Genetic localization of epicoccamide biosynthetic gene cluster in Epicoccum nigrum KACC 40642

  • Epicoccum nigrum produces epipyrone A (orevactaene), a yellow polyketide pigment. Its biosynthetic gene cluster was previously characterized in E. nigrum KACC 40642. The YES liquid culture of this strain revealed high-level production of epicoccamide (EPC), with an identity that was determined using liquid chromatography-mass spectrometry analysis and molecular mass search using the SuperNatural database V2 webserver. The production of EPC was further confirmed by compound isolation and nuclear magnetic resonance spectroscopy. EPC is a highly reduced polyketide with tetramic acid and mannosyl moieties. The EPC structure guided us to localize the hypothetical EPC biosynthetic gene cluster (BGC) in E. nigrum ICMP 19927 genome sequence. The BGC contains genes encoding highly reducing (HR)-fungal polyketide synthase (fPKS)-nonribosomal peptide synthetase (NRPS), glycosyltransferase (GT), enoylreductase, cytochrome P450, and N-methyltrasnferase. Targeted inactivation of the HR-fPKS-NRPS and GT genes abolished EPC production, supporting the successful localization of EPC BGC. This study provides a platform to explore the hidden biological activities of EPC, a bolaamphiphilic compound.

Disaster Prevention Service Design Framework (방재서비스디자인 프레임워크 연구)

  • Many hidden aspects of society around the world that has been lurking as risk factors have been ultimately connected to disasters. Such connection did not just stop with massive damages to properties and loss of lives but became a serious social issue. Especially in the case for Korea, because the country has gone through abrupt economic growth which has resulted in rapid urbanization and expansion of economic scale, Korea has been caught in a vicious cycle of experiencing human and social calamities repeatedly. Due to the seriousness of such problem, there's been continued research in various fields for managing disasters, and the domain of knowledge has been expanding with complexity. The following study went beyond the perspectives of disaster prevention study, disaster prevention engineering and social sciences suggested from previous research directions in disaster and disaster prevention, and attempted an alternative research method by approaching from a service design perspective. During the process, the paper looked over the summary on disaster and service design. By applying it on disaster prevention projects, the research sets forth and discusses effective and practical disaster prevention service design. Through specific case studies, the research methodology applied was verified, and the purpose of the study was carried out in the perspective of real world applications different from textbook type discussions. It is expected that through the disaster prevention service design process and platform that were discovered during the research result, defining and understanding disaster prevention service design will be made apparent. Additionally, the research is expected to be used as a basic building block for building a visualized plan for tangible and intangible factors related to disasters before the disaster process through such proposed disaster prevention service design.

Effect of Treatment with Docosahexaenoic Acid into N-3 Fatty Acid Adequate Diet on Learning Related Brain Function in Rat (N-3계 지방산 적절 함량 식이의 docosahexaenoic acid 첨가가 기억력 관련 뇌 기능에 미치는 영향)

  • The effect of adding docosahexaenoic acid into an n-3 fatty acid adequate diet on the improvement of learning related brain function was investigated. On the second day after conception, Sprague Dawley strain dams were subjected to a diet containing either n-3 fatty acid adequate (Adq, 3.4% linolenic acid) or n-3 fatty acid adequate+docosahexaenoic acid (Adq+DHA, 3.31%linolenic acid plus 9.65% DHA). After weaning, male pups were fed on the same diet of their respective dams until adulthood. Motor activity and Morris water maze tests were measured at 10 weeks. In the motor activity test, there were no statistically significant differences in moving time and moving distance between the Adq and Adq+DHA diet groups. The n-3 fatty acid adequate with DHA (Adq+DHA) group tended to show a shorter escape latency, swimming time and swimming distance compared to the n-3 fatty acid adequate group (Adq), but the differences were not statistically significant. There was no difference in resting time, but the Adq+DHA group showed a higher swimming speed compared to the Adq group. In memory retention trials, the numbers of crossing of the platform position (region A), in which the hidden platform was placed, were significantly greater than those of other regions for both Adq and Adq+DHA groups. Based on these results, adding DHA into the n-3 fatty acid adequate diet from gestation to adulthood tended to induce better spatial learning performance in Sprague Dawley rats as assessed by the Morris water maze test, although the difference was not significant.

