Antitumor effect of Careya arborea against Ehrlich ascites carcinoma with reference to lipid peroxidation and enzymatic and non enzymatic antioxidant system in Swiss albino mice

  • The methanol extract of stem barks of Careya arborea Roxb. (MECA) (Family- Myrtaceae) was evaluated for antitumor activity and antioxidant status against Ehrlich's Ascites Carcinoma (EAC) bearing Swiss albino mice. After 24 h of tumor inoculation the MECA was administered at the doses of 50, 100 and 200 mg/kg body weight/mice/day for 14 days. After the last dose and 18 h fasting mice were sacrificed. The effect of MECA on the growth of transplantable murine tumor, life span of EAC bearing hosts, hematological profiles, serum and liver biochemical parameters were estimated. The MECA showed significant (P < 0.01) decrease in ascites volume, packed cell volume and viable cell count and prolonged the life span of EAC tumor bearing mice. Hematological profiles reverted to more or less normal levels in extract treated mice. The MECA also produced protective effect by decreasing the activity of serum enzymes, bilirubin and increase the protein and uric acid levels. MECA significantly (P < 0.05) decreased the levels of lipid peroxidation, while significantly (P < 0.05) increased the levels of glutathione content, vitamin C, vitamin E, superoxide dismutase and catalase CAT. The results indicate that MECA exhibited significant antitumor and antioxidant activity in EAC bearing mice.

한방생약부산물 급여가 육계의 생산성, 장관 미생물 균총, 혈액 생화학적 성상 및 면역학적 지표에 미치는 영향 (Effects of By-Products of Herbal Medicine on Performance, Intestinal Microbial Population, Blood Biochemical Profiles and Immunological Parameters in Broiler Chicks)

  • 본 연구는 한방생약부산물을 이용한 육계의 사료첨가제를 개발하기 위해 대조군(CON), HM1(한방생약부산물 0.15%), HM2(한방생약부산물 0.3%) 및 HM3(한방생약부산물 0.5%) 등 4개군에 육계 96수를 완전임의 배치하여 생산성, 장내 미생물 균총, 혈액 생화학적 지표, 면역글로블린 및 백혈구 성상의 변화에 대하여 조사하였다. 한방생약부산물 0.15% 첨가군(HM1)에서 대조군에 비해 육계전기(3~21일령) 사료요구율이 유의적(p<0.05)으로 개선되었으나, 전 기간에서는 차이가 없었다. 장내 소화물의 E. coli 및 Lactobacili 총 균수에서도 한방생약부산물 급여는 영향을 미치지 않았다. 혈액 생화학적 지표에서는 한방생약부산물 급여 시 AST 및 glucose 수준을 유의하게(p<0.05) 감소시켰다. 면역글로불린(IgG 및 IgA)과 백혈구 감별계수에서는 처리에 따른 차이는 없었다. 결론적으로 육계에서 0.15~0.3% 수준의 한방생약 부산물 첨가는 육계전기에 사료이용성 및 간 조직의 기능을 개선하는 효과가 있는 것으로 사료된다.

조각자가 만성 비세균성 전립선염 Rat 모델에서 혈액 및 세포조직의 변화에 미치는 영향 (The Effects of GLS(Gleditsiae spina) Treatment on Hematological and Cytopathological Alterations in Non-Bacterial Prostatitis Rat Model)

  • Objective : Although chronic non-bacterial prostatitis is a common disease, it is very difficult to treat effectively. GLS (Gleditsiae Spina) has traditionally been used in treatment of urinary tract inflammation and voiding disturbance. In this study, we investigated the therapeutic effects and action mechanism of GLS in the rat model of non-bacterial prostatitis induced by castration and testosterone treatment. Method : We observed four experimental objects of normal group, control group, testosterone group, and GLS group. Rats were treated with 17$\beta$-estradiol after castration for induction of experimental non-bacteral prostatitis, which is similar to human chronic prostatitis in histophatological profiles. GLS and testosterone were administered as an experimental specimen and a positive control, respectively. The prostates were evaluated by histopahological parameters including the epithelial score and epithelio-stromal ratio for glandular damage. Also, the prostates were observed by hematological alterations of WBC, RBC, hemoglobin, hematocrit and platelet. Results : While prostates of control rats revealed severe acinar gland atrophy and stromal proliferation, the rats treated with GLS showed a diminished range of tissue damage. Epithelial score was improved in GLS over that of the control. The epithelial-stromal ratio was lower in GLS when compared to that of the control. Conclusion : These findings suggest that GLS may protect the glandular epithelial cells. We concluded that GLS could be a useful remedy agent for treating chronic non-bacterial prostatitis.

