생약복합제 GCSB-5의 품질 표준화를 위한 흑두 및 두충의 함량 분석 (Quantitative Analysis of Glycine Semen Nigra and Eucommiae Cortex for Standardization of GCSB-5 Preparation)

  • GCSB-5 preparation is a purified extract from a mixture six herbal medicines (Acanthopanacis Cortex, Achyranthis Radix, Saposhnikoviae Radix, Cibotii Rhizoma, Glycine Semen Nigra, Eucommiae Cortex) that have been widely used in traditional medicine to treat various bone disorders. This study was carried out to obtain the HPLC analysis method that can be used to establish quantitative analysis of Glycine Semen Nigra and Eucommiae Cortex for standardization of GCSB-5 preparation. HPLC analysis methods for the simultaneous determination of genistin (Glycine Semen Nigra) and geniposide (Eucommiae Cortex) were established for the quality control of herbal medicinal raw material and preparation. And validation of HPLC analysis methods were conformed for verification of HPLC methods by check to specificity, linearity, intra-day precision, inter-day precision and accuracy following ICH guideline. As the result of quantitative analysis, the contents of genistin and geniposide in the raw material of GCSB-5 preparation were 0.0426-0.0427 mg/g and 0.431-0.432 mg/g. And GCSB-5 preparation contained genistin of 0.0202-0.0203 mg/capsule and geniposide of 0.211-0.212 mg/capsule, respectively.

버섯차 개발을 위한 로스팅 식용버섯류와 곡물첨가물의 혼합비율에 따른 추출온도 및 시간별 생리활성 및 영양성분 변화 (Changes of biological activities and nutrition contents by different extraction conditions in the mixtures of roasted edible mushrooms and grain additives for the development of mushroom tea)

  • 본 연구에서는 느타리(Pleurotus ostreatus)와 표고(Lentinula edodes) 버섯차 개발을 위하여 귀리(oat)와 현미(brown rice)를 부재료로 사용함으로서 버섯 특유의 향미를 저감시킴과 동시에 곡물류 특유의 고소함과 영양학적 가치 향상을 기대하였다. 이에 버섯시료의 가공방법 중 열풍건조 및 로스팅 처리에 의한 생리활성 및 영양성분의 변화를 분석한 결과, 느타리와 표고의 열풍건조 및 로스팅 시료 사이의 DPPH 라디컬 소거능과 아질산염 소거능의 차이는 크게 없었으나, 총 폴리페놀 함량과 베타글루칸 함량은 로스팅 처리에 의하여 함량치가 증가하는 것으로 나타났다. 또한 로스팅 처리된 귀리와 현미의 생리활성과 영양성분을 비교한 결과, 귀리의 DPPH 라디컬 소거능 및 베타글루칸 함량은 현미에 비하여 높은 것으로 나타났으며, 귀리는 높은 글루탐산(Glu) 성분 등을 포함하여 모든 아미노산 성분이 현미보다 높게 나타났다. 로스팅 버섯 시료와 부재료의 혼합비율별 추출온도 및 추출시간 조건에 따른 생리활성 분석결과, DPPH 라디컬 소거능은 로스팅 표고와 귀리 1:1 혼합물(LE+O)의 100℃, 3분 추출조건에서 33.5%로 가장 높았으며, 아질산염 소거능은 느타리와 현미 3:1 혼합물(PO+B)의 100℃, 10분 추출조건에서 49.9%로 가장 높은 소거활성을 보였다. 총 폴리페놀 함량은 70℃의 10분 추출시간 조건에서 느타리와 현미 3:1 혼합추출물(PO+B)의 함량치가 16.2 mg GAE/g로 가장 높았으며, 베타글루칸 함량은 표고와 현미(LE+B) 3:1 혼합물에서 34.4%로 다른 혼합비율의 함량치에 비하여 높았다. 아미노산 분석결과, 느타리와 현미 혼합물(PO+B) 중 1:1 혼합비율에서 필수 아미노산 성분함량이 다른 혼합비율에 비하여 높았으며, 표고와 현미(LE+B) 혼합물 3:1의 비율에서 필수아미노산 성분 함량이 가장 높았다. 아미노산 성분 중 단맛과 감칠맛을 나타내는 성분함량이 월등히 높았던 귀리를 혼합함에 따른 느타리, 표고와 귀리 혼합물에서의 아미노산 성분함량의 상승효과는 나타나지 않았다.

