Effects of Solvent Extraction by Immersion on the Quality and Storage Stability of Rice (용매침지(溶媒浸漬)에 의한 탈지(脫脂)처리가 쌀의 품질(品質) 및 저장성(貯臧性)에 미치는 영향)

  • Effects of solvent extraction by immersion on the quality and storage stability of Korean rice were studied. Proportions of lipid extracted from whole grain of rice by immersing into two volumes(v/wt) of hexane and ethanol for 72 hours at room temperature were 0.41% and 0.38% respectively. Small changes of water content and hardness of rice were observed by solvent treatment. Cooking characteristics; that is, water-uptake ratio. extended volume, total solid, and starch-iodine blue test of rice was markedly changed by ethanol treatment, while little changes were observed by hexane treatment. No considerable differences in moisture sorption isotherm of rice were observed by both solvent treatments. Changes in TBA number and stale flavor appearance of rice treated with or without solvent immersion during storage at $60^{\circ}C$ showed that rice treated with hexane had best storage stability compared to ethanol treatment, while ethanol treatment of rice had better storage stability than no treatment. Similar results were noted in changes of the flavor score of cooked rice samples which were freeze dried.

Growth enhancement and cytotoxicity of Korean mistletoe fractions on human cell lines (한국산 겨우살이 분획물의 면역세포의 생육증진 및 세포독성)

  • The biological activities on human immune and cancer cell lines of the four kinds of Korean mistletoes (Korean Viscum album, var. coloratum, : Korean Viscum sp. in Quercus acutissima Carr., Korean Viscum sp. in Castanea crenata, Korean Viscum sp. in Betula platyphylla, and Korean Viscum sp. in Salix koreensis) extracts were investigated. The extracts were preparated with ethanol, and fractionated with n-butanol, ethyl acetate, chloroform, hexane, and second distilled water. Cytotoxic potencies of the fractions on human normal lung cell line (HEL 299) showed under 28% in the concentration of 0.5 mg/ml. Growth inhibition effect of the Korean mistletoe extracts on the several human cancer cell lines depends on the concentration of the extracts, and extracting solvent. The hexane, chloroform, and ethyl acetate fractions indicated a strong anticancer activity, but not in aqueous and butanol fractions. Some mistletoe fractions have a different characteristic on the cancer cell lines. Stimulation on the growth of human immuno cell lines(B cell : Raji, T cell: Jurkat) of the extracts were confirmed in the ethyl acetate, chloroform, hexane fractions, but not in aqueous system.

Growth Characteristic, Mono-strain Mass Culture and Antioxidant Effects of Two Benthic Diatoms Amphora coffeaeformis and Achnanthes longipes from Korea

  • Amphora coffeaeformis and Achnanthes longipes are commonly found as dominant benthic microalgae in Jeju coastal water throughout the year. In order to investigate pharmaceutical uses of these diatoms, each single species was isolated with micropipette under phase contrast microscope and subcultured with synthetic seawater media which was enriched with F/2 media, trace metal solution and $Na_2SiO_3$). Growth characteristics of these species were also determined with different combination of salinity, nutrients concentration and temperature. Thereafter, mass culture of each species was done based on the maximum growth condition. Biomass was collected after two weeks of mass culture and freeze dried for antioxidant study. The antioxidant properties of different fractions (n-hexane, chloroform and ethylacetate) obtained by solvent fractionation of 80% methanolic extract of two microalgae were investigated for free radical, reactive oxygen species scavenging (Super oxide, Hydrogen peroxide, Hydroxyl radical and Nitric oxide), metal chelating and lipid peroxidation inhibition activities. All fractions of A. longipes showed higher $DPPH^{\cdot}$ (free radical) scavenging activities (n-hexane: 89.0%, Chloroform: 76.0%, Ethylacetate: 66.0%, Methanol: 90.6% and aqueous residue: 63.0%). N-hexane fraction of A. longipes showed significantly higher activity (49.0%) on nitric-oxide. Ethylacetate fraction of A. longipes and aqueous residue of A. coffeaeformis exhibited 64.0% and 75.6% metal chelating activity which was higher than commercial antioxidants (${\alpha}$-tocopherol: 18.0% and BHT: 16.0%). The n-hexane fraction of A. coffeaeformis had 67.5% activity on $DPPH^{\cdot}$. Chloroform and n-hexane fractions of A. coffeaeformis exhibited 46.2% and 47.6% $H_2O_2$ scavenging effects which were closely similar to commercial antioxidants (${\alpha}$-tocopherol: 49.2% and BHT: 58.6%). Chloroform and ethylacetate fractions of A. longipes and fraction of n-hexane and chloroform of A. coffeaeformis showed better lipid peroxidation activities than ${\alpha}$-tocopherol. These data suggest that both organic and aqueous fractions have good antioxidative compounds with different antioxidant properties.

