Development of Bialaphos-Resistant Transgenic Rice Using Agrobacterium tumefaciens (Agrobacterium tumefaciens를 이용한 bialaphos 저항성 형질전환 벼의 개발)

  • The bialaphos is a potent inhibitor of glutamine synthease in higher plants and is used as a non-selective herbicide. We have used the bialaphos resistant gene(Bar) encoding for an acetyltransferase isolated from Streptomyces hygroscopicus SF1293. Callus derived from mature seeds of rice(Oryza sativa L. cv. Dong Jin) were co-cultivated with Agrobacterium tumefaciens EHA101 carring a plasmid pGPTV-HB containing genes for hygromycin resistance (HygR) and Bar. Transgenic plants showing in vitro resistance to 50 mg/L hygromycin and 10 mg/L bialaphos were obtained by using a two-step selection/regeneration procedure. Transformation efficiency of rice was about 30% which was as high as reported in other dicotyledons. Progenies ($\textrm{T}_{1}$ generation) derived from primary transformant of 17 lines were segregated with a 3 resistant : 1 sensitive ratio in medium containing hygromycin and bialaphos. Stable integration of Bar gene into chromosomal DNA was proven by Southern blot analysis of genomic DNA isolated from $\textrm{T}_{2}$ progenies. Transgenic plants ($\textrm{T}_{3}$) grown in the field were resistant to bialaphos (Basta) at a dosage lethal to wild type plants.

  • This study was conducted to investigated the major characteristics of genetically modified rice of 'Milyang 204' originated from Dongjinbyeo compared to a non-transgenic rice varieties Dongjinbyeo and Jun-ambyeo. Basta resistant transgenic rice lines carrying bar gene produced by the Yeongnam Agricultural Research Institute were evaluated for their agronomic characters. The transgenic Japonica rice of 'Milyang 204' showed inferior phenotypic traits compared to a non-transgenic rice variety Dongjinbyeo and Junambyeo. On the basis of UPOV (Union Internationale Pour la Protaection des Obtentions Vegetables) and NSMO(National Seed Management Office) the transgenic 'Milyang 204' showed difference in some traits out of some agronomic traits, such as leaf color, angle of flag leaf, number of spikelets, culm length, white core and white belly compared to the non-transgenic varieties rice.

  • This research aims to contribute the characterization of acetolactate synthase (Ec 4.1.3.18; ALS) and the resistance mechanism by sequence analysis of ALS gene of the sulfonylurea-resistant and -susceptible Monochoria vaginalis. The ALS gene was obtained from susceptible (S) and resistant (R) M. vaginalis to sulfonylurea herbicides (SUs). The 815 bp the fragment and the genomic DNA sequence coding for acetolactate synthase (ALS) of S and R biotypes of M. vaginalis were cloned and sequenced. Nineteen clones were divided greatly into 4 groups as result of sequencing. The first group was not difference to S type, the second group was amino acid of P197S which found point mutations causing substitution of serine for proline at amino acid 197, the third group was observed greatly other part of 6 places than group 1, and the fourth group appeared the intergrade of group 1 and 3. Therefore, it could be assumed what ALS gene of various types can be one plant. The peptide of the 13 amino acid Domain A region for ALS genes from R biotype of M. vaginalis differed from that of the S biotype by one base substitution at proline codon of Domain A. It could also be confirmed that point mutation of serine for proline at amino acid 197.

Progeny Analysis and Selection of Tomato Transformants with patII Gene linked to Inherent Disease Resistance Gene (제초제 저항성 유전자와 기존 병 저항성 유전자가 연관된 형질전환 토마토 개체 선발 및 후대분석)

  • This study was carried out to develop a model system using selection method for disease resistant plant breeding programs using a herbicide bialaphos-resistant patII gene as a gene-based marker. Spraying bialaphos could eliminate the susceptible plants from the segregating populations such as ${F_2}^{\prime}s$ and thereafter. Tomato cv. Momotaro-yoke was transformed with patII gene 60 independent transformants were acquired. Total 42 transformants were analyzed in transgene copy numbers by Southern blotting and the segregation ratios for the bialaphos resistance. Statistical analysis revealed that the transgene copy numbers and the segregation ratios were not always coincided, especially having the tendency of underestimating the real numbers of the transgenes in the multicopy lines. A two-stepwise screening method was applied to select $T_1$ tomato plants which linked the transgenic patII to a disease resistance gene (I2 and Ve). Based on the resistant to susceptible ratios, T-20 plant was finally selected due to the estimated linkage 12-13 cM between the patII gene to the I2 gene on chromosome 11. This newly developed system could be applied to any economical crop in breeding programs.

