도토리추출물이 흰쥐의 체내 항산화효소계에 미치는 영향 (Effect of Acron Extracts on the Antioxidative Enzyme System)

  • This study was conducted to investigate the effect of acorn extracts on the antioxidative enzyme system. Male Sprague-Dawley rats(110$\pm$10g) were fed on containing normal and high fat diets. They were orally administrated (0.02g/100g B.W) of acorn ethylacetate-extract and water-extract at the same time once a day, respectively. Net weight gain and feed efficiency ratio were increased in high fat diet groups and decreased by acorn extracts administration. The effect of acorn extracts on hepatic glutathione peroxidase(GSH-Px), glutathione S-transferase(GST) and catalase activities did not show significancy in normal fat diet groups. GST and catalase activities and lipid peroxidation content(LPO) were significantly increased in high fat diet groups and this increment were decreased by acorn extracts administration. However GSH-Px activity and GSH content were decreased in high fat diet groups but increased by acorn extracts administration. The activities of lactate dehydrogenase(LDH), alkaline phosphatase(ALP) and aminotransferase in serum were significantly increased in high fat diet group but these increment reduced in acorn extracts administration groups. These results indicate that acorn extracts could improve the liver function and prevent the metabolic diseases by hyperlipidemia.

Early Diet Dilution with 40% Rice Hull Induces Lower Body Fat and Lipid Metabolic Programming in Peking Ducks

  • This study was conducted to evaluate the effect of early diet dilution with 40% rice hull on growth performance, carcass characteristic and composition of meat-type ducks, and to reveal the possible mechanism for decreased body fat deposition. 160 1-day-old White Peking ducks with initial body weight of $44.5{\pm}1.0$ g were allotted to two treatments with 8 replicate pens per treatment and 10 ducks per pen (5 male and 5 female). Ducks were fed with the experimental starter diets diluted with 0% (control, RH0), 40% rice hull (RH40) during 8 to 14 d of age, respectively. Thereafter, all ducks were fed with grower diet. Ducks fed with RH40 diet from 8 to 14 d of age increased (p<0.05) feed intake, decreased (p<0.05) body weight, body weight gain and adjusted feed intake (excluded rice hull), abdominal fat, skin with fat, and fat content in carcass, and reduced (p<0.05) activities of hepatic malic dehydrogenase, glucose-6-phosphate dehydrogenase and fatty acid synthetase. When diet dilution was withdrawn in the re-fed period from 15 to 42 d of age, full compensatory growth of body weight, breast meat and leg meat weight were attained. However, ducks were still less (p<0.05) carcass fat content and showed continually lower (p<0.05) hepatic lipogenic enzyme activities at the market age in RH40 ducks than the control. These results indicated that diluting diet with 40% rice hull during 8 to 14 d of age might be a suitable method to improve feed efficiency, and to reduce carcass fat deposition in the production of meat-type ducks.

포도박이 고지방식이를 섭취한 흰쥐의 지질 산화와 항산화 효소 활성에 미치는 영향 (Effect of Dietary Grape Pomace on Lipid Oxidation and Related Enzyme Activities in Rats Fed High Fat Diet)

