기체크로마토그래피/질량분석기에 의한 수질, 토양 및 저질 시료중의 benzophenone 분석법에 관한 연구 (Determination of benzophenone in water, soil and sediment by gas chromatography/mass spectrometry)

  • 본 연구는 GC/MS를 이용하여 수질, 토양 및 저질 시료 중의 benzophenone (BP)을 분석하는 방법을 확립하고자 하였다. BP는 수질 시료 (100 mL)에서 n-hexane으로 추출하였으며, 토양 및 저질 시료 (10 g)에서는 먼저 메탄올로 추출한 후 hexane으로 다시 추출하여 농축시켜 분석하였다. 수질 시료 중의 BP 회수율은 71.4% 이상이었으며 토양에서의 회수율은 86.5-94.7%를 보였고 재현성은 19.8% 이하였다. 검정곡선은 상관계수 ($r^2$) 값이 수질과 토양 모두에서 0.998이상의 좋은 직선성을 보여주었다. 수질 시료의 경우 43지역 중 3지역의 수질에서 30-200 ng/L 농도 범위로 BP가 검출되었으며 토양과 저질 시료에서는 모든 지역에서 검출되지 않았다. 이 분석방법은 환경 중에 미량으로 존재하는 BP의 분석과 모니터링에 유용하게 사용할 수 있는 적합한 방법이라 사료된다.

청파활성분획이 허혈성 뇌손상에 미치는 실험적 연구 (Experimental Study of Fraction of Citri Reticulatae Viride Pericarpium extract on the Brain Damage in Cerebral Ischemia)

  • Citri Reticulatae Viride Pericarpium extract(CRVP) have been used in oriental medicine for many centuries as a therapeutic agent for smoothing the liver and regulating the circulation of qi, and promoting digestion and removing stagnated food. The effects of CRVP on the inhibition of brain damage in cerebral ischemia is not known. Therefore, this Study was designed to investigate the cerebral protective effects of CRVP on the transient cerebral ischemia using modern techniques, and further to provide the possibility of scientification of oriental medicine. The size of cerebral infarct size was measured by morphometry, and brain edema was measured by morphometry and brain water content determination. The results were a$ follows ; 1. Water fraction of CRVP was reduced infect area of rats brain slices which were subjected to a transient cerebral ischemia in a dose-dependent manner. 2. Methylene chloride fraction and hexane fraction of CRVP was significantly reduced infarct area of rats brain slices which were subjected to a transient cerebral ischemia in a dose-dependent manner. 3. Methylene chloride fraction and hexane fraction of CRVP was significantly reduced infarct volume of rats brain which was subjected to a transient cerebral ischemia in a dose-dependent manner. 4. Methylene Chloride fraction and hexane fraction of CRVP was significantly decreased brain edema induced by a transient cerebral ischemia in a dose-dependent manner. 5. Methylene chloride fraction and hexane fraction of CRVP was significantly decreased brain water content of rats which were subjected to a transient cerebral ischemia. It is suggested that CRVP has an anti-ischemic effect through the inhibition of brain damage in a transient cerebral ischemia, and that in future further development of main effective constituent in CRVP can provide a novel therapeutic strategy for cerebral ischemia.

Immunomodulatory Effects of Hexane Insoluble Fraction of Ficus septica Burm. F. in Doxorubicin-treated Rats

