Cardiac physiologic regulation of sub-type specific adrenergic receptors in transgenic mice overexpressing β1- and β2-adrenergic receptors.

  • Objective Combination of ${\beta}_1-adrenergic$ receptor (AR) blockade and ${\beta}_2-AR$ activation might be a potential novel therapy for treating heart failure. However, use of ${\beta}-AR$ agonists and/or antagonists in the clinical setting is controversial because of the lack of information on cardiac inotropic or chronotropic regulation by AR signaling. Methods In this study, we performed hemodynamic evaluation by examining force frequency response (FFR), Frank-Starling relationship, and response to a non-selective ${\beta}-AR$ agonist (isoproterenol) in hearts isolated from 6-month-old transgenic (TG) mice overexpressing ${\beta}_1-$ and ${\beta}_2-ARs$ (${\beta}_1-$ and ${\beta}_2-AR$ TG mice, respectively). Results Cardiac physiologic consequences of ${\beta}_1-$ and ${\beta}_2-AR$ overexpression resulted in similar maximal response to isoproterenol and faster temporary decline of positive inotropic response in ${\beta}_2-AR$ TG mice. ${\beta}_1-AR$ TG mice showed a pronounced negative limb of FFR, whereas ${\beta}_2-AR$ TG mice showed high stimulation frequencies with low contractile depression during FFR. In contrast, Frank-Starling relationship was equally enhanced in both ${\beta}_1-$ and ${\beta}_2-AR$ TG mice. Conclusion Hemodynamic evaluation performed in the present showed a difference in ${\beta}_1-$ and ${\beta}_2-AR$ signaling, which may be due to the difference in the desensitization of ${\beta}_1-$ and ${\beta}_2-ARs$.

  • The aim of investigated the role of FABP5 in the hepatic lipogenesis and lipid metabolisms. Mice were overexpressed and silenced liver FABP5 using virus particles. Mice were fed a Western-type diet or regular chow for 1week and then sacrificed mouse after 24hr fasted. Liver homogenates were used for protein analysis by Western blot and mRNA levels by RT-PCR. Hepatic and serum lipids were analysed by thin-layer chromatography. Mice fed a Western-type or high saturated fat diet revealed large increases in FABP5 expression. However, FABP5 mRNA levels were drastically reduced under fasted. Hepatic TG was significantly increased FABP5-OEAV mice, but a significantly decreased hepatic free cholesterol under fed. The discovered a substantial decrease in hepatic TG mass with FABP5 silencing. In these data, presented evidence for an important role of FABP5 in hepatic lipogenesis and hepatic TG storage. FABP5 may also be a potential target in the treatment of NAFLD, metabolic syndrome, and obesity. Furthermore, studies to which transcription factors are involved in FABP5 expression and regulation.

Study on the in vitro and in vivo anti-obesity effects of a combination of Syzygium aromaticum L. and Sorbus commixta Hedl. (정향과 마가목 복합물의 in vitro와 in vivo 항비만 효과 연구)

  • Purpose: This study investigated the anti-obesity effects of a combination of Syzygium aromaticum L. and Sorbus commixta Hedl. (SS) in vitro and in vivo. Methods: The extracts of Syzygium aromaticum extract (SA) and Sorbus commixta extract (SC) were prepared individually using distilled water. They were mixed in a 1:2 ratio for use in the experiment. To assess the anti-obesity potential of SS in vitro, we examined cell proliferation, cellular triglyceride (TG), and total cholesterol (TC) levels, as well as lipogenesis and β-oxidation in 3T3-L1 cells. To confirm its anti-obesity potential in vivo, C57BL/6J mice were fed a 60% high-fat diet (HFD) to induce obesity. SA alone, SC alone, and their combination compound, SS (at a dosage of 200 mg/kg) were orally administered for 6 weeks. Thereafter, to conduct a comparative evaluation, serum analysis, western blotting of liver tissues, and histopathological analysis were performed. Results: Both SS200 and SS400 significantly inhibited the cellular TG and TC contents in the 3T3-L1 cells. Furthermore, treatment of the cells with SS (at a dose 200 and 400 ㎍/mL) also led to a noticeable regulation of key lipogenic and β-oxidation factors. Treatment of obese mice with SS resulted in a greater reduction in serum leptin and TG levels compared to treatment with the individual compounds (SA and SC). Furthermore, activation of AMP-activated protein kinase α by SS treatment resulted in the suppression of sterol regulatory element-binding proteins (SREBP)-1, leading to the inhibition of acetyl-CoA carboxylase (ACC) expression. Conclusion: Our results suggest that SS may have the potential to prevent obesity through a reduction in the TG and TC levels and regulation of lipogenesis and β-oxidation.

