Enhanced Expression of Foreign Gene in Baculovirus-Infected Insect Cells Using a Silkworm Hemolymph (누에 체액을 이용한 외래 유전자의 발현효율 증대)

  • To enhance expression of foreign gene by the novel expression vector, pBmKSK1, of Bombyx mori nuclear polyhedrosis virus, E. coli $\beta$-galactosidase gene expressing recombinant virus was infected in BmN-4 cells and various concentrations of silkworm hemolymph were added to the recombinant virus-infected BmN-4 cells containing fetal bovine serum. The expression efficiency of foreign gene was determined by $\beta$-galactosidase activity in the culture media. The results showed that the silkworm hemolymph was effective to expression of foreign gene in the BmN-4 cells, suggesting that the silkworm hemolymph could be substituted for fetal bovine serum in the BmN-4 cells to enhance expression of foreign gene.

  • The book lung in the wolf spider, Pardosa astrigera was consisted of a series of flattened triangular-shaped air sacs, stacked with about 70 sheets, and was located in the ventrolateral region of opisthosoma. Each hemolymph spaces (average $8{\mu}m$ in thickness) surrounded by the air sacs (average $6{\mu}m$ in thickness). The air sacs was supported by cylindrical cuticular spikes of microfibril bundles. Epithelial cell processes surrounded the hemolymph spaces. The nuclei of the epithelial cells were concentrated near the atrium. In the middle portion of air sac, the epithelial cells formed pillars across the hemolymph spaces and spot desmosome and zonula adherens were seen between the plasma membranes. In the hemolymph space of this spider, granular hemocytes (average diameter $8{\mu}m$) were the most dominant type of hemocytes. In the medial sinus, the hemolymph flow between the air sacs of a paired book lungs and then flow out of the lung vein. The air comes in the atrium through the ventral lung slit and makes a tidal wave in and out of the air sacs.

  • Animal cell culture media are usually supplemented with fetal bovine serum (FBS); however, the use of FBS presents certain problems including high cost. By using an adaptation process and the addition of silkworm hemolymph, the FBS concentration can be reduced without causing a significant decrease in cell growth.

  • As a previous step for the development of immunostimulating substance from marine natural products the two new lectins, MLA-II and MLA-III, have been isolated and purified from the hemolymph of Meretrix lusoria in addition to the previously reported MLA-I. The new lectins molecules were characterized as pure by electrophoretic studies.

  • Soluble proteins of silkworm hemolymph were investigated by means of isoelectric focusing (IEF). The protein spectra during ontogenesis of races and inter-races hybrids kept in Bulgaria was studied. A total of 51 protein bands in the hemolymph from fourth larval instar to imago were ascertained. Stage specific expression was established. The specific expression of some protein bands in the individual spectra manifest phenotype of gene determinate polymorphism (HP F, HP J, HP K, HP L, HP Q - in the zone with pH gradient 3.5-6.2 and HP K, HP L, HP N, HP P, HP T - in the zone with pH gradient 9.5 - 6.2). Breed specific expression was observed. On the basis of the obtained results, it was established that the investigated breeds are heterogeneous and the isoelectric focusing method is successful when specifying the inner-race and inter-race polymorphism in silkworm.

  • Male specific protein (MSP) was identified and purified from the hemolymph of Galleria melloneffa L. by electrophoresis and anion exchange chromatography. MSP has a native molecular weight of 55 kDa as determined by gel filtration chromatography and consists of a single unit with the apparent molecular weight of 27 kDa and has the pl of approximately 5.8. MSP is present in the hemolymph from day 8 pupae throughout the male adult. MSP was also found in pupal fat body, adult fat body, and adult testis.

  • Juvenile hormone binding protein was identified in the hemolymph of fifth instar larvae and purified using column chromatography. Hemolymph was mixed with [3H] JH-III and electrophoresed on 691 NON-SDS gel, indicating that radioactivity peak appears at Rf value of 0.55. Gel filtration showed two radioactivity peaks equivalent to bound and free [3H]JH-III, respectively. JHBP was purified from hemolymph through gel filtration (Sephadex G-100), anion exchange chromatosraphv (DEAE Sepharose CL-6B), chromatofocusing chromatographv (PBE 94) and preparative electrophoresis.

  • Changes in the protein concentration in the hemolymph and the oxidation of respiratory substrates at various developmental stages in cabbage worm, Pieris rapae Linne were measured using Biuret method and Warburg manometric method, respectively. The results are summarized as follows: 1. The hemolymph protein contentration decreased gradually until the six-day pupa and increased thereafter. In the fifth larval instar the concentration reached a maximum and was about two times the value for the six-day pupa. 2. Endogenous respiration was very high at the prepupal stage, decreasing at two-day pupa, followed by an increase at the late pupal stage. 3. Glucose showed the marked activity throughout the whole stages, and had the striking influence on both the prepupal and adult stages. 4. The changes in protein concentration and the rate of oxygen consumption exhibited a general pattern of U-shaped curve through the process of life cycle according to histogenesis of the adult organ and histolysis of the larval organ.

Lipid and Carbohydrate Contents in the Adult Hemolymph during Flight of the Oriental Tobacco Budworm (Helicoverpa assulta (Guenee)) (비행중인 담배나방의 혈림프내 지질과 탄수화물의 함량변화)

  • Studies were carried out to investigate changes of lipid and carbohydrate contents in the hemolymph of the Oriental tobacco budworm(Helicoverpa assulta (Guenee» adults during flight and hormonal effects on mobilization of energy sources in the hemolymph. During a few minutes after flight, both sexes showed a rapid increase in lipid content and the high level was maintained for about 2 hours. But carbohydrate content in the hemolymph during flight showed almost no change but a slight increase seen during the first 10 min of flight in males only. Synthetic adipokinetic(Lom-AKH- n), hypertrehalosemic(Bld-HrTH) hormones and brain/ corpora cardiaca extract of H. assulta adult elevated lipid and carbohydrate contents in hemolymph and the effect was much more pronounced for lipid. These results suggested that lipid is a main fuel for flight activity and lipid mobilization is under the hormonal control. And this study showed that both adipokinetic and hypertrehalosemic factors may exist in H. assulta and these factors may have similar structures to those of Mas-AKH, Hez-HrTH, Lom-AKH- n or Pea-HrTH.

  • We have investigated the expression of early growth response protein 1 (EGR1) in INS-1 pancreatic β-cells treated with Allomyrina dichotoma hemolymph. The Korean rhinoceros beetle, A. dichotoma (Coleoptera: Scarabaeidae), is important in the insect industry for medical applications. We have already established a method for purification of A. dichotoma hemolymph that can be used in many experiments. EGR1 is reported as a multifunctional transcription factor that is implicated in virus infections. EGR1 has therefore been revealed as a major mediator and regulator in the physiological and pathological conditions of several cell and tissue types. New findings in this study are that A. dichotoma hemolymph, which promotes a dose- and time-dependent upregulation of EGR1 gene expression, shows an enhancement of this gene expression when combined with hypothermia or endoplasmic reticulum (ER) stress. These results suggest that A. dichotoma hemolymph may provide clues to EGR1-associated disease therapies involving gene regulation of EGR1.
