Effect of Inorganic Mercury on Hematological and Antioxidant Parameters on Olive Flounder Paralichthys olivaceus

  • The effects of inorganic mercury on hematological parameters and hepatic oxidative stress enzyme activity were studied in olive flounder Paralichthys olivaceus. Fish were injected twice intraperitoneally with mercuric chloride (2, 4, or 8 mg Hg/kg BW). The major hematological findings were significant decreases in the red blood cell count, hematocrit value, and hemoglobin level in olive flounder exposed to 8 mg Hg/kg BW. Remarkably low levels of calcium and chloride, and reduced osmolality, were also observed at 8 mg Hg/kg BW. In hepatic tissue, significant increases in glutathione peroxidase and catalase activity were observed above 4 mg Hg/kg BW Inorganic mercury also increased glutathione S-transferase and glutathione reductase activity at 8 mg Hg/kg BW in hepatic tissue. The present findings suggest that exposure to a low concentration (${\geq}4$ mg Hg/kg BW) of inorganic mercury can cause significant changes in hematological and antioxidant parameters.

Effects of ultraviolet light B irradiation on nitric oxide activity in the sprague-dawley rat in vivo (I) (흰쥐에 조사한 자외선B가 Nitric Oxide의 활성에 미치는 효과 (I))

  • Recent studies have revealed that Nitric oxide(NO) was one of the demonstration for the physiological regulator, endothelial derived relaxing factor(EDRF) and that NO was produced by ultraviolet irradiation in human. Thus, the present author have carried out a experimental study on the change of hematological, histological value of ultraviolet irradiation in sprague-dawley rats. The subjects were divided into four groups of ten rats each selected at random. There were 4 groups: 1. no irradiation control; 2. ultraviolet $75mJ/cm^2$ irradiation group; 3. ultraviolet $150mJ/cm^2$ irradiation group; 4. ultraviolet $225mJ/cm^2$ group. After a irradiation, hematological and histological tests were performed to observe erythrocyte, hemoglobin, hematocrit, MCH, MCHC, MCV, $O_2$ saturation, pH, $PO_2,\;PCO_2$ value and to observe histological changes. In hematological tests, erythrocyte, hemoglobin, hematocrit significantly increased in $75mJ/cm^2$ than control group and more $150mJ/cm^2$ ultraviolet irradiation group respectively. Also In blood gas tests, $PO_2$ significantly increased in $75mJ/cm^2$ and more $150mJ/cm^2$ group than control group. Whereas $PCO_2$ significantly decreased in $75mJ/cm^2$ and more $150mJ/cm^2$ group than control group (Duncan-Tukey test, P<0.05). In histological tests, control and $75mJ/cm^2$ group unchanged, but more $150mJ/cm^2$ group changed that it was cytolysis, cytotoxic effect, acanthosis, proliferation of keratinocyte, appearance of amorphous cell and pyknotic nucleus, production of sunburn cell. In conclusion, the present author results support the importance of the relation between NO effect and hematological, histological value by ultraviolet B irradiation.

Total Cholesterol and Alkaline Phosphatase are Increased in D/D Type of Angiotensin Converting Enzyme

  • The polymorphism (insertion, I or deletion, D) of angiotensin converting enzyme (ACE) gene is designated as the presence of a 287 bp Alu repeat. The D/D homozygote carrier is associated with high ACE activity, and this high activity has been implicated with hypertension, coronary artery disease, or diabetic nephropathy. We studied the clinical candidate marker in ACE gene polymorphism using chemical and hematological analysis. The subjects are divided into normotensive and hypertensive groups and ACE genotype in the group was confirmed by PCR method. Chemical analysis was preceded with Hitachi7060, and hematological analysis was performed using Mythic 22. In 116 targeted people, 17 (38.64%) of 44 I/I genotype group are hypertension, 15 (34.09%) in 44 with D/I, but, D/D type in the 28 cases is 15 patients (53.57%) in hypertension. In hypertension group, biochemical analysis (triglyceride, and alkaline phosphatase) and hematological analysis (white blood cell, platelet) are showed high value in D/D genotype of ACE gene. The relationship between hypertension and ACE genotype is the same results as previously reported and we thought that the high laboratory value of white blood cell, platelet, triglycerides, and alkaline phosphatase are also indicator of hypertension in D/D type of ACE.

