Lack of connexin 32 does not enhance the benzene-induced hematotoxicity and hemopoietic tumor incidence in mice

  • This study was performed to evaluate using wild-type (WT) and $C{\times}32$ knockout (KO) mice if lack of cell to cell communication by connexin 32 gap junction enhances the benzene-induced hematotoxicity and hemopoietic tumor development. The WT and $C{\times}32$ KO mice were exposed to 300 ppm of benzene for 6 hours/day, 5 days/week for a total of 26 weeks by inhalation, and then sacrificed to evaluate the toxicities of hemopoietic organs or allowed to live out their life span to evaluate the hemopoietic tumor incidence. The significant increase and decrease of organ weight were respectively noted in spleen and thymus of both WT and $C{\times}32$ KO mice without significant difference between the genotypes. Histopathologically, benzene exposure for 26 weeks induced the morphological changes in hemopoietic organs, characterized by fat cell accumulation in the bone marrow and extramedullary hemopoiesis in the spleen. The fat cell accumulation was, compared with that of WT mice, considerably exacerbated in the $C{\times}32$ KO mice. However, no significant difference was observed in the changes of hematological values and bone marrow cellularity as well as in the onset and incidence of hemopoietic tumors between WT and $C{\times}32$ KO mice. In conclusion, this study indicated little significant role of the cellular communication by $C{\times}32$ gap junction in the action mechanism of benzene hematotoxicity and leukemogenicity.

Distribution of HLA-DQA1*01, *03, *05 and DQB1*02 Subtypes and the Associated Haplotypes in the Korean Population

  • Background: As all HLA class II genes, the DQ genes show their polymorphic variation mainly in the second exon, which encodes the first extracellular domain of the molecule. PCR-SSOP (Polymerase chain reaction-Sequence specific oligonucleotide probe) techniques were frequently used for HLA-DQA1 and DQB1 typing but certain alleles, $DQA1^*0101/0104/0105$, $^*302/0303$, $*0501/0505$ and $DQB1^*0201/^*0202$ which differ from each other in segment other than exon 2, could not be unequivocally assigned. Methods: To overcome this problem, we applied additional PCR-SSP (PCR-Sequence specific primer) method to analyze DQA1 exons 1, 3 and 4 and DQB1 exon 3. And we investigated the distributions and haplotypes of HLA-DRB1, DQA1 and DQB1 alleles in 406 unrelated Korean healthy individuals. Results: Using this method the indistinguishable alleles of DQA1 and DQB1 in PCR-SSOP were typed definitively. We also found several important associations between DQA1 and DQB1 alleles in the Korean population; $DQA1^*0101-DQB1^*0501$, $DQA1^*0104-DQB1^*0502$ or $-^*0503$, $DQA1^*0105-DQB1^*0501$, $DQA1^*0302-DQB1^*0303$, $DQA1^*0303-DQB1^*0401$ or $-^*0402$, $DQA1^*0501-DQB1^*0201$, $DQA1^*0505-DQB1^*0301$, and $DQA1^*0201-DQB1^*0202$. The haplotypes of DRB1-DQA1-DQB1 associated with $DQA1^*01$, $^*03$, $^*05$, and $DQB1^*02$ subtypes were investigated. Several haplotypes associated with these alleles were observed in the Korean population. Conclusion: Our results can be helpful to find potential unrelated donors for bone marrow registries and study the HLA-associated disease and anthropology at high-resolution allelic level.