  • 본 연구에서는 포도박이 고지방식이를 섭취한 흰쥐의 효소 활성과 지질과산화 수준에 미치는 영향을 조사함으로써 포도박의 생리활성과 자원화에 필요한 기초자료를 얻고자하였다. 고지방식이를 섭취한 흰쥐에게 포도박 실험식이를 급여한 후 혈청, 간조직 중의 지질과산화물 함량, glutathione 함량과 간조직 효소 활성을 측정하였다. 포도박 첨가군의 식이섭취량은 대조군에 비하여 감소하였고, 체중증가량의 실험군 간 유의적인 차이는 없었다. 식이효율은 고지방식이군이 정상식이군 보다 유의적으로 증가하였다. 지질과산화물 함량은 혈청의 경우 정상식이군에서는 포도박 첨가에 의한 변화가 없었으나 고지방식이군에서 함량이 증가되었고 포도박 첨가에 의해 감소되었다. 간조직과 간 microsome에서는 포도박을 첨가한 군이 각각의 대조군에 비하여 유의적으로 감소하였다. 간조직 내의 총glutathione 함량과 GSH/GSSG 비는 포도박 첨가군이 대조군에 비하여 모두 유의적으로 증가하였다. 간조직의 SOD 활성은 정상식이군에서는 차이가 없었으나 고지방식이에 포도박을 첨가한 군이 고지방대조군에 비하여 활성이 유도되었다. 간조직의 catalase 활성은 대조군에 비하여 포도박 첨가군이 유의적으로 증가하였다. G6Pase 활성은 포도박의 첨가로 인하여 대조군보다 활성이 증가하였지만 실험군간 유의적인 차이는 없었다. GST와 GSH-Px 활성은 정상 식이군에서는 변화되지 않았으나 고지방식이에 포도박을 첨가한 군이 고지방대조군 보다 효소 활성이 유의적으로 증가되었다. 이상의 결과를 종합해 보면 식이섬유와 폴리페놀 성분을 다량 함유한 포도박 식이는 체내 항산화계를 활성화함으로써 지질 산화와 관련성이 높은 심혈관계 질환의 예방효과를 가져올 수 있는 것으로 기대되었다. 포도박의 이러한 생리활성에 대한 연구결과는 향후 포도 가공 중에 얻어지는 포도박 폐기물을 자원화 할 수 있는 기초자료로 이용될 것으로 사료된다.

식이(食餌)에 의한 흰쥐 간지질(肝脂質) 및 효소활성(酵素活性) 변화(變化)에 관한 연구(硏究) (Changes in the Lipid Composition and Some Enzyme Activities in the Rat Liver as Affected by Diets)

  • Albio rats right after weaning, weighing $50{\sim}55g$, were divided into the control, high-carbohydrate-, high-lipid-, and high-protein-fed groups. and were fed for 12 weeks with the respective diets to observe the increase in body weights as well as changes in the chemical constituentes and enzyme activities in the liver tissue, with the following results. (1) There was little difference in the rate of increase in the body weights among the groups, showing normal growth, except the high-protein-fed group which showed decrease in rate of body weight increase from the 7th week after feeding. (2) The liver weight was either increased after 12 weeks of feeding with the high-carbohydrate and high-Lipid diets, or showed no difference with the high-protein diet, as compared to the control weight. (3) The liver cytosol protein content was increased when fed with the high-protein diet, but decreased when fed with the high-carbohydrate and high lipid diets, as compared to the control content. (4) The triglyceride and cholesterol contents in the liver were decreased in the high-protein-fed group, but increased markedly in the high-carbohydrate- and high-lipid-fed groups as compared to the control values. (5) The hepatic glucokinase, G6PD, LDH, and fatty acid synthetase activities were increased in the high-carbohydrate and high-lipid-fed groups, and GOT and CPT activities were increased in the high-protein-fed group. From the above results. it was known that the high-carbohydrate and high-lipid diets stimulated the hepatic lipid metabolism, giving rise to lipogenesis, but the high- protein diet could prevent the lipogeuesis leading to the body weight increase.

갈근, 갈화, 갈근과죽력의 추출물이 알코올을 투여한 생쥐에 미치는 영향 (Effects of Radix Puerariae, Flos Puerariae and Bamboo+Radix Puerariae Water Extracts on the Ethanol-administered Mice)

  • This study was performed to investigate the effects of Radix Puerariae (RP), Flos Puerariae (FP) and Caules in Liquamen Phyllostachyos+Radix Puerariae water extracts on the alcohol dehydrogenase (ADH). transaminase (GOT, GPT) activities, and two hepatic antioxidant enzyme (SOD, catalase) activities in acute ethanol administered mice, and we have investigated the morphological changes that occur in hepatocytes of the experimental mice. The activities of ADH decreased compared with control group in the A1(66%), C1(57%), C2(54%) groups. The transaminase activites increased in the control groups compared with experimental groups. Ethanol treatment group without the RP or FP administration significantly lowered the activities of hepatic SOD and catalase, whereas MnSOD increased in the A1(27%) and B2(43%)groups, CuZnSOD increased in the B2(25%) and C2 groups. The catalase activites were increased in the A1(270%), A2(478%), B2(487%) and B1(770%) compared with control group, A lot of PAS-positive granules were observed in the A1, A2, C1 and C2 groups compared with the other groups. These results suggested that RP, FP and RP+Bamboo extracts administration may be prevent from liver damage in the alcohol treatment mice.