Studies on the hematological and blood chemical findings for dry and lactating periods (건유(乾乳)와 필유(泌乳)에 따른 유우(乳牛)의 혈액(血液) 및 혈액화학상(血液化學像)의 변화에 관한 연구)

  • This experiment was carried out to investigate the hematological and blood chemical values in dairy cattle during the dry and lactating period. Blood was collected from six healthy dairy cattle in Gyeongnam province. Leucocytes count, hemoglobin concentrations and the percentage of packed cell volume were lowest at the early lactation period. Leucocytes count was not affected during dry the to lactating period. The differential count of eosinophilic leucocytes was low at the late lactation period, while the basophilic leucocytes was high at the dry period. Inorganic phosphorus value was below under normal level. Creatinine value was from 1 to 2mg/100ml of serum. Positive reaction to CRP was shown in normal dairy cattle. The values of AST and ALT were higher during the dry than lactating period. r-GTP and total cholesterol tend to be decreased during the lactating period. CPK value was not affected for dry and lactating period.

Protein quality, hematological properties and nutritional status of albino rats fed complementary foods with fermented popcorn, African locust bean, and bambara groundnut flour blends

  • The objective of this study was to determine protein quality and hematological properties of infant diets formulated from local food materials. The food materials were obtained locally, fermented, and milled into flour. The flours were mixed as 70% popcorn and 30% African locust bean (FPA), 70% popcorn and 30% bambara groundnut (FPB), and 70% popcorn, 20% bambara groundnut, and 10% African locust bean (FPAB). Proximate analysis, protein quality, hematological properties, and anthropometric measurements of the animals fed with the formulations were investigated. The protein contents of the formulated diets were significantly higher than that of Cerelac (a commercial preparation) ($15.75{\pm}0.01g$/100 g) and ogi (traditional complementary food) ($6.52{\pm}0.31g$/100 g). The energy value of FPAB ($464.94{\pm}1.22\;kcal$) was higher than those of FPA ($441.41{\pm}3.05\;kcal$) and FPB ($441.48{\pm}3.05\;kcal$). The biological value (BV) of FPAB (60.20%) was the highest followed by FPB (44.24%) and FPA (41.15%); however, BV of the diets was higher than that of ogi (10.03%) but lower than that of Cerelac (70.43%). Net protein utilization (NPU) of the formulations was 41.16-60.20%, whereas true protein digestibility was 41.05-60.05%. Metabolizable energy (232.98 kcal) and digestible energy (83.69 kcal) of FPAB were the highest, whereas that of FPA had the lowest values. The protein digestibility values corrected for amino acid score of the diets (0.22-0.44) were lower than that of Cerelac (0.52), but higher than that of ogi (0.21). The growth patterns and hematological properties (packed cell volume, red blood cells, hemoglobin, mean corpuscular hemoglobin concentration, mean corpuscular hemoglobin, and mean corpuscular volume) of the formulated diets were higher than those of ogi, but lower than those of Cerelac. In conclusion, we established that the FPAB food sample was rated best in terms of protein quality over the other formulated diets. Therefore, a FPAB blend may be used as a substitute for ogi.

  • Hematological analysis can provide crucial information for monitoring the health of fish. However, there is no current information available regarding hematological changes in olive flounder following infection by Vibrio scophthalmi. In this study, hematological and biochemical alterations were determined in olive flounder infected by the high virulence strain (HVS) and low virulence strain (LVS) of V. scophthalmi. Survival in serum, skin mucus, and macrophages of olive flounder was also compared between the HVS and LVS. The results demonstrated that the hematocrit value in infected fish declined from 23.4% at 0 h to 18.0% at 168 h post infection. The total protein concentration in fish infected with the HVS was significantly higher than in fish infected with the LVS and a non-infected control. Lysozyme activity was significantly different between infected and control fish. The HVS survived in serum and cell numbers increased substantially, while cell numbers of the LVS in serum decreased. These changes in hematological characteristics in fish infected by V. scophthalmi can be used as an effective and sensitive index to monitor the physiological and pathological conditions of fish. The survival and reproduction of V. scophthalmi in host serum, skin mucus, and macrophages play a major role in systemic infection and can serve as a virulence indicator for different strains.

Research on the Hematological Changes in Accordance with Radiation Dose and Radiation Exposure period of the Medical Radiation Workers (의료 방사선 종사자의 피폭기간 및 피폭선량과 혈액성분 변화에 대한 조사)

  • In this study, we analyzed the effects of radiation exposure, as compared to the hematological parameters change of medical radiation workers and the public. The mean value of all hematological parameters were in the normal range. Eosin mean value of the radiation workers($2.52{\pm}1.79%$) showed that a significantly lower than the control group($2.92{\pm}1.39%$). In the comparison of the results depending on the occupation period, it showed high value that the mean of the radiation workers group WBC, platelet, Lymph, Mono, Baso. Over 20 years of radiation workers WBC, Mono showed low values and less than 10 years of radiation workers mean value of Baso showed low values, there was no statistical significance. In the comparison of the results depending on the 4 years cumulative radiation dose, Over 5.0 mSv of Radiation works RBC($4.61{\pm}0.53$ vs $4.91{\pm}0.38$), Hct($41.51{\pm}4.07$ vs $43.97{\pm}3.40$), Eosin($1.74{\pm}1.14$ vs $2.92{\pm}1.39$) showed low value, it was statistical significance. 0.5~1.0 mSv radiation exposure workers Hb ($13.93{\pm}1.75$) showed a significantly lower value than that of the control group ($14.90{\pm}1.29$).