Stress, Coping, and Depression in Patients Following Hemopoietic Stem Cell Transplantation (조혈모세포이식 환자의 스트레스, 대처 및 우울)

  • Purpose: This study was done to identify the level of stress, coping, and depression among hemopoietic stem cell transplantation patients who received care in an outpatient center. Method: Data were collected from 81 patients who underwent hemopoietic stem cell transplantation at C University S Hospital between August 2005 and February 2006. Results: Stress and depression were significantly higher following hemopoietic stem cell transplantation in women, and in patients who were worse off financially or who were in bad health. The highest item of stress was 'economic burden for treatment'. There were significantly higher levels of emotion-focused coping among patients who had a spouse and who received motivation from the doctor. The highest item for problem-focused coping was 'try to look at the bright side of life'. The highest items for emotion-focused coping were to 'have faith that treatment will be finished quickly' and 'believe that your situation will improve'. Stress was significantly correlated with depression among these patients. Conclusion: It is necessary to develop nursing interventions to enhance positive coping and to decrease stress and depression among patients who have a hemopoietic stem cell transplantation.

  • Ultrastructure of the hemopoietic organ in the grasshopper, Euprepocnemis shirakii, was studied using light and electron microscopes. The hemopoietic organs located on the upper surface of the dorsal diaphragm between 1st and 8th segments were consisted of 3 kinds of cells; the reticular cells, stem cells and two types of granulocytes. The reticular cells had numerous cytoplasmic processes and forming a complex network. Characteristically, pinocytotic vesicles and rough endoplasmic reticulum were well developed in this cell. The stem cells give rise to differentiating hemocytes of the different cell lineages. Granulocytes within the hemopoietic organs were originated from the stern cells and differentiated into two types of hemocytes ; granulocyte( I ), granulocyte( II ).

Sleep Disturbance and Related Variables during Hospitalization for Hemopoietic Stem Cell Transplantation (조혈모세포이식 환자의 이식단계별 수면장애 영향요인)

  • Purpose: The present study was to investigate sleep disturbance and related variables in patients undergoing hemopoietic stem cell transplantation(HSCT) on the day of admission, before HSCT, and of discharge, respectively. Method: A total of 52 HSCT patients were recruited from an university hospital from August 2002 to August 2003. The patients were asked to complete a set of sleep disturbance questionnaires. Result: The scores of sleep disturbance-related physical and emotional subscales on the day before HSCT and on the day of discharge were significantly higher than that on the day of admission. However, the score of a emotional subscale was not significantly different during the hospitalization for HSCT. About 10% of the variance of sleep disturbance was explained by the length after diagnosis on the day of admission, and 59.4% was by the type of disease, religion, gender, educational level on the day before HSCT. About 40.7% was by family income, gender, and the frequency of administration on the day of discharge. Conclusion: Nursing staffs should care for HSCT patients with more interest to reduce variables disturbing sleep. Especially, more careful nursing interventions have to be considered at the time of the day before HSCT and the time of discharge.

Anxiety in Patients Undergone Hemopoietic Stem Cell Transplantation (조혈모세포이식을 받은 환자의 불안)

  • Purpose: The purpose of this study was to identify the influencing factors on anxiety of patients who had received hemopoietic stem cell transplantation(HSCT). Method: A total of 81 patients who had undergone a HSCT at a university hospital were recruited from July of 2001 to June of 2002. They were asked to complete questionnaires about anxiety, self-esteem, family support, doctor's support and nurse's support. Result: The mean score of anxiety, self-esteem, family support, doctor's support and nurse's support was 41.9, 3.0, 4.6, 7.6 and 7.3, respectively. A significant negative correlation was found between anxiety and self-esteem. Self-esteem was positively correlated with family support, doctor's support and nurse's support. The major variable that influenced anxiety was the self-esteem, explaining 25.4% of the anxiety. Conclusion: On the basis of these results, it is necessary for HSCT patients to develop a nursing intervention for the enhancement of self esteem. Also, family support, doctor's support and nurse's support should be considered as a main support system in the nursing strategy for improving the quality of life of patients undergone HSCT.

