Nine compounds were isolated from the fruit Citrus junos. Their structures were elucidated as 9-hydroxy-4-methoxypsoralen, auraptene, limonin, deacetylnomilin, cirsimaritin, narirutin, naringin, hesperidin and neohesperidin by physico-chemical evidences. 9-Hydroxy-4-methoxypsoralen and auraptene have not been reported from C. junos yet.
The antibacterial and antifungal effect of grapefruit seed extract(GFSE) was investigated for its purpose of application to a diverse spectrum of field as sanitizers, disinfectants and preservatives. GFSE showed coparatively high content of such flavoniods as naringin and hesperidin and ascorbic acid. GFSE containing a low level of organic acids is a heavy viscous and water-soluble liquid. As a result of the antimicrobial test of GFSE, Bacillus subtilis and Aspergillus oryzae did not survive at detectable levels when treated with more than 100 ppm of GFSE. The minimum inhibitory concentrations of GFSE for a wide variety of pathogenic and putrefactive bacteria, fungi and yeasts were 100 ppm and 250 ppm, respectively. In the comparable electron micrograph of microbial cells treated with GFSE or not, we could conclude that GFSE destroy microorganisms by disrupting the functions of the cell wall membrane and microbial spores.
The essential oil composition by means of gas liquid chromatography, and the occurrence and distribution of flavonoid glycosides in leaves, peels and barks of citrus plants native to Je Ju island were investigated. Results indicate that the occurrence of p-cymene, d,l-limonene, linalool, geraniol and linalyl acetate in the essential oils, and of hesperidin in leaves, peels and barks are fairly common to these species.
The relationships of inhibitory activities of inflammation and wound healing of flavonoids were studied in vitro and in vivo. Generally flavonoids have not only significantly anti-inflammatory activity in carrageenin-induced paw edema and Freund's complete adjuvant-induced arthritis, but also inhibitory activity of wound healing. The more inhibitory activities of wound healing flavonoids have, the more they have the anti-inflammatory activities; apigenin > guercetin > flovone > rutin > hesperidin > naringin. Their inhibitory mechanism seems to be inhibition of the inflammatory cell infiltration and fibroblast proliferation, and so they decreased the granulomatous activity and tensile strength.
The functional characteristics of a ${\beta}$-cyclodextrin glucanotransferase (CGTase) excreted from alkalophilic Bacillus sp. BL-31 that is highly specific for the intermolecular transglycosylation of bioflavonoids were investigated. The new ${\beta}$-CGTase showed high specificities for glycosyl acceptor bioflavonoids, including naringin, rutin, and hesperidin, and especially naringin. The transglycosylation of naringin into glycosyl naringin was then carried out under the conditions of 80 units of CGTase per gram of maltodextrin, 5 g/l of naringin, 25 g/l of maltodextrin, and 1 mM $Mn^{2+}$ ion at $40^{\circ}C$ for 6 h, resulting in a high conversion yield of 92.1%.
Traditional Chinese Medicines (TCM) have attracted great interest in recent researchers as alternative medicines for incurable diseases. This review focuses on qualitative and quantitative analytical approaches for bioactive metabolites of components flavonoids and saponins of traditional Chinese medicines by TLC system, although various methods have been introduced. Emphasis will be put on the processes of metabolite extraction from intestinal bacterial cultures or urines, separation (mobile phase) and detection. The identified metabolites by selection of extraction solvent and detection methods are also discussed. In addition, metabolite determinations of flavonoids (baicalin, apiin, rutin, quercetin, quercitrin, kaempferol, diosmin, hesperidin, poncirin, naringin, puerarin, daidzin, daidzein, tectoridin) and saponins (ginsenosides, kalopanaxsaponins, glycyrrhizin, chiisanoside, saikosaponins, soyasaponins) in culture fluid, in urine and in some herbal formula extracts are summarized. These bioactive metabolites of these components by intestinal microflora should be connected to pharmacological actions.
Processing of medicinal plants is one of the processing methods for reducing of toxicity or improving of effect on medicinal plants. In this study, we studied the changes in the constituents and antioxidant activity in accordance with the processing conditions by the salt water of Citrus unshiu Markovich. Changes in antioxidant activity was measured by the DPPH method and changes in the components were analyzed by high performance liquid chromatography. As a result, the content of the main constituents (narirutin, hesperidin) and total polyphenol were increased by increasing the concentration of salt water. Moreover, antioxidant activity was increased gradually in proportion to the increase of the total polyphenol content.
