Effects of vitamin E and selenium on the ovariectomized rat (Vitamin E와 selenium이 난소가 제거된 흰쥐에 미치는 영향)

  • The present study was devised to determine the effects of body weight, organ weight, hematological values and biochemical parameters by vitamin E and selenium (Selevit) in the ovariectomized (OVX) rats. The animals were divided into 4 groups. Intact group (n=10) received no treatment and operation. Sham group (n=10) received only sham operation and no treatment. OVX group received operation and no treatment. OVX+Selevit received operation and Selevit. The body weights of the all group increased, and that of OVX+Selevit group was the lowest rates. There were significant differences (P<0.01, P<0.001) of body weights between OVX+Selevit group and all other groups. Also, organ weights such as heart, liver, spleen and kidney were measured. The heart and liver weight were significantly lower (P<0.01, P<0.001) in the OVX+Selevit group than in the Intact and Sham group. Also, the kidney weight was significantly lower (P<0.05, P<0.01) in the OVX+Selevit group than in the all other group. On the other hand, there was no significantly difference in the organ weight of spleen between the OVX+Selevit group and the other groups. The number of WBC was significantly higher (P<0.05) in the OVX+Selevit group than in the all other groups. The hematological values of RBC, MCV, MCH and MCHC were no significant differences in any groups. The biochemical parameters of serum total protein and alkaline phosphatase increased significantly in the OVX+Selevit group as compared to that in the OVX group. But, there were no significant differences in AST and ALT in any groups. We conclude that vitamin E and selenium were significantly decreased the body weights in the ovariectomized rats. Our findings suggest that vitamin E and selenium may influence the process of lipid packaging and absorption in the ovariectomized rats.

Effects of testosterone on the orchidectomized rats

Orch group received operation and no treatment. Orch+Testos received operation and testosterone. The body weights of each group increased, but that of Orch+Testos group was significantly lower in Orch+Testos group than in all the other groups. The hematological values of white blood cell (WBC), red blood cell (RBC), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC) were no significant differences in any other groups. The concentrations of serum total protein and albumin increased significantly (P<0.05) in the Orch+Testos group as compared to that in the Orch group. However, there were no significant differences in Ca, IP and Mg in any other groups. We conclude that testosterone was significantly decreased the body weight in the orchidectomized rats. Our findings suggest that testosterone may influence the process of lipid packaging and absorption in the orchidectomized rats.

The Effects of GLS(Gleditsiae spina) Treatment on Hematological and Cytopathological Alterations in Non-Bacterial Prostatitis Rat Model (조각자가 만성 비세균성 전립선염 Rat 모델에서 혈액 및 세포조직의 변화에 미치는 영향)

  • Objective : Although chronic non-bacterial prostatitis is a common disease, it is very difficult to treat effectively. GLS (Gleditsiae Spina) has traditionally been used in treatment of urinary tract inflammation and voiding disturbance. In this study, we investigated the therapeutic effects and action mechanism of GLS in the rat model of non-bacterial prostatitis induced by castration and testosterone treatment. Method : We observed four experimental objects of normal group, control group, testosterone group, and GLS group. Rats were treated with 17$\beta$-estradiol after castration for induction of experimental non-bacteral prostatitis, which is similar to human chronic prostatitis in histophatological profiles. GLS and testosterone were administered as an experimental specimen and a positive control, respectively. The prostates were evaluated by histopahological parameters including the epithelial score and epithelio-stromal ratio for glandular damage. Also, the prostates were observed by hematological alterations of WBC, RBC, hemoglobin, hematocrit and platelet. Results : While prostates of control rats revealed severe acinar gland atrophy and stromal proliferation, the rats treated with GLS showed a diminished range of tissue damage. Epithelial score was improved in GLS over that of the control. The epithelial-stromal ratio was lower in GLS when compared to that of the control. Conclusion : These findings suggest that GLS may protect the glandular epithelial cells. We concluded that GLS could be a useful remedy agent for treating chronic non-bacterial prostatitis.

