종류가 다른 식용유지에 첨가된 고DHA(Docosahexaenoic Acid) 어유가 흰쥐의 지질대사에 미치는 영향 (The Effect of Docosahexanoic Acid-Rich Fish oil Added to Different Dietary Fats on Lipid Metabolism in Rat)

  • This study was undertaken to elucidate the effect of DHA-rich fish oil (DHA-rich oil) added to different dietary fats on lipid metabolism. Rats were fed perilla oil, sesame oil and beef tallow with or without DHA-rich oil for 12 weeks. The weight gain was higher in groups with DHA-rich oil than that of groups without DHA-rich oil, with DHA-rich oil, while weight of epididymal fat pad was lower in perilla oil and beef tallow groups with DHA-rich oil. The contents of total lipid and triglyceride in plasma were not affected by dietary fat types, but that of total and HDL cholesterol in plasma were higher in sesame oil group than perilla oil and beef tallow groups without DHA-rich oil. The contents of total lipid, triglyceride, total cholesterol, HDL cholestrol and LDL cholesterol in plasma were decreased by DHL-rich oil addition. The contents of total lipid, total cholesterol and triglyceride in the liver were not affected by dietary fat type. The contents of total cholesterol and triglyceride in the liver were not affected by dietary fat type. The contents of total lipid and TG in liver were not affected by DHA-rich oil addition while hepatic cholesterol increased by DHA-rich oil addition. The activities of glucose 6-phosphate dehydrogenase and malic enzyme were highest in beef tallow group without DHA-rich oil and decreased by DHA-rich oil addition. Peroxisomal ${\beta}$-oxidation had an inverse relationship against the activities of lipogenic enzymes. In conclusion, dietary DHA-rich oil decreased fat accumulation and had hypolipidemic effect, especially in beef tallow group. Also groups with DHA-rich oil showed more hypolipidemic effect than perilla oil group. And DHA-rich oil addition to diets resulted in increasing dietary n-3/n-6 ratio. Therefore increase in n-3/n-6 ratio as well as dietary DHA were considered to be responsible for the hypolipidemic effect resulted from DHA-rich oil addition.

Anti-obesity Effect of Steamed Soybean and Fermented Steamed Soybean in High-fat Diet-induced Obese ICR Mice

  • This study was performed to investigate the ameliorating effects of steamed soybeans (SS) and fermented SS (FSS) on lipid metabolism in high-fat diet-induced obese mice. ICR mice were divided into four groups and given the following different diets: normal diet (ND), high-fat diet (HFD), HFD with 1% SS (HFD + SS), and HFD with 1% FSS (HFD + FSS). After 14 weeks, the body weight gain was higher in the HFD group compared with the ND group but lower in the HFD + FSS group compared with the HFD group. Plasma levels of triglyceride (TG), total cholesterol (TC), low density lipoprotein-cholesterol, aspartate aminotransferase (AST), and alanine aminotransferase (ALT) were significantly higher in the HFD group compared to the ND group, but lower in the HFD + SS and HFD + FSS groups compared with the HFD group. In addition, leptin concentration in plasma was lower in the groups fed HFD + SS and HFD + FSS compared with the HFD group. The accumulation of hepatic TG and TC was significantly inhibited in the HFD + SS and HFD + FSS groups. Furthermore, SS and FSS attenuated lipid peroxidation and nitric oxide formation in the liver induced by the high-fat diet. These results suggest that soybeans, especially FSS, may be useful in preventing obesity-induced abnormalities in lipid metabolism.

발효당귀가 Orotic acid 유발 흰쥐 지질 대사에 미치는 영향 (Effect of Fermented Angelica gigas Nakai on Lipid Metabolism in Orotic Acid Model Rats)