  • The surface tension of PO added sodium poly(oxyethylene(EO), oxyisopropylene(PO)) dodecyl ether sulfate firstly were slightly lower than EO added sulfate in the concentration range of $10^{-6}{\sim}10^{-3}mol/{\ell}$. And they had lower critical micelle concentration ($10^{-4}{\sim}9{\times}10^{-5}mol/{\ell}$) than general anionic surfactants. The adsorptivity ($2.2{\times}10^{-10}mol/cm^2$) of sodium $(PO)_{10}(EO)_5$ dodecyl ether(compound of PO addition firstly) calculated by Gibbs' adsorption isotherm were higher than that of sodium $(EO)_{10}(PO)_5$, dodecyl ether(compound of EO addition firstly), but were lower than that of sodium dodecyl sulfate (${\Gamma}=3.2{\times}10^{-10}mol/cm^2$). These could be understood that the adsorption areas of compounds were very large because of their high molecular weight. Moreover, PO compounds showed better properties than EO compounds in foamability, emulsifying power for organics (n-hexane, benzene), detergency for the lard, tallow oil mixture and dispersability for iron oxide. It was interpreted in terms of surface properties of the PO compounds. These showed that the interfacial activity become higher when hydrophilic and hydrophobic portion existed in aggoromerated state respectively. The test results of emulsifying power for organics (n-hexane, benzene) showed better for benzene than n-hexane. Eight kinds of sodium (EO, PO) dodecyl ether derivatives showed irregular dispersibilities for polar iron oxide in water dispersed media.

Effect of Exogenous Trehalose on the Solvent Tolerance of Pseudomonas sp. BCNU 106 (유기용매 내성 Pseudomonas sp. BCNU 106 균주의 외인성 트레할로스의 영향)

  • To some extent, the growth of solvent-tolerant Pseudomonas sp. BCNU 106 is limited by toxic solvents. Therefore, various strategies to overcome this limitation need to be investigated. One such strategy is to use exogenous trehalose. The highest intracellular trehalose content of 181.88 mM was measured at 12 hr. The extracellular trehalose content decreased rapidly within 12 to 16 hr in the presence of cyclohexane. Moreover, the number of Pseudomonas sp. BCNU 106 cells grown in Luria-Bertani (LB) broth supplemented with 0.1 M trehalose in the presence of 1%(v/v) cyclohexane, hexane, propylbenzene, and m-xylene increased 89.94-, 89.72-, 91.25-, and 118.9-fold, respectively, in comparison to the control level. High survival rates of 80% and 90% were observed in the presence of cyclohexane and hexane by the addition of 0.05 M trehalose for up to 4 hr, respectively. Exogenously-added trehalose was transported into the cells, and it conferred protection against cyclohexane, hexane, propylbenzene, and m-xylene. Adding exogenous trehalose to the growth medium improved the tolerance of Pseudomonas sp. BCNU 106; thus, it is a potential biocatalyst for biotransformation and biodegradation.

Full-scale Soil Washing and Non-discharged Washing Water Treatment Process of Soil Contaminated With Petroleum Hydrocarbon (현장규모의 유류오염 토양세척 및 무방류 세척 유출수 처리 공정)

  • A non-discharged system of sequentially physico-chemical water treatment was used to treat the contaminated water produced from washing system of soils according to full-scale soil washing. After washing the TPH contaminated soils, the remaining concentrations of COD$_{Mn}$, SS, and n-hexane were analyzed for each compartment to estimate the treatment efficiencies of non-discharged system. Three times of sampling events were conducted for 4 different compartments (sediment tank, flocculation tank, oil/water separator, and process-water tank). In addition, soil washing efficiencies and concentrations of each parameter (COD$_{Mn}$, SS, and n-hexane) for process-water tank were analyzed for about 8 months. As results, the average efficiency of soil washing was high to have 95.9%, regardless of the condition of TPH contamination level for soils, as well as the concentrations of COD$_{Mn}$, SS, and n-hexane in the process-water tank were below the regulation limits of the Water Environmental Conserveation Act. Accordingly, the full-scale washing treatment system in this study could make the washing water 100% recycled which lead the system to be environmentally-friendly and economical.