Longitudinal Study on Changes in Nutritional and Health Status of Young Adults on Two-Year Diet

  • Changes in nutritional and health status brought on by a two-year balanced diet were assessed with anthropometric measurements as well as hematological and lipid profiles in 56 healthy young men. Recommended dietary allowances (RDA) were adjusted with estimated daily weighted activity factor. The weighted resting energy expenditure factor of the subjects was 2.37 $\pm$ 0.05. Compared with RDA, all nutrient intakes were adequate and 56% of energy was supplied by carbohydrates, 18% by protein and 26% by fat. The vitamin and mineral intakes except vitamin B-2 were higher (26.46-129.88%) than RDA. Vitamin B-2 intake was 92.15 \ulcorner 14.16% of RDA. There was no seasonal variation on nutrient intakes. Height was increased and systolic blood pressure was decreased by balanced diet for two years. Body weight, diastolic blood pressure and body mass index (BMI) were unchanged. The level of hemoglobin and hematocrit was not changed, the level of plasma protein was decreased and the level of plasma total cholesterol and albumin was increased. These results suggest that a balanced diet can increase height and complement health status achieved through vigorous exercise, even in adults, and that the level of Korean RDA for energy is adequate to maintain existing body weight.

Black soldier fly as feed ingredient for ruminants

  • This paper is a review of some experiments using black soldier fly (BSF) and its by-product to explore their nutritional value, production potential in Indonesia and its application in the ration of ruminants. Evaluation on the effect of milk replacer, creep feed containing BSF, BSF frass and the possibility to use lactic acid bacteria from BSF as probiotics are presented. Utilization of BSF larvae in milk replacer as skim and cream milk substitute showed that there were similarity on physiological, hematological status and performance of goat kids compared to those offered goat milk or commercial milk replacer. In addition, BSF larvae can be used to substitute soybean meal in the creep feed for post weaning goat kids without any differences in weight gain and blood profiles. However, utilization of BSF frass in the fattening goat ration resulted lower digestibility of dry matter and organic matter due to the chitin content in the frass. Black soldier fly larvae grown on chicken manure harbour lactic acid bacteria (LAB) which have potential as probiotics for ruminants. In general, BSF larvae has potential as ingredient for milk replacer, creep feed, fattening ration, and source of LAB for probiotics.

건강한 지원자에서 홍삼농축액의 혈행 개선 효과: 무작위, 이중맹검, 위약-대조 시험 (Effect of Korean Red Ginseng Extract on Blood Circulation in Healthy Volunteers: A Randomized, Double-Blind, Placebo-Controlled Trial)

  • Korean red ginseng has broad efficacious effects against hypertension, diabetes, nociception, and cancer, and it counteracts weakness. It has been reported that Korean red ginseng is able to normalize blood pressure, improve cholesterol and lower blood glucose levels. We have recently reported that Korean red ginseng extract (KRGE) significantly prevented rat carotid arterial thrombosis in vivo, and inhibited platelet aggregation ex vivo and in vitro in a dose-dependent manner. The purpose of this study was to examine the effects of KRGE on blood circulation in human by measuring ex vivo platelet aggregation, plasma coagulation and serum lipid profiles in healthy volunteers. Subjects were randomly divided into three groups (placebo-group, KRGE-low dose group, KRGE-high dose group). Administration of KRGE to subjects significantly inhibited ADP-induced platelet aggregations both in KRGE-low dose group from $72.79{\pm}20.53$ to $62.00{\pm}23.06%$ (p=0.0009), and in KRGE-high dose group from $75.14{\pm}21.86$ to $64.52{\pm}24.72%$ (p=0.0039), respectively. Administration of KRGE to subjects also significantly inhibited collagen-induced platelet aggregations both in KRGE-low dose group from $85.52{\pm}12.57$ to $79.62{\pm}20.47%$ (p=0.0916), and in KRGE-high dose group from $80.24{\pm}18.11$ to $70.31{\pm}25.93%$ (p=0.0565), respectively. Whereas, KRGE has no significant effects on coagulation system, such as prothrombin time (PT) and activated partial thromboplastin time (APTT), and serum lipid profiles, such as total cholesterol, low density lipoprotein cholesterol, high density lipoprotein cholesterol and triglyceride. KRGE also has no significant effects on hematological and serum biochemical profiles. These results suggest that KRGE has a potential to improve blood circulation through antiplatelet activity in human, and KRGE intake may be beneficial for the individuals with high risks of thrombotic and cardiovascular diseases.