  • Orotic acid 1% 수준으로 지방간을 유발시킨 흰쥐에 Monascus purpureus 균주 발효 당귀 5%를 식이 첨가하여 간 조직 및 혈중의 지질대사에 미치는 영향을 검토하였다. 발효 참당귀 분말을 10일간 식이 급여한 결과, orotic acid 유발 지방간 흰쥐의 간 조직 내 중성지질 농도를 낮추고, 혈청 지질 농도가 정상 수준으로 회복되었음을 확인하였다. 간 조직 내 MTP mRNA 및 PDI mRNA 발현 정도는 정상군에 비해 발효 당귀군에서 약간 증가하는 경향을 보이거나, 유의적으로 증가된 수치를 나타내었다. 간 조직의 형태학적 관찰에서는 세포 간극이 좁고 잘 구성된 소엽구조를 하고 있는 정상군에 반해 OA 대조군은 orotic acid 투여로 소포성 지방 변성이 소엽 중심대에 나타나 있고, 지방세포의 크기와 수가 증가하여 전형적인 지방간 형태의 양상을 보였다. 한편, 발효 당귀군의 간장은 간세포의 소포성 지방세포들이 현저히 감소함으로써, 정상 수준의 형태학적 간 성상을 나타내어 지방간 개선에 긍정적인 영향을 미치는 것으로 판단되며, 간 조직의 지질 축적 억제로 인한 지질대사 개선에도 효과가 있는 것으로 사료된다.

Monoacylglycerol O-acyltransferase 1 (MGAT1) localizes to the ER and lipid droplets promoting triacylglycerol synthesis

  • Monoacylglycerol acyltransferase 1 (MGAT) is a microsomal enzyme that catalyzes the synthesis of diacylglycerol (DAG) and triacylglycerol (TAG). However, the subcellular localization and catalytic function domain of this enzyme is poorly understood. In this report, we identified that murine MGAT1 localizes to the endoplasmic reticulum (ER) under normal conditions, whereas MGAT1 co-localize to the lipid droplets (LD) under conditions of enriching fatty acids, contributing to TAG synthesis and LD expansion. For the enzyme activity, both the N-terminal transmembrane domain and catalytic HPHG motif are required. We also show that the transmembrane domain of MGAT1 consists of two hydrophobic regions in the N-terminus, and the consensus sequence FLXLXXXn, a putative neutral lipid-binding domain, exists in the first transmembrane domain. Finally, MGAT1 interacts with DGAT2, which serves to synergistically increase the TAG biosynthesis and LD expansion, leading to enhancement of lipid accumulation in the liver and fat.

Effects of Natural Alternative Sweeteners on Metabolic Diseases

  • The rising prevalence of obesity and diabetes is a significant health concern both in globally and is now regarded as a worldwide epidemic. Added sugars like sucrose and high-fructose corn syrup (HFCS) are a major concern due to their link with an increased incidence of diet-induced obesity and diabetes. The purpose of this review is to provide insight into the effects of natural sweeteners as alternatives to sucrose and HFCS, which are known to have negative impacts on metabolic diseases and to promote further research on sugar consumption with a focus on improving metabolic health. The collective evidences suggest that natural alternative sweeteners have positive impacts on various markers associated with obesity and diabetes, including body weight gain, hepatic fat accumulation, abnormal blood glucose or lipid homeostasis, and insulin resistance. Taken together, natural alternative sweeteners can be useful substitutes to decrease the risk of obesity and diabetes compared with sucrose and HFCS.

고지방식이를 섭취한 마우스에서 느티만가닥버섯의 항비만 효과 (Anti-obesity Effect of Hypsizigus marmoreus in High Fat-fed Mice)

  • 본 연구는 느티만가닥버섯의 항비만 효과를 알아보기 위해 4주령된 C57BL/6 수컷 쥐를 30마리를 1주일간 적응시킨 후, 정상식이군, 고지방식이군, 5% 느티만가닥버섯 분말 첨가 식이군으로 나누어 8주간 실시하였다. 체중 및 체중증가량은 고지방식이군에서 유의적으로 증가하였으며, 느티만 가닥버섯 분말 첨가 식이군에서 감소하였다. 간 및 지방조직의 무게도 고지방식이군에 비하여 느티만가닥버섯 처리군에서 낮았다. 고지방 식이에 의한 혈청 중 총콜레스테롤, 인슐린 및 렙틴 수준 증가는 느티만가닥버섯 분말 섭취로 인하여 낮아졌다. 또한, 간 중 지질 함량도 느티만가닥버섯 분말 첨가군에서 감소하였다. HE staining으로 관찰한 간내 지방축적과 지방세포의 크기도 느티만가닥버섯 분말 첨가 식이군에서 현저히 감소하였다. 부고환 지방조직에서 관찰한 지방생성 관련 유전자인 $PPAR{\gamma}$, SREBP-1c, SCD-1의 단백질 발현은 고지방식이군과 비교 시, 느티만가닥버섯 분말첨가 식이군에서 억제되었다. 이상의 실험결과, 고지방 식이와 함께 공급한 느티만가닥버섯 분말은 체중 감소 및 체지방 축적 억제와 더불어 혈청 지방 수준 개선에도 긍정적 영향을 미치는 것으로 나타났다.