제주도(濟州道) 재래감귤인 병귤(甁橘) Citrus platymamma $H_{ORT}$. ex $T_{ANAKA}$의 엽(葉), 과피(果皮), 수피(樹皮)의 클로로포롬 가용부(可容部)에서 무색침장정(無色針狀晶), $C_{10}H_{10}O_{4}$, mp $139{\sim}140^{\circ}c$을 단리(單利)하고 이화학적(理化學的) 성상(性狀)및 UV, IR, NMR, Mass spectra를 이용하여 그 화학구조(化學構造)를 구명(究明)한바 1,4-benzenedicar-boxylic acid dimethyl ester이었다. 또 과피(果皮)의 수용부(水溶部)에서 단리(單利)한 2개의 flavonoid 배당체(配糖體)는 그 이화학적(理化學的)인 성상(性狀) 및 IR-spectrum으로 보아 naringin hesperidin이었다.
Previous studies have suggested that Citrus fruits might suppress the proliferation of various cancer cells. However, little is known about any specific ingredients in the extract of Citrus fruits to exert its anti-proliferative activity in cancer cells. The present study aimed to identify the active ingredients in Citrus fruits to suppress the proliferation of rat hepatocellular carcinoma cells. Among tested compounds, two polymethoxylated flavones (nobiletin and tangeritin) showed significant anti-proliferative activity whereas other compounds (synephrine, rutin, hesperidin) did not. Interestingly, nobiletin as well as tangeritin also decreased the protein amount of gluconeogenic enzymes, PEPCK and G6Pase. The possible involvement of gluconeogenic activity in the proliferation of hepatocellulacarcinoma cells are further to be investigated.
감귤류와 감귤류의 과피는 오랜 기간 동양의학의 약재로 사용되어 왔고 최근에는 생과와 주스로 많이 소비되고 있다. 하지만 감귤류 가공 공정 시 많은 과피 부산물이 발생하므로 활용방안이 필요하다. 감귤류의 과피에는 플라바논(flavanone)이 풍부하며 이 플라바논의 형태 중 배당체보다 비배당체의 체내 흡수가 더 효과적이므로 비배당체가 생리적 효과가 더 뛰어나다. Bioconversion(물질전환)은 cytolase에 의해 narirutin과 naringin은 naringenin으로, hesperidin은 hesperetin으로 각각 전환되며, 본 연구는 감귤의 과피에 다량 존재하는 플라바논을 물질전환 한 뒤 면역세포에서 항산화 및 항염증 효과를 확인하였다. 물질전환 후에 감귤 과피 추출물의 전자공여능이 농도 의존적으로 높아지는 것을 확인하였고, 면역세포인 RAW264.7에 200, $500{\mu}g/mL$ 감귤 bioconversion 전(CU) 후(CU-C) 과피 추출물을 4시간 동안 처리한 후 LPS($1{\mu}g/mL$, 8시간)를 처리하였다. 염증 관련 효소인 iNOS와 COX-2의 mRNA와 단백질 발현을 확인한 결과 물질전환과 관계없이 감귤 과피 추출물이 LPS에 의해 유도된 iNOS와 COX-2의 mRNA와 단백질 발현을 농도 의존적으로 감소시켰지만 물질전환 전보다 후 추출물의 억제 효과가 더 높았다. iNOS에 의해 생성되는 NO 역시 감귤 과피 추출물이 LPS에 의해 유도된 NO 생성을 억제시켰다. 본 연구 결과는 감귤 과피 추출물이 항산화와 항염증 생리활성을 보였지만 cytolase를 이용하여 물질전환 한 감귤 과피 추출물이 물질전환 전보다 이러한 생리활성이 강화됨을 보임으로써 향후 감귤류 부산물로 폐기되어온 감귤류 과피가 산화적 스트레스와 염증반응에 기인하는 만성질환을 위한 기능성 소재로 활용이 가능할 것으로 기대된다. 감귤 과피 추출물의 in vitro 상에서 검증된 항염증 효능이 향후에 in vivo 상에서 여러 염증 조직에 미치는 영향에 대한 추가 검증이 필요하다고 사료된다.