A TBM data-based ground prediction using deep neural network (심층 신경망을 이용한 TBM 데이터 기반의 굴착 지반 예측 연구)

  • Tunnel boring machine (TBM) is widely used for tunnel excavation in hard rock and soft ground. In the perspective of TBM-based tunneling, one of the main challenges is to drive the machine optimally according to varying geological conditions, which could significantly lead to saving highly expensive costs by reducing the total operation time. Generally, drilling investigations are conducted to survey the geological ground before the TBM tunneling. However, it is difficult to provide the precise ground information over the whole tunnel path to operators because it acquires insufficient samples around the path sparsely and irregularly. To overcome this issue, in this study, we proposed a geological type classification system using the TBM operating data recorded in a 5 s sampling rate. We first categorized the various geological conditions (here, we limit to granite) as three geological types (i.e., rock, soil, and mixed type). Then, we applied the preprocessing methods including outlier rejection, normalization, and extracting input features, etc. We adopted a deep neural network (DNN), which has 6 hidden layers, to classify the geological types based on TBM operating data. We evaluated the classification system using the 10-fold cross-validation. Average classification accuracy presents the 75.4% (here, the total number of data were 388,639 samples). Our experimental results still need to improve accuracy but show that geology information classification technique based on TBM operating data could be utilized in the real environment to complement the sparse ground information.

  • Recently, systems biology has been increasingly applied to gain insights into the complexity of living organisms. Many inaccessible biological information and hidden evidences fur example flux distribution of the metabolites are simply revealed by investigation of artificial cell behaviors. Most bio-models are models of single cell organisms that cannot handle the multi-cellular organisms like plants. Herein, a structured and multi-cellular model of potato was developed to comprehend the root starch biosynthesis. On the basis of simplest plant cell biology, a potato structured model on the platform of Berkley Madonna was divided into three parts: photosynthetic (leaf), non-photosynthetic (tuber) and transportation (phloem) cells. The model of starch biosynthesis begins with the fixation of CO$_2$ from atmosphere to the Calvin cycle. Passing through a series of reactions, triose phosphate from Calvin cycle is converted to sucrose which is transported to sink cells and is eventually formed the amylose and amylopectin (starch constituents). After validating the model with data from a number of literatures, the results show that the structured model is a good representative of the studied system. The result of triose phosphate (DHAP and GAP) elevation due to lessening the aldolase activity is an illustration of the validation. Furthermore, the representative model was used to gain more understanding of starch production process such as the effect of CO$_2$ uptake on qualitative and quantitative aspects of starch biosynthesis.

Cypress Essential Oil Improves Scopolamine-induced Learning and Memory Deficit in C57BL/6 mice (사이프러스 에센셜 오일의 흡입이 전임상 실험동물의 손상된 학습능력과 기억력에 미치는 영향)

  • Objectives : Increasing evidence supports the biological and pharmacological activities of essential oils on the central nervous system such as pain, anxiety, attention, arousal, relaxation, sedation and learning and memory. The purpose of present work is to investigate the protective effect and molecular mechanism of cypress essential oil (CEO) against scopolamine (SCO)-induced cognitive impairments in C57BL/6 mice. Methods : A series of behavior tests such as Morris water maze, passive avoidance, and fear conditioning tests were conducted to monitor learning and memory functions. Immunoblotting and RT-PCR were also performed in the hippocampal tissue to determine the underlying mechanism of CEO. Results : SCO induced cognitive impairments as assessed by decreased step-through latency in passive avoidance test, relatively low freezing time in fear conditioning test, and increased time spent to find the hidden platform in Morris water maze test. Conversely, CEO inhalation significantly reversed the SCO-induced cognitive impairments in C57BL/6 mice comparable to control levels. To elucidate the molecular mechanisms of memory enhancing effect of CEO we have examined the expression of brain-derived neurotrophic factor (BDNF) in the hippocampus. CEO effectively elevated the protein as well as mRNA expression of BDNF via activation of cAMP response element binding protein (CREB). Conclusions : Our findings suggest that CEO inhalation effectively restored the SCO-impaired cognitive functions in C56BL/6 mice. This learning and memory enhancing effect of CEO was partly mediated by up-regulation of BDNF via activation of CREB.