오미자를 함유한 복합물의 항균, 발한억제 및 소취효과 (Antimicrobial, Antiperspirant and Deodorant Effect of Herbal Extract Mixtures Including Schisandra chinensis)

  • The aim of this study was to investigate the antimicrobial, antiperspirant and deodorant effect of herb mixtures including Schisandra chinensis(SC). This herb mixtures including Schisandra chinensis was made of Astragalus membranaceus(AM), Triticum aestivum(TA), Atractylodes japonica(AJ) and peppermint essential oil extracted from Mentha piperita(MP). We examined antimicrobial activity, antiperspirant activity and L-leucine dehydrogenase activity of the herbal extracts. We found that the mixture of SC and MP have the best antimicrobial effects on B. subtilis subsp. spizizenii and mixture of SC, AJ, AM and TA have best antiperspirant effect. Moreover, the combination of SC and AM showed the most inhibitory effect on L-leucine dehydrogenase activity. These results suggested that the herb mixtures Including Schisandra chinensis have good antimicrobial and antiperspirant effects.

삼황사심탕(三黃瀉心湯) 추출물의 lipopolysaccharide 유도에 의한 염증 조절과 대식세포 활성에 대한 연구 (Effect of Samhwangsashimtang Extract on Lipopolysaccharide-Stimulated Inflammatory Response and Macrophage Activity)

  • Objectives : Samhwangsashim-tang(SHSST), a mixture of Rhei radix et rhizoma, Scutellariae radix, and Coptidis rhizoma, has been regarded as being able to treat bleeding, gastric discomfort, dry mouth, insomnia and purpura due to Blood Heat. Currently, this herbal formula is applied to gastritis, gastric ulcer, hypertension, atherosclerosis or other types of vascular inflammatory disorders. Methods : We extracted this herbal mixture with 30% ethanol and examined for its effects on systemic inflammatory responses and in vitro macrophage activity. Mice were orally given to SHSST for 7 days and then lipopolysaccharide(LPS) was intraperitoneally injected. Tumor necrosis factor-${\alpha}$(TNF-${\alpha}$) levels in serum were measured 1 h after LPS challenge. Peritoneal macrophages were isolated from thioglycollate-injected mice and used for in vitro cellular activity. Cell death was measured using the MTT method and annexin V/propidium iodide staining. LPS-stimulated signaling molecules necessary for TNF-${\alpha}$ expression were determined by Western blotting. Results : Oral administration of SHSST for 7 days resulted in a significant reduction in LPS-stimulated TNF-${\alpha}$ release into serum. In vitro treatment of SHSST was cytotoxic in a concentration-dependent manner. However, SHSST caused a concentration-dependent reduction in necrosis and increase in apoptosis in mouse peritoneal macrophages. SHSST inhibited the activation of NF-${\kappa}B$, p38 and JNK signaling molecules in response to LPS. Conclusion : Taken together, our results demonstrated that SHSST was effective in lowering LPS-stimulated TNF-${\alpha}$ serum levels, possibly through its modulation of NF-${\kappa}B$, p38 and JNK in macrophages.

Evaluation of Estrogenic Activity of Extract from the Herbal Mixture Cynanchum wilfordii Hemsley, Phlomis umbrosa Turczaninow, and Angelica gigas Nakai

  • Hormone replacement therapy (HRT) consists of highly effective prescription medications for treating menopausal symptoms; however, these agents have exhibited side effects including the risk of estrogen-induced carcinogenesis. Therefore, interest in phytotherapy-based materials as a natural source of alternatives to estrogen therapy has increased. However, some of these herbal medicines have been reported to increase the risk of estrogen-induced cancer. Herbal formulations composed of a combination of Cynanchum wilfordii Hemsley (CW), Phlomis umbrosa Turczaninow (PU), and Angelica gigas Nakai (AG) extracts (CPAE) have been used for treating menopausal symptoms. Therefore, in this study, we aimed to examine the safety of CPAE by determining its potential adverse estrogenic activity using the Organization for Economic Cooperation and Development (OECD) test guideline 455 (TG455) in a stably transfected transcriptionally activated human estrogen receptor ${\alpha}$ ($hER{\alpha}$)-HeLa9903 cell model. We found that CPAE did not how any estrogenic activity or stimulate promoters containing estrogen response elements in MCF-7 cells. In addition, CPAE showed no significant selective activity against $hER{\alpha}$ and $hER{\beta}$, non-selective activity against the ER, or effects on ER target gene expression. Furthermore, CPAE did not significantly induce MCF-7 cell proliferation and uterine weight increase in ovariectomized rats. These results demonstrate that CPAE can be used as beneficial herbal drug for prevention and therapeutic intervention of estrogen carcinogenesis in menopausal women.