  • Cordgrass (Spartina spp.) is recognized as a highly invasive plant in estuaries throughout the world because of remarkable versatility and resiliency, significant reproduction, strong adaptability, rapid spreading, and vigorous growth. In this review, therefore, to provide insights on the effective management practices, the previous research works were summarized and discussed. Spartina spp. is a perennial halophyte, warm-season (C4) grass that reproduces both sexually through seeds and asexually by rhizomes. Management strategies for cordgrass have included various physical, biological, and chemical controls. Herbicides are usually the most cost-effective means of control. Currently, glyphosate, imazapyr, fluazifop and haloxyfop have been practically used. To improve the control efficacy, a combination of two more than methods (example, mowing-spraying) is needed to be applied consistently every year for at least 3 to 4 years and to be sprayed with enough dry time (>4-6 hr) at an early growth stage (before flowering). Consistently repeated application of same herbicide have to be avoided to prevent an unexpected emergence of herbicide-resistant lines. On the other hand, Spartina spp. have many positive functions for agricultural and eco-engineering purposes. Thus, we have to give more intensive research for effectively managing advantages and disadvantages of Spartina plantations.

Herbicide Resistance in Plant Tissue Culture (식물체(植物體) 조직배양(組織培養)과 제초제(除草劑) 저항성(抵抗性))

  • This study was conducted to evaluate herbicide resistant plant through tissue culture. Callus was induced from embryos of Echinochloa crusgalli Beauv. (var, oryzicola Ohwi, var. caudata Kitagawa and var, crusgalli). An optium medium for callus induction and succinate dehydrogenase activity in inducted callus were detected and callus growth of various varieties of Echinochloa crusgalli was assessed under the treatment of various rates of butachlor[N-(butoxymethyl)-2-chloro-N-(2,6-diethylphenyl)acetamide]. MS medium seemed to be the most appropriate to induce callus from the embryos of varieties of E. crusgalli by using 2,4-D about 5.5mg/l as a hormone source. The activity of succinate dehydrogenase in inducted callus showed positive reaction against to TTC(2,3,5-triphenyltetrazolium chloride) regardless of concentrations of butachlor and varieties of E. crusgalli, indicating that all the callus induced were alive. The callus growths derived from seeds of E, cnesgalli were greatly affected by various rates of butachlor and were completely inhibited at the highest concentration of butachlor, $10^{-3}M$, regardless of varieties of E. crasgalli. $10^{-6}M$ of butachlor inhibited 24.6% of the callus growth of E. crusgalli Beauv, var. oryzicola Ohwi, while E. crusgalli Beauv. var. crusgalli showed 42% of inhibition, showing that there was difference in response of varieties of E. crusgalli Beauv. to butachlor.

Risk assessment of genetically engineered rice Bt-9 resistant to Cnaphalocrocis medinalis: influence on above-ground arthropods in Korea

  • The effect of genetically engineered rice Bt-9 on the diversity and abundance of plant-dwelling insects and spiders was tested under field conditions. Genetically engineered rice Bt-9, expressing mCry1Ac1 from Bacillus thuringiensis, confers resistance to rice leaf roller (Cnaphalocrocis medinalis) and provides tolerance to the herbicide glufosinate (PPT). The study compared Bt-9 and two non-GM reference varieties, Ilmi-byeo and Dongjin-byeo, at LMO isolated fields in Gunwi (Kyungpook National University) and Jeonju (National Institute Agricultural Sciences) in Southern Korea in 2016 - 2017. A total of 40,817 individuals from 62 families and 11 orders were collected from the two living modified organism (LMO) isolated fields. From the three types of rice fields, a total of 13,982, 14,105, and 12,730 individuals from the Bt-9, Ilmi-byeo and Dongjin-byeo were collected, respectively. Throughout the study, the analysis of variance indicated no significant differences (p < 0.05). Multivariate analysis showed that the abundance and diversity of plant dwelling insects were similar. The data on insect species population densities were subjected to principal component analysis (PCA), which did not distinguish among the three varieties, Bt-9 and the non-GM, reference cultivars, during the cultivation years. However, the results of the PCA analysis were completely divided into four groups based on the yearly survey areas. Therefore, there was no evidence for a negative impact of Bt-9 on the above-ground insects and spiders.