Toxicity of Methylcyclohexane and Its Effect on the Reproductive System in SD Rats

  • Objectives: There is limited data regarding the toxicity of methylcyclohexane, despite its wide use in rubber adhesives, paint diluents, and cleansing agents. This study aimed to verify the toxicity and influence on the reproductive system of methylcyclohexane after its repeated injection in Sprague Dawley (SD) rats. Methods: Methylcyclohexane was injected subcutaneously into male and female SD rats once a day, five times a week, for 13 weeks at different doses (0, 10, 100, and 1,000 mg/kg/day) for each group. The toxicity of testing material was verified by observing the change in body and organ weight, hematological change, pathological findings, and effect on the reproductive system at each different concentration. Results: In the 1,000 mg/kg/day group, there were cases of animal deaths. In animals that survived, hematological changes, including a decrease in the red blood cell count, were observed. A considerable weight gain or loss and pathological abnormalities in the liver, kidney, and other organs were found. However, the 10 and 100 mg/kg/day groups did not cause deaths or other specific abnormalities. In terms of reproductive toxicity, there were changes in hormone levels, including a significant decrease in hormones such as estradiol and progesterone (p < 0.001) in male animals. Menstrual cycle change for female animals did not show concentration dependency. Conclusion: When injected repeatedly for 13 weeks, methylcyclohexane proved to be toxic for the liver, heart, and kidney at a high dose. The absolute toxic dose was 1,000 mg/kg/day, while the no observed adverse effect level was less than 100 mg/kg/day. The substance exerted little influence on the reproductive system.

Effects of Moschus, Bovis Calculus, Ursi Fel Aqua-acupuncture on Liver Damage Induced by Radix Aconiti (사향(麝香).우황(牛黃).웅담(熊膽) 약침(藥鍼)이 부자(附子)로 유발(誘發)된 간손상(肝損傷)의 회복(恢復)에 미치는 효과(效果)에 관한 관찰(觀察))

  • In order to investigate experimentally that effect of Moschus, Bovis Calculus, Ursi Fel aqua-acupuncture on acutely damaged liver of rats induced by radix aconiti, the author gave Moschus, Bovis Calculus, Ursi Fel aqua-acupuncture according to method of manufacture stimulation to corresponding points, Kan-su(B18) and Ki-mun(Liv14), and carried out hematological, serological observation. Experimental groups were divided into 2 group : the one group was injected with extracted solution prepared by distillation method (A aqua-acupuncture treatment group), the other group was injected with extracted solution prepared by alchol extraction method (B aqua-acupuncture treatment group). The following results were obtained : 1. WBC level was decreased significantly in the experimental groups at 12, 24 and 48 hours as compared with the control group. RBC and hemoglobin levels were not changed significantly in the experimental groups. 2. Total cholesterol level was decreased significantly in the experimental groups at 12, 24 and 48 hours as compared with the control group. ALP level was decreased significantly in the experimental groups at 6 and 24 hours as compared with the control group. ${\gamma}-GTP$ level was decreased significantly in the experimental groups at 6, 12, 24 and 48 hours as compared with the control group. 3. GOT level was decreased significantly in the experimental groups at 6 and 12 hours as compared with the control group. GPT level was decreased siginificantly in the experimental groups at 12, 24 and 96 hours as compared with the control group. According to the above findings, it is considered that Moschus, Bovis Calculus, Ursi Fel aqua-acupuncture has effects of recovery of acutely damaged liver.

The toxicity of Aceporol 460 as a novel high loading capacity solubilizer of paclitaxel

  • Previously, we reported a novel polymeric micellar solubilizer, Aceporol 330, that showed relatively low toxic effects when it was compared with that of Cremophor EL which is currently being used for paclitaxel. In this study, we have developed a new micellar solubilizer, Aceporol 460, that has 3-4 times higher loding capacity for paclitaxel than Aceporol 330. The single-dose and the repeated-dose toxicity of Aceporol 460 were evaluated in ICR mice. For single dose toxicity test, male and female mice were randomly assigned to one of five study groups to receive, and injected intravenously with dosages of 0, 3, 4mL Cremophor EL/kgbody weight, and 3, 4mL Aceporol 460/kg body weight, respectively. In both male and female mice, LD50 for Aceporol 460 can not he determined even at the maximal administrable dosage, 4mL/kg due to the high viscosity of chemical and there was no significant change in body weight, hematological and serum biochemical analysis, organ weight, and histopathological examination compared with that of Cremophor EL. For the repeated dose toxicity test, male and female mice were given the dosage of 0, 1.6mL Cremophor EL/kgbody weight/day, and 1.6mL Aceporol 460/kg body weight/day for 2 weeks. Results of repeated dose toxicity tests for 2 weeks suggested that Aceporol 460 treated group show no significant toxicological findings with body weight, hematological and serum biochemical analysis, organ weight, urinalysis, and ophthalmoscopic and histopathological examination compared with that of Cremophor EL. These results indicate that Aceporol 460 have higher paclitaxeL-loading capacity than Aceporol 330 and less toxic effects than Cremophor EL in male and female mice.