Effect of Genetic Predisposition on Blood Lipid Traits Using Cumulative Risk Assessment in the Korean Population

  • Dyslipidemia, mainly characterized by high triglyceride (TG) and low high-density lipoprotein cholesterol (HDL-C) levels, is an important etiological factor in the development of cardiovascular disease (CVD). Considering the relationship between childhood obesity and CVD risk, it would be worthwhile to evaluate whether previously identified lipid-related variants in adult subjects are associated with lipid variations in a childhood obesity study (n = 482). In an association analysis for 16 genome-wide association study (GWAS)-based candidate loci, we confirmed significant associations of a genetic predisposition to lipoprotein concentrations in a childhood obesity study. Having two loci (rs10503669 at LPL and rs16940212 at LIPC) that showed the strongest association with blood levels of TG and HDL-C, we calculated a genetic risk score (GRS), representing the sum of the risk alleles. It has been observed that increasing GRS is significantly associated with decreased HDL-C (effect size, $-1.13{\pm}0.07$) compared to single nucleotide polymorphism combinations without two risk variants. In addition, a positive correlation was observed between allelic dosage score and risk allele (rs10503669 at LPL) on high TG levels (effect size, $10.89{\pm}0.84$). These two loci yielded consistent associations in our previous meta-analysis. Taken together, our findings demonstrate that the genetic architecture of circulating lipid levels (TG and HDL-C) overlap to a large extent in childhood as well as in adulthood. Post-GWAS functional characterization of these variants is further required to elucidate their pathophysiological roles and biological mechanisms.

The Optimal Enrichment Condition of Rotifer Brachionus rotundiformis (소화효소 활성으로 본 rotifer Brachionus rotundiformis의 적정 영양강화 조건)

  • The purpose of the study was to suggest the optimal lipid enrichment conditions used digestive enzyme activity of rotifer changing due to water temperature and salinity. The high population growth appeared at the experiment temperature more than 28 degrees highly on the culture temperature(maximum 32 degrees, 1,453 individual/mL). The fecundity was low at high temperature, and the egg ratio was high at low temperature. Population growth of 10 and 15 ppt appeared in most highly, but the fecundity and the egg ratio were high most significantly appeared in natural seawater(32 psu). The digestive enzyme activity by the culture environment mainly showed high activity in natural seawater(amylase exclusion, 15 psu). However, the TAP activity by the water temperature showed highly at the more high temperature, but the amylase and the lipase appeared at low temperature. We carried out the lipid enrichment at 20 degrees and 26 degrees in a condition of the natural seawater. Total protein, the total essential amino acids differed not significantly. The methionine content that was essential amino acids, a total lipid content, unsaturated index of fatty acids, DHA and the DHA/EPA ratio were high significantly each in $20^{\circ}C$ enrichment trial. Therefore, we could suggest the $20^{\circ}C$ and natural seawater for the optimal lipid enrichment condition in aquaculture, because methionine contents, several indexes by the lipid, TG-lipase activity, fecundity and egg ratio are high.

Carbohydrate Intake Associated with Risk Factors of Coronary Heart Disease in the Adults: NHANES III (성인의 만성질환관련 탄수화물 식사지침 연구)

  • Recent research reported health risks associate with high carbohydrates diets. Objectives of this study were to evaluate in a cross-sectional study if high carbohydrate diet is associated with coronary heart disease(CHD) risk factors: examined blood concnetration of triglyceride(TG), total cholesterol, high density lipoprotein cholesterol(HDL-C), plasma glucose, systolic blood pressure(BP), body mas index(BMI), wasit-hip ratio(WHR) and waist-stature ratio(WSR). Using the most recent US National Health and Nutrition Examination Survey(IIINHANES III) data, the nationally representative US population (3772 men, 4095 women of 25-64 years of age) was divided into low vs. high carbohydrate diet groups(below 40% vs. above 60% energy intake from cab carbohydrates) and compared by the CHD risk factors. Triglyceride was higher(p<0.001) in the high carbohydrate group, whereas high density-lipoprotein cholesterol(HDL-C)was lower(p<0.01) in the high carbohydrate diet group. In plasma glucose, there was no significant differences between high carbohydrate diet and low carbohydrate diet. In adiposity(BMI, WHR and WSR), it also showed no significant differences, After adjustment for age, ethnicity, alcohol and smoking in upper 60%-carbohydrate diet, Odds Ratio of TG and HDL-C were 1.42 and 1.23 in men and 1.22 and 1.17 in women. 50-60% carbohydrate diet was associated with decreased risk of CHD. Dietary guidelines for Koreans recommend 60-70% of total energy from carbohydrate, as Koreans traditionally consumed high carbohydrate diets. In a cross-sectional population of adults, diets containing 55-60% energy from carbohydrate were suggested as a dietary guideline of carbohydrate intake for Koreans. Correlation between Serum Lipid Level and Neurophysiological Findings in Patients with Carpal Tunnel Syndrome (손목굴증후군 환자의 신경생리학적 소견과 혈청 지질 수치와의 상관관계)

  • Background: Carpal tunnel syndrome (CTS) is the most common entrapment neuropathy caused by compression of the median nerve beneath the transverse carpal ligament. CTS can be correctly diagnosed by the patients' description of symptoms and electrophysiological tests that measure nerve conduction through the wrist. Many previous studies reported various risk factors of CTS, such as obesity, diabetes mellitus, thyroid disease and trauma. Obesity is associated with both hyperlipidemia and CTS. This study focused on the relationship between severity of CTS and serum lipid level. Methods: One hundred fourteen patients with CTS and 74 controls were divided into four groups according to the severity; normal, mild, moderate and severe. And then serum total cholesterol (TC), triglyceride (TG), low-density lipoprotein (LDL-C) and high-density lipoprotein (HDL-C) were measured in each group. Results: There was a positive correlation between TG and CTS severity (p<0.001). But TC, LDL-C and HDL-C were not correlated with CTS severity. Conclusions: These results suggest that high serum TG may act as an aggravating factor of CTS.