Rapid Diagnosis of Resistance to Glufosinate-ammonium in Transgenic Sweet Potato (형질전환 고구마에 대한 Glufosinate-ammonium 저항성 간이진단법)

  • Transgenic herbicide-resistant sweet potato plants [Ipomoea batatas (L.) Lam.] produced through a biolistic transformation were used in this study. The objective of this research was to find out a rapid and reliable assay method for confirming glufosinate-ammonium resistance. The techniques tested include whole-plant bioassay, one leaf bioassay, and leaf disk bioassay. Parameters investigated in this study were leaf injury and ammonium accumulation at 1 and 5 days after treatment of glufosinate-ammonium. In the leaf disk bioassay, leaf injury of the transgenic line 7171 was 1.9-fold less affected by glufosinate-ammonium than the wild type. The leaf injury of 7171 in one leaf and whole-plant bioassays was 59- and 92-fold less affected by glufosinate-ammonium, respectively, compared with that of the wild type. Leaf disk, one leaf, and whole-plant bioassays showed that ammonium accumulation of the 7171 was 2 to 20-, 4 to 43-, and 6 to 115-fold less affected by 0.5-5 mM glufosinate-ammonium than that of the wild type. All three bioassays successfully distinguished the resistance from the transgenic lines, but one leaf bioassay is the simplest and quickest. Leaf injury and ammonium accumulation were the same in leaves 1, 3, 5, 7, and 10 of 3 mM glufosinate-ammonium treated plants or nontreated plants. The one leaf bioassay was chosen as the standard procedure for future confirmation of resistance in transgenic sweet potato because it is a rapid and reliable assay.

  • The study was conducted to screen paraquat-tolerant plant species among crops and weeds, using the response of plant like leaf disc discoloration, visual injury and dry weight in the presence of paraquat. Mechanism of paraquat-tolerance was investigated in strains of soybean through evaluating activities of superoxide dismutase and peroxidase and the multiplication of callus derived from soybean cotyledon. In crops, Kwanggyo has been selected as a paraquat-tolerant variety among soybean cultivars tested, and Hood as a susceptible one. In weeds, Polygonum aviculare, Chenopodium album and Pinellia ternata were evaluated as the paraquat resistant species, providing the possibility for the donor plant species for paraquat resistance. Activity of superoxide dismutase known to detoxify paraquat was markedly greater in Kwanggyo, a paraquat-tolerant cultivar than in Hood, a susceptible one. In addition, the similar response like superoxide dismutase was observed in peroxidese activity. The greater inhibition of callus multiplication was determined in Hood, a susceptible one than a tolerant one, Kwnggyo. Based on all the informations, it is strongly proposed that paraquat tolerance in soybean is due to destruction of $O_2^-$ by elevated concentration of superoxide dismutase in the tolerant cultivar.

Evaluation of Pollen Viability of Nakdongbyeo, Two Transgenic Rice Lines, Its Hybrids with Weedy Rice, and Subsequent Selfed Progenies: F2 and F3 (낙동벼, 2개의 promoter를 각각 삽입한 유전자변형 계통과 잡초성벼(Oryza sativa)인공수정 한 후 다음세대인 F1, F2, F3의 화분활력 평가)

  • This experiment was conducted to evaluate pollen viability of Nakdongbyeo, transgenic rice lines, an F$_1$ hybrid from a cross between Milyang weedy rice and ABC-promoter transgenic rice line containing basta-resistant (bar) gene and subsequent selfed progenies, F$_2$ and F$_3$. The reaction of pollen with 3-{4,5 dimethylthiazolyl-2}-2,5-diphenyl monotetrazolium bromide (MTT) as a staining chemical immediately after pollen shedding showed maximum pollen viability of 86% in Nakdongbeyo, 75% in ABC-promoter transgenic rice line, 62% in ubiquitin-promoter transgenic line, 68% in F$_1$, 79% in F$_2$ and 78% in F$_3$. Viability gradually declined during subsequent observations at 20-minute intervals. However, there was a drastic decline in pollen viability after 40 minutes of pollen shedding. The mean difference of pollen viability among rice lines and time was highly significant, indicating significantly different pollen viabilities at different time intervals. Maximum viability of 36.2% was observed in F$_3$ and minimum viability of 3.5% was found in F$_2$ at 90 min after pollen shedding. Results of this experiment on pollen viability and longevity elucidate potential risks of pollen-mediated flow of herbicide-resistant gene from transgenic rice lines and possible integration of it into the weedy rice population.