  • The use of chemotherapeutics induces cardiotoxicity and affects immune functions, therefore development of combinatorial agents against cardiotoxicity and immunosuppression needs to be explored. Previous studies of the hexane insoluble fraction (HIF) of an ethanolic extract of Ficus septica leaves showed anticancer effects singly and in combination with doxorubicin on T47D breast cancer cells. In this present study, it was evaluated for its immunomodulatory activities in doxorubicin-treated rats. Thirty male Sprague Dawley rats were divided into five groups consisting of six rats each as follows: Group 1, receiving oral saline 10 ml/kg BW (control group); Group 2, receiving HIF dose 750 mg/kg BW orally, once daily; Group 3, receiving HIF dose 1.500 mg/kg BW orally, once daily; Group 4, given oral saline 10 ml/kg BW (normal group); Group 5, receiving HIF dose 1.500 mg/kg BW orally, once daily. The rats of group 1-3 were intramuscularly administered with doxorubicin at a dose of 4.67 mg/kg BW at the days 1 and 4 to suppress immune functions. Concomitantly, the rats were treated with saline or HIF for seven consecutive days (1 to 7). Treatment of HIF succeeded in reducing side effects of doxorubicin based on increasing lymphocyte density and phagocytosis activity and capacity of macrophages, as well as increasing the CD8+ blood level and decreasing spleen IL-10 expression. Hexane insoluble fraction of of ethanolic extract of Ficus septica leaves has potential as a protective agent combined with doxorubicin.

Frontal Analysis와 Pulse Input Method를 이용한 Loxoprofen의 등온흡착식 결정 (Determination of Loxoprofen Adsorption Isotherms by Frontal Analysis and Pulse Input Method)

  • 본 실험의 목적은 frontal analysis(FA)와 pluse input method(PIM) 방법으로 흡착분리공정에 있어서 중요한 전 단계의 하나인 등온흡착식을 결정하는데 있다. FA를 수행하기에 앞서 대상물질이었던 loxoprofen의 용해도를 측정하여 hexane/ethanol=95/5일 때 시료의 농도를 $0.2{\sim}3mg/ml$로 결정하고, pH조정을 위한 acetic acid는 0.5%로 결정하였다. 이동상의 주성분인 hexane의 조성이 증가함에 따라 용해도는 감소하였지만 분리도는 증가하는 경향을 확인하였다. 이런 용해도와 분리도 사이의 trade-off 또한 흡착분리과정에서의 조작변수가 될 수 있음을 의미한다. FA로 구한 이성분계 등온흡착식은 $$C_{S,re}=\frac{5.282C_{M,re}}{1+0.1412C_{M,re}+0.2139C_{M,ex}}$$, $$C_{S,ex}=\frac{6.413C_{M,ex}}{1+0.2139C_{M,ex}+0.1412C_{M,re}}$$로 결정하였고, PIM으로 구한 이성분계 등온흡착식은 $$C_{S,re}=\frac{5.245C_{M,re}}{1+0.1667C_{M,re}+0.2054C_{M,ex}}$$, $$C_{S,ex}=\frac{6.061C_{M,ex}}{1+0.2054C_{M,ex}+0.1667C_{M,re}}$$로 결정 하였다.

목향 추출물의 항산화 및 항암활성 (Antioxidative and Antitumor Activity of Extracts from Saussurea lappa)

  • This study was carried out to investigate the antioxidative and antitumor activities of Saussurea lappa for the purpose of developing a functional food. The methanol extract of Saussurea lappa was fractionated with five solvents (hexane, chloroform, EtOAc, BuOH, water) and examined for antioxidative activities and xanthine oxidase inhibitory activity in addition to growth inhibitory activity of human cancer cell (HT-29, SNU-1, HeLa). Total phenol compound contents were higher in EtOAC fraction than other fractions. Also, electron donating ability was over 90% at $500{\mu}g/ml$ (93.1 %) and $1000{\mu}g/ml$ (94.3%). The hexane fraction revealed stronger nitrite scavenging ability than the positive control (ascorbic acid) and its abilities were 22.4% and 42.8% at $500{\mu}g/ml$ and $1000{\mu}g/ml$, respectively. Xanthine oxidase inhibitory activity had similar tendency to electron donating ability. The EtOAc fraction showed high inhibition to xanthien oxidase activities at $500{\mu}g/ml$(81.9%) and $1000{\mu}g/ml$(90.4%). In the antitumor activity test, the hexane fraction exhibited potent growth inhibition activity against HT-29, SNU-1 and HeLa cells. Therefore, we believe that Saussurea lappa can be developed into a functional food with antioxidant activity. Additional studies are required for the hexane and chloroform fractions of Saussurea lappa to develop them into therapeutic supplements for stomach cancer, colon cancer, and cervical cancer.