Dietary Lipid, Plasma Lipoprotein and Fatty Acid Composition of Young Korean Women (일부 젊은 여성의 지질 섭취와 혈장 지단백 및 지방산 조성에 관한 연구)

  • This study was conducted to investigate dietary lipid intakes and the plasma lipoprotein levels and fatty acid composition of eight healthy young Korean women for 7 days. After an overnight fast, blood samples were taken on the last day of dietary survey. The plasma were fractinated to very low-density lipoprotein(VLDL), low-density lipoprotein(LDL), and high-density lipoprotein(HDL) by ultracentrifugation. From each fraction, the content of triglyceride (TG), phospholipid(PL), cholesterol(CHOL), free fatty acid(FFA) and protein were determined. Fatty acid composition of total plasma lipid was alos analyzed. The subjects consumed 34.7$\pm$2.8g of fat daily. The ingested amounts of EPA and DHA were 0.2 and 0.4g/day, respectively. They also consumed 112.2$\pm$12.9mg of cholesterol per day. The concentrations of VLDL, LDL and HDL in the plasma were 66.5$\pm$6.1, 114.0$\pm$8.8 and 129.4$\pm$5.1mg/dL, respectively, so the percentages of VLDL, LDL, and HDL were 20.7, 44.8 and 34.5%, respectively. The VLDL was composed of 27.5mg/dL of TG, 39.7 of PL, 58.8 of CHOL, 3.1 of FFA, and 22.4 of protein. The HDL had 10.7mg/dL of TG, 23.7 of PL, 18.7 of CHOL, 2.6 of FFA, and 73.7 of protein. In the plasma, linoleic acid was the most abundant fatty acid, followed by palmitic and oleic acid, the percentages of these 3 fatty acids were 30.8, 23.4 and 17.7%, respectively. The result indicated that the plasma levels of lipoprotein in the healthy young Korean women were slightly low and the levels of EPA and DHA were relatively high, compared to the data of other investigatiors. This might be due to the fact that they consumed less fat and cholesterol and higher EPA and DHA.

Effect of Cheonggukjang Made from Sword Bean on Serum Lipid Profile and Tissue Change in Rats

  • This study was to investigate serum triglyceride (TG), phospholipid (PL), and cholesterol concentrations after feeding the combined diets of prepared traditional Cheonggukjang (TC) or Cheonggukjang made from sword bean powder (CS), the feed, and high-fat cholesterols at certain ratio for 8 weeks in rats, and then confirmed the effects of the diets based on the changes in the tissues of livers, kidneys, and spleens. The serum TG concentrations were lower in CS group compared to that of in TC group. The increase of PL concentrations was greater in CS group than that of TC group. FChCS6 group (92% feed+2% cholesterol+6% CS) showed the lowest total-cholesterol (T-C), $60.1{\pm}8.5mg/dL$, indicating that CS was effective to lower the T-C. Both TG/PL and T-C/PL ratio tended to be decreased. As CS intake was effective in the reduction of low density lipoprotein-cholesterol level as well as the increase of high density lipoprotein-cholesterol level, it would be effective in the prevention of adult diseases and circulatory diseases. It was confirmed that there were no changes in organ appearance and tissues in the rats with CS intake. Therefore, CS intake showed superior functionalities than that of TC intake.

Effects of the Ethanol-extract of Allium wageki on the Fructose-induced Hypertensive Rats (쪽파 에탄올 추출물이 과당-유도 고혈압 흰쥐에 미치는 영향)

  • The effects of the ethanol (EtOH)-extract of Allium wageki were studied on the changes of body weight (BW), systolic blood pressure (SBP), triacylglycerol (TG), cholesterol, and insulin levels in Sprague-Dawley rats in which high fructose feeding elicited hyperinsulinemia, hypertriglyceridemia, and hypertension. SBP and BW were measured for 7 weeks on the fructose diet, fructose/EtOH-extract, vehicle- treated, or vehicle/EtOH-treated rats. The EtOH-extract of Allium wageki blocked the increases of SBP ($119.4{\pm}3.7\;mmHg$ in fructose/EtOH-extract of Allium wageki; $141{\pm}4.3 \;mmHg$ in fructose diet, p<0.01) and BW ($368{\pm}11.4\;g$ in fructose/EtOH-extract of Allium wageki; $402{\pm}15.7\;g$ in fructose diet, p<0.05) by high fructose-diet. The increases of TG, total cholesterol, and insulin levels were also blocked by administration of EtOH-extract of Allium wageki in fructose-induced hypertensive rats. These results suggest that EtOH-extract of Allium wageki has protect effects on the increases of TG, total cholesterol, insulin level, and blood pressure in fructose-induced hypertensive rats.