표고버섯이 고지방식이로 유도한 비만 흰쥐에 미치는 항비만 효과 (Anti-Obesity Effects of Lentinus edodes on Obese Mice Induced by High Fat Diet)

  • 본 연구는 표고버섯 물추출물의 항비만 효과를 알아보기 위하여 6주령의 수컷 C57BL/6 마우스를 이용하여 고지방식이와 함께 표고버섯 물추출 분말을 1%, 3%, 5% 수준으로 첨가하여 12주간 공급하였다. 고지방 식이섭취는 체중, 혈청지질 농도 및 지방조직 무게를 증가시켰다. 표고버섯 물추출물 섭취는 고지방식이로 증가된 혈청 총콜레스테롤, 중성지방, LDL-콜레스테롤 농도를 현저히 감소시켜 대조군보다 더 낮은 값을 나타냈다. 또한 5% 표고버섯 물추출물 첨가군은 고지방 식이군보다 체중 및 피하지방량이 현저히 감소하였다. 변배설량은 3%와 5% 표고버섯 물추출물 첨가군에서 고지방 식이군보다 현저히 증가하였다. 표고버섯 물추출물 섭취는 고지방식이로 인해 증가된 간조직의 지방축적과 지질과산화물 형성을 억제하였으며, 특히 5% 표고버섯 물추출물 섭취군에서 지질과산화물 형성이 유의적으로 감소되었고 총 항산화 활성도 유의적으로 가장 높게 나타났다. 고지방식이군은 간조직의 조직염색에서 지방간이 심하게 진행된 것이 발견되었고, 표고버섯 물추출물 섭취군은 지방세포에 지방이 침투하지 않은 간지방증 소견을 보였다. 위의 결과를 통하여 볼 때 표고버섯은 체중 및 체지방 감소, 그리고 혈청지방 농도 개선에 효과가 있어 비만관리에 도움을 주는 것으로 사료된다.

탈지대두 grits가 고지방식이를 투여한 흰쥐의 지질대사에 미치는 영향 (Effects of Defatted Soybean Grits on Lipid Metabolism in Rats Fed with High-fat Diet)

  • DSG가 고지방식이를 급여한 흰쥐의 체내 지질대사 개선에 미치는 영향을 살펴보고자, 기본식이군, DSG 첨가군, 고지방식이군 그리고 고지방식이와 DSG 첨가군으로 각각 나누어 4주간 사육하였다. 그 결과, 혈장 내 총 콜레스테롤, LDL-콜레스테롤 함량은 고지방식이군에서 증가하였다가 고지방식이와 DSG 첨가군에서 유의적으로 감소하였고, HDL-콜레스테롤 함량은 고지방식이군에서 감소되었다가 고지방식이와 DSG 첨가군에서 유의적인 증가를 보였다. 간 조직 내의 총 지질, 총 콜레스테롤 및 중성지질 함량은 DSG 투여로 인해서 고지방식이군에 비해 감소되었고, 분변 내의 콜레스테롤 및 중성지질 배설량은 DSG 투여로 인해서 고지방식이군에 비해 증가하였다. 또한 간 내 HMG-CoA reductase 활성은 고지방식이군에서 증가되었다가, 고지방식이와 DSG 첨가군에서 유의성 있게 감소되었다. 따라서 DSG는 혈중 콜레스테롤 함량, 조직 내의 지질 및 콜레스테롤 축적을 감소시켜 체내 콜레스테롤 개선에 효과적인 것으로 생각된다.

Alcohol과 쌀 곰팡이의 Toxicity가 간(肝)의 지방(脂肪) 축적(蓄積)에 미치는 영향(影響) (Effects of Alcohol Intoxication and Moldy Rice Consumption on the Liver Lipid Content in Rat)