Prognostic and Predictive Value of Hematologic Parameters in Patients with Metastatic Renal Cell Carcinoma: Second Line Sunitinib Treatment Following IFN-alpha

  • Background: Long-term survival is a problem with locally advanced and metastatic renal cell carcinomas. Sunitinib malate is an oral multitargeted tyrosine kinase inhibitor, but data on sunitinib use as a second line treatment in metastatic renal cell carcinoma (mRCC) are limited. Prognostic and predictive value of peripheral blood markers has been shown for many cancers. Materials and Methods: Efficacy and safety profiles of sunitinib after interferon alpha (IFN-${\alpha}$) were evaluated based on retrospective data for 23 patients with mRCC. Hematological parameters (neutrophils, lymphocytes, platelets, mean platelet volume, neutrophil/lymphocyte ratio, platelet/lymphocyte ratio) were recorded at the time of metastasis. It was evaluated whether hematological parameters were prognostic and predictive factors. Results: Median progression-free survival (PFS) time was 16.5 months (95%CI: 0-34.5). Median overall survival (OS) time was 25.7 months (95%CI: 10.8-40.0). Most common side effects were neutropenia (52.2%), stomatitis (26.1%) and hand-food syndrome (26.1%). PFS was found 3.13 vs 17.1 months in patients with neutrophil / lymphocyte ratio (NLR)>3 vs $NLR{\leq}3$ (p:0.012). Median OS was 6.96 vs 27.1 months in patients with NLR>3 vs $NLR{\leq}3$ (p:0.001).While 75% of patients who responded to sunitinib had $NLR{\leq}3$, in 72% of patients with no response to sunitinib NLR>3 was detected (p:0.036). The association between the Memorial Sloan-Kettering Cancer Center (MSKCC) criteria and NLR was statistically significant (p:0.022). Conclusions: Data on second line sunitinib treatment following cytokine in mRCC are limited. In our study, we observed second line sunitinib treatment following IFN-${\alpha}$ to be effective and tolerable. NLRappeared to have prognostic and predictive value.

Liver Involvement in Multiple Myeloma: A Hospital Based Retrospective Study

  • Objective: This study was to assess liver involvement in multiple myeloma with the aid of liver function tests. Materials and Methods: A hospital based retrospective study was undertaken using data retrieved of multiple myeloma from the register maintained in the Department of Biochemistry of the Manipal Teaching Hospital, Pokhara, Nepal between $1^{st}$ January, 2007 and $28^{th}$ February, 2012. We collected biomarkers of liver profiles including bilirubin (Total, Direct and Indirect), total protein, albumin, AG ratio, SGOT, SGPT, ALP, ${\gamma}GT$, LDH, ferritin, renal profile and hematological profile. Descriptive statistics and testing of hypothesis were used for the analysis using EPI INFO and SPSS 16 software. Results: Out of 37 cases of multiple myeloma, serum level of AST, ALT, ALP, ${\gamma}GT$ and LDH were increased above the cut-off point in 22 (59.5%), 24 (64.86%), 13 (35.13%), 9 (24.3%) and 11 (29.7%) respectively. The mean values of AST ($65.5{\pm}28.18$ U/L), ALT ($68.37{\pm}29.74$ U/L), ALP ($328.0{\pm}148.4$ U/L), ${\gamma}GT$ ($44.5{\pm}29.6$ U/L) and LDH ($361.7{\pm}116.5$ U/L), total protein ($9.79{\pm}1.03$ gm/dl) were significantly increased when compared with controls. In contrast, albumin ($3.68{\pm}0.43$ gm/dl) and the AG ratio ($0.62{\pm}0.15$) were significantly decreased. Similarly, anemia, hyperuricemia, azotemia, hypercalcaemia and Bence Jones proteinuria were found in 30 (78.9%), 27 (71.1%), 19 (51.5%), 15 (39.5%) and 16 (42.1%) respectively, in cases of multiple myeloma. Conclusions: While clinical manifestation of liver disease among the multiple myeloma was not common, abnormalities in liver function were characteristic.

