Rat의 hepatocyte에서 $amyloid-{\beta$}$로 유발된 세포사, 지질과산화 및 세포산화에 대한 귀전우 열수 추출물의 보호효과 (Physiological Activity and Effects on Lipid Peroxidation of Hot Water-Extract Obtained from Euonymus alatus in Cultured Rat Hepatocyte)

  • 목적 : 귀전우 Euonymus alatus (Thunb.) Sieb로부터 얻은 추출물에 대하여 생리활성(항산화 활성)을 검토하였고, $amyloid-{\beta}{\;}(A{\beta})$가 지니는 Rat의 배양 Hepatocyte 세포독성에 대한 세포보호효과 및 세포막 지질의 과산화에 미치는 영향을 검토하였다. 방법 : 귀전우 추출물과 항산화제인 BHT. ascorbic acid 및 ${\alpha}-tocopherol$을 각각 linoleic acid에 소정량 첨가하고 $50^{\circ}C$에서 일정기간 저장하여 linoleic acid의 산화 안정성을 비교 검토하였으며, 또한 유지 자동산화의 촉진인자인 금속이온($Fe^{3+},{\;}Zn^{2+}$) 을 첨가하여 귀전우 추출물의 금속이온봉쇄효과를 조사하였다. 결과 :세포성지질의 과산화의 지표인 malondialdehyde(MDA)생성이 A{\beta}처리로 크게 증가하였으나 세포막파괴에 의한 세포파괴의 전형적인 현상이 귀전우 열수 추출물의 전처리와 후처리로 크게 감소되었다. 귀전우 추출물의 농도에 따른 항산화 효과는 전 농도별에서 높은 항산화 효과를 나타내었다. 그리고 시간 경과에 따른 항산화 효과에 있어서도 정도의 차이는 있지만 각 농도(5mg, 10mg 및 25mg)에서 유도기의 연장효과가 상당히 우수한 것으로 나타났다. 귀전우 추출물과 항산화제와의 비교에 있어서는 합성 항산화제인 BHT 및 ascorbic acid 만큼 우수한 항산화력을 나타내었으며, 천연 항산화제인 ${\alpha}-tocopherol$ 보다는 아주 우수한 것으로 나타났다. 귀전우 추출물의 금속이온 봉쇄효과에 있어서는 linoleic acid에 귀전우 추출물과 $Fe^{3+}$$Zn^{2+}$을 첨가한 군이 귀전우 추출물을 첨가하지 않은 군보다 항산화 효과가 아주 우수한 것으로 나타나 금속이온 봉쇄력이 강한 것으로 나타났다. 결론 : 귀전우 열수 추출물은 $A{\beta}$ 의한 세포파괴를 감소시키는 활성을 가지며 예방 및 보호효과를 나타내었고, hepatocyte에 대한 보호효과와 세포막지질 과산화의 저해 및 $A{\beta}$처리와 같은 독성에 대한 적응능력 향상을 통한 세포보호효과를 주는 것으로 간세포 보호 등의 임상적 응용에 그 효과가 기대된다.

  • The present study was designed to estimate the protective effect of (-) epigallocatechin gallate (EGCG) on ethanol-induced liver injury in rats. Chronic ethanol administration (6 g/kg/day ${\times}$ 60 days) caused liver damage that was manifested by the elevation of markers of liver dysfunction - aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, lactate dehydrogenase, bilirubin and ${\gamma}$-glutamyl transferase in plasma and reduction in liver glycogen. The activities of alcohol metabolizing enzymes such as alcohol dehydrogenase and aldehyde dehydrogenase were found to be altered in alcohol-treated group. Ethanol administration resulted in the induction of cytochrome p450 and cytochrome-$b_{5}$ activities and reduction of cytochrome-c reductase and glutathione-S-transferase, a phase II drug metabolizing enzyme. Further, ethanol reduced the viability of isolated hepatocytes (ex vivo) as assessed by trypan blue exclusion test and induced hepatocyte apoptosis as assessed by propidium iodide staining. Treatment of alcoholic rats with EGCG restored the levels of markers of liver injury and mitigated the alterations in alcohol metabolizing and drug metabolizing enzymes and cyt-c-reductase. Increased hepatocyte viability and reduced apoptotic nuclei were observed in alcohol + EGCG-treated rats. These findings suggest that EGCG acts as a hepatoprotective agent against alcoholic liver injury.

