Anti-inflammatory Effect of Broccoli Leaf Hexane Fraction in LPS-stimulated RAW264.7 Cells

  • 본 연구에서는 브로콜리 잎 헥산 분획물의 항염 효과를 평가하여 기능성 식품 및 화장품 소재로의 적용 가능성을 확인하였다. LPS-자극된 RAW264.7 세포에서 전염증성 사이토카인의 생성, iNOS와 COX-2의 발현, MAPK (ERK, JNK, p38) 및 브로콜리 잎 헥산 분획을 사용한 NF-κB의 인산화를 분석하였다. 브로콜리 잎 헥산 분획은 TNF-α, IL-4, IL-6, IL-1β 등의 전염증성 사이토카인의 분비와 iNOS와 COX-2의 발현을 억제했습니다. 또한, 브로콜리 잎 헥산 분획물은 MAPK와 NFκB의 인산화를 감소시켰다. 따라서 브로콜리 잎 헥산 분획물은 식품 및 화장품에서 천연 항염증 소재로 적용 가능성이 있는 것으로 판단된다. 향후 항염증 기전 및 주요 생리활성 물질의 규명에 대한 연구가 필요할 것으로 생각된다.

추출 용매에 따른 오죽(Phyllostachys nigra Munro) 추출물의 생리활성 효과 (Effect of Solvents of Extraction on the Biological Activities of Phyllostachys Nigra Munro)

  • 오죽(Phyllostachys nigra Munro)을 이용하여 추출용매에 따른 총 페놀함량, 항산화 활성, superoxide dismutase (SOD) 유사활성, 아질산 소거능, tyrosinase활성 저해능, angiotensin-converting enzyme (ACE) 저해활성을 in vitro에서 검토 했다. Ethyl acetate와 hexane을 추출용매로 이용할 때 총 페놀화합물 함량은 각각 $44.1{\pm}2.3mg/kg$$47.3{\pm}2.4mg/kg$였다. 항산화 효과는 추출용매 종류에 따라 각각 다르게 나타났고 hexane > ethyl acetate > n-butanol > methanol > water순이었다. 그러나 SOD 활성은 물 > methanol > n-butanol> hexane> ethyl acetate 추출 순이었다. 아질산 소거능은 pH 1.2에서 추출용매에 따라 약간의 차이는 있었으나 54~69.2% 범위였다. 그러나 pH가 증가함에 따라 추출용매에 관계없이 모두 감소하였다. 특히 pH가 1.2에서 6.0으로 증가 할 경우 ethyl acetate 추출물의 아질산 소가능은 $69.2{\pm}3.1%$에서 $7.8{\pm}2.4%$로 감소하였다. 오죽 추출물의 tyrosinase의 저해 활성은 추출용매에 따라 약간 차이가 있으나 전반적으로 15.2~21.3% 범위였다. 추출용매에 따른 tyrosinase의 저해 활성은 water > methanol > n-butanol > hexane > ethyl acetate 추출 순이었다.

n-Hexane 노출에 의한 신경행동학적 증상에 monoamine oxidase(B)와 serotonin transporter receptor 유전자다양성의 영향 (Genetic polymorphisms of monoamine oxidase(MAO) B and serotonin transporter receptor(5-HTTR) influence variability in neurobehavioral symptoms by n-hexane exposur)