초석잠의 잎 추출물의 항균 활성 (Antimicrobial activity of the hexane extract of Stachys sieboldii MIQ leaf)

  • 초석잠 헥산 추출물이 식중독균에 대한 천연 항균 활성 가능성을 검토하였다. 초석잠 헥산 추출물의 항균 작용을 알아보기 위하여 공시 균주인 Bacillus cereus, Staphylococcus aureus, E, coli 및 Listeria monocytogenes등에 대하여 disc법으로 실험한 결과 그람음성 및 그람 양성균에 대하여 15~20 mm의 clear zone이 나타났다. 초석잠 헥산 추출물의 MIC는 Bacillus cereus에 대해 250${\mu}g$/$m\ell$ Listeria monocytogenes에 대해 250~500/$m\ell$그리고 Staphylococcus aureua와 Pseudomonoas aeruginosa에 대해 각각 500${\mu}g$/$m\ell$ 나타내었다. 초석잠 추출물을 처리한 전자현미경(TEM)상에서는 처리균주들의 세포막이 파괴되어 세포내용물이 용출된 것을 볼 수 있었다. 초석잠 추출물을 처리한 균주의 세포막 손상의 정도를 알아보기 위하여 균체내 효소인 $\beta$-galactosidase 활성을 측정한 결과 클로로포름보다 세포막을 더 손상시키는 것이 확인되었다. 그리고 초석잠 추출물의 pH 및 열 안정성을 실험한 결과 매우 안정하였다.

Hexane and Chloroform Fractions of Laetiporus sulphrueus var. miniatus Inhibit Thrombin-treated Matrix Metalloproteinase-2/9 Expression in Human Oral Squamous Carcinoma YD-10B Cells

  • Laetiporus sulphrueus var. miniatus is widely distributed worldwide, and has commonly been used as a medicinal mushroom. In the present study, we investigated the effects of water extract and solvent fractions from the Laetiporus miniatus as possible antioxidant, anti-thrombin and anti-invasive agents against phorbol 12-myristate 13-acetate (PMA)- or thrombin-induced matrix metalloproteinase-2 (MMP-2) and MMP-9 activities. Samples were fractionated into n-hexane, $CHCl_3$, ethyl acetate, n-butanol, and water fractions, and individually analysed. The water fraction had the highest extraction yield at 34.90% (w/w), while the n-butanol fraction demonstrated the highest anti-oxidative activity at 81.44%. In the thrombin inhibitory activity test, the water fraction exhibited the highest activity at 94.64%. Even at the concentration of $40{\mu}g/mL$, evaluation of anti-proliferating activity in YD-10B cells did not reveal any cytotoxic effects. Although MMP-9 expression in YD-10B cells increased after the addition of PMA and thrombin, MMP-2 did not. Additionally, MMP-2/-9 levels in PMA-treated YD-10B cells (i.e., both mRNA expression and protein activation) were highly inhibited in the hexane and chloroform fractions. Compared with MMP-2 levels, MMP-9 mRNA expression and proteolytic activity were inhibited to a greater extent by the hexane and chloroform fractions in thrombin-treated YD-10B cells. Taken together, these results support that thrombin induces tumor invasion through MMP-2/9 and suggest that the L. miniatus may act as an effective functional food, conferring anti-oxidative, anti-thrombotic and anti-cancer activities.