Hematological and Serum Biochemical Analyses in Experimental Caprine Besnoitiosis

  • This study was undertaken to investigate the hematological and biochemical changes in experimentally infected goats with Besnoitia caprae from the time of infection till 360 days post-infection (PI). Six male goats were inoculated subcutaneously with $13{\times}10^7$ bradyzoites of B. caprae, and blood samples were collected from the jugular vein. The total erythrocyte and total leukocyte counts, hematocrit value, and differential leukocyte counts were determined. Serum biochemical analysis, including the total protein, albumin, total globulin, cholesterol, triglyceride, chloride, testosterone, calcium ($Ca^{2+}$), inorganic phosphorus, sodium ($Na^+$), potassium ($K^+$), iron ($Fe^{2+}$), glucose, serum amyloid A (SAA), haptoglobin (Hp), fibrinogen, ceruloplasmin, aspartate aminotransferase, alanine aminotransferase, creatine kinase, lactate dehydrogenase, and alkaline phosphatase, was undertaken. Skin biopsy from the limbs were collected at weekly intervals and histologically examined for Besnoitia cysts. Cysts were present in the skin biopsies of the leg of the infected goats from day 28 PI. There were variations in hematological analyses, but no significant difference was seen. From day 30 to 360 PI, results showed that SAA, Hp, fibrinogen, and ceruloplasmin concentrations increased, whereas testosterone concentrations decreased. Infected goats exhibited decrease of albumin and increase of serum total protein and globulin concentrations. By contrast, there were no significant differences in the remained analyses concentrations.

Toxicity Screening of Single Dose of Inorganic and Organic Arsenics on Hematological and Serum Biochemical Parameters in Male Cynomolgus Monkeys

  • A screening study of the acute toxicity of organic arsenics such as arsenobetaine and arsenocholine, a product of arsenic methylation metabolite, and inorganic arsenic was carried out to examine hematological and serum biochemical parameters in cynomolgus monkeys(Macaca fascicularis). We found soft and liquid feces, and vomiting in all treated groups with inorganic and organic arsenics. The monkeys in inorganic arsenic-treated group showed a significant increase in vomiting frequency compared with those in three organic arsenics-treated groups. These results suggest that inorganic arsenic might be more toxic than three other organic arsenics tested. The monkeys in inorganic arsenic-treated group showed a decrease in platelet and an increase in monocyte on day 4 and the monkeys in arsenocholine-treated group showed an increase in reticulocyte percentage on day 8. The monkeys in inorganic-treated group also showed decreases in AST and ALT values and the monkeys in arsenobetaine-treated group showed a decrease in AST value and an increase in T-CHO value. However, these hematological and biochemical changes were within the physiological ranges, showing that the single dose of inorganic and organic arsenics did not affect at least hematological and serum biochemical parameters. The present study of toxicity with single dose of arsenics provides valuable indicators for longer term study of toxicity of repeated doses of arsenics in primates.

  • Using aged Wistar rat living body change by aging Dokhwaljihwang was each orally administrated and achieved research about aging control. In Wistar rat 10, 30, 50 week and 40 week Dokhwaljihwang between 10 weeks form condition change of weight, change of intestine weight, hematology, blood chemistry, research result about serum content following conclusion get. 1. Observed gain in weight than control group form of Dokhwaljihwang to aged Wistar rat. 2. Is thought to promote activation of living body action gaining intestine weight along with gain in weight. 3. Displayed decrease of MDA's content of serum than control group form of Dokhwaljihwang to aged Wistar rat. 4. Change that is Wistar rat's hematological value by aging according to 10, 30, 50 week WBC, RBC, Hgb, monocytes, eosinophil etc. increase, and HCT, PLT etc. showed tendency that decrease according to old-week, and observed improvement that is hematological value than control group form of Dokhwaljihwang 5. Change that is Wistar rat's biochemical value by aging was measured highest in 50 week because ALT, AST, BUN, CRN, T-bili., T-chol, TG, TP, ALB, A/G, P etc. increase according to 10, 30, 50 week, and observed improvement that is biochemical value than control group form of Dokhwaljihwang. Is considered by being effect that Dokhwaljihwangimprove living body function decline by aging by this result.