Effect of Zinc-enriched Yeast FF-10 Strain on the Alcoholic Hepatotoxicity in Alcohol Feeding Rats

  • The possible protective effects of highly zinc-containing yeast Saccharomyces cerevisiae, FF-10 strain, isolated from tropical fruit rambutan on acute alcoholic liver injury in rats were evaluated. Zinc concentration in this strain was 30.6mg%. The activities of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), and $\gamma$-glutamyl transpeptidase ($\gamma$-GTP) were highly increased when alcohol was treated, relative to the normal rats. Also, a highly significant increase in the blood alcohol and acetaldehyde levels by alcohol treatment was observed. Administration of FF-10 strain markedly prevented alcohol-induced elevation of the activities of serum ALT, AST, and $\gamma$-GTP, and the levels of blood alcohol and acetaldehyde, and these reduced levels reached to that of normal rats. As compared with alcohol treated control rats, the FF-10 strain supplementation showed highly decreased the triglyceride concentration in serum. Alcohol treatment induced the marked accumulation of small lipid droplets, hepatocytes necrosis, and inflammation, but FF-10 strain administration attenuated to alcohol-induced accumulation of small lipid droplets and hepatocyte necrosis in the liver. Therefore, the current finding suggests that zinc-enriched yeast FF-10 strain isolated from tropical fruit rambutan may have protective effect against alcohol-induced hepatotoxicity.

사염화탄소를 투여한 흰쥐의 간세포에 대한 식물 추출물들의 보호효과 (Protective Effects of Plant Extracts on the Hepatocytes of Rat Treated with Carbon Tetrachloride)

  • 본 실험은 사염화탄소 독성에 대한 몇 가지의 식물소재들- 솔잎, 콩나물, 인진쑥, 표고버섯 - 의 간보호 효과를 조사하기 위하여 수행되었다. 우선적으로 일차 간세포 배양실험에서 식물추출물(열수 또는 에탄올)을 첨가(20 mg/mL 배지)하고 2시간 후에 사염화탄소를 처리(50$\mu$L)한 결과, 배지내의 AST와 ALT 효소활성은 다음과 같았다. 솔잎은 에탄올 및 열수 추출물(P<0.05)에서, 표고버섯은 열수 추출물(p<0.01∼0.05)에서, 그리고 콩나물과 인진쑥은 추출방법에 따라 유의성 있는(p<0.01) 감소를 보였다. 또한, 3주간의 식물 추출물을 첨가한 사료를 사전급여하고 사염화탄소를 2일간 복강주사(1 mg/kg 체중)한 동물실험에서 혈청내의 AST 및 ALT 활성은 간세포 배양의 결과와 유사하게 솔잎과 콩나물에서 낮은 활성효과(p<0.01)를 보였다. 정상군에 대한 사염화탄소의 투여는 지질과산화물과 cytochrome P-450의 함량, 그리고 xanthine oxidase(XOD) 활성을 크게 증가시켰다. 그러나 식물소재 중에서 솔잎, 콩나물, 인진쑥 등을 추출물을 첨가한 실험군에 대한 사염화탄소 처리는 일반적으로 지질과 산화와 XOD 활성증가를 억제하는 것(p<0.01)으로 나타난 반면에, cytochrome P-450의 함량 증가는 솔잎과 인진쑥의 첨가군에서 관찰되었다. 따라서 본 실험에서 cytochrome P-430의 함량증가에 대한 식물 추출물들의 효과는 확실하지 않았지만, 솔잎과 콩나물의 추출물은 항산화관련 실험에서 대부분 탁월한 효과를 나타내었다.

인진청간탕가미방(茵陳淸肝湯加味方)이 간세포활성(肝細胞活性), 세포주기(細胞週期) 및 APOPTOSIS에 미치는 영향(影響) (The Effects of Injinchunggantang-derivative on Cell Viability, Cell Cycle Progression and Apoptosis of Hepatocytes)

