Effect of Gam-Tea on the Metabolizing Enzyme Activity of Some Free Radical and Alcohol in Rats

  • To investigate an effect of Gam-Roa tea on the free radical or alcohol detoxicating enzyme activities, the rats got a drink at the Gam-Roa tea instead of water for 3 months, and then the animals were sacrificed and obtained the following findings. The animals receiving Gam-Roa tea showed a decreasing tendency of hepatic xanthine oxithine oxidase activity and significantly incresed content of cytochrome P-450 compared with the control. Furthermore, hepatic superoxide dismutase and glutathione S-transferase activities were also more increased in rats received Gam-Roa tea than in the control group, those receiving water. On the other hand, alcohol or aldehyde dehydrogenase activities were more increased in rats receiving Gam-Roa tea than the control. In conclusion, it is likely that the liver of rats receiving Gam-Roa tea may have the oxygen free radical or alcohol detoxication potential.

표고버섯 열수 추출물이 발암원을 급여한 흰쥐의 간 기능 관련 효소활성에 미치는 영향 (Effects of Hot Water Extracts from Lentinus edodes on Hepatic Functional Enzyme Activities in the Rat Fed Butter Yellow(p-Dimethylaminoazobenzene))

  • 표고버섯의 열수 가용성 다당류(PS)와 간 장해물질이며 발암성을 갖는 butter yellow를 흰쥐의 식이에 6주간 투여하여 혈장과 간의 효소활성 및 과산화지질의 함량을 측정한 결과는 다음과 같다. 1. 혈장 중 GOT 활성은 better yellow 첨가군(BO군)보다 PS의 투여군(BP군 : BO+PS)이 비교적 낮은 수준을 나타내었으며 ${\gamma}$-GTP와 GPT 활성에서도 BO군에 비하여 BP군에서 유의하게 낮은 활성을 나타냈다. 2. 간장 중 glutathion peroxidase, catalase 및 lactate dephdregenase 활성 또한 기초식이군(NO군)에 PS를 토여한 군(NP군)은 NO군과 유의한 차이가 없었으나 BP군은 BO군에 비하여 유의하게 낮은 수준이었다. SOD활성은 각 군간에 유사한 수치로 유의적인 차리를 보이지 않았다. 3. 간장의 microsomal 분획분 및 cytosol 분획분 중의 glutathione S-transferase 활성도는 BP군이 BO군보다 유의하게 낮은 수준을 보였으나 better yellow를 첨가하지 않은 NO군에 비하여 상당히 높은 수준을 나타내었다. 4. 간장 내 glutathione과 malondialdehyde의 함량 또한 PS의 투여로 생성이 저하되는 것으로 나타났다. 이상의 결과를 보아 표고버섯의 열수 추출물은 생체조직의 지질의 과산화를 방지하므로써 better yellow에 의한 간장장애의 치료 또는 독성을 완화하는 효과가 있는 것으로 사료된다.

Effects of Patriniae Radix and Melandrii Herba on Enzyme Activities in Mice

  • Effect of various fractions from the roots of Patrinia scabiosaefolia (Valerianaceae) and whole plants of Melandryum firmum (Caryophyllaceae) on enzyme activities in mice was investigated. The butanol fractions from both plants caused a significant elevation of serum transaminase activities when administered intraperitoneally, but did not, orally. Prolonged exposure by oral administration of both plants elevated hepatic cytochrome p-450 content, indicating the induction of drug metabolizing enzymes in liver.

식이성 Tungstate가 사염화탄소 투여에 의한 흰쥐 간 손상에 미치는 영향 (Effect of Dietary Tungstate on the Liver Damage in $CCl_4$-treated Rats)

  • To evaluate the role of xanthine oxidase in liver damage by CCl4, a group of rats were fed tungstate for a month, which suppressed the activities of xanthine oxidase in serum and liver. Control group of rats were fed standard diet without tungstate. Liver damage was induced both in tungstate fed and control groups by two intraperitoneal injections of CCl4 at the level of 0.1ml/100g body weight at intervals of 24 hours. Increases in the levels of serum alanine aminotransferase by CCl4 were significantly smaller in tungstate fed rats than in control rats. Concomitantly, histopathologic changes were less in tungstate fed rats than in control ones. In rats either treated with CCl4 or not, hepatic type O xanthine oxidase activities were remarkably reduced by tungstate feeding. Hepatic aniline hydroxylase activities were higher in rats fed tungstate than control rats when animals were not treated with CCl4, but the enzyme activities were lower in tungstate fed rats than control when they were treated with CCl4. Neither tungstate feeding nor CCl4 treatment caused any significant changes in hepatic glutathione contents, and activities of hepatic glutathione S-transferase, glutathione peroxidase and superoxide dismutase. It is concluded xanthine oxidase reaction augment CCl4 induced liver damage via oxygen free radical system.