배암차즈기 추출물의 세포독성과 항균효과 (Cytotoxicity and Antimicrobial Effect of the Extract of Salvia plebeia)

  • The comparison of $IC_{50}$ values of Salvia plebeia R. Br. extracts on L1210, $P388D_1$ cancer and Vero normal cell lines showed that the n-hexane soluble extract of S. plebeia R. Br. retains the most growth-inhibitory activity against tumor cell lines. The minimum inhibitory concentration (MIC) of the extract against microorganisms were also examined. Antimicrobial activity of amocla and ketoconazole as references was compared to those of other solvent extracts such as $H_2O$, n-hexane, chloroform, ethyl acetate and methanol. The antimicrobial activity of extract had growth inhibition activity against gramnegative bacteria, gram-positive bacteria and fungi $(MIC\;>\;200\;{\mu}g/ml)$ except for n-hexane extract. Seven bacterial strains were tested for in vitro susceptibility to the extract of S. plebeia R. Br. However the n-hexane extract of S. plebeia R. Br. inhibited the growth of several bacterial strains (MIC values between 100 and $200\;{\mu}g/ml$ for gram positive bacteria, $25\;{\mu}g/ml$ for P. putida).

두유박과 사과쥬스박을 이용한 식이섬유원의 제조 (Preparation of dietary fiber sources using apple pomace and soymilk residue)

  • 식물성 섬유질 함유 자원으로부터 식품의 가공적성을 떨어뜨리지 않으면서 우수한 기능특성을 보이는 식이섬유원을 제조할 목적으로 대량 수급이 용이한 사과쥬스박과 두유박에 식이섬유의 함량을 높이고 기능성을 향상시키기 위한 각종 처리를 행하여 식이섬유원을 제조하였다. 사과쥬스박은 마쇄, 수세 및 사별에 의하여 사과육과 사과피로 분리할 수 있었는데 사과육이 사과피보다 기능성이 우수하였고 사과육은 수세, n-hexane 처리 등에 의해 WHC 11.27 g/g, FHC 2.01 g/g, CEC 64.96 meq/100g의 기능성을 보이며 60.94%의 NDF를 함유한 제품을 제조할 수 있었다. 두유박을 이용한 식이섬유원 제조에서는 수세, n-hexane처리, 산처리, 알칼리처리 등의 처리로 WHC 10.15 g/g, FHC 1.52 g/g, CEC 63.72 meq/100g의 기능성을 보이며 52.66%의 NDF 함량을 갖는 식이섬유원이 제조되었다. 이때 원료로부터의 회수율은 사과육의 경우 약 30%, 처리두유박의 경우는 38.2%이었다. 식이섬유질원의 색도는 수세시 명도를 효과적으로 증가시킬 수 있었으나 유기용매, 산 및 알칼리 처리에 의해서는 명도가 감소하였다.

사과와 토양 중에서 Difenoconazole의 잔류성에 대한 기체 크로마토그래피 분석 (Gas Chromatographic Analysis on Residual Difenoconazole in Apple and Soil)

  • 기체 크로마토그래피법을 이용하여 과수 재배용 살균제인 difenoconazole의 분석을 위한 최적 조건을 구하고, 사과와 토양 중에서의 잔류성을 조사하였다. 사과사료는 5% NaCl과 n-hexane액으로 분리, 농축하여 florisil column상에서 acetone과 n-hexane 혼합액으로 정제하여 GLC(ECD)로 분석하였다. 0.20과 1.0ppm의 표준물 첨가실험결과 평균회수율은 86.0~92.0% 였고, 검출한계는 0.01ppm 이었다. 사과에 대한 안정 사용기간은 수확 15일 전 3회 사용이 적당한 것으로 나타났으며, 이 경우 difenoconazole의 잔류량은 0.037ppm에서 0.044ppm 이었다. 토양시료를 methanol과 ammonium hydroxide 의 혼합액으로 추출하여 여과한 후 포화 NaCl 용액과 dichloromethane층으로 분배하였다. 유기층을 농축하여 toluene 용매로 재용해하였고, Sep-Pak column으로 정제한 후 GLC(FID)로 분석하였다. 0.10, 0.50과 1.0ppm 표준물 첨가실험 결과 평균 회수율은 101.2~103.7% 였고, 검출한계는 0.025ppm 이었다.