  • Human occupational exposure to n-hexane has been associated with neurobehavioral symptoms such as depression, irritablity, acute irritation symptom, concentration disturbance and fatigue. Effects of monoamine oxidase (MAO) B and serotonin transporter receptor (5-HTTR) polymorphisms on the neurobehavioral symptoms were investigated in 70 male workers from TV and computer monitor manufacturing plants exposed to n-hexane. Neurobehavioral symptoms were assessed through a self-reported questionnaire and ambient level of n-hexane was measured by NIOSH method. Blood and urine were collected from each workers to determine the MAO(B), 5-HTTR and urinary 2,5-hexanedione(2,5-HD). The mean concentration of volatile n-hexane was $18.8{\pm}28.8ppm$ and that of urinary 2,5-HD was $1.07{\pm}1.47mg/g$ creatinine. Statistically significant associations with sexual disturbance were age and smoking. The frequencies of MAO(B) AA, AG and GG were 18.6%, 45.7% and 35.7%, respectively, and the frequencies of 5-HTTR ll, ls and ss genotype were 82.9%, 15.7% and 1.4%, respectively. MAO (B) gene polymorphisms had susceptibility to the neurobehavioral symptoms such as fatigue, concentration disturbance, irritability and acute irritation symptom and 5-HTTR gene polymorphism had susceptibility to the sleep disturbance and acute irritation symptom. On multiple logistic regression analysis for the neurobehavioral symptoms, memory disturbance was significantly associated with smoking(OR=6.752, 95% CI=37.46) and drinking(OR=4.033, 95% CI=1.252-12.98), emotional lability was MAO(B) genotype(OR=0.412, 95% CI=0.170-0.996), fatigue (OR=1.011, 95% CI=1.000-1.021) and acute irritation(OR=0.990, 95% CI=0.981-1.000) were working duration and sexual disturbance were significantly associated with age(OR=1.208, 95% CI=1.042-1.399), ambient n-hexane(OR=1.077, 95% CI=1.005-1.154) and 2,5-HD(OR=0.186, 95% CI=0.041-0.841). This finding implies that the MAO (B) and 5-HTTR polymorphisms may affect susceptibility for specific neurobehavioral symptoms associated with n-hexane exposure in workers.

모과 에탄올 추출물의 항산화 효과 (Antioxidative Effect of Chaenomelis Fructus Ethanol Extract)

  • 본 연구에서는 in vitro에서 모과의 항산화 활성을 평가하기 위하여 용매별 분획물의 항산화능을 검토하였다. 모과에탄올 추출물의 수율은 9.23%이었고, 이를 n-hexane, chloroform, ethylacetate, n-butanol, water로 계통 분획한 수율은 water분획물이 1.52%로 가장 높았으며 그 다음으로 ethylacetate 분획물 1.46%의 순이었다. 모과 에탄을 추출물의 총 flavonoid 함량은 $0.331{\pm}0.021\;mg/mL$이었고, 각 분획물의 총 flavonoid 함량은 n-hexane 분획 물이 $0.653{\pm}0.046mg/mL$로 가장 많이 검출되었다. 모과 에탄올 추출물의 총 polyphenol 함량은 0.279mg/mL이었고, 각 분획물중 n-hexane 분획물이 $0.712{\pm}0.017\;mg/mL$로 가장 많이 함유된 것으로 나타났다. In vitro에서 모과 분획물별 항산화 효과를 측정하기 위해 DPPH radical에 대한 자유기 소거능을 측정 한 결과 n-hexane 분획물이 $27.3{\mu}g/mL$로 가장 강한 항산화 활성을 나타내었으며, 이는 양성 대조군인 합성 항산화제 BHT의 $26.5{\mu}g/mL$와 유사한 전자공여능을 나타냈다. 또한 rancimat으로 측정한 항산화지수도 n-hexane 분획물이 BHT와 유사한 수치를 나타냈으며, 지질과산화물 생성 억제효과도 n-hexane 분획물이 62.5%로 가장 항산화력이 우수하였다. 또한 본 연구팀은 in vivo에서 모과 에탄을 추출물의 항산화 효과를 관찰하였는데, 모과에탄을 추출물과 알코올을4주간 경구 투여한 후 흰쥐의 간 조직 손상에 대한 보호효과를 검토한 결과 알코올 투여로 증가된 유리기 해독계 효소인 superoxide dismutase, catalase 및 glutathione peroxidase 활성은 억제시키고, glutathione 함량은 증가시켰으며 지질과산화물 함량을 저하시켰다. 따라서 모과는 알코올 투여로 손상된 간조직의 보호에 도움을 줄 수 있을 것이라는 가능성을 제시해 주었다(32). 이상의 실험결과 모과에탄올 추출물은 in vivo와 in vitro 연구 모두에서 항산화 효과를 나타내었으며, 이는 모과가 우수한 천연 항산화제로서의 개발가능성을 제시해 주었다.