ECPS: Efficient Cloud Processing Scheme for Massive Contents (클라우드 환경에서 대규모 콘텐츠를 위한 효율적인 자원처리 기법)

  • Major IT vendors expect that cloud computing technology makes it possible to reduce the contents service cycle, speed up application deployment and skip the installation process, reducing operational costs, proactive management etc. However, cloud computing environment for massive content service solutions requires high-performance data processing to reduce the time of data processing and analysis. In this study, Efficient_Cloud_Processing_Scheme(ECPS) is proposed for allocation of resources for massive content services. For high-performance services, optimized resource allocation plan is presented using MapReduce programming techniques and association rules that is used to detect hidden patterns in data mining, based on levels of Hadoop platform(Infrastructure as a service). The proposed ECPS has brought more than 20% improvement in performance and speed compared to the traditional methods.

  • Due to the rapid development and dissemination of 5G communication and IoT technologies, there are increasing demands for big data analysis techniques and service systems. In particular, explosively growing demands on AI technology adoption are also causing high competitions to take advantages of machine/deep-learning models to extract novel values from enormously collected data. In order to adopt AI technology to various research and application domains, it is necessary to prepare high-performance GPU-equipped systems and perform complicated settings to utilze deep learning models. To relieve the efforts and lower the barrier to utilize AI techniques, AIaaS(AI as a service) platform is attracting a great deal of attention as a promising on-line service, where the complexity of preparation and operation can be hidden behind the cloud side and service developers only need to utilize the high-level AI services easily. In this paper, we propose an AIaaS system which can support the creation of AI services based on Docker and OpenFaaS from the registration of models to the on-line operation. We also describe a case study to show how AI services can be easily generated by the proposed system.

  • Recently, there are rapid development of information and communication and rapid growth of e-business users. Therefore we try to solve security problem on the internet environment which charges from wire internet to wireless internet or wire/wireless internet. Since the wireless mobile environment is limited, researches such as small size, end-to-end and privacy security are performed by many people. Wireless e-business adopts credit card WPP protocol and AIP protocol proposed by ASPeCT. WAP, one of the protocol used by WPP has weakness of leaking out information from WG which conned wire and wireless communication. certification chain based AIP protocol requires a lot of computation time and user IDs are known to others. We propose a Micro-Payment protocol based on credit card. Our protocol use the encryption techniques of the public key with ID to ensure the secret of transaction in the step of session key generation. IDs are generated using ECC based Weil Paring. We also use the certification with hidden electronic sign to transmit the payment result. The proposed protocol solves the privacy protection and Non-repudiation p개blem. We solve not only the safety and efficiency problem but also independent of specific wireless platform. The protocol requires the certification organization attent the certification process of payment. Therefore, other domain provide also receive an efficient and safe service.

Cognitive improvement effects of Momordica charantia in amyloid beta-induced Alzheimer's disease mouse model (여주의 amyloid beta 유도 알츠하이머질환 동물 모델에서 인지능력 개선 효과)

  • Accumulation of amyloid beta (Aβ) and oxidative stress are the most common reason of Alzheimer's disease (AD). In the present study, we investigated the cognitive improvement effects of butanol (BuOH) fraction from Momordica charantia in Aβ25-35-induced AD mouse model. To develop an AD mouse model, mice were received injection of Aβ25-35, and then orally administered BuOH fraction from M. charantia at doses of 100 and 200 mg/kg/day during 14 days. In the T-maze and novel object recognition test, administration of BuOH fraction from M. charantia L. at doses of 100 and 200 mg/kg/day improved spatial ability and novel object recognition by increased explorations of novel route and new object. In addition, BuOH fraction of M. charantia-administered groups improved learning and memory abilities by decreased time to reach hidden platform in Morris water maze test. Oral administration of BuOH fraction from M. charantia significantly inhibited lipid peroxidation and nitric oxide levels in the brain, liver, and kidney compared with Aβ25-35-induced control group. These results indicated that BuOH fraction of M. charantia improved Aβ25-35-induced cognitive impairment by attenuating oxidative stress. Therefore, M. charantia could be useful for protection from Aβ25-35-induced cognitive impairment.
