Malignant catarrhal fever (MCF) is a systemic disease of ruminants caused by a gamma herpesvirus, ovine herpesvirus 2 (OvHV-2). Dot blot hybridization (DBH) protocols for detecting and differentiating this MCF virus were developed. OvHV-2 specific primer pairs, 556/555, were used for the amplification of target DNA. Then, the amplified DNA was labeled with incorporation of digoxigenin (DIG). The Dig-labeled probe was able to detect and differentiate specifically OvHV-2 DNA. This DBH technique can be applied to confirm the presence of MCF virus on clinical samples and to differentiate specifically between OvHV-2 infection and other viral infections.

Four 1-year old goats (Capra hircus), which were infected with MCF virus, OvHV-2, by being housed together with MCF virus-infected seep, were referred with a I-month history of chronic dermatitis. On the other hand, MCF virus-negative goats, which were isolated for negative control, had not those kinds of skin problems. Examination of the affected goats revealed generalized alopecia, patchy erythema, and superficial erosions with histologic evidence of mural folliculitis. Fungal culture tests and external parasite tests with the scraping skin samples were negative. However, polymerase chain reaction revealed the existence of MCF virus DNAs in the lesion. These results suggested that MCF virus may induce mural folliculitis and alopecia in goat.
We previously reported that the equine herpesvirus type 1(EHV-1) immediate-early protein(IE protein) physically interacts with the general transcription factor human TFIIB(Jang et al, J Virol 75:10219-10230, 2001). The interaction between the IE protein and TFIIB is necessary for the IE protein to efficiently transactivate the early TK and late IR5 EHV-1 promoters. A panel of deletion and truncation mutants of the TFIIB gene was constructed and employed in protein-binding assays to map the IE protein-binding domain within TFIIB. Evidence is presented that the first direct repeat of TFIIB interacts specifically with the EHV-1 IE protein.
Herpesvirus saimiri (HVS), a member of the $\delta$-herpesvirus family, encodes an oncoprotein called Saimiri Transforming Protein (STP) which is required for lymphoma induction in non-human primates. Previous study has shown that STP-C, an oncoprotein of HVS, activates NF-$\kappa$B signaling pathway. However, the detailed mechanism of STP-Cmediated NF-$\kappa$B activation has not been reported yet. We first report that STP-C interacts with TRAF6 protein in vivo and in vitro and further investigation shows that $Glu_{12}$ residue of STP-C is critical for binding to TRAF6. Introduction of ubiquitin together with STP-C augments NF-$\kappa$B activity compared to that of STP-C expression alone. STP-C expression further induces ubiquitination of endogenous TRAF6. In addition, either a deubiquitination enzyme, CYLD or a dominant negative E2-conjugation enzyme reduced NF-$\kappa$B activity in spite of the presence of STP-C, supporting that the interaction between STP-C and TRAF6 induces ubiquitination of TRAF6. NF-$\kappa$B activation by STP-C through the ubiquitinated TRAF6 causes the increased production of IL-8, an inflammatory chemokine and the enhanced expression of costimulatory molecule ICAM, which might ultimately contribute cellular transformation by the exposure of HVS-infected cells with inflammatory microenvironment and chronic activation.

The presence of ostreid herpesvirus 1 (OsHV-1) and the percentage of viral DNA detected in Pacific oyster Crassostrea gigas adults were investigated monthly between May and November 2012 at three locations along the southern coast of Korea. Among 210 oysters examined by polymerase chain reaction (PCR) analysis, OsHV-1 DNA was detected in only one oyster collected in August. The low detection rate of OsHV-1 DNA was consistent with the lack of reported OsHV-1-associated disease in C. gigas cultured in Korea. The sequence of the present PCR product amplified with the C2/C6 primer pair was identical to that of OsHV-1 ${\mu}Var$ except for one nucleotide, and the sequence amplified with Del36-37F2/Del36-37R showed a 605-bp deletion as in OsHV-1 ${\mu}Var$. Although these sequence data are insufficient to determine genotype, the results suggest that the herpesvirus detected was similar to OsHV-1 ${\mu}Var$. This is the first report on the presence of OsHV-1 in adult Pacific oysters cultured in Korea.
The infection status of domestic farmed eels Anguilla japonica, Anguilla bicolor and Anguilla marmorata with Japanese eel endothelial cell-infecting virus (JEECV) and anguillid herpesvirus 1 (AnHV) was examined at the major eel farming areas in Korea. These viruses were detected in all areas examined, regardless of the eel species or age. Any farm with a history of viral infection in adult fish confirmed the infection to be transmitted to stocked fry within 3 to 5 months. It is proposed that both viruses are horizontally transmitted within a given farm. The primary symptoms and histopathological lesions produced by the two viral infections are similar, making it difficult to distinguish the two diseases through clinical symptoms. Both viruses displayed 100% detection in the gills, suggesting that the gills are an optimal tissue for JEECV and AnHV monitoring. This study concluded that JEECV and AnHV were prevalent on eel farms across the country and caused very high mortality when the two viruses co-infected fry. Additional studies, including experimental infections, are needed to clearly understand the pathogenicity of each virus and the risk of co-infection.
An equine herpesvirus-4 (EHV-4) was isolated in nasal swabs collected in a horse showing respiratory clinical signs. Equine dermis cells inoculated with the sample were observed with characteristic viral cytopathic effects after 3 days of postinoculation and the infected cells exhibited bright intracelluar fluorescence by indirect immunofluorescence assay. At the nucleotide level, the partial glycoprotein B gene of the Korean EHV-4 isolate (K001) had 99.9% identity to 1942 strain (GenBank No. M26171). To author's knowledge, the report describes the first isolation and partial characterization of EHV-4 in Korea. The virus can be used for further study of EHV-4.
Pacheco's disease (PD) is a highly contagious, acute disease of psittacines caused by a herpesvirus. Five 56-day-old dead parakeets (Neophema bourkii) were submitted to the National. Veterinary Research & Quarantine Service for diagnosis. At necropsy, principle lesions were seen in the pancreas, which were hard, whitish, nodular and atrophic resulting in distortion of the duodenal loop. Histologically, lymphocytic pancreatitis with eosinophilic intranuclear inclusions syncytium-formed acinar epithelium was prominent. In the brain, marked focal gliosis and perivascular lymphocytic cuffing were observed. On the electron microscopy examination, various forms of viral particles were found in both the nucleus and cytoplasm of acinar epithelium of pancreas, identified as a herpesvirus. These results were suggested that the cases were diagnosed as PD in the parrakeet.
Pleural effusion lymphoma (PEL) is a rare B-cell lymphoma that has a very poor prognosis with a median survival time of around 6 months. PEL is caused by Kaposi's sarcoma-associated herpesvirus, and is often co-infected with the Epstein Barr virus. The complement system is fundamental in the innate immune system against pathogen invasion and tumor development. In the present study, we investigated the activation of the complement system in PEL cells using human serum complements. Interestingly, two widely used PEL cell lines, BCP-1 and BCBL-1, showed different susceptibility to the complement system, which may be due to CD46 expression on their cell membranes. Complement activation did not induce apoptosis but supported cell survival considerably. Our results demonstrated the susceptibility of PEL to the complement system and its underlying mechanisms, which would provide insight into understanding the pathogenesis of PEL.