Antimicrobial Activity and Antimutagenesis of Cinnamon (Cinnamomum cassia Blume) Bark Extract (계피추출물의 항균 작용과 항돌연변이원성)

  • In order to develop antimicrobial substances, many kinds of medicinal herbs were extracted with absolute ethanol and then antimicrobial activities against various microorganisms were investigated. Ethanol extract from cinnamon bark showed the strongest antimicrobial activity on the growth of almost all submitted microorganisms. Specially, molds such as Aspergillus sp. and Pencillium sp. were inhibited strongly. Therefore, the crude antimicrobial substance from the ethanol extract was fractionated with various solvents such as n-hexane, chloroform, ethyl acetate, and n-butyl alcohol and then their antimicrobial activities were tested. Among the various solvent fractions from the ethanol extract, n-hexane fraction was the best in antimicrobial activity especially against molds. There were no significant changes in antimicrobial activity of the n-hexane fraction by heat treatment at $100^{\circ}C$ for 60 min or $121^{\circ}C$ for 15 min and by the change of pH 4.0~10.0. We could get the results that the n-hexane fraction of cinnamon bark extract showed not only antimutagenicity but also no mutagenicity by Ames test with Salmonella typhimurium TA 98 and TA 100.

Antioxidant and Mixture Effects of Curry Spices Extracts Obtained by Solvent Extraction (용매에 따른 카레 향신료 추출물의 항산화 효과 및 혼합효과)

  • This study was performed to investigate the antioxidative effects of solvent extracts of eighteen curry spices. Yields of the curry spices extracted by methanol, ethylacetate and hexane were $6.4{\sim}42.9%$, $3.9{\sim}26.2%$ and $1.6{\sim}29.2%$, respectively. Methanol, ethylacetate and hexane extracts of spices were added up to 1,500 ppm in the rice bran oil and antioxidative effects of extracts were tested by Rancimat. Induction periods of spices extracted by methanol were in the order of rosemary(33.5hr)>sage(29.1hr)>ginger(28.6hr)>turmeric(26.9hr)>nutmeg(25.8hr)>oregano(25.6hr)>thyme(25.3hr)>BHT(24.5hr)>control(22.6hr). Those of ethylacetate extracts were as follows: rosemary(43.9hr)>sage(30.9hr)>oregano(29.0hr)>thyme(27.5hr)>ginger(27.1hr)>BHT(24.5hr)>control(22.6hr). Those of hexane extracts were in the order of rosemary(47.4hr)>sage(31.4hr)>oregano(27.3hr)>ginger(25.5hr)>thyme(25.1hr)>control(22.6hr). Solvent extracts of rosemary, sage, oregano, ginger and thyme showed significant antioxidative effects. Two thousands ppm hexane-extract of rosemary was the most effective. Addition of hexane-extracts of rosemary didn't show any synergism with the hexane extracts of sage, oregano, ginger, and thyme.

Antioxidative and Free Radical Scavenging Effects of Rhus javanica Linne (오배자의 항산화 및 자유라디칼 소거효과)

  • This study was carried out to investigate the antioxidative activities of medicinal plants. Through the examination of methanol extracts from 30 species for radical scavenging effects using DPPH method, the extracts from Rhus javanica Linne, Smilax china Linne and Polygonum cuspidatum Siebold et Zucarinii showed strong antioxidative activity. Because of its highest antioxidative activity among 30 medicinal plants, radical scavenging effects of 4 different extract compartments (n- Hexane, EtOAc, BuOH and $H_2O$ extracts) from Galla Rhois MeOH extract of Rhus javanica Linne were examined by DPPH method and antioxidant effects on the 4 different extract compartments were tested by Ferric-Thiocyanate method. Antioxidative activities of n- Hexane, EtOAc and BuOH extracts were similar or even higher than that of natural (tocopherol) or synthetic antioxidants (BHA), suggesting that major fractions for the antioxidative activity of Rhus javanica Linne were the n- Hexane, EtOAc and BuOH extract compartments.

Cholesterol-Lowering Effect of Pine Nut in Plasma of Rats (잣 성분의 혈중 콜레스테롤 저하효과)

  • Cholesterol-lowering effect of pine nut on mice was investigated by examining changes in body weight, feed intake, and triglyceride, total cholesterol, high density liporotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), and very low density lipoprotein cholesterol (VLDL-C) contents in mice plasma. Changes in body weight and feed intake between pre-designed experimental animal groups fed various contents of cholesterol and pine nut and control group fed basal diet were not significantly different. Food efficiency ratio of experimental groups fed higher than 10% pine nut was significantly higher than that of control group. Apparent differences in atherogenic index representing cholesterol-lowering effect in plasma were not found among all groups fed 0-20% pine nut. Inhibition activities of water and hexane extracts of pine nut on HMG-CoA reductase and cholesterol esterase were examined in vitro. Hexane extract showed 66% inhibition effect on HMG-CoA reductase, whereas none was observed with water extract.