Effect of Diets Supplemented with Pharbitis Seed Powder on Serum and Hepatic Lipid Levels, and Enzyme Activities of Rats Administered with Ethanol Chronically

  • The levels of $\gamma$-aminobutyric acid (GAGA) have been analyzed from pharbitis seeds by an AccQ-Tag amino acid analysis procedure. The GABA level of the pharbitis seeds was 125 nmole per gram fresh weight. To investigate the effects of pharbitis seed diets on serum and hepatic lipid levels, as well as enzyme activities of rats administered with ethanol chronically, Sprague-Dawley male rats were fed with either a AIN-76 diet (control), a control diet plus ethanol, a control plus pharbitis seed diet, or a control plus pharbitis seed diet plus ethanol for 30 days. Pharbitis seed diets decreased the serum total cholesterol, triglyceride, LDL-cholesterol, and $\gamma$-GTP levels that were increased by the chronic ethanol administration. In addition, pharbitis seed diets decreased the liver triglyceride and total lipid levels that were increased by the ethanol administration. However, ethanol metabolism was not retarded by the pharbitis seed supplemented diets. The present Endings, plus previous data showing the differences in the effects of cabbage diets having a high or a low level of GABA on the lipid levels and the enzyme activities of rats (Cha and Oh [2000] J. Korean Soc. Food Sci. Nutr. 29, 500-505), raise the possibility that GABA in plants could have a nutraceutical role in the recovery of chronic alcohol-related diseases.

Effect of Structured Lipids Containing CLA on Hepatic Antioxidant Enzyme Activity in Rats Fed a Normal Diet

  • Conjugated linoleic acid (CLA) has been shown to have a range of biological activities, including anti-carcinogenic, anti-atherosclerotic, anti-adipogenic and anti-diabetogenic effects. Recent reports also showed that CLA has free radical scavenging capacity, which may have health benefits for human beings. The current study was performed to investigate the effect of structured lipid (SL)-containing CLA on plasma lipids and hepatic antioxidant enzyme activity. Sprague-Dawley mts were fed 5% and 10% SL-containing normal diet for 6 wks and these groups were compared to rats fed 5% and 10% corn oil. In plasma lipids, total-cholesterol was not affected by fat source or dietary fat level while triglyceride level decreased significantly in groups fed 10% fat diet compared to the other groups. Plasma thiobarbituric acid reactive substances (TBARS) level decreased significantly in the S5 and S10 groups compared to the C5 and C10 groups, although hepatic TBARS level was not altered by fat source. On the other hand, in terms of hepatic antioxidant enzyme activity, superoxide dismutase activity increased in the S10 group, whereas catalase activity decreased in the S10 group. Glutathione peroxidase activity decreased significantly in the SL groups compared to the C5 group. Glutathione reductase activity increased and glucose-6-phosphate dehydrogenase activity decreased in the C10 group compared to the C5 and C5 groups. In conclusion, the free radical scavenging activity of CLA seemed to suppress oxidative stress, which reduced lipid peroxidation resulting in lower hepatic antioxidant enzyme activity.

Inhibition of Tumor Formation and Changes in Hepatic Enzyme Activities by Kimchi Extracts in Sarcoma-180 Cell Transplanted Mice

  • Inhibitory effects of the methanol extract, hexane extract, methanol soluble fraction (MSF) and juice from 3 weeks fermented Kimchi on the tumor formation in sarcoma-180 cell transplanted mice were studied. Effects of the solvent extracts and juice of the Kimchi on the levels of lipid peroxide, glutathione, and the enzyme activities of the liver were also investigated in normal and sarcoma-180 cell transplanted mice. At 32 days following trans-plantation, MSF reduced the tumor formation by 54% compared with the control group, resulting in the smallest tumor weight. Lipid peroxided content in liver increased by the transplantation of sarcoma-180 cells. However, it decreased when MSF of Kimchi was treated to the mice. MSF also suppressed xanthine oxidase activity in cytosol of the liver cells in mice transplanted by sarcoma-180 cells. Kimchi extracts had no inhibitory effect on hepatic aminopyrine-N-demethylase activity in sarcoma-180 cell transplanted or normal mice. Methanol extract and hexane extract of Kimchi slightly increased hepatic glutathione contents in sarcoma-180 treated mice. The injection of MSF from Kimchi markedly increased glutathione levels in the liver of sarcoma-180 treated mice. The injection of MSF from Kimchi markedly increased glutathione levels in the liver of sarcoma-180 treated mice compared to the controls. The MSF recovered the activities of hepatic glutathione reductase and glutathione S-transferase that decreased by the injection of sarcoma-180 cells. These results showed that MSF of Kimchi could suppress the growth of tumors, inhibiting lipid peroxide production and xanthine oxidase activity, in mice. We also suggested that Kimchi extract might play an important role in the prevention of cancer by enhancement of the glutathione level itself as well as via glutathione reductase and glutathione S-transferase.