고혈압 환자에 도움을 줄 수 있는 한방 다류 개발 (Development of Functional Tea Products for Hypertension Patients with the Mixed Medicinal Herbal Extracts)

  • 본 연구는 두충을 주 원료로 고안된 한방 혼합 다류가 고혈압 질환자의 혈압, 혈청지질 및 혈액학적 변화에 미치는 영향을 평가하기 위하여 수행하였다. 한방 혼합 다류를 시험 4주간 복용한 그룹과 시험 전 그룹을 비교한 결과, 수축기 혈압은 167.14 mmHg에서 134.86 mmHg로, 이완기 혈압은 100.00 mmHg에서 87.10 mmHg로 뚜렷하게 저하되었다. 혈청지질 중 총콜레스테롤(TC), 고밀도-콜레스테롤(HDL-C), 저밀도-콜레스테롤(LDL-C)의 농도 및 동맥경화지수(AI)는 유의적인 차이가 없었다. 반면, 한방 혼합 다류를 복용 시 중성지방(TG) 농도가 237.10 mg/dL에서 145.00 mg/dL로 강하게 감소되었다. 시험 4주 후와 시험개시 전의 혈액학적 지표(적혈구, 백혈구, 헤마토크리트, 헤모글로빈, 혈소판 및 호모시스테인)를 비교한 경우, 유의적 변화는 관찰되지 않는 반면 평균 적혈구 혈색소 농도(MCHC)는 유의적으로 2.70% 증가되었다. 이상의 결과로 부터 두충을 주 원료로 고안된 한방 혼합 다류는 중성지방 농도를 감소시킴으로써 혈압을 저하시키는 것으로 추정하며, 평균 적혈 수혈 색소 농도를 증가시킴을 확인하였다.

Significance of varying concentrations of T-2 toxin on growth performance, serum biochemical and hematological parameters in broiler chickens

  • The study was to determine the effects of diverse concentrations of T-2 toxin in broiler diet. Three hundred 1-day-old chicks with initial body weight of 46 ± 0.52 g were chosen and randomly assigned into five dietary treatments with 5 replicate cages and 12 broilers per cage for 42 d feeding trial. Dietary treatments were prepared with basal diets containing 0 (T1), 50 (T2), 100 (T3), 150 (T4), 200 (T5) ppb T2-toxin. Significant results were observed in the decreased intake of feed, feed conversion ratio (FCR), body weight gain (BWG), level of serum protein, cholesterol and hemoglobulin of broilers in increased concentration of the T-2 toxin in diet (150 and 200 ppb) groups than control. Also, observed that the uric acid, serum glutamic pyruvic transferase (SGPT), serum glutamic oxaloacetic transferase (SGOT) and Heterophil/Lymphocyte (H/L) ratio value were significantly higher (p < 0.05) in groups T4 and T5 than control. However, the BWG, feed intake and FCR, as well blood biochemical profiles of serum protein, cholesterol, hemoglobulin, uric acid, SGPT, SGOT and H/L ratio in groups T2 and T3 were statistically similar to control diet of broilers. It was concluded that the results showed that no adverse effects on growth performance and blood biochemical parameters in broilers feed with T-2 toxin (50 and 100 ppb) during the entire trial.