  • To evaluate the effects of Injinchunggantang-derivative on cell viability, cell cycle progression, and apoptosis, MTT assay, cell cycle analysis, Cpp32 protease assay, DNA fragnemtation assay, quantitative RT-PCR, and Western blotting were performed. The results were as followes. In MTT assay, etoposide+Injinchunggantang-derivative-treated cells as well as Injinchunggantang-derivative-treated cells showed higher viability than etoposide-treated cells with no time-concentration-dependence, which implied that Injinchunggantang-derivative has hepato-protective effect Cell cycle analysis showed that Injinchunggantang-derivative has no significant effect on the cell cycle. Cpp32 protease assav and DNA fragmentation assay Injinchunggantang-derivative carry inhibitory effects on apoptosis induction. It was suggested that Injinchunggantang-delivative might regulate the cell cycle, in particular $G_1$ checkpoint by blocking p53 and Watl pathway. Injinchunggantang-derivative inhibited the mRNA expressions of Cpp32, Fas, and Bcl-2, which could result in inhibition of apoptosis. These results imply that Injinchunggantang-derivative increases hepatocyte viability, and protects hepatocyte from damage by regulating the expression of genes associated with cell cycle and apoptosis, which explains the mechanism of the clinical effect of Injinchunggantang-derivative on liver diseases.

모델 랫드에 간 허혈/재관류로 유발된 손상에 대한 항산화제의 보호 효과에 관한 연구 (A Study on the Protective Effect of Antioxidants on Damage Induced by Liver Ischemia/Repefusion in a Rat Model)

  • 허혈 재관류 손상은 기관 이식, 외과적 혈관 재개통 및 출혈시에 발생하며 조직 및 기관 기능 장애를 유발한다. 최근 허혈 재관류 손상의 기전적 연구를 위해 간장 허혈 모델을 많이 이용하고 있다. 본 연구는 허혈 재관류 랫드 모델을 이용하여 항산화와 항염증 효과를 가진 것으로 알려진 Vanillin에 의한 간장 및 신장 손상에 대한 보호 효과를 알아보고 관련된 기전을 조사하기 위하여 실시하였다. 시험물질은 각각 100 mg/kg의 농도로 3일간 투여한 후, 60분동안 간을 결찰하여 허혈 재관류를 유도하여 관찰하였으며, 음성대조군, sham대조군 및 허혈재관류만 실시한 허혈 재관류대조군을 따로 두어, 약물투여군과 비교하였다. Vanillin 처치군에서는 AST, ALT 활성이 허혈 재관류대조군에 비해 유의하게 억제되었고, 조직병리학적 관찰에서도 염증 부분과 괴사부분이 현저하게 감소하였다. MDA와 SOD는 허혈 재관류군에 비해 유의적인 변화를 보였다. 이상의 결과를 종합하면 Vanillin은 간장 허혈 재관류에 의한 세포염증 및 세포괴사를 완화시켜 간세포 보호작용을 나타내었고, 신장의 사구체 및 원위세뇨관에 염증 변화를 완화 시키고 있어 세포 손상을 방어하는 것으로 생각되며, 이러한 방어효과는 항산화 기능에 의한 영향으로 사료된다.

Effects of Soybean Embryo on Liver Protection and Lipid Metabolism of Alcohol-Fed Rats

  • In this study, the ameliorating effect of soybean embryos on the impact of alcohol consumption was investigated on rat hepatocytes and in reducing total serum cholesterol levels and total serum lipid levels. Liver histology and two clinically important enzyme markers, aspartate aminotransferase (AST) and alanine aminotransferase (ALT), of rats administered with both alcohol and soybean embryo were compared with a control group. The treatment regimen of soybean embryo significantly reduced the serum ALT and AST levels of the subjects, demonstrating the hepato-protective effects of soybean embryo. Electron microscopy indicated that the administration of soybean embryo preserved the important hepatocyte structures and prevented the presence of lipid droplets and secondary lysosomes. Furthermore, total cholesterol and total lipid levels were significantly reduced. These results indicate that treatment with soybean embryo can positively mediate the effects of alcohol on hepatocytes and general liver functions.

AMPK 활성화를 통한 소형화(素馨花) (Jasminum officinale L. var. grandiflorum) 열수 추출물의 HepG2 간세포 보호 활성 (Hepato-Protective Activities of Jasminum officinale L. var. grandiflorum Aqueous Extract via Activation of AMPK in HepG2 Cells)