월국환(越鞠丸) 메탄올 추출물이 산화적 간손상에 미치는 효과 (Effects of Wolgukwhan Methanol Extract on Oxidative Liver Injury)

  • Objectives: In traditional medicine, Wolgukwhan has been used for the treatment of digestive system disease, such as indigestion, brash, ructation, nausea and vomiting. This study was purposed to investigate the effects of Wolgukwhan methnol extract (WGWM) on oxidative liver cell injury. Methods: In vivo assay, we administerated acetaminophen(500mg/kg, i.p.) to starved mice 24hrs after pretreatment of WGWM for 6days. In the liver homogenates, lipid peroxide and glutathione(GSH) levels were measured. In addition, activities of hepatic enzyme, such as catalase, glutathione peroxidase(GPX), glutathione S-transferase(GST) were measured in the hepatic mitochondrial and cytosolic fractions. Results: In vivo administeration of WGWM showed effective inhibition of acetaminophen induced lipid peroxidation and elevations of glutathione level. The acetaminophen treatment resulted in a decrease of catalase, GPX and GST activities. By contrast, WGWM pretreatment increased compare to those of untreated groups. Conclusions: These results suggested that WGWM might protect against lipid peroxidation by free radicals, destruction of hepatic cell membranes.

Inhibitory Effect of Ligularia fischeri var. spiciformis and Its Active Component, 3,4-Dicaffeoylquinic Acid on the Hepatic Lipid Peroxidation in Acetaminophen-Treated Rat

  • To find the action mechanism of the MeOH extract (LFS) of Ligularia fischeri var. spiciformis herbs (Compositae) and its active component, 3,4-dicaffeoylquinic acid (DCQA) on antihepatotoxicity, the effect was investigated on hepatic lipid perxodation and drug-metabolizing enzyme activities in acetaminophen-treated rat. Pretreatment with 250 mg/kg LFS (p.o.) and 10 mg/kg DCQA (p.o.) significantly decreased hepatic lipid peroxidation caused by acetaminophen injection. Further, LFS and DCQA inhibited hepatic microsomal enzyme activation such as hepatic P-450 cytochrome $b_5$, aniline hydroxylase and aminopyrine N-demethylase, suggesting that the two substances might effectively prevent the metabolic activation or scavenge electrophilic intermediates capable of causing hepatotoxicity. Both LFS and DCQA increased hepatic glutathione content and glutathione reductase activity, indicating that both resultantly prevented hepatotoxicity via antioxidative mechanism. Therefore, it was found that LFS had antihepatotoxicity based on the antioxidative action of DCQA.

Effect of Oolong Tea Extracts on Plasma Glucose Level and Antioxidant System in Diabetic Rats

  • The present study was conducted to investigate the effect of oolong tea extract on blood glucose level and antioxidant system in diabetic rats. The Sprague-Dawley rats were fed on AIN-76 based experimental diets containing 1 % oolong tea extract for 6 weeks. They were induced to be diabetic by receiving streptozotocin (45mg/kg BW) intramuscularly. Blood glucose, blood and hepatic concentration of vitamins A and E, and antioxidant enzyme activities were measured. Oolong tea extract feeding decreased the plasma glucose in diabetic rats. Dietary supplementation of oolong tea extract did not affect antioxidative enzyme activities such as superoxide dismutase, glutathione peroxidase and catalase in diabetic rats. The plasma level of retinol was increased in diabetic rats by feeding oolong tea extract. Plasma and hepatic levels of ${\alpha}$-tocopherol were higher in diabetic rats fed oolong tea extract. In conclusion, these results suggest that oolong tea extract consumption might reduce the plasma glucose in diabetic rats and protect the oxidative damage from diabetic stress to some extent.