하늘타리(Trichosanthes kirilowii Max) 재분획물이 Steptozotocin 유발 당뇨 흰쥐의 인슐린 활성에 미치는 영향 및 급성독성에 관한 연구 (The Effect of Trichosanithes kirilowii Max. Subfractions on the Insulin Activity in Streptozotocin Induced Diabetic Rats and Their Acute Toxicity)

  • The effects of Trichosanthes kirilowii Max. subfractions, which has long been used as a wild vegetable and folk medicine, on insulin activity and acute toxicity were investigated. Diabetes melitus was induced in male Sprague-Dawley rats by the injection of streptozotocin(STZ) into the tail vein at a dose of 45mg/kg. The hexane fraction of the plant was subfractioned by silica gel column chromatography and were administered orally for 14 days and the normal and STZ-control group were orally administered with tween 80. The body weight gain was monitored and plasma levels of glucose, cholesterol, triglyceride, free fatty acid HDL-cholesterol were determined. The plasma activities of aspartate aminotransferase(AST) and alanine aminotransferase (ALT) were analysed. The subjraction A of hexane fraction lowered plasma glucose levels significantly and increased insulin activity compared to that of STZ-control. The activities of AST and ALT were decreased by administration of subfractions A and C of hexane fraction. The intakes of the hexane fraction of Trichosanthes kirilowii Max. did not showed the acute toxicity. It is suggested from the results that the subfraction A of the hexane fraction of Trichosanthes kirilowii Max. showed a hypoglycemic effect on diabetic rats and that the subfraction may be beneficial for insulin secreation.

Monascus purpureus P-57 변이주가 생산하는 홍국색소의 항산화효과 (Antioxidant Activity of Monascus Pigment of Monascus purpureus P-57 Mutant)

  • Monascus purpureue KCCM 60016으로부터 색소생성 이 우수한 변이주 P-57이 생성한 홍국색소의 항산화 활성을 조사하였다. 홍국색소는 chloroform에 아주 잘 추출되며, 특히 hexane에서는 황색색소가 특이적으로 높게 추출되었다. DPPH radical소거 효과는 hexane분획, chloroform 분획, ethyl acetate분획, butanol 분획, water 분획의 순으로 높게 나타났다. Xanthine oxidase저해효과는 hexane분획, chloroform분획, ethyl acetate 분획의 순으로 높게 나타났으며, hexane 분획물의 경우 5 ppm에서 $41.2\%$, 50 ppm에서 $82.4\%$로 높은 저해율을 보였으며, 저해기작은 비경쟁적 저해였다.

Activities of Natural Plant Extracts against HIV-1

  • Anti-HIV-1 activities for the extracts (buthanol, hexane, chloroform, and water) of medicinal plants widely used in the folk medicine were evaluated for screening of anti-AIDS agents. The activities of the extracts to inhibit HIV-1 replication were also analyzed. The 50% effective concentration (EC50) of inhibition activity of the p24 production for chloroform extract of Saphora flavescens, chloroform extract of Herba ephedrae, and hexane extract of Pachyma hoelen Rumph showed 5.8, 29.9, and 37.3 2g/ml, respectively, as good activities. Hexane extract of Sophora flavescens, buthanol extract of Tulipa edulis, hexane extracts of Tulipa edulis, Herba ephedra, and Pachyma hoelen Rumph in the 50% cytotoxic concentration ($CC_{50}$) in inhibition activities of recombinant HIV-1 RT showed 12.9, 19.5, 11.6, 12.0, and 36.8 % at concentration of 200 ${\mu}g$/ml, respectively, as good activities. From these results, chloroform extract of Saphora flavescens, chloroform extract of Herba ephedrae, and hexane extract of Pachyma hoelen Rumph were very effective against HIV-1 among all extracts tested. Therefore, we expect these plants will be a useful for anti- HIV-1 therapeutics in future.