  • Objectives : Jasminum officinale L. var. grandiflorum is used as a traditional or folk remedy in China to treat arthritis, hepatitis, duodenitis, conjunctivitis, gastritis, and diarrhea. In this study, we aimed to study the hepatocyte protective activity and molecular mechanism of Jasminum officinale L. var. grandiflorum aqueous extract (JGW) using HepG2 hepatocyte cell lines. Methods : HepG2 cells were pretreated with diverse concentrations of JGW, and then the cells were exposed to tert-butyl hydroperoxide (tBHP) for inducing oxidative stress. Hydrogen peroxide (H2O2) production, glutathione (GSH) concentration, mitochondrial membrane potential (MMP) and cell viability were measured to investigate hepato-protective effects of JGW. Phosphorylation of AMP-activated protein kinases (AMPK), acetyl coenzyme A carboxylase (ACC) and effects of compound C on cell viability were examined to observe the role of AMPK on JGW-mediated cytoprotection. Results : Pretreatment with JGW (10-300 ㎍/mL) significantly suppressed cytotoxicity induced by tBHP in a concentration dependent manner and reduced the expression of cleaved PARP and cleaved caspase-3 proteins related to apoptosis in HepG2 cells. In addition, pretreatment with JGW significantly prevented the increase in H2O2 production, GSH depletion, and lower MMP induced by tBHP. Treatment with JGW (30 minutes of incubation and concentrations of 100 and 300 ㎍/mL) increased the phosphorylation of AMPK and ACC and treatment with compound C, a chemical inhibitor of AMPK, inhibited the cytoprotective effect of JGW. Conclusions : Our results demonstrated that JGW may protect hepatocytes from oxidative stress via activation of AMPK.

택사(澤瀉)가 유리지방산으로 유발된 HepG2 cell의 lipoapoptosis에 미치는 영향 (The Effect of Alisma orientale Extract on Free Fatty Acid-induced Lipoapoptosis in HepG2 Cells)

  • Objectives : This study was designed to investigate the effect on lipoapoptosis of Alisma orientale extract against free fatty acid-induced cellular injury. Methods : HepG2 cells were used in an vitro model. HepG2 cells were treated with free fatty acids to generate a cellular model of nonalcoholic fatty liver disease (NAFLD). Using this cellular model, the anti-apoptotic effect and reducing steatosis of Alisma orientale extract against free fatty acid-induced cellular injury was evaluated by measuring steatosis and apoptosis. Results : Alisma orientale extract significantly attenuated free fatty acid-induced intracellular steatosis. Alisma orientale extract inhibited free fatty acid-mediated activation of pJNK, PUMA, BAX, caspase-3, and -9, and apoptotic kinases that are correlated with NAFLD. Alisma orientale extract also promoted Bcl-2, a anti-apoptotic protein. Conclusions : From the above, the Alisma orientale extract decreased the hepatocyte steatosis and showed the hepatocelluar protective effect by the regulation of apoptosis-related protein. It proposes the possibility of Alisma orientale extract to the treatment of nonalcoholic fatty liver disease in clinics.

Protective effects of an ethanol extract of Angelica keiskei against acetaminophen-induced hepatotoxicity in HepG2 and HepaRG cells

  • BACKGROUND/OBJECTIVE: Although Angelica keiskei (AK) has widely been utilized for the purpose of general health improvement among Asian, its functionality and mechanism of action. The aim of this study was to determine the protective effect of ethanol extract of AK (AK-Ex) on acute hepatotoxicity induced by acetaminophen (AAP) in HepG2 human hepatocellular liver carcinoma cells and HepaRG human hepatic progenitor cells. MATERIALS/METHODS: AK-Ex was prepared HepG2 and HepaRG cells were cultured with various concentrations and 30 mM AAP. The protective effects of AK-Ex against AAP-induced hepatotoxicity in HepG2 and HepaRG cells were evaluated using 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide, lactate dehydrogenase (LDH) assay, flow cytometry, and Western blotting. RESULTS: AK-Ex, when administered prior to AAP, increased cell growth and decreased leakage of LDH in a dose-dependent manner in HepG2 and HepaRG cells against AAP-induced hepatotoxicity. AK-Ex increased the level of Bcl-2 and decreased the levels of Bax, Bok and Bik decreased the permeability of the mitochondrial membrane in HepG2 cells intoxicated with AAP. AK-Ex decreased the cleavage of poly (ADP-ribose) polymerase (PARP) and the activation of caspase-9, -7, and -3. CONCLUSIONS: These results demonstrate that AK-Ex downregulates apoptosis via intrinsic and extrinsic pathways against AAP-induced hepatotoxicity. We suggest that AK could be a useful preventive agent against AAP-induced apoptosis in hepatocytes.