Combined Treatment of Herbal Mixture Extract H9 with Trastuzumab Enhances Anti-tumor Growth Effect

  • Extracts from Asian medicinal herbs are known to be successful therapeutic agents against cancer. In this study, the effects of three types of herbal extracts on anti-tumor growth were examined. Among the three types of herbal extracts, H9 showed stronger anti-tumor growth effects than H5 and H11 in vivo. To find the molecular mechanism by which H9 inhibited the proliferation of breast cancer cell lines, the levels of apoptotic markers were examined. Proapoptotic markers, including cleaved PARP and cleaved caspases 3 and 9, were increased, whereas the anti-apoptotic marker Bcl-2 was decreased by H9 treatment. Next, the combined effect of H9 with the chemotherapeutic drugs doxorubicin/cyclophosphamide (AC) on tumor growth was examined using 4T1-tumor-bearing mice. The combined treatment of H9 with AC did not show additive or synergetic anti-tumor growth effects. However, when tumor-bearing mice were co-treated with H9 and the targeted anti-tumor drug trastuzumab, a delay in tumor growth was observed. The combined treatment of H9 and trastuzumab caused an increase of natural killer (NK) cells and a decrease of myeloid-derived suppressor cells (MDSC). Taken together, H9 induces the apoptotic death of tumor cells while increasing anti-tumor immune activity through the enhancement of NK activity and diminishment of MDSC.

Fermented Chaga-Cheonggukjang attenuates obesity condition and suppresses inflammatory response of the liver in high fat diet-induced mice

  • Chaga mushroom and Cheonggukjang have been used in alternative medicine. In this study, we determined the anti-obesity effects of fermented Chaga-Cheonggukjang (FCC), an extract prepared by secondary fermentation of a mixture of Cheonggukjang and Chaga by Lactobacillus acidophilus on highfat diet (HFD)-induced mice. Male ICR mice were fed a normal diet or HFD in the presence or absence of 3% and 5% FCC (FCC3 and FCC5). After 3 months, the mice were sacrificed, and serum and tissue samples were examined. Body weight and epididymal fat pad (EFP) weight were significantly lowered in FCC3 and FCC5 groups compared to those in the HFD control group. FCC supplementation suppressed serum triglyceride (TG) and increased serum HDL levels. Serum GOT, GPT, leptin levels and hepatic COX-2 mRNA expression were significantly higher in the HFD groups, and these increases were significantly attenuated by FCC supplementation. FCC suppressed body weight and EFP weight gain, as well as inflammatory responses in the liver in HFD-fed mice. Thus, FCC supplementation may have protective effects for obesity-related disease.

생약복합물 용매추출물과 조다당획분의 생리활성 (The Physiological Activity of Crude Polysaccharide Solvent Extracted from Herbal Medicine Mixture)

  • After ethanol (BM-E and RW-E) and hot-water (BM-HW and RW-HW) extracts were fractionated from two herbal mixtures (BM and RW), their physiological activities were investigated. All extracts consisted of more than 50% of neutral sugar, with their total polyphenol levels higher than flavonoid levels. Radical scavenging activities of EtOH extracts remained significantly higher compared to that of hot-water extracts, and in particular, RW-E showed consistently higher antioxidant activity than BM-E. When anti-inflammatory activities of the extracts were evaluated by LPS-stimulated RAW 264.7 cells at 10~500 μg/mL non-cytotoxicity doses, BM-E showed significantly higher levels of TNF-α, IL-1β, IL-6, and nitric oxide inhibitory activity than those of hot-water extracts and RW-E. Murine peritoneal macrophage cells were shown to be enhanced in crude polysaccharides (BM-CP and RW-CP fractionated from BM-HW and RW-HW) compared to hot-water extracts and polysaccharide K (PSK, positive control). Especially, RW-CP exhibited higher activity than BM-CP, and component sugar analysis showed that BM-CP mainly contained galacturonic acid, glucose, arabinose, galactose, and xylose (34.5%, 33.9%, 16.1%, 7.1%, and 6.3%, respectively), whereas RW-CP showed different measurements (29.5%, 59.2%, 5.0%, 4.5%, and 0.2%). In conclusion, two herbal mixtures could contain varying sets of physiological activities dependent on different extraction and fractionation methods.