Hope and Related Variables in Patients Undergoing Hemopoietic Stem Cell Transplantation for Leukemia (조혈모세포이식을 받은 백혈병환자의 희망정도와 영향요인)

  • Purpose: The purpose of the present study was variables in patients hemopoietic stem cell transplantation(HSCT) for leukemia. Method: A total of 88 participants were recruited at St. Mary's Hospital. The data were gathered from July, 2001 to June, 2002 using questionnaires. Result: The mean score of hope was $3.39{\pm}0.35$. Positive relationships were found between hope and self-esteem, physician's support, nurse's support, and satisfaction of life. And negative relationships were found between hope and depression, trait-anxiety, and state-anxiety. The major variable, that associated with hope, was the state-anxiety, explained 35.4% in the variance of the hope. Conclusion: Leukemic patients undergoing HSCT tended to have a high level of hope. Higher levels of self-esteem, physician's support, nurse's support and satisfaction of life were related with higher level of hope. And, higher the levels of depression and anxiety were related with a lower level of hope.

  • Ultrastructural characteristics of hemocytic differentiation pathway in Sericinus montetla Grey were observed with transmission electron microscope. Hemocytic differentiation took place in loose islets of hemopoietic organs as indicated by the presence of differentiating hemocytes; prohemocytes, plasmatocytes, granular cells and spherule cells. They are differentiatied from the stem cells through individual cell lineages. However, differentiating aspects of the oenocytoids were not observed. According to Brehelin and Zachary (1986), the premature hemocytes around the hemopoietic organs were classified into five types; prohemocytes, plasmatocytes, granular cells, spherule cells and oenocytoids.

Study on the Detection of In vivo and In vitro Hemocytic Pathway in Grasshopper, Euprepicnemis shirakii (메뚜기 체내와 체외에서 혈구생성 경로 추적에 관한 연구)

  • A Differentiating pathway of hemocytes in vitro and in vivo of grasshopper, Euprepocnemis shirakii was described using light and electron microscopes. In the interior of body, the stem cells of the hemopoietic organ differentiated into six types of cells respectively which are prohemoyte, plasmatocyte, granulocyte I, granulocyte II, spherulocyte and oenocytoid. The formation of these hemocytes was derived from the stem cells surrounded by a reticular cell. Hemopoietic tissue cultured in the insect media differentiated different hemocytes, but none of them underwent any mitotic division. Morphological features of the cultured cells in media were essentially the same as those of the hemocytes differentiated from the stem cells in vivo. These results were shown that each stem cell could differentiate into different types of hemocytes. It was confirmed that the stem cells possessed the pluripotent differentiation ability to directly each hemocyte, and that the once formed hemocytes in vivo and in vitro didn t undergo further transformation to other hemocytes. The maintenance of circulating hemocytes in grasshopper had been depended on the widely spreading hemopoietic organ situated in the upper surface of the dorsal alary muscle and located on the first to eighth segments.

Hemopoietic Effects of Deer Blood on Cyclophosphamide Induced Pernicious Anemia (녹혈이 Cyclophosphamide로 유도된 악성빈혈의 조혈기능 촉진에 미치는 영향)

  • The hemopoietic effects of deer blood (whole blood, blood cell, plasma, respectively) were examined using in vivo rat model. Experimental animals (Sprague-Dawley rat, male, 200 g) were divided into negative control group (injection of saline), positive control group (injection of Sipjeondaebotang) experimental groups (injection of whole blood, blood cell, plasma) and healthy control group. Cyclophosphamide(150mg/kg) was injected into experimental groups, negative and positive control group to induce bone marrow supression. After 8 days, freez dried deer blood (whole blood, blood cell, plasma respectively) and Sipjeondaebotang of 200 mg/kg in dose was administered orally into experimental groups and positive control group, once a day for 3 days (A group) and once a day for 12 days (B group) respectively. And then body weight and organ weight, biochemical profile (ALB, GOT, GPT, PRO, CRE), hematological values (WBC, RBC, HGB, HCT, MCV, MCH, MCHC, PLT) and CBC differentiation (Neutro, Lymph, Mono, Reticulo) were carried out. Finally, platelets were specially increased in the plasma treated A group and reticulocytes were specially increased in the plasma treated B group.