Effects of estrogen on the ovariectomized rat (에스트로겐이 난소가 제거된 흰쥐에 미치는 영향)

The heart weight was significantly lower(P<0.05) in the OVX+E2 group than the Intact group. The liver weight of the OVX+E2 group was significantly different in comparison with those of the other groups. On the other hand, there were no significantly differences in the organ weight of spleen and kidney between the OVX+E2 group and the other remaining groups. The hematological values of WBC, RBC, MeV, MCH and MCHC were not significantly different among the groups. The serum total protein and albumin decreased significantly in the OVX+E2 group as compared to those in the intact group. But, there were no significant differences in Ca, IP and Mg among the groups. We conclude that estrogen was significantly decreased the body weight in the ovariectomized rats. Our findings suggest that estrogen may influence the process of lipid packaging and absorption in the ovariectomized rats.

Single-dose Toxicity of Guseonwangdo-go Glucose 5% Intravenous Injection in a Rat Model

  • Objectives: The purpose of this study was to examine the single-dose intravenous toxicity of Guseonwangdo-go glucose 5% pharmacopuncture (GWG5). Methods: Forty Sprague-Dawley rats were divided into four groups of five males and five females per group: an intravenous (IV) injection of 1.0 mL of normal saline solution per animal was administered to the control group; IV injections of 0.1, 0.5, and 1.0 mL of GWG5 per animal were administered to the experimental groups (G: 0.1, G: 0.5, and G: 1.0). Observation of clinical signs and body weight measurements were carried out for 14 days following the injections. At the end of the observation period, hematological, biochemical, and histopathological tests, as well as necropsy examinations, were performed on the injected parts. Results: No mortalities or adverse clinical signs were observed in any of the groups. The body weights of all groups continuously increased. In the hematological and the biochemical tests, females in G-0.1 had minimal changes, but those changes were not dose dependent. On necropsy examination, no abnormalities were observed. In the histopathological test, focal inflammatory cell infiltrations were observed in two female rats, one in the control group and one in G-1.0. Also, one female rat in the control group had an epidermis crust. These changes were concluded to have been caused by the insertion of the needle into a vein. Conclusion: The above findings suggest that the lethal dose of GWG5 administered via IV injection is more than 1.0 mL per animal in both male and female rats. Further studies are needed to establish more detailed evidence of its toxicity.

A Study on the Nutritional Status of Children Living in Rural Area and City Orphan Asylum -II. Hematological view- (농어촌(農漁村)과 시설거주(施設居住) 아동(兒童)의 영양상태(營養狀態)에 관(關)한 연구(硏究) -제2보(第2報) : 혈액성장(血液性狀)을 중심(中心)으로-)

  • This study was designed to investigate the nutritional anemic state and hematological findings of children, aged 11 and 12 years, living in Haenam Koon (living at home) and in Mokpo City (living in an orphan asylum) located in Chonnam. The mean red blood cell number of male was higher than female in both groups of living in the rural area and in the city orphan asylum (p<0.01), but the white blood cell count was not significantly different. The levels of average hemoglobin, hematocrit and mean corpuscular hemoglobin concentration of the children in both groups were similar, and 19.8% of children living at home in the rural area and 32.1% of children living in the city orphan asylum were anemic. The mean levels of serum total protein, albumin and A/G ratio in children of both group were not statistically different, and 18.4% of children living at home in the rural area and 13.2% of children living in the city orphan asylum were insufficient in the serum total protein value. The average serum cholesterol level of children living at home in the rural area was higher than that of children living in the city orphan asylum(p<0.01), and that of female living at home was higher than that of male (p<0.05). The mean levels of serum iron, total iron binding capacity and transferrin saturation of children living at home in the rural area were significantly higher than those of children living in the city orphan asylum (p<0.01).