황기, 지치 복합물의 연골세포에서의 Matrix Metalloproteinases 저해 효과 및 유효성분의 분석 (Inhibitory Effect of Mixed Extracts Obtained from Astragali Radix and Lithospermi Radix on Matrix Metalloproteinases in IL-1β-induced SW1353 Cells and Quantitative Analysis of Active Compounds)

  • Background: Astragali radix (A) and Lithospermi radix (L) have long been used as traditional medicines due to their known anti-inflammatory effects. This study aimed at evaluating, their optimal mixing ratio and their functional compounds by investigating the inhibitory effects of mixed extracts of A and L and their active compounds on matrix metalloproteinases (MMPs). Methods and Results: A and L extracts were obtained by extraction at $80^{\circ}C$ using 50% and 70% fermented alcohol, respectively, and then mixed at a ratio of 5 : 5, 6 : 4, 7 : 3 and 8 : 2 (w/w). The activities of MMP-1, MMP-3, and MMP-13 were evaluated in interleukin-1beta ($IL-1{\beta}$)-induced SW1353 cells. The extract mixtures showed synergistic inhibitory effects on MMP-3 and MMP-13, higher than the effects of the individual A and L extracts. The 7 : 3 mixture (ALM16) showed the most effective MMPs inhibitory activity, while among the active ingredients, calycosin-7-O-${\beta}$-D-glucoside and lithospermic acid exhibited excellent MMPs inhibitory activity. Additionally, an HPLC method was established for simultaneous quantification of the effective components of the extract mixtures, and validated by measuring the linearity, precision and accuracy of the limit of detection (LOD) and limit of quantification (LOQ). Conclusions: ALM16 showed the most effective MMPs inhibitory activity. Calycosin-O-${\beta}$-D-glucoside, calycosin and lithospermic acid were identified as useful candidates, as they were the major functional compounds in the MMP inhibitory activity. Summarily, ALM16 might be a highly effective in osteoarthritis management, owing to its because it exhibits a protective effect on cartilage via excellent inhibition of MMPs.

Ethanol extract of Sinsun-yukza-hwan, a Korean medicinal prescription, promotes hair growth in C57BL/6 mice, an alopecia animal model

  • Objectives : In Korean medicine, a prescription of Sinsun-yukza-hwan (Shenxian-liuzi-wan, SSY) has been used in clinic for treatment of alopecia via oral. This study was performed to determine transdermal effects of the ethanol extract from SSY on hair growth and -related gene expressions in mice. Methods : We analyzed index compound, 5-hydroxy-methyl-2-furaldehyde (HMF), in SSY extract by ultra performance liquid chromatography (UPLC). 6 weeks old C57BL/6 mice with removed hair were used as an alopecia animal model. Mice were divided into 3 experimental groups including normal (3 water: 1 ethanol: 2 polyethylene glycol mixture as a vehicle), SSY extract and 5% minoxidil (as a positive control), treated groups. SSY was applied topically on the hair-shaved skin of C57BL/6 mice every day for 15 days. The color, thickness and density of hair were monitored every $5^{th}$ day by naked eye, photograph and phototrichogram using folliscope. Also hair growth-associated gene expressions were measured by immunoblotting assay. Results : Hair density of minoxidil or SSY-treated group was significantly increased compared to that of vehicle application on the $15^{th}$ day, respectively. And hair thickness of minoxidil and SSY groups was increased compared to that of vehicle treated group on the $15^{th}$ day, respectively. Induction of insulin-like-growth factor 1(IGF-1) and vascular endothelial growth factor (VEGF) were also significantly accelerated by SSY extract compared to those of vehicle-applied group. Conclusions : These results provide scientific evidence to support the potent multi-application of SSY as a cosmeceutical material for promoting hair growth.