Benzo(a)pyrene에 의해 유도된 간기능 장해에 미치는 당귀의 효과 (The Effect of Angelicae gigantis Radix on the Benzo(a)pyrene-induced Hepatotoxicity in Rats)

  • This study was performed to investigate the effect of Angelicae gigantis Radix extract oil the hepatic detoxifying enzyme activities and lipids. Male sprague-dawley rats were administrated the extracts with benzo(a)pyrene, a hepatotoxic agent, inducing liver damages. Results obtained from this study were as follows: 1. The markedly increased enzyme activities (AST, ALT, LDH, ALP, ${\gamma}$-GTP, GSH-Px) in B(a)P induced groups tended to be decreased by the treatment of the Angelicae gigantis Radix extract. 2. Liver GSH content and lipid peroxide activity were decreased by the administration of the extracts. 3. It tended that the curative effects were better than the protective effects of the extracts.

고려인삼의 폴리아세틸렌 성분이 과산화 지질 형성에 미치는 영향 (Effect of Polyacetylene Compounds from Korean Ginseng on Lipid Peroxidation)

  • 고려인삼에서 분리한 폴리아세틸렌 성분인 파낙시돌, 파낙시놀과 파낙시트리올의 사염화탄소로 유도된 마우스와 흰주의 지질과 산화와 효소적 또는 비효소적으로 유도된 시험관내 과산화 지질 형성에 미치는 영향을 관찰하였다. 정상 또는 사염화탄소 처리된 마우스와 흰쥐에 대한 폴리아세틸렌의 효과를 혈청과 간 과산화 지질수준과 혈청효소 (GOT, GPT, LDH) 활성을 측정함으로써 관찰하였다. 간 마이크로좀 내 cytochrome P-450 함량과 aniline hydroxylase와 aminopyrine demethylase 활성도 측정되었다. 정상 마우스에 폴리아세틸렌을 처리한 경우 파낙시놀의 경우를 제외하곤 간과 혈청의 과산화 지질 형성과 혈청효소들의 활성에 변화가 없었으며, 파낙시놀은 간의 지질 과산화를 억제하였다, 폴리아세틸렌 성분들은 사염화탄소로 유도된 간의 과산화지질형성에 대한 보호작용을 나타내었고, 혈청지질과산화 수준을 낮추었다. 또한 사염화탄소로 유도된 LDH의 혈액내 유출에 대한 보호작용이 있으나, 혈청 GOT와 GPT 수준엔 영향을 주지 않았다. 사염화탄소는 cytochrome P-450 함량과 aniline hydroxylase, aminopyrine demethylase 활성을 낮추었으며, 이 경우 폴리아세틸렌은 효과를 나타내지 못하였다. 반면, 사염화탄소 없이 폴리아세틸렌만 처리한경우, 파낙시돌과 파낙시놀은 aniline hydroxylase를 세폴리아세틸렌성분은 aminopyrine demethylase를 유도하였으며, cytochrome P-450엔 영향을 주지 않았다. 시험관내 간 마이크로좀의 지질 과산화는 폴리아세틸렌 첨가시 농도에 비례하여 억제되었다.