  • For the elucidation of the metabolic effects due to alcoholic drink and moldy rice intake author designed two experiments as follows. In the first experiment, sixty male and female rats, divided into six groups, 10 rats each in both sexes, were given 7.5% sucrose-30% alcohol solution (Jinro-Soju) and diets varied of fat content (2%, 7%, 30% of diet) and protein quality (casein, bean, anchovy) for 6 weeks. In the Second experiment, six groups of rats, 10 male rats each, were fed the diets containing zero, 5%, 10%, 25%, 50%, and 100% moldy rice contaminated Penicillium Islandicum and the experimental periods were 4 weeks and 6 weeks. The results of these studies due to the toxicity of alcohol consumption and moldy rice ingestion of the fatty liver production and the other effects are as follows. 1. Food intake of alcohol consumed groups decreased to 50%-70% of that of standard group. In the second study, there was no significant differences on food intake due to the different contents of moldy rice and experimental period. 2. On the view paint of body weight gain, the body weight of alcohol consumed groups gained much less than that of standard group, because food consumption was decreased due to alcohol ingestion. 3. In comparison of F.E.R. and P.E.R. between standard and experimental groups, there was no significant differences in this study. 4. As a matter of fact, there was no significance on the nitrogen balance in both studies. 5. From the result of hematology, R.B.C. counts, W.B.C. counts, hemoglobin, and hematocrit showed normal values in all groups including in this study. So we might conclude that the toxicity of alcohol and moldy rice do not effect significantly on blood picture. 6. The larger organs shrank on the range of 20%-70% of that against standard group in this study. The major reason for the shrinkage of organs might be account of decreased food consumption due to alcoholic drink. There was no great changes on the organ weight due to the ingestion of moldy rice. 7. The nitrogen content in various organs in both experiments was revealed at the normal level for the worst condition in terms of the least food intake. In other word, it was noteworthy that the concentration of nitrogen in various organs was kept at the normal ratio as standard groups under the circumstances of this study. 8. The lipid content in the liver of rats fed alcohol and diets containing either various lipid contents or protein quality did not increase. Hepatic lipids accumulation due to the dietary fat content was observed, but there wvas no significances among the compared groups. In the second experiment, the difference of hepatic lipid content between the moldy rice groups and standard group was not showed. In addition to the result of total lipid, hepatic free cholesterol, free fatty acid, and triglyceride did not change in both studies, we might conclude that the toxicity of alcohol and moldy rice could not effect on the hepatic lipid contents. 9. There was no significant differences on the serum glucose level between alcohol groups and standard group. In the second experiment, serum glucose level increase in 6th week compared with in 4th week, but there was no significant differences.

Anti-hyperglycemic and Anti-hyperlipidemic Activities of Acanthopanax Senticosus Herbal Acupuncture in C57BL/6J ob/ob Mice

  • Objectives : The aim of this study was to investigate the hypoglycemic and hypolipidemic activities and mechanisms of Acanthopanax senticosus (AS) herbal acupuncture. Methods : Anti-diabetic and anti-steatotic activity of the AS herbal acupuncture was investigated on C57BL/6J ob/ob mice. After random grouping at the age of 9 weeks, the herbal acupuncture groups were injected subcutaneously at the left and right Gansu (BL18) corresponding acupuncture points alternately on exactly the same time every day with 0.1ml of either 400 mg/kg or 800 mg/kg of AS (AS400 and AS800) for 8-week period. As a positive control, metformin was administrated at a dose of 300 mg/kg (MT300). Body weights were measured weekly, and on every other week blood was collected for blood glucose analysis. At the end of study, blood was also collected for determination of plasma insulin and lipid levels, after which they were killed and periepidydimal fat, liver, muscle, and pancreas were immediately removed. The removed tissues were instantly soaked in liquid nitrogen and stored at $-70^{\circ}C$ for morphological examination and mRNA analysis. Results : The AS herbal acupuncture significantly prevented weight gain on C57BL/6J ob/ob mice. The AS herbal acupuncture lowered blood glucose and improved glucose tolerance in C57BL/6J ob/ob mice. The increase of insulin response during the OGTT was inhibited by the AS herbal acupuncture. Insulin sensitivity of skeletal tissue was enhanced. Plasma lipid levels were significantly improved in the AS herbal acupuncture groups. The AS herbal acupuncture decreased hepatic lipogenesis and hepatic triglyceride production, and increased fatty acid (FA) transporter that involves in FA uptake. The AS herbal acupuncture inhibited the increase of liver mass by prevention of the accumulation of TG but did not inhibit weight gain of fat tissue on C57BL/6J ob/ob mice. Conclusion : In summary, we have demonstrated several unique properties of the AS herbal acupuncture in decreasing body weight, and reversing insulin resistance and hepatic steatosis in ob/ob mice. This AS herbal acupuncture acts as an insulin sensitizer and specifically decreases circulating glucose and lipids, and suppresses hepatic lipogenesis.