국소적 전도차단을 보인 n-Hexane induced neuropathy 1례 (A Case of n-Hexane Induced Neuropathy with Focal Conduction Block)

  • A 32-year-old man who had worked at aluminum processing plant for 4 months visited us. He complained of numbness and paresthesia of both foot and hands and weakness of all extremities. Electrophysiologic study showed motorsensory polyneuropathy of diffuse axonal type with focal conduction block. And we discovered higher concentration of n-Hexane in his workplace. On sural nerve biopsy, bubbly enlarged nerve fibers in light microscope and thick myelin sheath and axonal degeneration on electron microscope were found. We diagnosed it as n-Hexane induced neuropathy.

더덕으로부터 면역세포 활성 성분의 분리 (Isolation of Active Components on Immunocytes from Codonopsis Lanceolatae)

  • The purpose of this research was to isolate an active component on immunoytes from 70% MeOH extract of Codonopsis lanceolatae Radix(CLE). CLE was fractionated successively with hexane, methylene chloride, n-butyl alcohol and water, and then the water fraction was separated with molecularporous membrane tubing(m.w. 3,500). Each fraction(50mg/kg) was administered p.o. once a day for 7 days in BALB/c mice respectively. None of these fractions affected the apoptosis and mitochondrial transmembrane potential in thymocyte. Hexane and methylene chloride fractions decreased CD4$^{[-10]}$ CD8$^{+}$ single-positive cells, and the water fraction enhanced CD4$^{+}$ CD8$^{[-10]}$ single-positive cells in thymocyte. The proliferation of thymocytes was decreased by the fraction hexane, but was enhanced by the water fraction. Hexane, methylene chloride and butyl alcohol fractions suppressed the production of nitric oxide, which was not affected by the water fraction. Hexane and butyl alcohol fraction suppressed the phagocytic activity, but water fraction enhanced it. The components(m.w. 3,500 above) separated from the water fraction enhanced the proliferation of thymocyte, the population of CD4$^{+}$ CDB$^{[-10]}$ single-positive cells, and phagocytic activity in macrophage. These results suggest that the stimulative components of proliferation, TH population and phagorytc activity is in the water fraction, and the molecular weight is 3,500 above. Hepatoprotective Effect of Stamen Extracts of Mesua ferrea L. against Oxidative Stress induced by $CCl_4$ in Liver Slice Culture Model

  • Stamens of Mesua ferrea L. are a well-known herbal drug used in Indian System of Traditional Medicine to treat various diseases. The claimed activity of this plant part is necessitated to investigate antioxidant and hepatoprotective activity. Authenticated plant sample was extracted with hexane, ethanol (EtOH) and water (aq.) using ASE 100 accelerated solvent extractor. Antioxidant activity was evaluated by means of different in vitro assays. Hepatoprotective effect was investigated on carbon tetrachloride induced oxidative stress in liver slice culture model. Cytotoxic marker lactate dehydrogenase (LDH) released in culture medium and the activity of lipid peroxidation along with antioxidant enzymes (AOEs) namely superoxide dismutase (SOD), catalase (CAT) and glutathione reductase (GR) were estimated. Hexane and EtOH extracts were significantly inhibited DPPH, NO, SOD and $ABTS^+$ radical in dose dependent manner. The trade of phenol content was: aq. extract < hexane extract < EtOH extract. A significant correlation was shown by total phenol content and free radical scavenging activity of extracts. The culture system treated with hexane extract, EtOH extract or ascorbic acid exhibited significant depletion in LDH, lipid peroxidation, antioxidative enzymes SOD, CAT and GR. Hexane extract and EtOH extracts of stamen of M. ferrea protected liver slice culture cells by alleviating oxidative stress induced damage to liver cells.