오미자 Schizandrin C 유도체 DDB 복합물 DWP-04가 Acetaminophen 해독계에 미치는 영향 (Evaluation of a Schzandrin C Derivative DDB-mixed Preparation(DWP-04) on Acetaminophen Detoxification Enzyme System in the Animal Model)

  • The effects of the DWP-04 [DDB:selenium yeast:glutathione (31.1 : 6.8 : 62.1 (w/w%)] on acetaminophen detoxification enzyme system were studied in rats. Treatment with DWP-04 was prevented againt acetaminophen-induiced hepatotoxicity in rat as evidenced by the decreased formation of lipid peroxide. Effect of DWP-04 on the activities of free radical-generating enzymes, free radical scavenging enzymes and glutathione-related enzymes as well as detoxification mechanism of DWP-04 against acetaminophen-treated was investigated in rat. Activities of cytochrome p450, cytochrome b5, aminopyrine demethylase and aniline hydroxylase as free radical-generating enzymes activities were decreased by the treatment with DWP-04 against acetaminophen treated. Although acetaminophen-induced hepatotoxicity results in the significantly decrease in the level of hepatic glutathione and activities of glutathine S-transferase, quinone reductase, glutathione reductase and ${\gamma}-glutamyl-$cysteine synthetase, these decreasing effects were markedly lowered in the DWP-04-treated rat. Therefore, it was concluded that the mechanism for the observed preventive effect of DWP-04 against the acetaminophen-induced hepatotoxicity was associated with the decreased activities in the free radical-generating enzyme system.

고지혈증 유발 흰쥐에서 녹두가 혈청 BUN 및 효소활성에 미치는 효과 (Effects of Mung Bean on Serum BUN and Enzyme Activities in Hyperlipidemic Rats Induced by High Cholesterol Diet)

  • 본 연구의 목적은 5% 녹두(Phaseolus aureus L.)의 급여가 고지혈증 유발 Sprague-Dawley(SD)계 흰쥐의 혈청 blood urea nitrogen(BUN), creatinine 및 요산(uric acid)의 농도와 aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphatase(ALP), lactate dehydrogenase(LDH), amylase, lipase 및 catalase 활성에 미치는 영향을 규명하고자 시행하였다. 연구결과 녹두는 고지혈증으로 유발된 흰쥐의 혈청 BUN, creatinine, 요산 농도 및 AST, ALT, ALP, LDH, amylase, lipase의 활성 감소와 catalase 활성을 증가시키는 것으로 나타났다. 따라서 녹두는 신장과 간 등의 기능 개선과 예방에 효과적일 것으로 판단되어 기능성 소재로서의 이용 가능성이 있을 것으로 기대된다.

운동으로 유발된 산화 스트레스와 마늘의 항산화 작용 (Antioxidant Effect of Garlic Supplement against Exercise-Induced Oxidative Stress in Rats)

  • This study was to investigate lipid peroxidation, antioxidant enzyme activity and DNA damage after exercise, and the protective effect of garlic against exercise-induced oxidative stress. Male Sprague-Dawley rats(4 weeks old) were randomly divided into three groups of 6 rats each; control group(Con) without garlic and exercise, Ex group with exercise alone, and Ex-G group with 2% garlic and exercise. For 4 weeks, rats were given diets containing 15% corn oil and 1% cholesterol with or without garlic. The swimming was selected as a model for exercise performance. Rats swam for 40 min a day, for 5 days a week. Group Ex and Ex-G showed significant lowering in body weight gain and fat accumulation compared to control. No significant changes were observed in levels of plasma cholesterol and triglyceride among three groups, demonstrating that exercise and garlic had no effects on changes of blood lipid. This finding of blood lipid seems to be due to higher plant sterol content in corn oil. The DNA tail moment of lymphocytes showed greater tendency in Ex and Ex-G than in control, but garlic supplements failed to suppress DNA damages. Compared to control, Ex had higher plasma TBARS which was lowered to the control's level in Ex-G with 2% garlic supplementation(p<0.05). Ex-G led to a higher hepatic superoxide dismutase(SOD) activity than control and Ex(p<0.05). Activity of hepatic catalase was also increased in Ex-G, while in Ex it was significantly low(p<0.05). It seemed that TBARS levels were related to the activities of SOD and catalase, and that garlic contributed to increasing the enzyme activities and led to decrease of TBARS. These results demonstrate that lipid peroxidation and DNA damage occur as a consequences of oxidative stress after exercise, and that antioxidant defense against oxidative stress could be enhanced by garlic supplementation through the induction of antioxidant enzymes. However, further investigations should be done on the garlic effect on DNA damage.
