm thick (10 mm diameter) ceramic injection molded disk samples within 5 hours.
